  • In normal cells tau protein is associated with axonal microtubules, whereas in Alxheimer's disease it is immobilized in the somatodendritic compartment of certain nerve cells as a major component of the paired helical filament. As a part of the study to analyze the nature of the paired helical filament (PHF) deposits and some related factors in brain, we have cloned and expressed a human tau gene cDNA in Escherichia coli to obtain the recombinant human tau protein in abundance.

  • Oyster mushroom, Pleurotus ostreatus is bifactorial heterothallism. Single basidiospore isolates from fruiting bodies are homokaryotic and self-sterile. However, we found that homokaryons derived from some strains of P. ostreatus could develop fruiting bodies with two different types. One hundred and two isolates out of 155 monospore isolates formed fruiting bodies (65.8%). First group did not have only mature or sporulating fruiting bodies but also clamp connections, which initial isolate also did not present clamp connections (Abortive homokaryotic fruiting, AHF). Second group had developed fruiting bodies with clamp connections even though initial homokaryotic colony did not form clamp connections (Pseudo- homokaryotic fruiting, PHF). The mycelial colonies derived from PHF by tissue culture formed clamp connections, while mycelial colonies of AHF lacked them. We obtained 535 PHF and 79 AHF inter-strain hybrids among 8 strains of P. ostreatus by hyphal anastomosis. The fruiting body yield of PHF group is higher than that of AHF group in bottle cultivation. A preselection of single spore isolates for fertility would save labour in strain improvement of P. ostreatus.

  • Tau protein is one of the microtubule-associated proteins in the mammalian brain. In Alzheimer's disease, tau protein is immobilized in the somatodendritic compartment of certain nerve cells, where it forms a part of the paired helical filament (PHF). To understand the role of tau protein in the formation of PHF, a recombinant human tau protein expressed in Escherichia coli and five synthetic peptide fragments (peptide 1 to peptide 5), corresponding to the C-terminal region of tau protein, were prepared and their ability in self-assembly to form filamentous structures was examined. The recombinant human tau protein formed short rod-like structures in 0.1M MES buffer containing 1 mM $MgCI_2$, while a synthetic peptide fragment 1 containing 55 amino acid residues could assemble into a lot of long filamentous structures in water and particularly twisted helical structures in 0.1M MES buffer containing 1 mM $MgCI_2$. This suggests that the C-terminal region possesses a filament-forming ability and may be related to the formation of the helical structure by providing a powerful filament-forming driving force.

  • The shrinkage and the mechanical properties of polypropylene hybrid fiber reinforced mortar PHFRM were investigated in this study. Mortars were prepared with limestone crushing sand, Portland cement and polypropylene hybrid fibers PHF. Two types of virgin fibers, having the same length (30 mm) were used for reinforcing test mortars, fibers in diameter of 0.45 mm, used by PLAST BROS factory of Bordj Bou Arreridj (Algeria) for the fabrication of brooms (for household cleaning) and fibers in diameter of 0.25 mm, available on the market, having multiple applications. In this investigation, it was aimed to study the total and autogenous shrinkage, the flexural and compressive strength of mortars based on hybrid fibers. As a result, PHF have negatively affected the mortar workability. However, shrinkage risk was reduced and coarser fibers (PF45) were most effective for reducing shrinkage risk. The mechanical performances and the ductility of PHFRM were also enhanced.

  • The purpose of this study was to identify novel extracts of brawn algae with the high deodorant capacity and antioxidant activity. To achieves this, an composition, deodorant performance and free radical scavenging activity were analyzed in powder and water extracts of Ecklonia cava(PEC/EEC), Saccharina japonica(PSJ/ESJ), Eisenia bicyclis(PEB/EEB), Sargassum spp.(PSS/ESS) and Hizikia fusiformis(PHF/EHF) using several analytical methods. Major component of five powders was verified as PEC, phlorotannin; PSJ, alginic acid and glutamic acid; PEB, carotenoid, chlorophyll, phlorotannin, tocopherol and fucoxantin; PSS, ${\beta}$-carotene and polyphenol; PHF, tannin. The highest level of DPPH scavenging activity was detected in EEC($IC_{50}=23.018{\mu}g/ml$), followed by EEB, ESJ, EHF and ESS. The deodorizing performance to ammonia was maintained high level in PEC, PSJ, PEB and PSS, while PHF showed low level in the same condition. But, the deodorizing performance to acetic acid was slightly higher in PSJ and PEC than those of PEB, PSS and PHF. Based on the results of DPPH scavenging activity and deodorizing capacity, we selected fractions of S. japonica as leading extracts and further prepared the 70% acetone extract (AESJ) and 70% ethanol extract (EESJ) from PSJ. The contents of total flavonoid, total phenol and total condensed tannin was significantly increased with 112-209% in AESJ and EESJ compared to those of ESJ. Also, a similar enhancement was observed on the level of DPPH scavenging activity and deodorizing performance against ammonia/acetic acid in AESJ and EESJ. Therefore, these results suggest that AESJ and EESJ contained with high total phenol and total condensed tannin contents may exhibit high deodorant capacity and antioxidant activity as well as has the potential for use as a powerful biomaterial for antibacterial deodorant.

