Synthesis and Characterization of Sodium Acrylate and 2-Acrylamido-2- Methylpropane Sulphonate (AMPS) Copolymer Gels

  • A series of superabsorbents based on acrylic acid (AA), sodium acrylate, 2-acrylamido-2-methylpropane sulphonic acid, N,N'-methylene bis-acrylamide (MBA) were prepared by inverse suspension polymerization. These hydrogels were further crosslinked on the surface with polyethylene glycol-600 (PEG-600). The water absorbency or swelling behaviors for these xerogels in water and 0.9% saline solutions, both under free condition and under load were investigated. Absorption characteristics of these hydrogels were found to depend on nature and concentration of crosslinker in the system. It was also found that the saline absorption was significantly improved as the incorporation of AMPS in the polymer was increased. The surface crosslinking introduced in the polymers was found to improve the absorption under load characteristics without lowering the free water absorption capacities of the polymer to a considerable extent.

Preparation of Crack-free YBCO Films by EPD on Silver

  • Electrophoretic deposition (EPD) of alcohol YBCO suspensions on the Ag wire electrode is studied. Poly(ethylene glycol) was coordinated to a structure formed by the EPD process with YBCO particles. The suspension is characterized in terms of zeta potential and conductivity. The d.c electric fields of 200-300 V/cm are applied for 1-10 min. The optimal condition for the EPD allows modifying the properties and microstructure of the deposited films. Superconducting coatings with nanometer-sized pores and a preferred orientation along the c-axis were prepared from the result with chemically modified precursor solution. In contrast, YBCO coatings of submicrometer-sized pores and randomly orientated grains were prepared from the solution without PEG.

Amphotericin B Aggregation Inhibition with Novel Nanoparticles Prepared with Poly(${\varepsilon}$-caprolactone)/Poly(N,N-dimethylamino-2-ethyl methacrylate) Diblock Copolymer

  • Diblock copolymers composed of poly(${\varepsilon}$-caprolactone) (PCL) and poly(N,N-dimethylamino-2-ethyl methacrylate) (PDMAEMA), or methoxy polyethylene glycol(PEG), were synthesized via a combination of ring-opening polymerization and atom-transfer radical polymerization in order to prepare polymeric nanoparticles as an antifungal drug carrier. Amphotericin B (AmB), a natural antibiotic, was incorporated into the polymeric nanoparticles. The physical properties of AmB-incorporated polymeric nanoparticles with PCL-b-PDMAEMA and PCL-b-PEG were studied in relation to morphology and particle size. In the aggregation state study, AmB-incorporated PCL-b- PDMAEMA nanoparticles exhibited a monomeric state pattern of free AmB, whereas AmB-incorporated PCL-b- PEG nanoparticles displayed an aggregated pattern. In in vitro hemolysis tests with human red blood cells, AmBincorporated PCL-b-PDMAEMA nanoparticles were seen to be 10 times less cytotoxic than free AmB (5 ${\mu}g$/ml). In addition, an improved antifungal activity of AmBincorporated polymeric nanoparticles was observed through antifungal activity tests using Candida albicans, whereas polymeric nanoparticles themselves were seen not to affect activity. Finally, in vitro AmB release studies were conducted, proving the potential of AmB-incorporated PCL-b-PDMAEMA nanoparticles as a new formulation candidate for AmB.

