Changes of estrus signs and genital organs by hormomal induction of estrus in the bitch (인공발정유도견의 발정기 변화와 생식기의 변화)

  • To investigate changes of estrus signs and genital organs in the bitch by hormonal induction of estrus, fourteen bitches of nulliparous and multiparous(2nd-5th) were grouped into diestrus and anestrus according to their estrus cycle. The hormonal treatments were divided into four groups: group A($PGF_2{\alpha}+PMSG+hCG$) and group B(PMSG+hCG) in diestrus bitches and group C(GnRH+FSH+hCG) and group D(PMSG+hCG) in anestrus bitches. The external signs of proestrus and estrus as well as the vaginal smear findings and natural breeding as estrus detection were investigated in all the experimental groups. Also, genital organs were examined at two months after the hormone treatment. The bitches in anestrus showed 100% of male attraction, vaginal bleeding and vulvar swelling as proestrus signs after the hormonal treatment for estrus induction and they showed higher numerical value of signs than the bitches in diestrus. The group A showed the lowest value in proestrous signs of all the groups. The bitches in anestrus treated with GnRH+FSH showed 100% of positive estrus by vaginal smear findings and 75% of natural breeding as estrus detection index and these values were the highest of all the groups. Pregnancy was recognized in only group C and the conception rate was 7.14% in al the experimental animals. Of the side effects after the hormone treatment, external findings of continous male attraction, continous external swelling and purulent exudate were recognized in all the experimental groups and the bitches in diestrus showed higher value of the findings than the bitches in anestrus. Of the changes of genital organs after the hormone treatment, hypertrophy of uterine horn, sanguineous exudate and purulent exudate as uterine findings were recognized in all the groups and these findings were shown more in the bitches in diestrus than in those in anestrus. These results indicated that group C showed the highest value of all the experimental groups in external signs of estrus and estrus detection and also pregnancy was recognized only in that group, consequently, that the hormonal treatment of group C would be the most effective for estrus induction, and also indicated that bitches in anestrus were more suitable than bitches in diestrus for the induction of estrus. In addition, side effects in external genital organs and uteri after hormone treatment were shown more in the bitches in diestrus than in those in anestrus, indicating that bitches in anestrus would be of choice for estrus induction.

Relationship between Transferable Embryos and Major Metabolite Concentrations in Holstein Donor Cows

  • This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.

Relationship between Estrous Expression Rate, BCS and Transferable Embryos in Holstein Donor Cows

  • This research was investigated the relationship between the number of the transferable embryos and estrus expression rate, BCS (Body Condition Score), which affect the nutritional state of the cow, in Holstein donor cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. When BCS was $$\leq_-$$2.5, the total number of collected ova was 7.3 + 1.9, which is significantly lower (p<0.05) than the numbers 15.4 + 2.8 and 15.4 + 2.1 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively. Whereas the numbers of transferable embryos were 5.2 + 1.4 when BCS was $$\leq_-$$2.5, which was smaller than the numbers 6.0 + 2.1 and 8.5 + 1.8 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively; however, the differences were not significant. As for estrus induction rate, the cow groups whose BCSs were 2.75 and $$\geq_-$$3.0 showed 100.0% and 95.0%, respectively. Whereas the cow group whose BCS was $$\leq_-$$2.5 showed 57.1%, and the differences were significant (p< 0.05). As for estrous expression rate, the cow groups whose BCSs were $$\leq_-$$2.5, 2.75 and $$\geq_-$$3.0 showed 100.0%, 100.0% and 85.7%, respectively; however, the differences were not significant. According to the result of this research, it is considered that the total number of collected ova and the number of transferable embryos will be affected by the nutritional state before and after in vivo embryo production and superovulation treatment, and that although the mechanism is not clear, poor stockbreeding management and nutritional level would cause the decrease of ovum recovery rate and the number of transferable embryos in high-producing cows. On the other hand, diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.

