The objectives of this study were to optimize submerged culture conditions of a new fungal isolate, Ganorderma resinaceum, and to enhance the production of bioactive mycelial biomass and exopolysaccharides (EPS) by fed-batch culture. The maximum mycelial growth and EPS production in batch culture were achieved in a medium containing 10 g/l glucose, 8 g/l soy peptone, and 5 mM $MnCl_2$ at an initial pH 6.0 and temperature $31^{\circ}C$. After optimization of culture medium and environmental conditions in batch cultures, a fed-batch culture strategy was employed to enhance production of mycelial biomass and EPS. Five different EPS with molecular weights ranging from 53,000 to 5,257,000 g/mole were obtained from either top or bottom fractions of ethanol precipitate of culture filtrate. A fed-batch culture of G. resinaceum led to enhanced production of both mycelial biomass and EPS. The maximum concentrations of mycelial biomass (42.2 g/l) and EPS (4.6 g/l) were obtained when 50 g/l of glucose was fed at day 6 into an initial 10 g/l of glucose medium. It may be worth attempting with other mushroom fermentation processes for enhanced production of mushroom polysaccharides, particularly those with industrial potential.

A psychrotrophic bacterium, Arthrobacter sp. ON14, isolated from Antarctica, was shown to exhibit a high ${\beta}$-galactosidase activity at a low temperature. A genomic library of ON14 was constructed and screened for ${\beta}$-galactosidase genes on functional plates containing 5-bromo-4-chloro-3-indolyl-${\beta}$-D-galactopyranoside (X-gal) as the substrate. Two different ${\beta}$-galactosidase genes, named as galA, galB, were found in ON14. Computational analyses of the genes revealed that the encoded protein GalA belongs to family 2 of glycosyl hydrolysases and is a cold-active protein, whereas GalB belongs to family 42 of glycosyl hydrolysases and is a mesophilic protein. Reverse transcription analyses revealed that the expression of galA is highly induced at a low temperature ($4^{\circ}C$ ) and repressed at a high temperature ($28^{\circ}C$ ) when lactose is used as the sole carbon source. Conversely, the expression of galB is inhibited at a low temperature and induced at a high temperature. The purified GalA showed its peak activity at $15^{\circ}C$ and pH 8. The mineral ions $Na^+$, $K^+$, $Mg^{2+}$, and $Mn^{2+}$ were identified as enzyme activators, whereas $Ca^{2+}$ had no influence on the enzyme activity. An enzyme stability assay revealed that the activity of GalA is significantly decreased when it is incubated at $45^{\circ}C$ for 2 h, and all its activity is lost when it is incubated at $50^{\circ}C$.
The objective of this study was to examine Jeonju residents' recognition towards their native local foods. The results indicate that their degree of interest in the native foods was above average. As age and education increased, the degree of interest was also increased. Also, married people showed greater interest as compared to single people (p<$0.05{\sim}0.001$). There were more people who said they preferred native foods than people who said they didn't, where 213 people (41.3%) answered that they preferred the native foods. Of the people who said they like native foods, their main reason was because the native foods taste good. The main reason for the people who said they disliked native foods was that they didn't have many opportunities to try native foods. TV and radio were the first means, among others, from which people learned about native foods. The place where people ate native foods most often was native food restaurants (242 people, 46.95%). A frequency of a native food intake (including home and dining-out) of $1{\sim}2$ times/year was most prominent, with 137 people (26.6%). Also, a cooking frequency of "hardly ever cooks" was most prominent with 163 people (31.6%). The most prominent reason for the low cooking frequency at home was because they didn't know the recipes. Among 5 basic recipe characteristics for native foods (easy preparation of ingredients, simple recipe procedures, quick preparation, reasonable recipe procedures, sanitation), "sanitation" had the highest average score of 3.54, and "simple recipe procedure" had the lowest score of 2.44. To summarize, in order to develop Jeonju native local foods, it is necessary to implement various strategies such as frequently holding events where people can experience their native foods, running a native food training center, advertising through TV and radio, developing easy home recipes, and simplifying recipes.
