Objectives : This study was to evaluate the pharmacological effect of Korea Red Ginseng aqueous extract (KRGE) on serum-deprived apoptosis of neuronal-like pheochromocytoma PC12 cells and to investigate its underlying action mechanism. Methods : KRGE was prepared by extracting Korea Red Ginseng with hot water and concentrating using a vacuum evaporator. Cell viability was determined after incubation of cells with KRGE or chemical inhibitor in serum-deprived medium for 60 h by counting intact nuclei following lysing of the cell membrane. Caspase activities were measured using chromogenic substrates and signal-associated protein phosphorylation and cytochrome c release were determined by Western blot analyses using their specific antibodies. Results : Serum deprivation induced PC12 cell death, which was accompanied by typical morphological features of apoptotic cell, such as nuclear fragmentation, caspase-3 activation, and cytochrome c release. This apoptotic cell death was significantly inhibited by KRGE and caspase-3 inhibitor, but not by the addition of NMA, ODQ, and PD98059. KRGE promoted phosphorylation of Akt and Bad, and this phosphorylation was inhibited by the PI3K inhibitor LY92004. In addition, this inhibitor also reversed KRGE-mediated protection of PC 12 cells from serum deprivation. These results suggested that KRGE protects PC12 cells from serum deprivation-induced apoptosis through the activation of PI3K/Akt-dependent Bad phosphorylation and cytochrome c release, resulting in caspase-3 activation. Conclusions : KRGE should be considered as a potential therapeutic drug for brain diseases including stroke induced by apoptosis of neuronal cells.
전기방사공정에 의해 고분자의 나노 크기의 섬유를 만드는 기술로 널리 사용되어졌으며, 제작된 나노섬유는 그 높은 표면적과 형태학적 특성때문에 조직재생 공학분야에서 많이 사용되어져 왔다. 본 연구에서는 기존의 전기방사공정을 개선한 복합전기장을 이용하여 생분해성/생체적합성 poly(${\varepsilon}$-caprolactone) (PCL) 마이크로/나노섬유를 제작하였고, 기존의 나노섬유의 배열성보다 제어가 가능한 배열성을 갖는 공정시스템을 통하여 보다 우수한 배열성을 갖는 PCL 나노섬유를 제작하였다. 고배열된 PCL 나노섬유는 신경세포 재생을 위한 세포담체로서의 가능성을 확인하고자 신경세포(PC-12)를 배양하였으며 그 결과 높은 배열성을 갖은 PCL 나노섬유 매트에서 신경세포의 배열성이 얻어짐을 확인하였다.
목적 : 사람의 유방암 세포인 MCF-7 세포주를 대상으로 gultathione S-transferase K1 (hGSTK1) 유전자의 발현 정도 및 방사선 조사에 의한 hGSTK1 유전자의 발현 변화를 관찰하고 hGSTK1 유전자를 과발현시킴으로써 hGSTK1이 방사선 감수성에 어떤 영향을 미치는 지를 관찰하였다. 재료 및 방법 : 사람의 모유두 세포 pBluescript phagemid cDNA library로부터 선별한 hGSTK1 cDNA를 pcDNA3.1/Myc-His (+) vector에 결합시킨 후, 사람의 유방암 세포인 MCF극 세포에 이입시켰다. hGSTK1 유전자를 이입시키지 않은 MCF극 세포와 hGSTK1을 이입시킨 MCF-7 세포에 $2\~12\;Gy$의 엑스선을 조사하여 생존 분획을 비교하였다. hGSTK1 유전자를 이입시키지 않은 MCF-7 세포와 hGSTK1을 이입시킨 MCF-7 세포에서 방사선량, 분할조사 여부, 방사선 조사 후 경과 시간에 따른 hGSTK1 mRNA 발현의 차이를 보기 위하여 RT-PCR 분석을 시행하였다. 결과 : hGSTK1 유전자를 이입시키기 전의 MCF-7 세포보다 hGSTK1 유전자를 이입시킨 MCF-7 세포에서 생존 분획이 유의하게 높은 것으로 나타났다. hGSTK1 유전자를 이입시키지 않은 MCF-7 세포에서 2 Gy 생존 분획은 $0.3250{\pm}0.0319$였고, hGSTK1 유전자를 이입시킨 MCF-7 세포에서 2 Gy 생존 분획은 $0.4125{\pm}0.0325$였다 (p<0.05). 그러나 RT-PCR에 의한 hGSTK1 mRNA 분석에서는 방사선량, 분할조사 여부, 방사선 조사 후 경과 시간에 따른 발현의 차이를 볼 수 없었다. 결론 : MCF-7 세포주에서 hGSTK1의 과발현은 방사선 감수성에 영향을 미쳐서 MCF-7 세포의 생존 분획을 증가 시켰다고 볼 수 있으나 이에 대한 정확한 기전을 알기 위해서는 더 많은 연구가 필요할 것이다.