동해 남동해역 다중채널 해양탄성파 탐사자료의 해저면, 페그-레그 및 자유해수면 다중반사파 제거 최적화 전산처리 (Optimal Rejection of Sea Bottom, Peg-leg and Free-surface Multiples for Multichannel Seismic Data on South-eastern Sea, Korea)

  • 동해 남동해역에서 2008년 취득된 탄성파 탐사자료에 포함된 다중반사파를 억제하기 위한 전산처리를 수행하였다. 다중반사파는 수심이 얕은 지역의 해저면 하부 1,750 ms까지 250 ms 간격으로 발달하였으며 해저면 다중반사파, 페그레그(Peg-leg) 다중반사파 및 자유해수면 다중반사파로 구분하였다. 다중반사파를 제거하기 위하여 속도분석을 통한 중합, 최소지연 예측 디콘볼루션 필터와 파동방정식을 이용한 다중반사파 제거모듈(Wave Equation Multiple Rejection, WEMR)을 이용하였다. 주요한 다중반사파들은 예측 디콘볼루션을 통하여 제거되었으며 잔존하는 다중반사파의 제거에 WEMR이 유용하였다. 남동해역 탄성파 자료의 특성에 적합한 전산처리 방법을 조합하였고 이에 필요한 매개변수를 정리하여 향후 인근지역의 전산처리 과정에서 응용할 수 있는 결과를 얻었다.

폴리에톡시레이티드 아스코르빈산의 개발 (DEVELOPMENT OF POLYETHOXYLATED ASCORBIC ACID AS A WHITENING AGENT)

  • A series of novel ascorbic acid derivatives, polyethoxylated ascorbic acid (PEAA) were synthesized by coupling ascorbic acid with polyethylene glycol (PEG) of two molecular weights (MW: 350 and 550) at the C-2 or C-3 hydroxyl group (2PEAA350, 3PEAA350, 2PEAA550, 3PEAA550) to increase the stability and retain the activity, as a skin whitening agent. Their stability, scavenging activity against free radical, inhibitory activity against tyroxinase and inhibitory activity of melanin synthesis in Bl6 melanoma cell of PEAAs were evaluated in viかo and compared with those of ascorbic acid and 3-O-ethyl ascorbic acid (3OEAA), a Con stable vitamin C derivative. Among PEAAs, 2PEAA350 and 2PEAA550 tad high scaveniging activity against See radical, inhibitory activity against tyrosinase and inhibitory activity if melanogenesis but low stability, 3PEAA350 had high stability and moderate scavenging activity against free radical, infibitory activity against tyrosinase and inhibitory activity of melanogenesis. The stability, scavenging activity against free radical and inhibitory activity of melanogenesis of 3PEAA350 were higher than those of 30EAA. The most stable 3PEAA350 among PEAAs was nontoxic in various toxicological tests. These results suggest that PEAA would be a good whitening agent far enhancing stability and bioavailability.

  • 본 실험에서는 일련의 poly(ethylene glycol)-poly(D,L-lactide)(또는 -poly(D,L-lactide-co-glycolide))이중블록 공중합체들을 D,L-lactide(또는 glycolide)의 고리열림 중합에 의해 합성하고, 당뇨병 치료제 일종인 rosiglitazone의 가용화를 위한 고분자 미셀 제형으로서의 특성을 평가하였다. 고체분산법을 이용하여 높은 봉입률의 약물함유 미셀 제형을 효과적으로 제조하였고, 미셀 제형의 수용액 내 안정성을 hydrotropic agent의 일종인 2-hydroxy-N-picolylnitinamide(HPNA)의 사용으로 더욱 향상시킬 수 있었다. 생체 외 세포생존평가에서 생체적합성과 낮은 독성을 보였고, 쥐를 이용한 동물실험에서도 약물을 함유한 고분자 미셀이 약물 대조군보다 혈당을 보다 효과적으로 감소시켰다.