The purpose of this study was to provide baseline data for revising the recommended energy intake for Korean adults. We recruited 290 adults so as to determine their resting energy expenditure (REE) and energy intake. The REE was measured by indirect calorimetry. We also calculated the REE from prediction equations formulated by World Health Organization (WHO), The energy intake for two consecutive days was assessed using the 24 hour recall method. The body weight, lean body mass (LBM) and percentage body fat were measured using the INBODY 3.0 system. We compared the results of three age groups ; 20 to 29 years,30 to 49 years and 50 years or more. The average energy intake of each age group was below the 7th Korean Recommended Dietary Allowances (RDA). The average energy intake was lower in the older age groups. However, no difference was observed among the age groups when the energy intake as a percentage of the Korean RDA was compared. Our measurement of the REE was higher than the REE calculated by the WHO's method. Correlation coefficients between the measured and the calculated values of REE for all age groups showed significant correlations (r=0.475-0.672). As the ages of all the subjects increased, the REE/kg of body weight decreased. There were no significant differences in the REE / kg of the LBM between the different age groups; however, the REE/kg of the LBM was higher in the female group than in the male group. Negative correlations of the REE with the age (r=-0.242) and body fat ratio (r=-0.313) were observed; positive correlations of the REE with the BMI (r=0.265), height (r=0.570), weight (r=0.562) and LBM (r=0.586) were also found (p<0.01).
Objective : This study is aimed to evaluate the effects of Evodiae Fructus herbal-acupuncture (EF-HA) at KI10 on osteoporosis induced by ovariectomy in mice. Method : Mice underwent bilateral ovariectomy. After recovering, the ovariectomized (OVX) mice were treated by needle prick, saline injection, herbal acupuncture with Evodiae Fructus (EF-HA) at KI10 for 8 weeks. Result : 1. EF-HA at KI10 significantly inhibited the overgrowth of tibia in ovariectomized mice. 2. NP at KI10 significantly restored the tibial BMD (bone mineral density) in ovariectomized mice. 3. EF-HA at KI10 significantly restored the phosphorus and creatinine levels in ovariectomized mice serum. 4. EF-HA at KI10 significantly restored the tibial Ca and P levels in ovariectomized mice. 5. EF-HA at KI10 significantly reduced the tibial osteoclast-like cells in ovariectomized mice. 6. EF-HA at KI10 significantly inhibited the overgrowth of tibial GPL (growth plate length) in ovariectomized mice. Conclusion : EF-HA at KI10 has protective and therapeutic effect for osteoporosis in ovariectomized mice. Thus, it is suggested that EF-HA can be an useful therapeutics in clinical field after further researches.
Use of electrical stimulation for diagnosis and treatment of pyhsiolgocial impairment on the auditory system was reported by many researchers since 1800. Unfortunately, there were no standard methods or theoretical bakground for choosing stimulus conditions because of the lack of understanding on the transmission of sound through the auditory pathways. Therefore, if we could find the mechanisms of normal and abnormal functions for the system objectively, it may be possible to establish the conditions for the appropriate treatment This study was devised with three experimental conditions of electrical stimulation(0.1, 1, and 10mA) to verify the effect of electrical stimulation on the auditory system. ABRs and ECochGs were obtained from 3 Guinea Pigs before and after each stimulation, and five parameters were extracted from ABR and ECochG waveforms. The results showed that the mean differences of the first peaks of ABR, I, before and after all the stimulus conditions were significant at the significant level of p<0.05. Also, amplitude of ECochGs peak(AP) showed higher degrees of differences for the electrical stimulation than that of ABRs. Based on the results of this experiment establishment of an electrical model which characterizes the auditory system could be achieved by computer simulation. This will lead to the design of most accurate model for the real auditory system, which provide the quantitive information regarding diagnosis and treatment.
Background: S-2-(3 aminoprophlamino) ethylphosphorothioic acid(WR-2721) is one of the radical scavenging thiols. We tested its protective effects in the reperfused heart. Material and Method: The experimental setup was the constant pressure Langendorffs perfusion system. We investigated the radical scavenging properties of this compound in isolated rat hearts which were exposed to 20 minutes ischemia and 20 minutes reperfusion. Four experimental groups were used:group I, control, Amifostine 50 mg(1 mL) peritoneal injection 30 minutes before ischemia(group II), Amifostine 10 mg(0.2 mL) injection during ischemia through coronary artery(group III),and Amifostine 50 mg(1 mL) peritoneal injection 2 hrs before ischemia(group IV). The experimental parameters were the levels of latate, CK-MB, and adenosine deaminase(ADA) in frozen myocardium, the quantity of coronary flow,and left ventricular developed pressure, and it's dp/dt. Statistical analysis was performed using repeated measured analysis of variance and student t-test. Result: The coronary flow of group II and IV were less than group I and III at equilibrium state but recovery of coronary flow at reperfusion state of group II, III, and IV were more increased compared with group I. The change of systolic left ventricular devoloping pressure of group II and IV were less than control group at equilibrium state, which seemed to be the influence of the pharmacological hypotensive effect of amifostine. But it was higher compared with group I at reperfusion state. The lactic acid contents of group II were less than control group in frozen myocardium.(Group I was 0.20 0.29 mM/g vs Group II, which was 0.10 0.11 mM/g). The quantity of CK-MB in myocardial tissue was highest in group IV (P=0.026 I: 120.0 97.8 U/L vs IV: 242.2 79.15 U/L). The adenosine deaminase contents in the coronary flow and frozen myocardium were not significantly different among each group. Conclusion: Amifostine seemed to have significant cardioprotective effect during ischemia and reperfusion injuries of myocardium.
