Variance component analysis of growth and production traits in Vanaraja male line chickens using animal model

  • Objective: A comprehensive study was conducted to study the effects of partition of variance on accuracy of genetic parameters and genetic trends of economic traits in Vanaraja male line/project directorate-1 (PD-1) chicken. Methods: Variance component analysis utilizing restricted maximum likelihood animal model was carried out with five generations data to delineate the population status, direct additive, maternal genetic, permanent environmental effects, besides genetic trends and performance of economic traits in PD-1 chickens. Genetic trend was estimated by regression of the estimated average breeding values (BV) on generations. Results: The body weight (BW) and shank length (SL) varied significantly (p≤0.01) among the generations, hatches and sexes. The least squares mean of SL at six weeks, the primary trait was 77.44±0.05 mm. All the production traits, viz., BWs, age at sexual maturity, egg production (EP) and egg weight were significantly influenced by generation. Model four with additive, maternal permanent environmental and residual effects was the best model for juvenile growth traits, except for zero-day BW. The heritability estimates for BW and SL at six weeks (SL6) were 0.20±0.03 and 0.17±0.03, respectively. The BV of SL6 in the population increased linearly from 0.03 to 3.62 mm due to selection. Genetic trend was significant (p≤0.05) for SL6, BW6, and production traits. The average genetic gain of EP40 for each generation was significant (p≤0.05) with an average increase of 0.38 eggs per generation. The average inbreeding coefficient was 0.02 in PD-1 line. Conclusion: The population was in ideal condition with negligible inbreeding and the selection was quite effective with significant genetic gains in each generation for primary trait of selection. The animal model minimized the over-estimation of genetic parameters and improved the accuracy of the BV, thus enabling the breeder to select the suitable breeding strategy for genetic improvement.

Synthesis of 2-(5,6-dimethoxy-l-indenyl)ethyl amine (2-(5,6-디메톡시-1-인데닐)에틸 아민의 합성)

  • In order to search the new serotonin bioisoster, 2-(5,6-dimethoxy-l-indenyl)ethyl amine(1) was synthesized. 3,4-Dimethoxybenzaldehyde, as starting material, was condensed with malonic acid in the presence of pyridine and piperidine to form 3,4-dimethoxycinnamic acid(2). Compound 2 was performed catalytic hydrogenation with 10% Pd-C to give propanoic acid derivative 3, which was cyclized by Friedel-Crafts acylation to afford 5,6-dimethoxyindan-l-one(4). Compound 4 was reduced with NaBH$_4$ in ethanol to obtain l-indanol 5, and it was dehydrated to give 5,6-dimethoxy-l-indene(6). This compound was lithiated with 2.5M n-butyllithium and reacted with 1,2-dibromoethane to give 2-(5,6-dimethoxy-l-indenyl)ethyl bromide(7), and which was treated with anhydrous ammonia to synthesize compound 1.

Clinical Characteristics and Treatment of Immune-Related Adverse Events of Immune Checkpoint Inhibitors

  • Immune checkpoint inhibitors (ICIs) have been changing the paradigm of cancer treatment. However, immune-related adverse effects (irAEs) have also increased with the exponential increase in the use of ICIs. ICIs can break up the immunologic homeostasis and reduce T-cell tolerance. Therefore, inhibition of immune checkpoint can lead to the activation of autoreactive T-cells, resulting in various irAEs similar to autoimmune diseases. Gastrointestinal toxicity, endocrine toxicity, and dermatologic toxicity are common side effects. Neurotoxicity, cardiotoxicity, and pulmonary toxicity are relatively rare but can be fatal. ICI-related gastrointestinal toxicity, dermatologic toxicity, and hypophysitis are more common with anti- CTLA-4 agents. ICI-related pulmonary toxicity, thyroid dysfunction, and myasthenia gravis are more common with PD-1/PD-L1 inhibitors. Treatment with systemic steroids is the principal strategy against irAEs. The use of immune-modulatory agents should be considered in case of no response to the steroid therapy. Treatment under the supervision of multidisciplinary specialists is also essential, because the symptoms and treatments of irAEs could involve many organs. Thus, this review focuses on the mechanism, clinical presentation, incidence, and treatment of various irAEs.

