Cultural Characteristics of Ophiocordyceps heteropoda Collected from Korea

  • Isolates of Ophiocordyceps heteropoda (Kobayasi) collected from Mt. Halla on Jeju-do, Korea were tested for mycelial growth on different agar media and in the presence of different carbon and nitrogen sources. Similarly, isolates were also incubated at different temperatures as well as under continuous light and dark conditions. Growth was better on Hamada agar, basal medium, and malt-yeast agar, but poor on Czapek-Dox agar. Different carbon sources such as dextrin, saccharose, starch, lactose, maltose, fructose, and dextrose resulted in better growth. Complex organic nitrogen sources such as yeast extract and peptone revealed the most effective growth. Mycelial growth was best at $25^{\circ}C$. The growth rate was faster in the dark than the light, but mycelial density was less compact in the dark.

  • Chemotaxis is the directed movement of cells in gradients of signaling molecules, an essential biological process that underlies morhpogenesis during development, and the recruitment of immune cells to sites of infection. Especially, bacterial chemotaxis has utilized as an important prelude to study metabolism, prey-predator relationship, symbiosis, other ecological interactions in microbial communities. Recently, novel analytical formats integrated with microfluidics were introduced to investigate the chemotaxis of the cells with the precise control of chemical gradient and small volume of cells. In this study, we present a method to detect bacterial chemotaxis by direct fluidic contacting. The developed fluidic-handling method is driven by capillary force, hydrophobic barrier and a cohesion force between fluids. We have investigated the chemotactic response of E Coli. and Pseudomonas aeruginosa to three kinds of chemoeffectors such as HEPES buffer, peptone and chloroform.

Production of Laccase and Bioremediation of Pentachlorophenol by Wood-Degrading Fungus Trichophyton sp. LKY-7 immobilized in Ca-Alginate Beads

  • The wood-degrading fungus Trichophyton sp. LKY-7 (T. LKY-7) was immobilized in ca-alginate beads for laccase production and PCP bioremediation. The immobilized T. LKY-7 enabled the repeated use of this fungus for laccase production and produced high amount of laccase throughout 5 cycles incubation. As a laccase inducer. oak wood meal (Quercus variabilis) seemed to be effective laccase inducer for T. LKY-7, and the optimum addition amount was 1% (W/W) in glucose-peptone medium. Bioremediation of pentachlorophenol by the immobilized T. LKY-7 reached an efficency of up to 90% without toxic inhibition. The immobilized T. LKY-7 might thus be applicable for semicontinuous laccase production and bioremediation to serve inoculum for reactor system.

Preparation and Characteristics of Alkaline -active Cellulases from Coprinaceae

  • Coprinus cinereus 2249 producing alkaline-active cellulase was screened from 29 species of Corpinaceae and constitutively produced alkaline carboxymethyl cellulase (CMCase) and filter paper cellulase (Fpase). When cultivated at pH 9.0, 25$^{\circ}C$ and 5 days, copnnus cinereus 2249 produced higher alkaline activity on 0.5% CMC, 2% wheat bran as carbon source and 0.5% peptone, 0.05% yeast extract as nitrongen source compared with other culture conditions. The level of cellulase production was higher in the presence of wheat bran than in the presence of CMC. The optimum temperature and pH for alkaline -active cellulase activity weas 50$^{\circ}C$ and 9, 0, respectively.

  • Effects of the plant growth regulator (P. G. R.)on the reaction of proteinase, $\gamma-amylase$ and acid phosphatase were investigated, and also were the conditions of production of P. G. R. by Stroptomyces sp. 445. The P. G. R. had no effect on the act ivities of such enzymes in mung bean seedling. But in germinating seed previously treated with P. G. R. it effected the activity of protease in cotyledon. In the conditions of production of P. G. R., the maxim, activity was appeared in shaking cutlure at $30^{\circ}C$ for 5 days, and by the addition of peptone or casein hydrolysate as nitrogen source, soluble starch as carbon source, and sulfur as metal ion.

