Validation of an HPLC Method for the Pharmacokinetic Study of Glipizide in Human (글리피짓 체내동태 연구를 위한 혈청 중 글리피짓의 HPLC 정량법 검증)

  • A rapid, selective and sensitive reversed-phase HPLC method for the determination of glipizide in human serum was validated and applied to the pharmacokinetic study of glipizide. Glipizide and internal standard, tolbutamide, were extracted from human serum by liquid-liquid extraction with benzene and analyzed on a Nova Pak $C_{18}\;60{\AA}$ column with the mobile phase of acetonitrile-potassium dihydrogen phosphate (10 mM, pH 3.5) (4:6, v/v). Detection wavelength of 275 nm and flow rate of 0.7 ml/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^3$ factorial design using a fixed glipizide concentration (500 ng/ ml) with respect to its peak area and retention time. And also, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of 10-1000 ng/ml with correlation coefficient greater than 0.999. The lower limit of quantitation using 0.5 ml of serum was 10.0 ng/ml, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the quality control samples ranged from 82.6 to 105.0% for glipizide with overall precision (% C.V.) being 1.13-13.20%. The percent recovery for human serum was in the range of 85.2 93.5%. Stability studies showed that glipizide was stable during storage, or during the assay procedure in human serum. The peak area and retention time of glipizide were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of glipizide in human serum samples for the pharmacokinetic studies at three different laboratories, demonstrating the suitability of the method.

A Study of the Relationship between the Nutritional Quality of Life and Stature Sexual Dimorphism (영양 수준의 향상에 따른 신장 성적이형성의 변화: 탈북자 자료를 이용한 남북한 비교연구)

  • We tested the hypothesis that an improved nutritional environment increases the size of stature sexual dimorphism (SSD) by comparing mean height changes in North and South Korean adults. For the first round of sampling, those North Korean refugees who had reached the age of 20 years before leaving North Korea were selected for the study. Then, two birth-year cohorts born between 1955 and 1959 and between 1980 and 1984 were finally selected for the analysis (383 males and 529 females). North Korean refugee mean height data were compared to the mean height of the comparable South Korean population derived from 2004 Korean size results (373 males and 429 females). The results of the analysis showed that there was no increase in mean height among the refugees, either in males or females, between the two birth-year cohorts. As a result, no change in SSD was observed between the two birth-year cohorts. In contrast, South Koreans showed a remarkable increase in height, with males having a greater increase during the corresponding 25 years. Consequently, South Korean SSD increased significantly with time. These results support the hypothesis that improved nutritional quality increases SSD.

Partial Budget Modeling of Economic Losses of Aujeszky's Disease (부분예산분석을 이용한 오제스키병 발생 농가의 경제적 손실 추정)

  • Aujeszky's disease (AD) is a respiratory, infectious viral illness associated with high mortality, especially in neonatal piglets and has frequently been considered an economically important disease in many endemic countries. Although AD is still occurring in a geographically defined region in Korea, little attention has been paid to the economics of AD. In this study, partial budget technique was used to develop a simulation model to measure financial losses following the disease epidemic in a swine operation utilizing stochastic or deterministic parameters from the literatures and the index case herd of AD occurred in 2005, where available and applicable. For the infected case herd with a 12500-pig, the total economic loss for this operation was estimated to be about 199 million Korean won (95% confidence interval [CI] 148,645,000-250,741,000). Given net loss due to death of a pig at sow level was 119,000 won, total loss for the case herd with 1200 sows accounted for 143 million won (95% CI 92,599,000-193,729,000). The net loss of the death of one pig at growing and fattening level resulted in loss of 46,000 won (95% CI 40,000-53,000) and 126,000 won (95% CI 122,000-131,000), respectively. Taking into account for the number of pigs raised in the case herd, total loss amounted to 8 million won (95% CI 7,167,000-9,347,000) and 12 million won (95% CI 11,959,000-12,891,000), for growers and fatteners, respectively, assuming 63% of saved feed intake when a pig dies halfway through the respective period. Under the model's assumptions, suckling pig mortality was the major factors of loss in estimating the economic consequences (approximately 71.8% of the total loss). The high economic losses of a herd infected with AD suggest that the effective and region-specific control measures should be implemented in disease endemic foci.

