P53 Expression in Adenoid Cystic Carcinoma of the Head and Neck (두경부 영역에 발생한 선양낭포암에서의 p53 단백 발현)

  • Overexpression of the mutant p53, derived from mutation of the p53 gene which is located on the short arm of chromosome 17 and plays a role in suppression of the tumor, was reported in some human malignancy such as breast or colon carcinoma and suggested to be a prognostic factor. The authors investigated expression of p53 protein by immunohistochemical staining using anti-p53 monoclonal antibody in the paraffin embedded blocks of the 23 patients who were diagnosed at Seoul National University Hospital as adenoid cystic carcinoma of the head and neck between 1982 and 1991, and could be followed-up. 7 cases(34%) out of the 23 cases showed p53 expression, and there was no significant association between p53 positivity and local recurrence(p=0.31) or distant metastasis(p=0.16)

Expression of Tiam1 in Lung Cancer and its Clinical Significance

  • The aim of this study was to ana1yze T-cell lymphoma invasion and metastasis-inducing factor 1 (Tiam1) expression in 1ung cancer patients. A total of 204 patients with lung cancer tissue lesions were enrolled in the present study, along with 40 cases of normal lung tissue and 40 of normal fetal lung tissue. Tiam1 protein expression level was determined using intensity quantitative analysis, for comparison in lung cancer, metastatic, normal lung, and fetal lung tissue. The positive unit (PU) of Tiam1 was $13.5{\pm}5.42$ in lung cancer,$5.67{\pm}1.56$ in norma1 epithelial cells, and $5.89{\pm}1.45$ in fetal lung epithelial cells. The value in the lung cancer tissue was significantly higher than that in the normal lung tissue and the fetal lung tissue (P<0.01). The Tiam1 PU values with lymph node metastasis and without 1ymph node metastasis were $15.2{\pm}4.34$ and $12.5{\pm}4.23$, respectively, and the difference was statistically significant (P<0.05). The Tiam1 PU values in different tumor, nodes, metastasis (TNM) stages, III-IV period, and I-II phase were $14.7{\pm}4.14$ and $11.0{\pm}5.34$ (P<0.05). A correlation was found between Tiam1 expression and the age of patient, tumor size, tumor type, and tumor differentiation. Tiam1 protein expression in the lung tumor tissue is significantly higher than that in the normal lung tissue and fetal lung tissue. Tiam1 expression may be closely related to lung cancer development and metastasis.

Purification and Some Properties of Cyclodextrin Hydrolase (Cyclodextrin분해효소의 정제 및 그 특성)

  • Cyclodextrin hydrolase from Bacillus stearothermophilus KFCC 21203 was purified and the properties of the purified enzyme were investigated. The enzyme was purified 15 folds with 77 % recovery by ammonium sulfate fractionation, DEAE-cellulose chromatography, and Ultro AcA 34 gel filtration. The specific activity and the molecular weight of the enzyme were 1.30 units/mg protein and about 29,500, respectively, The maximum activity of the enzyme was shown at $55^{\circ}C$ and pH 5.5. However, stable temperature and pH were $40^{\circ}C$ and $5.0{\sim}8.0$, respectively. The Km value for ${\gamma}-cyclodextrin$ was $3.78{\times}10^{-3}$ M. The degradation activity of the enzyme was selectively high for ${\gamma}-cyclodextrin$, and very low for ${\beta}-cyclodextrin$, but not for ${\alpha}-cyclodextrin$. The decomposed products of ${\gamma}-cyclodextrin$ were mainly glucose and maltose, and a little mlatotriose. The activity of the enzyme was very high for amylose, potato starch, corn starch, amylopectin and maltooligomer, and relatively high for glycogen and dextrin. The decomposed products of them were mainly glucose and maltose.

