Protein Quality Evaluation of Cooked Monkfish (Lophiomus setigerus) Meats

  • To investigate the effect of cooking methods on protein quality of domestic fresh monkfish meat (FMM) and imported frozen monkfish meat (IMM), in vitro protein qualities were determined by amino acid anlysis, trypsin indigestible substrate (TIS) formation, and protein digestibility using the four-enzyme method. Crude protein contents of the boiled FMM and IMM were $90\%$ of the dry base, which were higher than fresh FMM $(82\%)$ and IMM $(84\%)$. Profiles of total amino acid in FMM and IMM were not changed by cooking methods. Total free amino acid contents decreased to $ 29.0-33.6\%$ for boiled $(l00^{\circ}C,\;10 min)\;and\;24\%$ for steamed $(100^{\circ}C,\;10\;min)$ samples. In vitro protein digestibilities of boiled and steamed FMM incnased $86.6-86.8\%$, compared to raw IMM $(82.9\%)$, boiled and steamed IMM $85.1-85.5\%$ and raw IMM $(83.6\%)$. TIS of FMM (23.6 mg/g solid) and IMM (15.9 mg/g solid) showed no significant (p<0.05) difference in cooking methods. The C-PERs (computed protein efficiency ratio) of boiled FMM (2.63) and IMM (2.50) were significantly higher (<0.05) than raw (1.97) and steamed FMM(1.97) and IMM(1.94). These results demonstrate that boiling of FMM and IMM improves protein digestibility and C-PER when compared to steamed FMM and IMM. Therefore, boiling could be an excellent means to maintain high-protein quality of monkfish meat. Also, the cooking method may be applicable to the preparation of monkfish stew without any loss of free amino acids.

Mode of Antiviral Activity of Water Soluble Components Isolated from Elfvingia applanata on Vesicular Stomatitis Virus

  • A preparation of water soluble components (EA) was made from carpophores of Elfvingia applanata (Pers.) Karst and its in vitro antiviral activity on vesicular stomatitis virus [(Indiana serotype, VSV(IND)] was investigated by plaque reduction assay. EA exhibited potent antiviral activity on VSV(IND) growth and negligible cytotoxicity on Vero cells, 50% effective concentration ($EC_{50}C$/) of 104$ug\textrm\/ml$ and 50% cytotoxic concentration ($CC_{50}C$) of 3,793$ug\textrm\/ml$, respectively. Selectivity index (Sl $CC_{50}C$/$EC_{50}C$) of EA on Vero cell and VSV(IND) was about 36.5. EA did not display either a direct virucidal effect on V5V(IND) or induction of antiviral substance by Vero cells upon its treatment. Thus, the mode of antiviral activity of EA was studied at steps of viral adsorption onto cell. When both EA and virus were added to cell monolayers, titer of cell-free virus in culture supernatant increased in ca. 30-40% compared with that of control group and titer of cell-associated virus was 60-100% higher than that of control group. These results suggested that antiviral activity of EA on VSV(IND) might be due to the hindrance of viral entry to cells at eITher endocytosis or loss of envelope.

  • Botrytis cinerea Pers. was found to be highly virulent to the grapevine plant, especially in greenhouse condition. Pseudomonas species play key roles for the biocontrol of many plant diseases especially in soil. Of the 83 isolates of Pseudomonas spp., a bacterial strain P84, isolated from tomato rhizosphere, was shown to suppress a wide range of phytopathogenic fungi in vitro. The isolate was identified as Pseudomonas putida on the basis of its bacteriological and genetic characteristics. The P. putida P84 strain carry the phlD gene for 2,4-diacetylphloroglucinol biosynthesis and may produce the antibiotics as an antagonistic mechanism involved in biocontrol. The antagonistic activity of the bacterium has a promising implication for its use as a biocontrol agent to control grapevine gray mold.

