Red Cell Sedimentation Rates of Reshuffled Packed Cell Volume in Chicken (PCV수치를 변경시킨 닭적혈구 침강속도)

  • The packed cell volume(PCV) of chicken, volume percentage of erythrocytes in whole blood, was reshuffled of 20%, 40% and 60% using autoplasma, and the red blood cell sedimentation rate was measured in Westergren tubes at room temperature($27{\pm}1^{\circ}C$) and low temperature ($8{\pm}1^{\circ}C$). The results obtained were summarized as follows. 1. The sedimentation rates of chicken red blood cell were settled faster at low PCV than higher PCV, i,e., there was a reverse relationship between the sedimentation rate and PCV. 2. The sedimentation rate of chicken red blood cell was accelerated more at high temperature than low temperature. 3. The sedimentation rate of reshuffled chicken red blood cell by time was almost linear for several hours.

Porcine circovirus: detection of antibodies and virus antigen in Chungbdk area (Porcine circovirus에 대한 항체가 조사 및 바이러스 항원 확인)

  • Porcine circoviruses(PCV) are the smallest nonenveloped DNA viruses containing a unique single-stranded circular genome. No recognized link was found between PCV infection of pig and disease. But the PCV consistently identified from postweaning multisystemic wasting syndrome(PMWS) and researches indicate that there are strong relationships between PCV and PMWS. Clinical signs were emaciation, dyspnea, high fever with normal appetite. Necropsy findings showed respiratory disease complex lesion and lymph node anomalities. An indirect-immunofluorescent antibody procedure was used to assay swine sera for the presence of PCV atibodies. Antibodies against PCV were found in an average of 20% of the samples tested. The PCV DNA was amplified from lymph nodes collected from pigs. PCV specific primers were successfully amplified PCV DNAs. Further studies are needed to determine the possible role this virus might have in disease.

  • New emerging post weaning multisystemic wasting syndrome (PMWS) threatening swine industry worldwide and investigation of the etiological agent is underway. The porcine circovirus (PCV) consistently identified from PMWS pigs and research result indicate that there is strong relationship between PCV and PMWS. Fauns with PMWS submitted pigs suffered from various PMWS typical signs and necropsy finding showed lymph node anomalies. The PCV DNA was amplified from inguinal lymph node collected from pigs with PMWS. PCV specific primers were successfully amplified PCV DNAs and were able to differentiate PCV type I and II. We have identified noble PCV virus with genetic variation. The virus showed insertion of the nucleic acid at the 5' of the genome but did not have PCR product with primer set corresponding to PCV type II virus.

  • Porcine circovirus (PCV) is the smallest virus that replicates autonomously in mammalian cells. PCV2 is recongized as the casual agent of postweaning multisystemic wasting syndrome (PMWS).[1] Although the role of PCV2 in reproductive failure requires further clarification, there have been several reports of PCV2-associated reproductive failure. Consistent clinical signs of affected farms include elevated abortion, stillbirths and fetal mummification. The objective of this study was to determine the prevalence of PCV2 in aborted fetuses and stillborn piglets. Second objective was to determine the distribution of PCV2 DNA and antigen in aborted fetuses and stillborn piglets by in situ hybridisation and immunohistochemistry. (omitted)

Multifocal interstitial nephritis of pigs slaughtered in Jeju (제주지역 도축돈의 간질성 신염)

  • Total 160 head of porcine kidneys were examined for gross and histopathological lesions and polymerase chain reaction (PCR) for porcine circovirus type 2 (PCV-2), porcine parvovirus (PPV), Leptospira species and porcine reproductive and respiratory syndrome virus (PRRSV). Grossly, 137 kidneys (85.6%) had lesions characterized by the presence of the scattered white foci. Microscopically, multifocal interstitial nephritis, which classified into 4 grades such as, no lesion (Score 0), mild lesion (Score 1), moderate lesion (Score 2) and severe chronic lesion (Score 3) with fibrosis, was observed in 159 cases (99.4%). The histopathologic mean score for multifocal interstitial nephritis was significantly different (P<0.05) between the cases of PCV-2 single infection and the cases of co-infection with PCV-2 and PPV. According to PCR evaluation, PCV-2 were detected in 73.8% (118 cases), PPV were in 66.9% (107 cases), however Leptospira spp. and PRRSV were negative in all kidneys. Both PCV-2 and PPV were detected in 52.5% (84 cases). In 84 cases co-infected with PCV-2 and PPV, the occurrence of lymphoid follicle and vasculitis were observed as 65.5% (55 cases) and 26.2% (22 cases), respectively. These results revealed that PCV-2 and PPV were major infectious agents for interstitial nephritis in slaughtered pigs, Jeju. And the histopathologic lesions of multifocal interstitial nephritis were more severe in the case co-infected with PCV-2 and PPV.