  • This study investigated the frequency, amount and consumption patterns of 50 potentially hazardous foods (PHF) along with consumers' risk perceptions towards PHF in Korea. A quantitative survey was performed from May through August by trained interviewers, surveying 1,000 adults aged over 18 who were randomly selected from six major provinces in Korea. Consumers perceived seafood, including shellfish, mussel, sashimi and sushi, as the top foods with the highest risk, followed by raw sliced beef. The food with the highest frequency intake per month was leafy vegetables, which is used to wrap other foods, followed by blanched vegetables, fried chicken, etc. The group of middle aged individuals with economic stability had the highest frequency intake of sashimi and sushi. Respondents living in small regions consume greater PHF portions at once. Food safety education with regard to the risk of PHF is essential for consumers, with high frequent intake of PHF.

  • Objective: The aim of the paper was to compare the fit of data derived from daily automatic milking systems (AMS) and monthly test-day records with the use of lactation curves; data was analysed separately for primiparas and multiparas. Methods: The study was carried out on three Polish Holstein-Friesians (PHF) dairy herds. The farms were equipped with an automatic milking system which provided information on milking performance throughout lactation. Once a month cows were also subjected to test-day milkings (method A4). Most studies described in the literature are based on test-day data; therefore, we aimed to compare models based on both test-day and AMS data to determine which mathematical model (Wood or Wilmink) would be the better fit. Results: Results show that lactation curves constructed from data derived from the AMS were better adjusted to the actual milk yield (MY) data regardless of the lactation number and model. Also, we found that the Wilmink model may be a better fit for modelling the lactation curve of PHF cows milked by an AMS as it had the lowest values of Akaike information criterion, Bayesian information criterion, mean square error, the highest coefficient of determination values, and was more accurate in estimating MY than the Wood model. Although both models underestimated peak MY, mean, and total MY, the Wilmink model was closer to the real values. Conclusion: Models of lactation curves may have an economic impact and may be helpful in terms of herd management and decision-making as they assist in forecasting MY at any moment of lactation. Also, data obtained from modelling can help with monitoring milk performance of each cow, diet planning, as well as monitoring the health of the cow.

  • Background: The execution of fibular allograft augmentation in unstable proximal humerus fractures (PHFs) was technically demanding. In this study, the authors evaluated the clinical and radiographic outcomes after tricortical iliac allograft (TIA) augmentation in PHFs. Methods: We retrospectively assessed 38 PHF patients treated with locking-plate fixation and TIA augmentation. Insertion of a TIA was indicated when an unstable PHF showed a large cavitary defect and poor medial column support after open reduction, regardless of the presence of medial cortical comminution in preoperative images. Radiographic imaging parameters (humeral head height, HHH; humeral neck-shaft angle, HNSA; head mediolateral offset, HMLO; and status of the union), Constant score, and range of motion were evaluated. Patients were grouped according to whether the medial column support after open reduction was poor or not (groups A and B, respectively); clinical outcomes were compared for all parameters. Results: All fractures healed radiologically (average duration to complete union, 5.8 months). At final evaluation, the average Constant score was 73 points and the mean active forward flexion was $148^{\circ}$. Based on the Paavolainen assessment method, 33 patients had good results and 5 patients showed fair results. The mean loss of reduction was 1.32 mm in HHH and 5.02% in HMLO. None of the parameters evaluated showed a statistically significant difference between the two groups (poor and not poor medial column support). Conclusions: In unstable PHFs, TIA augmentation can provide good clinical and radiological results when there are poor medial column support and a large cavitary defect after open reduction.
