Helicobacter pylori cag Pathogenicity Island cagL and orf17 Genotypes Predict Risk of Peptic Ulcerations but not Gastric Cancer in Iran

  • Background: Gastric cancer (GC) is the third most common cancer regarding mortality in the world. The cag pathogenicity island (PAI) of Helicobacter pylori which contains genes associated with a more aggressive phenotype may involve in the pathogenesis of gastrointestinal disease. We here aimed to examine the associations of cagH, cagL, orf17, and cagG genotypes of H. pylori cag PAI with severe gastrointestinal disease. Materials and Methods: A total of 242 H. pylori strains were genotyped. Histopathological examination and classification of subjects were performed. Results: The frequencies of the cagH, cagL, cagG, and orf17 genotypes were 40/54 (74.1%), 53/54 (98.1%), 38/54 (70.4%), and 43/54 (79.6%), respectively, in patients with peptidic ulceration (PU),while in the control group, the frequencies were 87/147 (59.6%) for cagH, 121/146 (82.9%) for cagL, 109/146 (74.7%) for cagG, and 89/146 (61.0%) for orf17. The results of simple logistic regression analysis showed that the cagL and orf17 genotypes were significantly associated with an increased risk of PU not GC; the ORs (95% CI) were 10.950 (1.446-82.935), and 2.504 (1.193-5.253), respectively. No significant association was found between the cagH and cagG genotypes and the risk of both the PU and the GC in Iran (P>0.05). Finally, multiple logistic regression analysis showed that the cagL genotype was independently and significantly associated with the age-and sex-adjusted risk for PU; the OR (95% CI) was 9.557 (1.219-17.185). Conclusions: We conclude that the orf17 and especially cagL genotypes of H. pylori cag PAI could be factors for risk prediction of PU, but not GC in Iran.

Prediction of Peak Back Compressive Forces as a Function of Lifting Speed and Compressive Forces at Lift Origin and Destination - A Pilot Study

  • Objectives: To determine the feasibility of predicting static and dynamic peak back-compressive forces based on (1) static back compressive force values at the lift origin and destination and (2) lifting speed. Methods: Ten male subjects performed symmetric mid-sagittal floor-to-shoulder, floor-to-waist, and waist-to-shoulder lifts at three different speeds (slow, medium, and fast), and with two different loads (light and heavy). Two-dimensional kinematics and kinetics were captured. Linear regression analyses were used to develop prediction equations, the amount of predictability, and significance for static and dynamic peak back-compressive forces based on a static origin and destination average (SODA) backcompressive force. Results: Static and dynamic peak back-compressive forces were highly predicted by the SODA, with R2 values ranging from 0.830 to 0.947. Slopes were significantly different between slow and fast lifting speeds (p < 0.05) for the dynamic peak prediction equations. The slope of the regression line for static prediction was significantly greater than one with a significant positive intercept value. Conclusion: SODA under-predict both static and dynamic peak back-compressive force values. Peak values are highly predictable and could be readily determined using back-compressive force assessments at the origin and destination of a lifting task. This could be valuable for enhancing job design and analysis in the workplace and for large-scale studies where a full analysis of each lifting task is not feasible.

The effect of medical service satisfaction of orthodontic treatment on life quality related to oral health(OHIP-14) (교정치료의 의료서비스 만족도가 구강건강관련 삶의 질(OHIP-14)에 미치는 영향)

  • Objectives: The purpose of this study is to investigate the relationship between perceived oral health, medical service satisfaction, and oral health impact profile (OHIP-14) and the factors affecting OHIP for orthodontic patients, and to provide basic data to improve the quality of life of orthodontic patients in accordance with oral health. Methods: The study explained the purpose and purport of this study to orthodontic patients who visit dental clinics located in Daejeon and Chungcheong province for about two months from July 01, 2019, and conducted a self‐reported questionnaire survey for 220 participants who agreed to participate in this study. of the collected questionnaires, data of 197 subjects were used for the final analysis except 23 that were inadequate. Results: As for orthodontic treatment satisfaction according to perceived oral health and OHIP, there was statistically significant difference between respondents who were 'very healthy' and 'not healthy' (p<0.05). The higher the supplementary service satisfaction, the higher the orthodontic treatment satisfaction, and the higher the perceived oral health, the OHIP was found to be the higher. The factors affecting OHIP were found to be significantly associated with orthodontic treatment satisfaction and perceived oral health. The explanatory power was 15.6%. Conclusions: Active and ongoing oral health education should be provided to dental hygienists to ensure that the patient's oral cavity is healthy, and although additional services need to be improved, it is more important to have management strategies for patients to improve orthodontic treatment satisfaction.