  • Wir $k\"{o}nnen$ die $Ged\"{a}chtnisforschungen$ mit Fremdsprachenlernen $verkn\"{u}pfen$. Ein Lernprozess ist gerade einer vom $Ged\"{a}chtnisprozess$. $F\"{u}r$ einen erfolgreichen Fremdsprachenunterricht ist es $n\"{o}tig$, dass beim Lernprozess ein Sprachwissen vom deklarativen $Ged\"{a}chtnis$ ins prozeduale $Ged\"{a}chtnis$ $\"{u}bertragen$ wird. Aber ein Lernerfolg ist je nach dem Individiuum anders. d.h. der unterschiedliche Lerneffekt zwischen Lemenden $h\"{a}ngt$ eventuelle von $pers\"{o}nlichen$ Erfahrung und von dem episodischen $Ged\"{a}chtnis$. Beim Lernprozess muss Sprachwissen vom deklarativen $Ged\"{a}chtnis$ zum prozedualen $Ged\"{a}chtnis$ $\"{u}bertragen$ werden, um Fremdsprachen zu beherrschen. Inzwischen spielen implizites $Ged\"{a}chtnis$ und episodisches $Ged\"{a}chnis$ wichtig Rolle beim $\"{U}bertragungsprozess$. $Daf\"{u}r$ zeigen wir Lernstrategie der Tempusverwendung in Bezug auf die $Ged\"{a}chtnisforschung$. Mit welchem Tempus will man seine Intention im Text $ausdr\"{u}cken$? $Daf\"{u}r$ zeigen wir ein einfaches Spiel $f\"{u}r$ den Fremdsprachenunterricht. Das ist das Spiel zur Strategie der Tempusverwendung bei der Textproduktion $f\"{u}r$ das reportiv-orientierte Thema. Unsere Untersuchung der Tempera kann besonders im Fremdsprachenunterricht dizu beitragen, dass der Lernende als Produzent einerseits unter Berucksichtigung der Textsorte, der zeitlichen Orientierung des Theams und der Sprecherabsicht bei der Textproduktion die Tempera ohne $Z\"{o}gern$ $ausw\"{a}hlen$ und benutzen kann und dass der Lernende als Rezipient auch durch das Erlernen der verschiedenen Funktionen der Tempera versteckte Absichten des Produzenten richtig verstehen kann. Unsere Untersuchung kann in diesem Sinne hinsichtlich der Tempusfunktion bei der erfolgreichen Erweiterung der kommunikativen $F\"{a}higkeiten$ im Unterricht behilflich sein.

A Study on the Amino Acid Contents of Edible Mushrooms (식용(食用)버섯류(類)의 아미노산(酸)에 관(關)한 연구(硏究))

  • Free amino acids in extracts and total amino acids in hydrolysates of eleven species of edible mushrooms were analyzed by amino acid autoanalyzer (Technicon PNC-1 Type). All these 11 species of mushroom can be repesented for convenience sake as follows. S-1; Agaricus campestris Fr. S-2: Agaricus campestris S-3; Pholiota nameko(I. Ito) S. Ito et Imai S-4; Auricularia auricula-judae(Fr.) $Qu{\acute{e}}l$ S-5; Tremella fuciformis Berk. S-6; Tricholoma matsutake(S. Ito et Imai) Sing. S-7; Pleurotus ostreatus Fr. $Qu{\acute{e}}l$ S-8; Lentinus edodes Berk Sing. S-9; Ramaria botrytis (Pers.) Ricken S-10; Coprinus comatus(Fr.) S.F, Gray S-11; Gyrophora esculenta The results obtained from this study are as follows. 1) 17 kinds of amino acid, including 7 kinds of essential amino acid in human nutrition except tryptophan were identified and quantified. 2) Of all free amino acids contained in mushrooms, glutamic acid is the richest, and then comes Ala, Thr, Pro and Lys in that order. There were no found Cys'and His in S-9;His in S-1; Met and Arg in S-11; Cys and Met in S-5;Pro, Cys, Met, Lys and Arg in S-4. Of all total amino acids which are closely related with nutritional valuation, glutamic acid is the richest, and then comes Asp, Ala, Arg, Leu, Thr, Gly in that order. Especially S-1 and S-2 contain high quantity $o{\acute{i}}$ proline in both free and total amino acids. 3) Cotents of ammonia in extracts of mushrooms in decreasing order in S-1, S-10, S-8, S-2, S-7, S-6, and S-2, S-6, S-8, S-9, S-1 in hydrolysates of mushrooms. 4) Gross Contents of free amino acid in extracts is high in decreasing order in S-10, S-1, S-7, S-6, S-8, and total amino acid in hydrolysates is high in S-10, S-2, S-2, S-8, S-1, S-9, S-6. 5) Besides 17 kinds of amino acid, 5 kinds of unknown amino acid are found in extracts and hydrolysates.