Teachers' Perception about 'Science Field Trip to Korean Historical Sites' ('한국 역사 속 과학탐방'에 대한 교사의 인식)

  • The purpose of this study was to investigate teachers' perception about 'Science Field Trip(SFT) to Korean historical sites'. Ninety-seven teachers were asked to reply the questionnaire about SFT to Korean historical sites. This questionnaire was divided into three parts: educational value, teaching method, and difficulty of management. Firstly, teachers indicated that SFT to Korean historical sites had much educational value in that it could give students chances to understand Korean culture, to acquire investigative process skills, to experience divergent investigation and cooperative learning, to raise integrated thinking skill concerning science and other subjects, and to learn contents related with science curriculum. It also had advantages of caring for students out of school and giving the information for future occupation. Secondly, teachers suggested the desirable teaching method: teachers' explanation about SFT to Korean historical sites with worksheets before it, students' group work during it, and the presentation and discussion after it. Finally, the most frequent answer in the difficulty of management was the lack of teaching materialsfor SFT to Korean historical sites.

THE INFRARED MEDIUM-DEEP SURVEY. V. A NEW SELECTION STRATEGY FOR QUASARS AT z > 5 BASED ON MEDIUM-BAND OBSERVATIONS WITH SQUEAN

  • Multiple color selection techniques are successful in identifying quasars from wide-field broadband imaging survey data. Among the quasars that have been discovered so far, however, there is a redshift gap at 5 ≲ z ≲ 5.7 due to the limitations of filter sets in previous studies. In this work, we present a new selection technique of high redshift quasars using a sequence of medium-band filters: nine filters with central wavelengths from 625 to 1025 nm and bandwidths of 50 nm. Photometry with these medium-bands traces the spectral energy distribution (SED) of a source, similar to spectroscopy with resolution R ~ 15. By conducting medium-band observations of high redshift quasars at 4.7 ≤ z ≤ 6.0 and brown dwarfs (the main contaminants in high redshift quasar selection) using the SED camera for QUasars in EArly uNiverse (SQUEAN) on the 2.1-m telescope at the McDonald Observatory, we show that these medium-band filters are superior to multi-color broad-band color section in separating high redshift quasars from brown dwarfs. In addition, we show that redshifts of high redshift quasars can be determined to an accuracy of Δz/(1 + z) = 0.002 - 0.026. The selection technique can be extended to z ~ 7, suggesting that the medium-band observation can be powerful in identifying quasars even at the re-ionization epoch.

Foliar ultrastructure of Korean Orostachys species (한국산(韓國産) 바위솔속(屬) 엽육조직(葉肉組織)의 미세구조(微細構造))

  • Ultrastructural characteristics were examined with leaves of three species, O. japonicus A. Berger, O. malacophyllus Fisch., and O. sikokianus Owhi that probably have CAM mode. The mesophyll cells of these Orostachys possessed vacuoles with precipitates, myelin-like figures, and plasmalemmasomes, along with typical chloroplasts, microbodies and darkly stained bodies in their thin peripheral cytoplasm. Separation of the plasmalemma from the cell wall, leaving a space between them, was a common phenomenon in these species. A complex array of small to large vacuoles which contain small, membrane-bounded vesicles or vacuole-like structures were frequently found. A well-developed thylakoid system was observed in the chloroplasts and this indicates that the photosynthetic capacity of these mesophyll cells is probably active. A peculiar configuration of cytoplasm, especially around the chloroplasts, was also encountered. The variety of cytoplasmic constituents and vacuoles suggest the water-storing mesophyll cells may be complex in function. Some cellular features detected in this study strongly suggest the possible occurrence of CAM mode in Orostachys species.