Correlation between GenoType MTBDRplus Assay and Phenotypic Susceptibility Test for Prothionamide in Patients with Genotypic Isoniazid Resistance

  • Background: The purpose of this study was to analyze the relationship between the gene mutation patterns by the GenoType MTBDRplus (MTBDRplus) assay and the phenotypic drug susceptibility test (pDST) results of isoniazid (INH) and prothionamide (Pto). Methods: A total of 206 patients whose MTBDRplus assay results revealed katG or inhA mutations were enrolled in the study. The pDST results were compared to mutation patterns on the MTBDRplus assay. Results: The katG and inhA mutations were identified in 68.0% and 35.0% of patients, respectively. Among the 134 isolated katG mutations, three (2.2%), 127 (94.8%) and 11 (8.2%) were phenotypically resistant to low-level INH, high-level INH, and Pto, respectively. Among the 66 isolated inhA mutations, 34 (51.5%), 18 (27.3%) and 21 (31.8%) were phenotypically resistant to low-level INH, high-level INH, and Pto, respectively. Of the 34 phenotypic Pto resistant isolates, 21 (61.8%), 11 (32.4%), and two (5.9%) had inhA, katG, and both gene mutations. Conclusion: It is noted that Pto may still be selected as one of the appropriate multidrug-resistant tuberculosis regimen, although inhA mutation is detected by the MTBDRplus assay until pDST confirms a Pto resistance. The reporting of detailed mutation patterns of the MTBDRplus assay may be important for clinical practice, rather than simply presenting resistance or susceptibility test results.

Genetic and Environmental Effects on the Lactation Yield and Milk Compositions in Holstein Cow (Holstein종 유우(乳牛)의 비유량(泌乳量) 및 유조성분(乳組成分)에 미치는 유전(遺傳) 및 환경(環境)의 효과(效果))

  • This study was conducted to estimate the genetic and environmental effects on lactation yield and milk compositions in Holstein cows. The data analysis were the records of 159 cows rearing at Nation Animal Breeding Institute from 1990 to 1991. The least square means were estimated on milk and fat yield in lactation yield, and the percent of fat, protein, solids-not-fat and total solid in milk composition. The results obtained are summarized as follows: 1. The average yield of milk and fat in 305 days were $7,26.56{\pm}3,57.24$ and $254.65{\pm}44.94Kg$ and the percent of fat, protein, solids-not-fat and total solid were $3.69{\pm}0.43$, $3.32{\pm}0.41$, $9.15{\pm}0.49$ and $12.75{\pm}0.96$ and the coefficients of variation were 18.68, 17.64, 11.88 and 12.34% for milk yield, fat yield, fat percent and protein percent, respectively. 2. The effect of sires was highly significant at 1% level in milk and fat yield and fat percent, and significant at 5% level in protein and total solid percent. Among the sires, B, L and O sire were superior in milk yield with 7,571.22, 7,499.11 and 7,420.58 Kg, and A, F and K sire were superior in protein percent with 3.75, 3.64 and 3.65, respectively. 3. The effect of parity was highly significant at 1% level in milk yield, and significant at 5% level in fat yield. Among the parities, the 3rd parity was superior in milk and fat yield with 7,634.54 and 274.98 Kg, and the 4th and over was superior in fat and protein percent with 3.90 and 3.50, respectively. 4. The effect of calving seasons was highly significant at 1% level in milk yield, and significant at 5% level in fat yield, and the percent of fat, protein and total solid. Among the calving seasons, spring and winter were superior in milk yield with 7,310.31 and 7,364.57 Kg, also spring and winter were superior in protein percent with 3.68 and 3.52, respectively.

Red Pine Bark Extract Alleviates Akt/GSK-3β Signaling Disruption in the Hippocampus of Streptozotocin-Induced Diabetic Sprague-Dawley Rats

  • This study investigates whether red pine (Pinus densiflora Sieb. et Zucc.) bark extract (PBE) can alleviate diabetes and abnormal apoptosis signaling pathways in the hippocampus of streptozotocin (STZ)-induced diabetic Sprague-Dawley (SD) rats. Two dosages of PBE (15 and 30 mg/kg of body weight/day) were administered orally to STZ-induced diabetic SD rats for 20 days. Blood glucose level and body weight were measured once per week. After 20 days of oral administration of PBE, the rat hippocampus was collected, and the production of Akt, p-Akt, GSK-3β, p-GSK-3β, tau, p-tau, Bax, and Bcl-2 proteins were determined by western blot analysis. A decrease in blood glucose level and recovery of body weight were observed in PBE-treated diabetic rats. In the Akt/GSK-3β/tau signaling pathway, PBE inhibited diabetes-induced Akt inactivation, GSK-3β inactivation, and tau hyperphosphorylation. The protein production ratio of Bax/Bcl-2 was restored to the control group level. These results suggest that PBE, rich in phenolic compounds, can be used as a functional food ingredient to ameliorate neuronal apoptosis in diabetes mellitus.