Physicochemical Quality Characteristics and Antioxidant Activity of Wasabi (Wasabia japonica) Leaf and Petiole Extracts (고추냉이 잎, 엽병 추출물의 이화학적 특성과 항산화 활성)

  • In this study, we investigated the physicochemical quality characteristics and antioxidant activities of water extracts ($20^{\circ}C$ and $95^{\circ}C$) against different parts (leaf and petiole) from Wasabia japonica (wasabi). Water extracts were divided into six types of wasabi powders: leaf hot air dried (LD), petiole hot air dried (PD), whole (leaf+petiole) hot air dried (WD), leaf steamed and hot air dried (LSD), petiole steamed and hot air dried (PSD), and whole steamed and hot air dried (WSD). Turbidity was higher in wasabi $20^{\circ}C$ water extracts than in $95^{\circ}C$ hot water extracts. Browning degree was higher in wasabi leaf extracts than in petiole extracts. The pH of hot water extraction was lower than that of room temperature extraction. Wasabi extracts did not show much difference in Hunter's color values according to extraction temperature, expected that b value of yellowness was significantly higher in leaf extracts than in petiole extracts. Carbazole pectin contents of leaf extracts were significantly higher than in petiole extracts, however water soluble pectin was higher in petiole extracts. The total polyphenol contents of LD20 and LD95 were 1,561.43 mg gallic acid equivalent (GAE)/100 g and 1,163.02 mg GAE/100 g, respectively, and total polyphenols decreased during hot water extraction. Extracts from different parts of wasabi showed a significant difference in total flavonoid contents. Total flavonoid contents of LD20, LD95, PD20, and PD95 were 554.44 mg/100 g, 396.65 mg/100 g, 55.42 mg/100 g, and 47.68 mg/100 g, respectively. In the sensory evaluation, LD95 extract showed significantly higher values than other samples in terms of color, flavor, taste, mouth feeling, and overall acceptability. In the analysis of sourness, saltiness, umami, sweetness, and bitterness taste by an electronic tongue, the sourness values of LD20 and PSD95, saltiness values of WSD20 and WSD95, and umami values of PD20 and PD95 were significantly higher than other extracts. The results of this study suggest that wasabi leaf and petiole extracts enhance qualities and antioxidant activities when used different parts together.

Synthesis and Biological Activity Test of the Sex Pheromone of the Summer Fruit Tortrix Moth (애모무늬잎말이나방의 성 페로몬 합성과 생물활성 시험)

  • Synthesis and biological activity test are described for the (Z)-9-tetradecen-1-yl acetate(1) and (Z)-11-tetradecen-1-yl acetate(1) and (Z)-11-tetradecen-1-yl acetate(2), the sex pheromone of the summer fruit tortrix moth, Adoxophyes orana. 8-Bromoctan-l-ol THP ether was prepared from 8-bromoctan-l-ol. The lithium anion of 1-hexyne was alkylated with 8-bromoctan-l-ol THP ether gave 9-tetradecyn-l-ol THP ether. Catalytic hydrogenation over Pd/BaSO4 followed by deprotection afforded (Z)-9-tetradecen-l-ol. Acetylation gave (Z)-9-tetradecen-1-yl acetate(1). l0-Bromodecan-l-ol THP ether was obtained from l0-bromodecan-l-ol. In liquid ammonia with THF and HMPA as cosolvents, sodium acetylide could be alkylated with 10-bromodecan-l-ol THP ether to give 11-dodecyn-l-ol THP ether. 11-Dodecyn-l-ol THP ether was then treated with n-BuLi in THF to give the lithium acetylide, which was alkylated with bromoethane to afford 11-tetradecyn-l-ol THP ether. Catalytic hydrogenation, deprotection, and acetylative gave (Z)-11-tetradecen-1-yl acetate(2). The synthetic pheromone thus obtained was attractive to the males of the summer fruit tortrix in the field.

Potential clinical utility of intraoperative fluid amylase measurement during pancreaticoduodenectomy

  • Backgrounds/Aims: Postoperative pancreatic fistula (POPF) after pancreaticoduodenectomy (PD) is a source of major morbidity and mortality. Early diagnosis and treatment of POPF is mandatory to improve patient outcomes and clinical risk scores may be ombined with postoperative drain fluid amylase (DFA) values to stratify patients. The aim of this pilot study was to etermine if intraoperative fluid amylase (IFA) values correlate with DFA1 and POPF. Methods: In patients undergoing PD from February to November 2020, intraoperative samples of intra-abdominal fluid adjacent to the pancreatic anastomosis were taken and sent for fluid amylase measurement prior to abdominal closure. Data regarding patient demographics, postoperative DFA values, complications, and mortality were prospectively collected. Results: Data were obtained for 52 patients with a median alternative Fistula Risk Score (aFRS) of 9.9. Postoperative complications occurred in 20 (38.5%) patients (five Clavien grade ≥ 3). There were eight POPFs and two patients died (pneumonia/sepsis). There was a significant correlation between IFA and DFA1 (R2 = 0.713; p < 0.001) and DFA3 (p < 0.001), and the median IFA was higher in patients with POPF than patients without (1,232.5 vs. 122; p = 0.0003). IFA > 260 U/L predicted POPF with sensitivity, specificity, positive and negative predictive values of 88.0%, 75.0%, 39.0%, and 97.0%, respectively. The incidence of POPF was 43.0% in high-risk (high aFRS/IFA) and 0% in lowrisk patients (low aFRS/IFA). Conclusions: IFA correlated with POPF and may be a useful adjunct to clinical risk scores to stratify patients during PD. Larger, prospective studies are needed to determine whether IFA has clinical utility.