UNG-based direct polymerase chain reaction (udPCR) for the detection of porcine circovirus 2 (PCV2) (UNG 기반 direct polymerase chain reaction (udPCR)을 이용한 돼지 써코바이러스 2형 진단법)

  • Porcine circovirus disease (PCVD) is a major problem of swine industry worldwide, and diagnosis of PCV2, causal agent of PCVD, has been doing in clinical laboratories of pig disease by polymerase chain reaction (PCR) methods. But the PCR analyses have a serious problem of misdiagnosis by contamination of DNA, in particular, from carryover contamination with previously amplified DNA or extracted DNA from field samples. In this study, an uracil DNA glycosylase (UNG)-based direct PCR (udPCR) without DNA extraction process and DNA carryover contamination was developed and evaluated on PCV2 culture and field pig samples. The sensitivity of the udPCR combined with dPCR and uPCR was same or better than that of the commercial PCR (cPCR) kit (Median diagnostics, Korea) on PCV2-positive serum, lymph node and lung samples of the pigs. In addition, the udPCR method confirmed to have a preventing ability of mis-amplification by contamination of pre-amplified PCV2 DNA from previous udPCR. In clinical application, 170 pig samples (86 tissues and 84 serum) were analysed by cPCR kit and resulted in 37% (63/170) of positive reaction, while the udPCR was able to detect the PCV2 DNA in 45.3% (77/170) with higher sensitivity than cPCR. In conclusion, the udPCR developed in the study is a time, labor and cost saving method for the detection of PCV2 and providing a preventing effect for DNA carryover contamination that can occurred in PCR process. Therefore, the udPCR assay could be an useful alternative method for the diagnosis of PCV2 in the swine disease diagnostic laboratories.

  • Porcine circovirus (PCV) is a small, nonenveloped virus that contains a single-stranded circular DNA genome of about 1.76 kb and belongs to the family circoviridae. The PCV-2 has been incriminated as the cause of post-weaning multisystemic wasting syndrome (PMWS) , an emerging disease in pigs. In the present study, a PCR assay was applied to detect PCV-2 in tissue samples. The presence of PCV-2 antigen in the porcine tissues was confirmed by indirect immunofluorescence (IIF) with PCV-2 specific monoclonal antibodies. And then DNA extracted from PCV-2 positive tissues was used as a template. One oligonucleotide primer suitable for PCR was selected from a published PCV-2 sequence (Genbank). Amplified PCR product was detected the same fragment lengths of 416 bp as a control. Based on these results, it was suggested that the PCR is a simple and sensitive method for support diagnostic purposes.

  • Each of twenty blood samples taken from apparently healthy Korean Cows was used to produce five different mixtures of autologous plasma and blood corpuscles such that their values of packed cell volume (PCV) lay between 10 to 50ml/l00ml. The measurements of erythrocyte sedimentation rate (ESR) using 45 degree-angled capillary hematocrit tube, 1.1-1.2mm bore, ($45^{\circ}-micro-ESR$) were practised for the blood of various levels of PCV under the ambient temperature of ${10^{\circ}C}$, ${20^{\circ}C}$ and ${30^{\circ}C}$. Correlation of ESR to ambient temperature showed linear regression each in five levels of PCV. ESR increased with ascending ambient temperature, and magnitude of the increase of ESR became greater as the level of PCV lowered. Correlation of ESR, of which values were transformed by the linear regression equation correlated to ambient temperature to PCV showed curvilinear regression each in three levels of ambient temperature, and ESR was increased with decreasing PCV. The data were statistically analysed and a list of relative anticipate $45^{\circ}-micro-ESR$ values for PCV and ambient temperature was presented.