Effects of Crude Protein Levels on Nitrogen Balance in Adult Jindo Dog (사료내 조단백질 수준이 진도성견의 질소균형에 미치는 영향)

  • This study was conducted to determine the protein requirement level in adult Jindo dog (Korea Jindo Dog) through nitrogen balance experiments. 12 female dogs aged 36~38 weeks old (for early stage) or aged 45~49 weeks old (for late stage) were fed one of 3 diets containing 17, 19 and 21% of crude protein. Body weight of dogs were measured during the 12 days of feeding trial, and feces and urine were collected last 3 days of the trial period. In early stage of Jindo dog, average daily gain of dogs fed experimental diets containing 17, 19 and 21% of crude protein were -38.33, -16.25 and -12.71g/d, respectively. There was a significant differences between 17 and 21% of crude protein treatments (p<0.05) and calculated retained nitrogen were -0.29, -0.04 and -0.03g/kg$BW^{.75}$/d, respectively. In late stage of Jindo dog, average daily gain of dogs fed experimental diets containing 17, 19 and 21% of crude protein were -34.05, -28.71 and -28.28g/d, and calculated retained nitrogen were -0.33, -0.06 and 0.09g/kg$BW^{.75}$/d, respectively, and were significantly higher (p<0.05) in dogs fed 21% of crude protein diet than those of other dogs. In early stage and late stage of adult Jindo dog, a calculated linear regression equation for nitrogen intake (x) and nitrogen retention (y) were y = 0.7484x-1.18 ($R^2$ = 0.9923) and y = 0.88231x-1.0894 ($R^2$ = 0.9982) and the requirement of crude protein were estimated as 9.85g/kg$BW^{.75}$/d and 7.72g/kg$BW^{.75}$/d from above equation.

Changes in Nutritional Composition of Gryllus bimaculatus Fermented by Bacillus sp. and Mycelium of Basidiomycetes (고초균 및 버섯 균사체를 이용하여 발효한 쌍별귀뚜라미 발효물의 단백질 및 무기질 성분 변화)

  • This study examined the changes in the protein and mineral composition of Gryllus bimaculatus fermented with Bacillus substilis and the mycelia of Basidiomycetes. Normal Gryllus bimaculatus (S) and experimental group data obtained after an inoculation of Bacillus substilis (SC) (KACC 19623), Pleurotus eryngii (SP) and Cordyceps millitaris (SC) were compared. The crude protein content of the Gryllus bimaculatus (control) was 75.48%, but it decreased to 64.55, 54.32, and 63.53% after fermentation with SB, SP and SC, respectively (p<0.05). An analysis of the organic elements showed that the contents of the carbon and nitrogen sources were also reduced after fermentation, and the most significant decrease was observed after fermentation with SP. In SDS-PAGE, a 120 kDa and a 48 kDa protein of Gryllus bimaculatus were found. On the other hand, protein bands faded after fermentation with SP and SC, respectively. Moreover, no visible band was observed after fermentation with SB. According to amino acid analysis, the total free amino acid content increased 3.84 and 1.74 times after fermentation with SB and SP, respectively, compared to the corresponding baseline data. In contrast, it decreased by 0.52 times after fermentation with SC. Among the essential amino acids found in crickets fermented with SB, the valine and isoleucine content was 3.57 and 2.64 times higher, respectively, than the recommended daily amount of essential amino acids.

Influence of Milk Yield, Parity, Stage of Lactation and Body Weight on Urea and Protein Concentration in Milk of Murrah Buffaloes

