Determination of Ursodeoxycholic Acid in Crude Drug Formulations by HPLC and SPE Using Selective Pre-column Derivatization with 2-Bromoacetyl-6-methoxynaphthalene (2-Bromoacetyl-6-methoxynaphthalene을 형광유도체화제로 HPLC와 SPE를 이용한 생약제제 중 Ursodeoxycholic acid의 정량)

  • A simple and sensitive high performance liquid chromatographic method to quantitate ursodeoxycholic acid in crude drug pharmaceuticals was investigated. Ursodeoxycholic acid react with 2-bromoacetyl-6-methoxynaphthalene (Br-AMN) in the presence of triethylamine to form highly fluorescent derivative. The derivatization procedure was performed at 7$0^{\circ}C$ and completed within 30 min. The optimal wavelength of the fluorescence detector are λ$_{ex}$=300 nm and λ$_{em}$ = 460 nm. The LOD of the ursodeoxycholic acid was 25 ng/mι based on the S/N =3, and the LOQ was 80 ng/mι based on S/N = 10. Crude drug pharmaceuticals pretreated by solid phase extraction (Sep-pak $C_{18}$ cartridge) which were shown very good separation and recovery values for the compound.d.

  • A high performance liquid chromatographic method was developed for the simultaneous determination of free and glycine conjugated bile acids. Free and glycine conjugated bile acids were extracted from bear gall bladder by methanol and from serum using a Sep-pak $C_{18}$ catridge. The extracted bile acids were labeled with 2-bromoacetyltriphenylene in acetonitrite using 18-crown-6-ether as a catalyst. Derivatized bile acids were separated from the individual bile acids on a reversedphase column (Chemcosorb 5-ODS-H) using acetonitrile-methanol-water(10:50:30) as a mobile phase and monitored by an UV-detector at 254nm. Linearities of calibration curve were obtained between 4 ng and 24 ng, and recoveries from bear gall bladder sample were higher than 94%.

  • Hot deformation behavior of A350 LF2 alloy was characterized by compression tests in the temperature range of 800-$1250^{\circ}C$ and the strain rate range of $0.001-10s^{-1}$. The microstructural evolution during hot compression was investigated and deformation mechanisms were analyzed by constructing processing map. Processing maps were generated using the dynamic material model (DMM). The combination of dynamic material model and Ziegler's instability criterion was applied to predict an optimum condition and unstable regions for hot forming.

  • After successful commissioning observations, IGRINS will soon begin its normal scientific operations on the 2.7m Harlan J. Smith telescope at the McDonald Observatory. We will present a working version of the IGRINS operation plan. A fraction of IGRINS guaranteed time will be devoted to a selection of strategic programs with legacy values, whereas the rest of the time will be used for normal programs. We will also describe the current status of the IGRINS data reduction pipeline package and its future development plan.

  • We observed selected super cluster regions in M82 with 5'5 arcsec field-of-view using near-IR high resolution echelle spectrometer, IRCS, at the SUBARU 8.2 m telescope. The slit width of 0.15 arcsec makes the high resolution (R ${\approx}$ 20,000) spectra in the H and K bands. In this poster, we present sample spectra of [FeII] lines and ro-vibration lines of $H_2$ which trace ionic shocks in the intercloud regions and molecular shocks. The line widths of $Br{\gamma}$ line are also measured to derive the velocity dispersion within the super star clusters.

  • In this study, the effects of the current pulsing conditions, on the spatter generation rate during the $CO_2$ gas metal arc welding (GMAW) were investigated. Normally using the inverter type power supply, of which the welding current waveform was regulated to reduce the spatter generation rate, but in this study pulsing was imposed on the welding current. Observation of the metal transfer phenomena during the pulsed current GMAS indicated that the droplet transfer from the electrode via the short circuit transfer and the repelling transfer mode could be minimized by selecting optimum combinations of pulsing parameters, which include base and peak current, base and pak duration. It was also demonstrated in this study that proper combinations of the pulsing parameters led to reduce generation of spatters during GMAW shielded by $CO_2$ gas.

Near Infrared Wavelength Calibration without Telluric OH Lines

  • Grating spectrograph observation in near-infrared bands requires wavelength calibrations. We first need to extract order-strips from the echellogram data using flat images, and then to correct the spatial distortions and wavelength scales of the order strips using calibration arc lamps or the telluric OH emission lines. For very high resolution spectrograph using echelle, however, the arc lamps do not have enough emission lines in the order-strips, so we usually use the OH lines for accurate wavelength calibrations in wavelengths shorter than 2.3 microns. In this poster, we present wavelength calibration methods for longer wavelength bands using the telluric absorption lines in early-type stellar spectra and the telluric atmospheric transmission models. This technique will be applied to the data reduction pipeline for the IGRINS.

  • This paper is concerned with the improvement of impact and tensile Properties of spheroidal graphite cast iron of the following duplex matricess which were heat treated in the eutectic transformation temperature range (that is, $({\alpha}+{\gamma})$ coexisting range) ; ferrite-martensite, ferrite-bainite and ferrite-pearlite. The absorbed energy and maximum load was measured by recording the load-deflection curve with instrumented Charpy impact testing machine in the temperature range from $+100^{\circ}C$ to $-196^{\circ}C$. It was found the ferrite-bainite duplex matrix showed the highest toughness among the above matrices in the room temperature and the low temperature range. Comparison of this matrix to ferrite-pearlite matrix(that is, as cast) showed a lowering of $27^{\circ}C$ in the nil-ductility transition temperature (NDT) and a lowering of $40^{\circ}C$ in the ductile-brittle transition temperature (TrE), Which seems to result from the finner dimple pattern observed using miorofractography.

  • The extraction procedure and HPLC condition were modified to analyze the residues of oxytetracycline, sulfamethoxazole and chloramphenicol in pork, simultaneously. The antibacterial agents in pork were extracted with 0.02M EDTA-Mcilivine buffer:ethanol:acetonitrile (5:3:2). After the removal of fat with n-hexane, the extracts were evaporated and purified with Sep-pak $C_{18}$ cartridge column using 0.01M oxalic acid 0.1% (v/v) triethylamine (TEA) in acetonitrile. The peak of antibacterial agents was detected with $\mu$ Bondapak C18 column, UV detector (280nm) and 0.01M oxalic acid: methanol: acetonitrile (7.5:2.0:0.5). Detection limits for three antibacterial standards were 0.03 ppm. Calibration curves were linear between 0.03 and 2.0 ppm (R$^2$>0.999). When spiked the level of 1.0 ppm of oxytetracycline, sulfamethoxazole and chloramphenicol into meats, the recoveries from meats were 77.3%, 79.7% and 59.3%, respectively. These results showed that the modified extraction method provided good analytical resolution and the recoveries of the above antibacterial agents in meats.

  • In this paper a further generalization of the theory of multilayer physical adsorption previously developed by the authors is attempted so that the effect of vertical interactions between adsorbent and adsorbate can be explicitly taken into account. In this attempt we have to discard the previously adopted assumption that the molecules in the second layer or above are all in the same physical state. In order to estimate the effect of vertical interactions on the adsorption isotherm the interaction energy between an adsorbed molecule and the adsorbent surface is assumd to vary as $r^{-3}$ where r is the distance that the molecule under consideration is separated from the adsorbent surface. Resulting adsorption isotherm is applied to interpret the adsorption data of tetramethylsilane vapor on iron film and good agreements between observed and calculated values are obtained over wide range of pressure.