Continuous Bio-hydrogen Production from Food Waste and Waste Activated Sludge (음식물 쓰레기와 폐활성 슬러지를 이용한 생물학적 수소생산 및 수소생산 미생물 군집분석)

  • Batch experiments were performed to investigate the effects of volumetric mixing ratio(v/v) of two substrates, food wastes(FW) and waste activated sludge(WAS). In batch experiments, optimum mixing ratio for hydrogen production was found at $10{\sim}20$ v/v % addition of WAS. CSTR(Continuous Stirred tank reactor) was operated to investigate the hydrogen productivity and the microbial community under various HRTs and volumetric mixing ratio(v/v) of two substrates. The maximum yield of specific hydrogen production, 140 mL/g VSS, was found at HRT of 2 day and the volumetric mixing ratio of 20:80(WAS:FW). The spatial distribution of hydrogen producing bacteria was observed in anaerobic fermentative reactor using fluorescent in situ hybridization(FISH) method.

A Study on the 6th Middle School Science Curriculum and Its Implementation (제 6차 중학교 과학 교육과정과 그 운영에 대한 조사 연구)

  • We have investigated the 6th middle school science curriculum and its implementation based upon the results of middle school science teachers' and professors' ratings and self-reportings. The national-wide survey was administrated to obtain the data from 213 teachers of 112 middle schools and 43 professors of colleges of education. The survey items correspond to four stages of science curriculum. In the stage of the planned science curriculum, 'goals', 'framework of contents' and 'contents' are relevant to the basic guideline for curriculum revision, 'the more emphasis on inquiry activities'. However, the amount of contents is not appropriate, and 'methods' are difficult to attain. In the stage of the written science curriculum, 'goals', 'framework of contents', 'contents', 'methods' and 'evaluations' are not useful for teachers to teach science. In the stage of the implemented science curriculum, a small number of teachers use the issues of local community, organize the different group activities, develop assessment tools cooperatively, and conduct a lesson and a program considering students' diversities. This is partially because of the large amount of learning contents, the lack of materials and the weakness of supporting system. Finally, in the stage of the attained science curriculum, the students' achievements under the 6th curriculum is not higher than those under the other curricula.

Grade 7th Pupils' Ideas about Identification and Control of Variables in Inquiry Problems (중학교 1학년 학생들의 탐구 문제에 대한 변인 판별 및 통제)

  • The purpose of this study was to compare the ideas of pupils with that of the scientists about controlling and identifying of variables, in the two cases: open or guided inquiry. The subjects were the 7th grade boys and girls in a school, in Seoul, Korea. For the guided inquiry, the problems were given by the experiments of pupils' text. Pupils were asked to identify the variables in the experiments. For the open inquiry, pupils set their own inquiry problem. The pupils whose marks are within upper one-third of three classes were chosen. Pupils' ideas on variables were investigated in the design of experiment for their problems. In that, questionnaire developed by researchers was used. In the former, many of the pupils identify just only one variable despite of the fact there were two independent or dependent variables in the experiments. In the latter, the number of independent variables increased two or three. However, pupils do not control independent variables: they vary two independent variables simultaneously in the design of experiment. From these, we compared the pupils' ideas on variables with the scientists'

The Characteristics of Science Teachers Participating in the Student Science Inquiry Olympic (학생과학탐구 올림픽 행사를 통해 본 혁신 지향 교사의 특징)

  • This study compared the innovative characteristics of science teachers who participated in the First Student Science Inquiry Olympic and those of science teachers who did not participate in the event. The subjects were science teachers in three groups: (1) the active participants in the Inquiry Olympic who supervised the students contestants, (2) the observers of the Inquiry Olympic who came to see the event, and (3) the ordinary teachers who did not come to the Inquiry Olympic and were sampled through stratified cluster sampling. The study instrument was a questionnaire; all the subjects received the survey questionnaire by mail. The return rate was 45%. In general, the Inquiry Olympic participants(both the student supervisors and the observers) demonstrated different characteristics from the nonparticipants in four categories. Firstly, the Inquiry Olympic participants showed higher level of self actualization, for example, interest in science education, higher inner motivation, stronger desire to innovate than did the nonparticipants. Secondly, the participants demonstrated more involvement in professional activities and greater degree of upward social mobility than the nonparticipants. Thirdly, the participants had communication behaviors different from nonparticipants, e. g.. greater leadership of public opinions, more experience of contact with the change agent, greater tendency to regard their school society as modern. Lastly, the participants had higher social status than the nonparticipants. Implications and suggestions are made for the utilization of the innovation-oriented science teachers to implement of innovations in the future.