Relationship between Nutritional Status and Transferable Embryos in Hanwoo Donors (한우 공란우의 영양 상태와 이식 가능 수정란과의 관계)

  • The objective of this study was to investigate the relationship between body weight, body condition score (BCS), blood urea nitrogen (BUN), glucose, cholesterol and number of transferable embryos for the purpose of improving reproductive performance in Hanwoo donors. Seventy five cows, at random stages of the estrous cycle, received a CIDR together with injection of 1mg estradiol benzoate and 50 mg progesterone, and gonadotropin treatment begann. Four days later, the animals were superovulated with a total of 28AU FSH (Antorin, 2AU = 1 ml) administered twice daily in constant doses over 4 days. On the 3rd administration of FSH, CIDR was withdrawn and 25 mg $PGF_2{\alpha}$ was administered. Cows were artificially inseminated twice after estrous detection at 12 hr intervals. The cows received $100{\mu}g$ GnRH at the time of 1st insemination. Embryos were recovered 7 days after the 1st insemination. In conclusion, cows with body weight < 400, 400~450 and > 450kg had number of transferable embryos of $4.2{\pm}1.7$, $6.1{\pm}2.7$ and $4.8{\pm}2.6$, cows with BCS <2.25, 2.25~2.75 and ${\geq}2.75$ had number of transferable embryos of $4.6{\pm}1.6$, $5.7{\pm}2.4$ and $5.1{\pm}2.7$ respectively. These data indicate that a body weight and BCS for superovulation of CIDR-treated Korean native cows does not affect the embryo yield.

Comparison of Oocyte Recovery Rates between Prepubertal and Adult Korean Native Goats (미성숙 및 성숙 재래 산양의 과배란 처리에 의한 난자의 회수율 비교)

  • This study was conducted to examine whether efficiency of oocyte production from superovulated prepubertal goats. Fifteen prepubertal and twenty adult goats, maintained in a pen under natural day length and fed hay ad libitum, were pretreated with progestagen implanted CIDR for 10 days. Superovulation treatment of the goats received twice daily intramuscular injection of a total of 70 mg FSH for 3 days from Day 8 of CIDR. All the gonadotrophin treated goats were injected with 10 mg $PGF_2{\alpha}$ on Day 8 and 400~600 IU hCG in the afternoon on Day 10. Oocytes were recovered by follicle aspiration or oviduct flushing at 35 to 40 h after hCG injection through mid-ventral incision. The in vivo matured oocytes was activated by ionomycin (5 min) and 6-DMAP (3.5~4 h). The activated oocytes were cultured in mSOF medium containing 0.8% BSA at 38.5$^{\circ}C$ in an atmosphere of 5% CO$_2$, 5% O$_2$, 90% N$_2$ for 7~8 days. There was no significant difference in the mean number of CL and in vivo matured and follicular oocytes recovered. But, quality of I + II grade follicular oocytes was lower (p<0.05) in the prepubertal goat (25.0%) than the adults (52.4%). The same results were also observed in the cleavage and blastocyst rate of activated oocytcs. The cleavage and blastocyst rate from prepubertal derived oocytes were lower (p<0.05) in the prepubertal goat (54.5%, 23.3%) than the adult goat (86.8%, 46.6%). Considering overall these results, we suggest that maturation of donor goats is a major factor affecting recovered oocytes quality and in vitro development of activated goat oocytes.

Studies on the Ovarian Respones of Rabbits Superovulated Repeatedly (반복과배란토끼의 난소반응에 관한 연구)