  • The need for insect and mite resistant turfgrass cultivars arose because of problems associated with pesticide use. Representative cultivars and genotypes of 18 warm-season turfgrass [Zoysia japonica Steud., Z. japonica${\times}$Z. metrella hybrids, Z. japonica${\times}$Z. tenuifotia hybrids, Z. matrella (L.) Merr., Cynodon dactylon (L.) Pers., C. dactylon${\times}$C. transvallensis hybrids, Paspalum notatum Flugge., P. vaginatum Swartz., Stenotaphrum secundatum (Walt.) Kuntze, Eremochloa ophiuroides (Munro.) and Buchloe dactyloides (Nutt.) Engelm.] and 20 cool-season turfgrasses [Poa pratensis L., Festuca arundinacea Schreb., F. rubra L., F. rubra var. commutata Gaud., F. ovina var. duriuscula L. Koch. Agrostis tenuis Sibth., A. palustris Huds., and Latium perenne L.] were evaluated for host resistance to feeding by the Spodoptera depravata (Butler) in the laboratory. Two experiments were set up in the laboratory using 8.5cm diameter${\times}$4.0cm deep plastic petri dishes as larvae feeding chambers. In experiment 1, one neonate larvae were place on the grass in each dish and the dishes were arranged with 5 replicates each within an environmental chamber maintained at $25^{\circ}C$ and 15h light: 9h dark Larval survival and larval weights at 7d and 14d, pupal weights, and days to pupation were compared among turfgrasses. In Experiment 2, 4cm sections of all grasses were oriented equidistant from each other in a pattern resembling the spokes of a wheel. Five one neonate larvae were introduced to the center of each dish. Dishes were immediately placed in an environmental chamber held at $25^{\circ}C$, 15h light: 9h dark Larvae were allowed to feed for 24h. Damage was rated from 0(no damage) to 9(completely consumed) were made for eachturfgrass. Resistance as antibiosis (high mortality, slowed growth, and least preference) was identified in Z. japonica${\times}$Z. tenuifolia hybirids ‘Emerald’, Z. japonica${\times}$Z. metrella hybirds ‘Miyako’ and Eremochloa ophiuroides (Munro.). Cool-season turfgrasses tested were susceptible to feeding by Spodoptera depravata (Butler).

Some Unrecorded Genera and Species in Korean Agaricales (II) (한국산 주름버섯목의 미기록속과 종(2))

  • The investigation on the species diversity for the Korean fungal flora was carried out by authors. Five species of the Agaricales recorded in the present paper are distributed Hygrocybe laeta (Pers.)P. Kummer var. laeta, Lyophyllum tylicolor (Fr.) M. Lange & Sivertsen, Clitopilus scyphoides (Fr.) Singer, var. scyphoides, Entoloma rusticoides (Gillet) Noordel. and Lactarius uyedae Singer were recorded for the first time and are described and illustrated in detail. Specimens cited here are deposited in the Herbarium Conservation Center of National Academy of Agricultural Sciences.