  • The present study was carried out to investigate the effect of test day milk yield, test day evening milk yield, parity, stage of lactation and body weight on milk urea and milk protein concentration. A total of 319 milk samples was collected from buffaloes over four month's period and subjected to urea and protein analysis. Milk urea concentration (mg/dl) was significantly (p<0.01) increased with increasing test day milk yield. The lowest value ($57.03{\pm}1.13$) was observed in the milk yield group ${\leq}4.5kg/day$ and the highest value ($64.15{\pm}1.13$) in the group 7.7-10.7 kg/day. However, test day evening milk yield had no significant effect on milk urea concentration. Milk protein did not vary significantly with the test day milk yield as well as test day evening milk yield. A clear decreasing trend of milk urea concentration (mg/dl) was found with the increasing parity. The highest MU concentration ($64.03{\pm}1.14$) was found in the first parity and the lowest ($55.67{\pm}1.22$) was found in the sixth and above parity. Whereas, stage of lactation had no effect on milk urea concentration. Moreover, parity and stage of lactation did not have any significant effect on milk protein concentration. Body weight (kg) was also found negatively (p<0.05) related with urea content (mg/dl) in milk. The highest mean MU concentration ($64.34{\pm}0.88$) was found when body weight was between 532 and 598 kg and lower mean values ($59.24{\pm}0.94$ and $59.33{\pm}1.23$) were observed in 599 to 665 kg and ${\geq}666kg$ group. Body weight also had significant (p<0.05) effect on milk protein content. The highest milk protein content (%) was found in ${\geq}666kg$ group and the lowest in <531 kg group. In conclusion, for proper interpretation of milk urea values to monitor protein nutrition status of the buffaloes parity, milk yield and body weight should be considered.

Change on the Food and Nutrient Intake Patterns of Men over Thirty Years Old in Jeon-ju Area(1991-1997) (전주지역 30세 이상 남성의 식품과 영양소 섭취 형태 변화 (1991-1997))

  • The objective of this study was to estimate changes on the flood and nutrient intake patterns of men over thirty years old in Jeon-Ju area. The first survey was conducted from December in 1991 to January in 1992, tile second one from January to February in 1994 and the third one from July to August in 1997. The nutrition survey using 24-hour recall method was executed to 303 subjects : 89, 82, 132 in 1991, 1994 and 1997, respectively. Results of the study are as follows : Kimchi, rice, garlic and onions were the most frequently eaten food items. Total daily intakes of foods were 85 : 15, 81 : 19 and 81 : 19 in 1991, 1994 and 1997, respectively. The average numbers of foods per person were 15.7, 20.1 and 21.9 daily in 1991, 1994 and 1997, respectively and tends to increase significantly(p<0.05). The minimum numbers of foods per person were 4, 7 and 9 and the maximum numbers of foods per person were 27, 35 and 39 in 1991, 1994 and 1997, respectively. KDDS(Korean\`s Dietary Diversity Score) is determined by how many among the five food groups (cereals, vegetables, meats, milks, oils groups)were consumed per day. Most subjects earned the KDDS "3" ; 61, 46 and 42% in 1991, 1994 and 1997, respectively. Average daily energy intakes wee 1,62㎉(72% of RDA), 2,063㎉(89% of RDA) and 1,818㎉ (79% of RDA) in 1991, 1994 and 1997, respectively. Energy intake rates of cereals : total energy intake were 65, 59, and 60% in 1991, 1994 and 1997, respectively, which were decreasing. Protein intakes were 58g(72% of RDA), 79g(107% of RDA) and 71g(97% of RDA), respectively and animal protein comprised 46, 53, and 59%, respectively ; which were increased. Fat intakes were 12g, 20g and 20g, respectively and animal protein comprised 38, 46, and 48% ; which were increased. Fat intakes were 12g, 20g and 20g, respectively, of which animal fat comprised 46, 53, and 59%, respectively ; which were increased, too. Malnourished (under 75% of RDA) rates were respectively 64, 34, and 47% in terms of energy ; 64, 31 and 33% in protein ; 67, 51, and 61% in calcium ; 53, 26, and 18% in iron ; 85, 74 and 84% in Vitamin A. Super-nourished(above 125% of RDA) rates were respectively 1, 13, and 3% in energy ; 1, 29, and 21% in protein ; 5, 18, and 7% in calcium ; 16, 31, and 7% in iron ; 16, 31, and 7% in Vitamin A, 42, 76, and 62% in Vitamin C. The percentages of calories from protein : fat : carbohydrate were 14 : 12 : 74, 15 : 16 : 69 and 16 : 17 : 67 in 1991, 1994 and 1997, respectively. KDDS(number of five food groups per day), Meal Balance(number of five food groups per meal), DVS(average numbers of foods per person), amount of foods correlated positively with all the nutrient intakes(p<0.05). KDDS was positively correlated with energy, protein, fat, calcium, thiamin, riboflavin and niacin(p<0.05)in(p<0.05)