PC12 세포에서 $CoCl_2$에 의한 hypoxia 유도는 HIF1 alpha의 상승 발현으로 확인하였다. 이때 apoptosis의 유도는 genomic DNA의 fragmentation과 apoptotic body는 Hoechst 염색으로 확인되었고, ER luminal chaperone의 발현 및 ER stress signal에 관여하는 ER membrane kinase인 IRE1, PERK, ATF6의 발현도 확인되었다. 이들이 apoptosis로 연결되는 고리 역할을 하는 IRE1-XBP1 mRNA splicing, PERK-eIF2 alpha, ATF6 protein cleavage도 반응하는 것으로 확인되었다. 위의 결과는 신경세포의 hypoxia상태는 ER stress signal pathway를 거쳐서 apoptosis가 된다는 것을 증명한 것으로 신경세포의 hypoxia치료를 위한 기초 자료가 될 것으로 생각한다.

Objectives : the effect of different sterilization methods on the surface morphology of PPDO-hybrid-PLGA nanofiber scaffold and attachments of PC12 cell were investigated. Methods : Poly (p-dioxone)-hybrid-Poly (lactide-glycolide) (PPDO-hybrid-PLGA) nanofiber scaffold, fabricated in a tube form with 1.5 mm internal diameter, 0.2 mm thickness and 5 mm length, was prepared using electrospinning method. To study the surface morphology using SEM, The study group and control group in respective were; Control:Non-sterilized scaffold, Group I:scaffold sterilized with 70% Alcohol, Group II: scaffold sterilized with Ethylene Oxide at $65^{\circ}C$, and Group III: scaffold sterilized with Ethylene Oxide at $37^{\circ}C$. To investigate viability of the PC12 cell on the scaffold, The study group and control group in respective were; Control: sterilized with 70% Alcohol, Group I: sterilized with Ethylene Oxide at $65^{\circ}C$, and Group II: sterilized with Ethylene Oxide at $37^{\circ}C$. Results : 1. The surface morphology was slightly changed in Group I, II and Group III, compared with control. 2. The attachment of PC12 cells in Group I, II was not higher than in control Discussion : The attachment of PC12 cell is not influenced by different sterilization methods.
Objectives : This Study was performed to assess the antioxidant and neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells and primary rat mesencephalic dopaminergic neurons. Methods : The anioxidant effect was investigated using the DPPH radical and ABTS cation scavenging assays and total polyphenol amout of Chunghyul-dan(Qingxuedan). The neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells was evaluated using MTT assay. The scavenging activity of Chunghyul-dan(Qingxuedan) on ROS production induced by 6-OHDA(6-hydroxydopamine) in PC12 cells was evaluated, as well as the attenuating effect on GSH reduction. Finally, we examined the neuroprotective effect of Chunghyul-dan(Qingxuedan) against 6-0HDA-induced toxicity in the primary culture of rat mesencephalic doperminergic neurons. Results : Chunghyul-dan(Qingxuedan) showed concentration-dependent scavenging activities in DPPH radical and ABTS cation scavenging assays and it was not cytotoxic to PC12 cells. In postand co-treatment, Chunghyul-dan(Qingxuedan) protected PC12 cells from the 6-OHDA induced toxicity at 50 and 100 ${\mu}$g/mL significantly. And Chunghyu!-dan(Qingxuedan) decreased the 6-OHDA induced ROS production at a dose dependent manner, while increaing the 6-OHDA induced GSH reduction at 50 and 100 ${\mu}$g/mL significantly. Finally, Chunghyul-dan(Qingxuedan) showed signicant protection of rat mescencephalic dopaminergic neurons from 6-OHDA at 1 ${\mu}$g/mL. Conclusions : These results demonstrate that Chunghyul-dan(Qingxuedan) has the antioxidant and neuroprotective effect against 6-0HDA induced cytotoxicity through decreasing ROS production and increasing GSH reduction.
Objectives : This study was performed to evaluate the neuroprotective effect of water extracts from Thujae Semen(TSW) in PC12 cells. Methods : We performed 2,2-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging assay, 2,2-azinobis- (3-ethyl-benzothiazoline-6-sulfonic acid(ABTS) cation scavenging assay, and determination of total polyphenolic content to examine the antioxidant effects of TSW. We also carried out 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay(MTT), reactve oxygen species(ROS) assay, and nitric oxide(NO) assay to examine neuroprotective effects against 6-hydroxydopamine(6-OHDA) in PC12 cells. Results : TSW showed $IC_{50}$ values of 404.3 and 219.9 ${\mu}g/mL$ in DPPH and in ABTS assays, respectively. TSW showed 9.74 ${\mu}g/mL$ of total polyphenol contents. TSW incresed cell viability in a dose dependent manner and it showed protective effect against 6-OHDA neurotoxicity at the concentration of 25-200 ${\mu}g/mL$. Moreover, it recovered 6-OHDA induced cell death at the same concentrations. The extract showed a dose dependent reduction of ROS and NO generation by 6-OHDA. Conclusions : We concluded that TSW has neuroprotective effect against 6-OHDA-induced toxicity in PC12 cells through ROS and NO inhibition.