고분자량 PEO 기반 분리막에 대한 다양한 고분자 첨가제의 영향 분석 (Polymeric Additive Influence on the Structure and Gas Separation Performance of High-Molecular-Weight PEO Blend Membranes)

  • 기체 분리막의 상업적 발전은 CO2 분리 효율을 향상시키는 데 중요한 역할을 한다. 고분자량 PEO (high-Mw PEO)는 높은 CO2 용해도, 가격 경쟁성 및 견고한 기계적 특성을 가져 분리막 제조용 고분자로 유력하지만 그 특유의 결정성으로 인해 기체 분리막에 응용이 어렵다. 본 연구에서는 결정성 감소를 위해 다양한 고분자 첨가제를 고분자량 PEO에 혼합하는 방법을 제시하였다. 폴리에틸렌글리콜(PEG), 폴리프로필렌글리콜(PPG), 폴리아크릴산(PAA) 및 폴리비닐피롤리돈(PVP)과 같은 상업적으로 이용 가능하고 섞임성이 좋은 수용성 고분자를 첨가제로 사용하여 PEO 결정성을 감소시킴으로써 가스 분리 성능을 향상시키고자 하였다. PEG 및 PPG의 경우 PEO의 결정 구조를 억제하지 못하고 분리막의 결함을 초래하였으나, PAA 및 PVP는 PEO의 결정 구조를 바꿔 결함이 없는 분리막을 제조하는 데 성공하였다. 고분자량 PEO 혼합막의 결정 구조 변화와 기체 분리 성능의 상관관계를 조사하여 본 연구의 결과와 이전에 기록된 결과를 바탕으로 고분자량 PEO에 대한 첨가제 고분자의 설계 및 선택에 대한 통찰력을 제공하며, 이를 통해 비용 효율적이고 상업적으로 실용적인 CO2 분리막을 제조하고자 하였다.

In Vivo Evaluation of Curcumin-loaded Nanoparticles in a A549 Xenograft Mice Model

  • Curcumin (Cum) has been reported to have potential chemo-preventive and chemotherapeutic activity through influencing various processes, inducing cell cycle arrest, differentiation and apoptosis in a series of cancers. However, the poor solubility of Cum limits its further applications in the treatment of cancer. We have previously reported Cum-loaded nanoparticles (Cum-NPs) prepared with amphilic methoxy poly(ethylene glycol)-polycaprolactone (mPEG-PCL) block copolymers. The current study demonstrated superior antitumor efficacy of Cum-NPs over free Cum in the treatment of lung cancer. In vivo evaluation further demonstrated superior anticancer effects of Cum-NPs by delaying tumor growth compared to free Cum in an established A549 transplanted mice model. Moreover, Cum-NPs showed little toxicity to normal tissues including bone marrow, liver and kidney at a therapeutic dose. These results suggest that Cum-NPs are effective to inhibit the growth of human lung cancer with little toxicity to normal tissues, and could provide a clinically useful therapeutic regimen. They thus merit more research to evaluate the feasibility of clinical application.

표지유전자로 형질전환된 연초와 감자로부터 원형질제의 유리 및 융합 (Protoplast Isolation and Fusion of Nicotiana glauca and Solanum tuberose Transformed by Selectable Marker Genes)

  • Protoplasts were isolated from mesophyll of tobacco(Nicotiana glauca) transformed with kanamycin-resistant gene (NPT II gene) and potato hairy root callus containing Ri plasmid of Agrobacterium rhiEogenes, and protoplasm fusion was made between the isolated protoplasts. The transgenic tobacco leaf tissue could grow on the media containing high concentrations of kanamycin, but not on the phytohormone-free media. On the other hand, the potato hairy root calli could be cultured on the phytohormone-free media but not on media containing more than 40 ㎍/ml kanamycin. In these conditions, the viability of both protoplasts were above 90%, These selection markers were used for the selection of protoplasts fused between the two, i.e. protoplast fusion was detected using selection media containing 100㎍/ml kanamycin and with no phytohormone. The mixture of 1.0% cellulase, 0.3% macerozyme, and 0.7M mannitol was best for the maximum protoplast production for tobacco, and that of 2.0% cellulase, 2.0% macerozyme, 1.0% dricelase, and 0.5M mannitol for potato. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution on the selectable medium. Cell walls were regenerated after 5 days in this medium, and colonies were alive until 4 weeks after cultural, but died after 6 weeks.