Protein Nutritional Qualities of Fish Meat Extracts and Their Residues (양식어류 고음과 잔사의 단백질 품질평가)

  • To confirm the food quality of conventionally processed fish extracts, protein quality of boiled crucian carp(Carassius carassius) and bastard halibut(Paralichthys olivaceus) extracts and their residues were evaluated. For the both fish extracts, some of the essential amino acids were lowered significantly but two times more proline and glycine were detected in extracts than those in raw fish meats. Boiling(100oC, 5 hours) caused 1.8(crucian carp)~2.4(bastard halibut) times more total free amino acid contents in fish extracts as compared to those in original fish meats. Taurine, glutamic acid, proline, lysine, and ammonia were the predominant free amino acids released in fish extracts. In vitro digestibility of boiled fish extracts were lower at a level of 4~6% than those of raw fish meats. Fish extraction residue had a higher in vitro digestibility and had a 60% lower level of TI than that of original fish meats. 18(bastard halibut)~ 24%(crucian carp) of available lysine was reduced in boiled fish extracts but a remarkable variation was not noted between extracts and residues. PERs and NPRs of fish extracts were significantly lower than those of casein, while those values of extraction residue were slightly higher as compared to those of control(ANRC casein). In vivo apparent digestibility exhibited a similar trend to in vitro digestibility. Hematological properties in serum of rat fed with fish extracts and residue were not changed significantly but the serum cholesterol concentration were reduced in rats fed fish extraction residue comparing with those of control. These results suggest that body weight loss due to fish extracts may not affect physiological changes.

Flora of Vascular Plants in Deokjeokdo (Ongjin-gun) and Its Adjacent Regions, Korea (덕적도(옹진군) 및 인근 도서지역의 관속식물상)

  • This study was carried out to inventory the vascular plant flora in Deokjeokdo and its adjacent regions (Mungapdo, Soyado), Ongjin-gun, South Korea, from April to October 2014. Based on the voucher specimens, vascular plants in this area consisted of 108 families, 362 genera, 578 species, 5 subspecies, 66 varieties and 4 forms, totally 653 taxa. In the flora of this area, 5 taxa of Korean endemic plants were found distributed in the sites, including Hepatica insularis Nakai and Asarum glabrata (C.S.Yook & J.G.Kim) B.U.Oh. Korean rare and endangered plants found in this area were 1 taxa of Critical Endangered Species (CR) and 5 taxa of Vulnerable Species (VU). The floristic regional indicator plants found in this area were 67 taxa comprising 3 taxa of grade V, 2 taxa of grade IV, 12 taxa of grade III, 3 taxa of grade II and 47 taxa of grade I. Naturalized plants consisted of 67 taxa, such as Chenopodium album L. and Erigeron annuus (L.) Pers. were recognized widely distributed in the area.

The Effects of Soil Particle Composition on Soil Physical Properties and the Growth of Woody Plants (토양의 입도조성이 토양의 물리성 및 목본식물의 생장에 미치는 영향)

  • This study has conducted to analyze the crelationship among soil properties and to investigate how they affect soil physical characteristics and plant growth. The experiment of woody plant growth was conducted as follows : Type I was the original soil. Type II, the soil particles smaller than 20${\mu}{\textrm}{m}$ was removed from the original soil. Type III, the soil particles is smaller than 75${\mu}{\textrm}{m}$ was removed from original soil. Wisteria floribunda A.P.DC and Celtis sinensisi Pers. were used for plant growth measurement. 1. Soil type II. the closest to Fuller's curved line, showed high dry bulk density and low in soil pores and saturated hydraulic conductivities. This created poor soil aeration and limited space for the root to growth. When the root did not have sufficient space to grow, there was a lot of physical stress, which hindered the root growth. 2. Soil typeIII was high saturated hydraulic conductivity and a lot of soil pores larger than 10 ${\mu}{\textrm}{m}$. As a result, there were more available spaces for root to spread. It was considered that there was less physical stress for root growth. Therefore, soil typeIII showed significantly greater root growth. 3. Because soil type III has less small particles and saturated hydraulic conductivity was high, and water infiltrates rapidly into the underground when there was rainfall or irrigation. The soil typeIII becomes much stronger soil mechanically due to the less small particles. Therefore, soil typeIII was a suitable material for applying on planting sites where soil compaction is expected.