  • Background : Pressure-controlled ventilation (PCV) is frequently used recently as the initial mode of mechanical ventilation in the patients with respiratory failure. Theoretically, because of its high initial inspiratory flow, pressure-controlled ventilation has lower peak inspiratory pressure and improved gas exchange than volume-controlled ventilation (VCV). But the data from previous studies showed controversial results about the gas exchange. Moreover, the comparison study between PCV and VCV with various inspiration : expiration time ratios (I : E ratios) is rare. So this study was performed to compare the respiratory mechanics and gas exchange between PCV and VCV with various I : E raitos. Methods : Nine patients receiving mechanical ventilation for respiratory failure were enrolled. They were ventilated by both PCV and VCV with various I : E ratios (1 : 2, 1 : 1.3 and 1.7 : 1). $FiO_2$, tidal volume, respiratory rate and external positive end-expiratory pressure (PEEP) were kept constant throughout the study. After 20 minutes of each ventilation mode, arterial blood gas, airway pressures, expired $CO_2$ were measured. Results : In both PCV and VCV, as the I : E ratio increased, the mean airway pressure was increased, and $PaCO_2$ and physiologic dead space fraction were decreased. But P(A-a)$O_2$ was not changed. In all three different I : E ratios, peak inspiratory pressure was lower during PCV, and mean airway pressure was higher during PCV. But $PaCO_2$ level, physiologic dead space fraction and P(A-a)$O_2$ were not different between PCV and VCV with three different I : E ratios. Conclusion : There was no difference in gas exchange between PCV and VCV under the same tidal volume, frequency and I : E ratio.

  • Background : Since the late 1960s, mechanical ventilation has been accomplished primarily using volume controlled ventilation(VCV). While VCV allows a set tidal volume to be guaranteed, VCV could bring about excessive airway pressures that may be lead to barotrauma in the patients with acute lung injury. With the increment of knowledge related to ventilator-induced lung injury, pressure controlled ventilation(PCV) has been frequently applied to these patients. But, PCV has a disadvantage of variable tidal volume delivery as pulmonary impedance changes. Since the concept of combining the positive attributes of VCV and PCV(dual control ventilation, DCV) was described firstly in 1992, a few DCV modes were introduced. Pressure-regulated volume control(PRVC) mode, a kind of DCV, is pressure-limited, time-cycled ventilation that uses tidal volume as a feedback control for continuously adjusting the pressure limit However, no clinical studies were published on the efficacy of PRVC until now. 'This investigation studied the efficacy of PRVC in the patients with unstable respiratory mechanics. Methods : The subjects were 8 mechanically ventilated patients(M : F=6 : 2, $56{\pm}26$ years) who showed unstable respiratory mechanics, which was defined by the coefficients of variation of peak inspiratory pressure for 15 minutes greater than 10% under VCV, or the coefficients of variation of tidal volume greater than 10% under PCV. The study was consisited of 3 modes application with VCV, PCV and PRVC for 15 minutes by random order. To obtain same tidal volume, inspiratory pressure setting was adjusted in PCV. Respiratory parameters were measured by pulmonary monitor(CP-100 pulmonary monitor, Bicore, Irvine, CA, USA). Results : 1) Mean tidal volumes($V_T$) in each mode were not different(VCV, $431{\pm}102ml$ ; PCV, $417{\pm}99ml$ ; PRVC, $414{\pm}97ml$) 2) The coefficient of variation(CV) of $V_T$ were $5.2{\pm}3.9%$ in VCV, $15.2{\pm}7.5%$ in PCV and $19.3{\pm}10.0%$ in PRVC. The CV of $V_T$ in PCV and PRVC were significantly greater than that in VCV(p<0.01). 3) Mean peak inspiratory pressure(PIP) in VCV($31.0{\pm}6.9cm$ $H_2O$) was higher than PIP in PCV($26.0{\pm}6.5cm$ $H_2O$) or PRVC($27.0{\pm}6.4cm$ $H_2O$)(p<0.05). 4) The CV of PIP were $13.9{\pm}3.7%$ in VCV, $4.9{\pm}2.6%$ in PVC and $12.2{\pm}7.0%$ in PRVC. The CV of PIP in VCV and PRVC were greater than that in PCV(p<0.01). Conclusions : Because of wide fluctuations of VT and PIP, PRVC mode did not seem to have advantages compared to VCV or PCV in the patients with unstable respiratory mechanics.