  • This study was carried out to investigate the changes in ovary in repeatedly superovulated rabbits. A total of 57 New Zealand White and Californian, 25 mature virgin and 32 immature does were used in this study. For induction of repeated superovulation, PMSG and HCG were injected at 17-day and 30-day intervals for mature does and 17-day intervals for immature ones. The repeatedly superovulated does at 17-day intervals were induced luteolysis of pseudopregnant corpus luteum with PGF2${\alpha}$ on Day 8 to 9 p.c. The effect of repeated superovulation on reproductive organs was investigated on Day 3 p.c. in mature does and on Day 3 and 6 p.c. in immature ones, respectively. 1. In mature virgin does, the number of ovulation points in the 2nd and 3rd superovulation period averaged 7.0 and 5.0 at 17-day intervals and 13.4 and 6.0 at 30-day intervals, respectively. These numbers were statistically similar to 9.5 ovulation points in the control. However, there were less (p<0.05) ovulation points in those periods compared with 22.1 ovulation points in the 1st superovulation period. 2. In immature does, the number of ovulation points in the 2nd and 3rd superovulation period averaged 5.3 and 2.3, respectively. These numbers were significantly (p<0.05) decreased than 17.1 ovulation points in the 1st periods. The number of ovulation points in the 2nd superovulation period was similar to that in the control, but there was a significant (p<0.05) decrease in the number of ovulation points in the 3rd period as compared to the control. 3. In mature virgin does, the number of visible normal and hemorrhagic follicles (>1.0mm diameter) on day 3 p.c. averaged 19.1 and 8.9 in the 1st superovulation period, respectively. In the 2nd 3rd superovulation period, the number of normal follicles averaged 8.3 and 15.5 at 17-day intervals and 17.8 and 14.5 at 30-day intervals. The number of hemorrhagic follicles in the 2nd and 3rd superovulation period averaged 6.3 and 2.0 at 17-day intervals and 5.2 and 7.8 at 30-day intervals, respectively. There was a slight decrease, although not significant, in the number of normal and hemorrhagic follicles in the 2nd and 3rd period at 17-day intervals compared to that in the 1st period. 4. In immature does, the number of visible normal follicles on day 3 and day 6 p.c. in the 1st superovulation period averaged 27:3 and 26.1, respectively. The follicles on day 3 p.c. tended to increase slightly more than that in the cortrol, but the average number of normal follicles on day 6 p.c. did not differ from that in the control. The number of visible hemorrhagic follicles on day 3 and day 6 p.c. in the 1st of follicles in the 1st superovulation period average 10.2 and 9.9, respectively. There was a slight increase in the number of follicles in the 1st period compared to that in the control. In the 2nd and 3rd superovulation period, the number of normal follicles revealed a slight decrease in the 3rd period, but the number of hemorrhagic follicles was not different between periods. 5. The number of growing follicles with incipient intral formation on day 3 p.c. in mature does of the 1st superovulaton period average 29.7 and the average number of growing follicles in the 3rd period was 26.7 at 17-day intervals and 31.0 at 30-day intervals, respectively. These numbers did not differ from that in the control. In immature does, the number of growing follicles averaged 57.7, 45.0 and 59.3 in the 1st, 2nd and 3rd superovulation period, respectively. There was a slight but not significant decrease in the number of growing follicles in the 3rd period compared to that in the control.

Effect of Multiple Superovulation and Parity on Embryo Production in Hanwoo (한우의 반복 과배란 및 산차가 수정란 생산에 미치는 영향)

  • This study was performed to investigate the effects of multiple superovulation and parity on embryo production in Hanwoo cows. Donors were superovulate 4 times $1\~2$ months interval and inserted CIDR plus (with the capsule of estradiol benzoate 10mg) on Day 10 from standing heat for 9 days and injected 2.5ml FSH (Antorin R-10) 2 times in a day on 6th day to 10th day from insertion of CIDR and the doses of FSH were decreasing 0.5ml on every 2 times. On 3th day of FSH injection, 25ml $PGF_2{\alpha}$ were injected i.m. and on 4th day, CIDR was removed. After 2 days from removing CIDR, AI was performed 2 times 12 hour apart with 2 straws of Korean Proved frozen Semen and simultaneously 200ug/ml GnRH was injected and embryos were recovered on 7th day from Al. The response rates of superovulated donors were $85.7\%,\;90.5\%,\;62.5\%,\;100\%$ from 1 to 4 times of superovulation, respectively. There were significant differences among No. of superovulation times (P<0.05). The results of transferable embryos were 3.7, 3.4, 3.4, 5.7 from 1 to 4 times of superovualtion, respectively. There were no differences among No. of superovulation times. The results of transferable embryos were 2.5, 3.0, 5.3, 3.0, 3.4 form heifer, first born to 4 the born, respectively. There were significant differences among the parities of donors (P<0.05). These results suggested that even 4 times of superovulations of Hanwoo donors could be able to recover transferable embryos, it might be used the donors maximally and improved the adaptation of embryo transfer to farms safely.