Yeasts Associated with Roots of the Endemic Plant Mankyua chejuense

  • BACKGROUND: Identification of endophytic yeasts inhabiting the internal roots of the Mankyua chejuense tree requires techniques involving biotechnology. There is a need for a culture-based method to isolate and identify yeast strains associated with M. chejuense.METHODS AND RESULTS: We spread homogenized M. chejuense root samples onto glucose-peptone- yeast agar containing antibiotics, Triton X-100, and L-sorbose. A total of 152 yeast isolates were obtained and identified via phylogenetic analysis based on ITS gene sequencing. The results revealed that the root-associated yeast species included the genera Cyberlindnera (140 isolates), Candida (11 isolates), and Kluyveromyces (one isolate). Additionally, three yeast isolates showed high bioethanol production.CONCLUSION: We identified the specific yeast community associated with M. chejuense roots. These yeast isolates may have industrial applications as bioethanol producers. Our findings revealed that Cyberlindnera isolates included C. suaverolens and C. satumus, while Kluyveromyces isolates showed high bioethanol production.

Monasucs anka의 적색조 생산 특성 (Characterization of Red Pigment Production by Monascus anka)

  • Optimal media and cultural conditions for the production of red pigment were established using Monasurs anka KFCC 4478. The optimal temperature and initial pH for the production of red pigment were 30$^{\circ}C$ and 7.0, respectively. Glucose turned out to be most suitable carbon source for red pigment production. Optimal glucose concentration was 3.0%. Addition combined of nitrogen sources of peptone and NaNo3 induced good red pigment production. Thiamine-HCI and nicotinic acid were increased the production of red pigment. Under optimal conditions, maximum red pigment production and cell growth were observed after 5 days of incubation.

방선균이 생성하는 단백질 가수분해효소 저해물질의 생산 (Production of Protease Inhibitor from Streptomyces sp. SK-862)

  • A inhibitor acting on substrate proteolytic enzyme was isolated from culture broth of Streptomyces sp. SK-862, which had been isolated from soil in Wonju City, by using the colloidal agar medium. The optimum culture temperature and initial pH for the production of the protease inhibitor was 28$^{\circ}C$ and pH 8.5, respectively. The optimum culture medium was composed of 1.5% glucose, 0.5% peptone, 0.1% K2PHO4, 0.05% CaCO3 and initial pH 8.5. The inhibitor production was maximum when the strain was incubated in shaking incubator at 70 strokes for 60 hours.

Growth of Clostridium thermobutyricum: a Cellulolytic Thermophile

  • Increased concentrations of yeast extract led to increased growth yields and faster growth rates of the newly isolated Clostridium thermobutyricum. This species produced butyrate as its main fermentation product from glucose as well as from yeast extract. In the presence of peptone or tyrptone and during growth on agar, up to 70% of the cells sporulated. Growth yields were 30 and 55 g per mole glucose in the presence of 0.05 and 2.0% yeast extract, respectively. The Arrhenius graph was biphasic, exhibiting an intermediary plateau around $38^{\circ}C$ with a concomitant change in the Arrhenius energy. The optimum temperature was $55^{\circ}C$. An unusually sharp decline in the growth rate occurred above $59^{\circ}C$ .

Streptomyces luteogriseus KT-10에 의한 Cathepsin B 저해물질의 발효생산 (Production of Cathepsin B Inhibitor by Steptomyces luteogriseus KT-10)

  • Streptomyces luteogriseus KT-10 isolated from Korean farm soil produced a strong cathepsin B inhibitor. Optimal conditions for the cathepsin B inhibitor production by s. luteogriseus KT-10 were evaluated. The cathepsin B inhibitor was produced with maximal yield in the cultural condition of pH 7.0 and $25^{\circ}C$ for 4 days. Optimal medium for the cathepsin B inhibitor production was determined to be a medium containing 20g, peptone 3g, yeast extract 1g, K2HPO4 0.5g, MgSO4.7H2O 0.5g, NaNO3 0.5g, NaCl 0.5g per l. The cathepsin B inhibitor produced by S. luteogriseus KT-10 could also inhibit the other proteinases such as trypsin, papain, and cathepsin D.