Polycomb group (PcG) proteins are conserved chromatin regulators involved in the control of key developmental programs in eukaryotes. They collectively provide the transcriptional memory unique to each cell identity by maintaining transcriptional states of developmental genes. PcG proteins form multi-protein complexes, known as Polycomb repressive complex 1 (PRC1) and Polycomb repressive complex 2 (PRC2). PRC1 and PRC2 contribute to the stable gene silencing in part through catalyzing covalent histone modifications. Components of PRC1 and PRC2 are well conserved from plants to animals. PcG-mediated gene silencing has been extensively investigated in efforts to understand molecular mechanisms underlying developmental programs in eukaryotes. Here, we describe our current knowledge on PcG-mediated gene repression which dictates developmental programs by dynamic layers of regulatory activities, with an emphasis given to the model plant Arabidopsis thaliana.
청국장 메탄올 추출물의 $H_2O_2$로 유도된 신경세포 독성에 대한 보호 효과를 조사하였다. 청국장 분말의 일반성분을 분석한 결과 조단백질 40.95%, 조지방 22.49%, 가용성 무질소물 15.99%, 수분 7.91%, 조회분 6.74% 및 조섬유 5.92% 순으로 나타났으며, 총 페놀성 화합물 함량은 28.43 mg/g이었다. $H_2O_2$로 유발된 산화적 손상에 의한 ROS 축적량을 조사한 결과 $H_2O_2$ 단독 처리구보다 청국장 메탄올 추출물 처리구에서 낮은 ROS 축적량을 보였다. $H_2O_2$로 유도된 PC12 신경세포에 대한 보호효과를 MTT 방법을 이용하여 측정한 결과 모든 시료에서 40%정도의 신경세포 보호효과를 보였고, LDH release 실험 결과 5 mg/mL 농도에서 15%정도의 LDH 방출량 저해 효과를 나타내었다. 청국장 메탄올추출물의 아세틸콜린에스터레이스 저해활성은 농도 의존적인 경향이었다. 본 연구 결과를 종합해 볼 때, 청국장 메탄올 추출물은 산화적 스트레스로부터 신경세포 보호효과 및 아세틸콜린에 스터레이스 저해활성을 나타내어 알츠하이머성 신경질환의 예방을 위한 식품으로서의 활용 가능성이 높다고 판단된다.
본 실험은 해수 미세 조류인 Chlorella capsuiata로부터 신경세포에 대한 활성을 증가시키는 기능성물질을 분리하여 해수자원의 생체 조절자원으로서의 가능성을 제시하고자 실시하였다. 선별된 해수 Chlorella를 이용하여 활성물질을 분리한 뒤, 그 물질의 신경활성을 탐색하였다. C. capsulata의 물 추출물로부터 분리된 분획물(CCE)의 분자량은 약 45KDa(data not shown)으로 기존에 연구된 60~100 KDa보다 더 낮은 범위에서 물질이 분리되었으며, 이는 현재 발표된 많은 연구결과에서 주장하는 물질들과는 다른 종류의 물질임을 제시하고 있어 이에 보다 심층적인 연구가 수행되어져야 한다고 생각된다. 실험 결과를 통해 볼 때 활성을나타낸 주된 물질은 C. capsulata의 수용성 성분으로 생각되어지며, 260 nm에서 최대 흡광도를 나타내는 물질로 C. capsulate에 존재하는 단백질이 열 변성에 의해 탄수화물과 결합한 glycoprotein의 형태로 존재하는 것으로 추측되지만 일부의 연구결과에서 280nm에 최대 흡광도를 보이는 활성물질이 glycoprotein이라고 주장하고 있어 이 분획물(CCE)에 대한 좀더 심도 깊은 연구가 수행되어져야 한다고 생각된다. 이는 최근에 발표된 Chiorella의 기능성과 관련한 논문들에서 언급한 내용들과 유사한 결과를 나타내지만 대부분이 담수조류에 대한 활성 탐색의 결과임을 감안한다면 본 실험을 통해 해수 미세조류로부터 분리된 물질의 새로운 활성물질로서의 가능성을 제시한 것이라 하겠다. 또한, 다른 유기용매를 통해 활성물질을 분리하는 것과 달리 순수한 물을 통해 Chlorella의 수용성 성분을 추출하는 것이 좀더 신경세포의 활성을 증가시킨다는 것을 확인하였다. 앞으로 이 수용성 물질에 대찬 면역활성과 in vivo 실험을 통해 좀더 깊은 연구가 수행되어지고, 나아가 대량배양기술, 분리정제 기술이 뒷받침된다면, 고비용의 동물세포를 이용한 신경활성물질을 대체할 새로운 신경 활성물질 개발은 물론 다방면에 걸친 생체 조절기능을 가진 기능성 소재로서 활용 범위가 점차 확되지 않을까 생각한다.