Risk Factors for the Probability of Pregnancy Following Synchronization Protocols in Dairy Cows (젖소에서 배란동기화 프로그램 적용 후 임신율에 영향을 미치는 요인 분석 연구)

  • The objective of this study was to determine the risk factors associated with pregnancy following 3 synchronization protocols in dairy cows. Data were collected on 1,952 cows from 22 dairy farms, including synchronization protocols ($PGF_{2{\alpha}}$ + estradiol benzoate [PG+EB], Ovsynch, and CIDR-ovsynch), cow parity, body condition score (BCS), and dates of previous calving, insemination and conception. The odds ratio (OR) for pregnancy were analyzed by logistic regression using the LOGISTIC procedure in SAS. The analysis revealed that farm (p = 0.005), cow parity (p = 0.0001), BCS (p < 0.005), and AI season (p < 0.05) significantly affected and calving to AI interval tended to affect (p < 0.1) the probability for pregnancy. Although synchronization protocols did not affect the probability for pregnancy (p > 0.05), cow parity and synchronization protocols showed a significant interaction (p < 0.005); the OR (0.60) was significantly lower (p < 0.0001) for multiparous cows compared to primiparous cows using PG+EB, whereas the OR (1.44) tended to be higher (p < 0.1) for multiparous cows compared to primiparous cows using the Ovsynch, and the probability for pregnancy did not differ between multiparous and primiparous cows using the CIDR-ovsynch (p > 0.05). Cows with BCS ${\geq}$ 3.00 were more likely pregnant (OR: 1.41) compared with cows having BCS ${\leq}$ 2.75, whereas cows inseminated during summer had a lower OR (0.73) compared with those inseminated during spring. Cows with a calving to AI interval > 150 days were more likely to be pregnant (OR: 1.20) compared with cows with a calving to AI interval ${\leq}$ 150 days. In conclusion, the OR for pregnancy following synchronization protocols in dairy cows was affected by farm, parity, BCS, calving to AI interval of the cow, and AI season, and there was a significant interaction between cow parity and synchronization protocols; the OR for pregnancy was lower for multiparous cows compared with primiparous cows using the PG+EB protocol.

A Statistical Analysis of the Fereign Bodies in the Food and air Passages (식도 및 기도이물의 임상통계학적 고찰)

  • The foreign bodies in the food and air passages are frequently observed in the field of otolaryngology, and the foreign bodies in the air passages have much significance in clinical practice because they may cause sudden death. A statistical study was done on 95 cases of foreign bodies in the food and air passages who had visited department of otolaryngology, Chung-Ang university from June, 1968 to April, 1978. The results were as follows; 1. The total cases of foreign bodies in the food and air passages was 95; 89 cases (93.7%) were in the food passage and the remaining (6.3%) were in the air passage. The ratio between the food passage and air passage was about 14. 8 to 1.0. 2. In distribution by sex, 64 cases (67.4%) were in male patients and the remaining 31 cases (32.6%) were in female patients. The ratio between male and female was 2.1 to 1.0. 3. The kinds of foreign bodies in the food in the order of their frequency, were coin, gogame stone and pebble. In the air passages, the peanut and bean were most frequently found. 4. In distribution by age, 64 cases (67.4%) of all foreign bodies were found in children under 5 year old, and coin was the most common kind of foreign body. Except for meat, almost all of foreign bodies were found in children under 10 year old. 5. There was chronologically no significant tendency in incidence; the incidence, however, had decreased during recent 2 years. 6. In the location of foreign bodies in the food passage, 70 cases (78.7%) were found at the first esophageal narrowing. Meat was more frequently found at second esophageal narrowing associated with cicatrical stenosis. Almost all of foreign bodies in the air passage was found in bronchi; 3 cases were in the right side of bronchi, and 2 cases were in the left side of bronchi and 1 case was in glottic region. 7. In duration of lodgement, 50 cases (52.6%) visited our hospital within 24 hours, and 3 cases after 30 days. 8. Under topical anesthesia, 83 cases (93.3%) of the foreign bodies in the food passage were removed by esophagoscopy, by 6 cases (6.7%) failed to be removed. All of the foreign bodies of the air passages (6 cases) were removed by bronchoscopy. Among them, 5 cases under 5 years old were removed by inferior bronchoscopy through tracheostomy site.