  • Isozyme variants of 15 enzyme systems were analyzed in megagametophytes of Ginkgo biloba L. Five enzyme systems (ADH, G6PD, IDH, MPI, and UGPP) appeared to be monomorphic. Only 11 isozyme zones observed in 10 enzyme systems were polymorphic : ACON-A, FST-B, GDH-A, GOT-B, MDH-B, MDH-C, MNR-A, PGI-B, PGM-A, 6PGD-B and SKDH-B. The segregation ratio and heterogeneity at most polymorphic zones suggested that each isozyme zone was controlled by a single locus with codominant alleles, but significant deviation from 1 : 1 segregation was observed at MDH-B in pooled data. Three pairs of isozyme loci (ACON-A : MDH-B, GOT-B : PGI-B, and MNR-A : SKDH-B) were found to be weakly linked. Recombination frequencies between them ranged from 0.38 to 0.40 (p<0.05).

Effects of Black Ginseng (9 Times-Steaming Ginseng) on Hypoglycemic Action and Changes in the Composition of Ginsenosides on the Steaming Process (흑삼(구증구포인삼)이 혈당 강하에 미치는 영향 및 증포별 ginsenoside 조성 변화)

  • This study examined the effects of black ginseng (9 times-steamed ginseng) on hypoglycemic action in streptozotocininduced diabetic rats as well as changes in ginsenoside composition by the steaming process. As the number of steaming cycles increased, the amounts of crude saponin and most ginsenoside contents decreased, while the amount of ginsenoside- Rg3 and the ratio of PD/PT (=[$Rb_1+Rb_2+Rc+Rd+Rg_3]/[Re+Rb_1+Rh_1]$) increased. This ginsenoside composition is a unique characteristic compared to other types of ginseng products. In order to investigate the hypoglycemic effect of the black ginseng extract, in vivo studies were performed in rats with streptozotocin-induced diabetes. The studies showed that the administration of the black ginseng extract decreased high blood glucose levels (more than 300 mg/dL) to a normal level (102 mg/dL). These results suggest that this black ginseng extract has a significant hypoglycemic effect and can be used as an anti-diabetic substance for dietary supplements or new drugs.

  • The thiophene or furan-containing hexadentate ligands 1, 12-bis (2-theophene )-2,5,8,ll-tetraazadodecane(Thiotrien ㆍ 4HCl) and 1,12-bis (2-furan)-2,5,8,11-tetraazadodecane(Furatrienㆍ4HC1) were synthesized as their tetrahydrochloride salt and characterized by EA, IR, NMR, and Mass spectrum. Their protonation constants (logK$_{nH}$) and stability constants (logK$^{M}_{L}$ for Cd$^{2+}$, Pb$^{2+}$, $Zn^{2+}$ and Cu$^{2+}$ ions were determined in aqueous solution by potentiometry and compared with 1, 12-bis(2-pyridyl)-2,5,8, 11-tetra-azadodecane(Pytrien) of pyridyl-containing ligand. The effect stability constants of ligands and metal ion for removal of heavy metals in aqueous solution were described.

Hitting the complexity of the TIGIT-CD96-CD112R-CD226 axis for next-generation cancer immunotherapy

  • Antibody-based therapeutics targeting the inhibitory receptors PD-1, PD-L1, or CTLA-4 have shown remarkable clinical progress on several cancers. However, most patients do not benefit from these therapies. Thus, many efforts are being made to identify new immune checkpoint receptor-ligand pathways that are alternative targets for cancer immunotherapies. Nectin and nectin-like molecules are widely expressed on several types of tumor cells and play regulatory roles in T- and NK-cell functions. TIGIT, CD226, CD96 and CD112R on lymphoid cells are a group of immunoglobulin superfamily receptors that interact with Nectin and nectin-like molecules with different affinities. These receptors transmit activating or inhibitory signals upon binding their cognate ligands to the immune cells. The integrated signals formed by their complex interactions contribute to regulating immune-cell functions. Several clinical trials are currently evaluating the efficacy of anti-TIGIT and anti-CD112R blockades for treating patients with solid tumors. However, many questions still need to be answered in order to fully understand the dynamics and functions of these receptor networks. This review addresses the rationale behind targeting TIGIT, CD226, CD96, and CD112R to regulate T- and NK-cell functions and discusses their potential application in cancer immunotherapy.