Smoking is one of the most serious but preventable causes of cardiovascular disease (CVD). Key aspects of pathological process associated with smoking include endothelial dysfunction, a prothrombotic state, inflammation, altered lipid metabolism, and hypoxia. Multiple molecular events are involved in smoking-induced CVD. However, the dysregulations of reactive oxygen species (ROS) generation and metabolism mainly contribute to the development of diverse CVDs, and NADPH oxidase (NOX) has been established as a source of ROS responsible for the pathogenesis of CVD. NOX activation and resultant ROS production by cigarette smoke (CS) treatment have been widely observed in isolated blood vessels and cultured vascular cells, including endothelial and smooth muscle cells. NOX-mediated oxidative stress has also been demonstrated in animal studies. Of the various NOX isoforms, NOX2 has been reported to mediate ROS generation by CS, but other isoforms were not tested thoroughly. Of the many CS constituents, nicotine, methyl vinyl ketone, and ${\alpha}$,${\beta}$-unsaturated aldehydes, such as, acrolein and crotonaldehyde, appear to be primarily responsible for NOX-mediated cytotoxicity, but additional validation will be needed. Human epidemiological studies have reported relationships between polymorphisms in the CYBA gene encoding p22phox, a catalytic subunit of NOX and susceptibility to smoking-related CVDs. In particular, G allele carriers of A640G and $-930^{A/G}$ polymorphisms were found to be vulnerable to smoking-induced cardiovascular toxicity, but results for C242T studies are conflicting. On the whole, evidence implicates the etiological role of NOX in smoking-induced CVD, but the clinical relevance of NOX activation by smoking and its contribution to CVD require further validation in human studies. A detailed understanding of the role of NOX would be helpful to assess the risk of smoking to human health, to define high-risk subgroups, and to develop strategies to prevent or treat smoking-induced CVD.
PURPOSE: The purpose of this study was to determine the correlation between the hip adductor muscles and abdominal muscles during bridge exercise. METHOD: Participants who met the criteria for this study(n=36) were divided into the three groups. The first experimental group performed normal bridge exercises and the second group performed bridge exercises with the contraction of the hip adductor muscles and the control group didn't perform any exercise. Transversus abdominis muscle thickness was measured by ultrasound imaging with a special transducer head device, at pre exercise, after 2 weeks, 4 weeks, and 6 weeks. RESULT: Data were analyzed using repeated ANOVA with the level of significance set at ${\alpha}=.05$. Transversus abdominis muscle thickness was influenced by contraction of the hip adductor muscles during bridge exercise in people without lower back pain. Compared with normal bridge exercise, transversus abdominis muscle thickness significantly increased in thickness during bridge exercise with contraction of the hip adductor muscles(p<.05). CONCLUSION: The results from this study showed that contraction of the hip adductor muscles during bridge exercise increased change in the transversus abdominis muscle thickness. These results can be a good source to prevent low back pain due to hip adductor weakness. Therefore, inducing activation of hip adductor with abdominal stabilizing exercise is more effective in patients with low back pain.

Background/Aims: Several epidemiological studies have validated the association of interleukin gene polymorphisms with acute pancreatitis (AP) in different populations. However, there have been few studies in Asian ethnic groups. We aimed to investigate the relationships between inflammatory cytokine polymorphisms and AP as pilot research in a Korean ethnic group. Methods: Patients who had been diagnosed with AP were prospectively enrolled. DNA was extracted from whole blood, and DNA sequencing was subsequently performed. Single-nucleotide polymorphisms (SNPs) of the interleukin $1{\beta}$ (IL1B), interleukin 1 receptor antagonist (IL1RN), and tumor necrosis factor ${\alpha}$ (TNFA) genes of patients with AP were compared to those of normal controls. Results: Between January 2011 and January 2013, a total of 65 subjects were enrolled (40 patients with AP vs. 25 healthy controls). One intronic SNP (IL1RN -1129T>C, rs4251961) was significantly associated with the risk of AP (odds ratio, 0.304; 95% confidence interval, 0.095 to 0.967; p = 0.043). However, in our study, AP was not found to be associated with polymorphisms in the promoter regions of inflammatory cytokine genes, including IL1B (-118C>T, c47+242C>T, +3954C/T, and -598T>C) and TNFA (-1211T>C, -1043C>A, -1037C>T, -488G>A, and -418G>A). Conclusions: IL1RN -1129T>C (rs4251961) genotypes might be associated with a significant increase of AP risk in a Korean ethnic group.
