양식참굴의 PAHs 노출에 대한 생리적 적응

  • 해양환경의 대표적 오염물질인 PAHs가 양식굴의 생리대사에 미치는 영향을 파악하고자, 서로 다른 PAHs 농도에 노출된 굴의 여수율, 호흡율, 배설율 및 동화율의 변화를 조사하였다. 부착생물을 깨끗하게 제거한 굴을 실험실로 옮겨 3일정도 순치시키고, 이후 각각의 0, 50, 100, 200ppb의 농도별로 약 10일간 미리 노출시켰다. 이후 각 개체별로 2L 들이 2$0^{\circ}C$항온수조에서 0.2$\mu\textrm{m}$ 여과해수를 사용하여 실험하였다. 여수율은 Chaetoceros simplex를 지표생물로 하여 수조내 시간당 감소율을 구하여 Kim(1995)의 식에 의해 구하였다. 호흡율은 유동파라핀으로 외부와의 공기접촉을 단절시키고, 수조내 단위시간당 DO meter로 산소변화를 측정하여 Widdows and Johnson(1988)의 식을 변형하여 계산하였다. 배설율은 단위시간동안 수조내 NH$_4$-N 농도 증가량으로 조사하였고, NH$_4$-N은 HACH Inc.에서 제공하는 측정법을 따랐다. 여수율은 500ppb에서 다소 영향을 받아 증가하였다. 호흡율과 배설율은 노출농도가 증가함에 따라 증가하였지만, 50ppb와 100ppb에서는 다소 차이가 없었다. 따라서 양식굴이 PAHs와 같은 오염물질에 노출되었을 경우, 초기에는 여수율의 변화는 없고 배설율과 호흡율이 상승하여 체내 에너지 요구량이 증가하는 것으로 생각된다.

Fluorescence Characteristic Spectra of Domestic Fuel Products through Laser Induced Fluorescence Detection

  • Traditional investigation procedures of soil and groundwater contamination are followed by soil gas sampling, soil sampling, groundwater sampling, establishment of monitoring wells, and groundwater monitoring. It often takes several weeks to obtain the analysis reports, and sometimes, it needs supplemental sampling and analysis to delineate the polluted area. Laser induced fluorescence (LIF) system is designed for the detection of free-phase petroleum pollutants, and it is suitable for on-site real-time site investigation when coupling with a direct push testing tool. Petroleum products always contain polycyclic aromatic hydrocarbon (PAH) compounds possessing fluorescence characteristics that make them detectable through LIF detection. In this study, LIF spectroscopy of 5 major fuel products was conducted to establish the databank of LIF fluorescence characteristic spectra, including gasoline, diesel, jet fuel, marine fuel and low-sulfur fuel. Multivariate statistical tools were also applied to distinguish LIF fluorescence characteristic spectra among the mixtures of selected fuel products. This study successfully demonstrated the feasibility of identifying fuel species based on LIF characteristic fluorescence spectra, also LIF seemed to be uncovered its powerful ability of tracing underground petroleum leakages.

Bioassays of Polycyclic Aromatic Hydrocarbons in ZR-75-1 Human Breast Cancer Cells

  • Polycyclic aromatic hydrocarbons (PAH) are frequently detected in food, water, soil, and sediment and are widespread environmental pollutants formed by the incomplete combustion of fossil fuels, woods and other organic matter. PAHs are considered to be probable human carcinogens. The mechanism of action of PAHs has been studied extensively, however it is not clear how PAHs turn on CYP1A1 in human breast cancer. Our laboratory have been studied the effect of PAHs in the human breast cancer cell MCF7. In this study, we examined the ZR-75-1 human breast cancer cells as a new system to evaluate bioactivity of PAHs. ZR-75-1 human breast cancer cell line responses to estrogen and progesteron. We have been able to estbilish long term culture system of this cells then used for the study to observe the effect of PAHs. We demonstrate that PAHs induced the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase (EROD) activity in a concentration-dependant manner. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1A1 mRNA. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD. Apparently, ZR-75-1 cells have Aryl hydrocarbon receptors, therefore it would be good experimental tool to study the cross-talk between PAHs and steroid actions.

Effect of Surfactant Molecules on the Aqueous Solubility of two PAHs (새로운 계면활성제들에 의한 두 PAH의 수중 용해도 증가)

  • Surfactants have been extensively considered for decontamination of the subsurface polluted with hydrophobic organic compounds. In order to investigate the effect of molecular structures on the solubilization of hydrophobic organic compounds, solubility enhancement of two PAHs in solutions of three different surfactants-conventional, dianionic, and gemini. The batch experimental results showed that the gemini was the most effective and the dianionic was the least, indicating that organic carbon content of the surfactants was the major factor which determines the sorption capacity of surfactant aggregates in water, unlike some of the previous reports.

Day and Night Distribution of Gas and Particle Phases Polycyclic Aromatic Hydrocarbons (PAHs) Concentrations in the Atmosphere of Seoul (서울 대기 중 기체 및 입자상 다환방향족탄화수소 (PAHs)의 낮·밤 분포 특성)

  • Day and night sampling for gas and particle phases PAHs were carried out in Seoul to characterize gas and particle phases PAHs concentrations in day and night times. There was no significant difference between day and night time for particle phase PAHs concentrations and phase distribution of PAHs, while, gas phase PAHs concentrations in daytime were about 1/2 of nighttime concentrations in both summer and winter due to photochemical reaction of gas phase PAHs during daytime. A high fraction of cancer risk for PAHs was attributed to particle phase PAHs and the excess cancer risk in winter was higher than in summer. The excess cancer risk level of total(gas+particle) PAHs in summer was partially observed when both gas and particle phase PAHs concentrations were considered as risk assessment. Based on the diagnostic ratios and factor analysis of PAHs concentrations, combustion(coal and natural gas) and vehicular emission might be the most significant contributors of PAHs and major factors for determining of PAHs concentration were different between day and night times.