Influence of Organic Ligands on Phytotoxicity of Paraquat (유기(有機)리간드가 Paraquat의 약해(藥害)에 미치는 영향)

  • Organic ligands in the environments are expected to play an important role in regulating the biotoxicity and fate of pesticides. Influences of dissolved humic and fulvic acids on the phytotoxicity of Paraquat were investigated using a bioassay with hydroponically grown rye as indicator species. Levels of Paraquat in water culture media were ranged from 0 to $12_{{\mu}M}$ and those of humic or fulvic acid were 1.0mM as a soluble carbon. Media were prepared in a factorial combination with pHs of 4.5, 6.5 and 8.5, Standard curves of necrosis days, fresh weight, and growth rates, as Phytotoxicity Indices, versus Paraquat concentrations were employed to evaluate the effects of organic ligands on phytotoxicity of paraquat. Organic ligand itself had little effect on rye growth, but Paraquat showed a high degree of toxicity. Paraquat started to show an intensive injury to rye at $0.4{\sim}0.6{{\mu}M}$ and upper critical phytotoxic concentration was estimated to be 11.0${{\mu}M}$ In the presence of organic ligands, times required to cause necrosis due to Paraquat were delayed upto 40%. Fresh weights and growth rates were upto 20% higher in treatments of organic ligands plus Paraquat than that of Paraquat alone. Results demonstrated that complexation of organic ligand with Paraquat reduced the bioabailability of Paraquat to rye.

Association between Single Nucleotide Polymorphisms of the Major Histocompatibility Complex Class II Gene and Newcastle Disease Virus Titre and Body Weight in Leung Hang Khao Chickens

  • The aim of the present study was to investigate the effect of single nucleotide polymorphisms in the major histocompatibility complex (MHC) class II gene on resistance to Newcastle disease virus and body weight of the Thai indigenous chicken, Leung Hang Khao (Gallus gallus domesticus). Blood samples were collected for single nucleotide polymorphism analysis from 485 chickens. Polymerase chain reaction sequencing was used to classify single nucleotide polymorphisms of class II MHC. Body weights were measured at the ages of 3, 4, 5, and 7 months. Titres of Newcastle disease virus at 2 weeks to 7 months were determined and the correlation between body weight and titre was analysed. The association between single nucleotide polymorphisms and body weight and titre were analysed by a generalized linear model. Seven single nucleotide polymorphisms were identified: C125T, A126T, C209G, C242T, A243T, C244T, and A254T. Significant correlations between log titre and body weight were found at 2 and 4 weeks. Associations between single nucleotide polymorphisms and titre were found for C209G and A254T, and between all single nucleotide polymorphisms (except A243T) and body weight. The results showed that class II MHC is associated with both titre of Newcastle disease virus and body weight in Leung Hang Khao chickens. This is of concern because improved growth traits are the main goal of breeding selection. Moreover, the results suggested that MHC has a pleiotropic effect on the titre and growth performance. This mechanism should be investigated in a future study.

The Nutritional Status of Antioxidant Vitamins in Relation to Serum MDA Level in Postmenopausal Women (폐경기 여성의 혈청 과산화지질 농도에 따른 항산화비타민의 영양상태)

  • The purpose of this study was to investigate the nutritional status of antioxidant vitamins in relation to serum malondialdehye(MDA) level in postmenopausal women with common occurance of cardiovascular disease(CVD). Data about general characteristics including smoking, drinking and exercise status, dietary intake and serum level of antioxidant vitamins, and serum MDA level were collected from eighty-five postmenopausal women. Mean serum MDA level was 1.62$\pm$0.03nmol/ml, and general characteristics and serum lipid profiles were not significantly different among the three group: low MDA(<1.45nmol/ml), midium MDA(1.45-1.74nmol/ml) and high MDA($\geq$1.74nmol/ml). Total mean vitamin A intake was 472.8$\pm$37.7RE, 68% of RDA, vitamin C intake was 134.3$\pm$8.7mg, 192% of RDA and vitamin E intake was 8.6$\pm$0.5mg, 86% of RDA for Korean. In takes of antioxidant vitamins from the diet were not significantly different among the three groups. However significant negative correlation(r=-0.242, p<0.05) was observed between vitamin E intake and serum MDA level in total subjects. Total mean serum vitamin A, $\beta$-carotene and vitamin C level were 0.59$\pm$0.01$\mu\textrm{g}$/ml, 0.25$\pm$0.01$\mu\textrm{g}$/ml and 9.02$\pm$0.28$\mu\textrm{g}$/ml, respectively. Total mean serum vitamin E and vitamin E/total cholesterol level were 9.15$\pm$0.42$\mu\textrm{g}$/ml and 4.09$\pm$0.17$\mu\textrm{g}$/mg, respectively. Serum antioxidant vitamins levels were not significantly different among the three groups. However serum vitamin C and E level were negatively correlated to serum MDA level. We can conclude that it will be helpful for postmenopausal women with common occurance of CVD to improve nutritional status of antioxidant vitamins by increasing intakes of antioxidant vitamins, especially vitamin C and E. Effects of Ethanol Extract of Onion on the Lipid Profiles in Patients with Hypercholesterolemia (양파의 알코올 추출물이 고콜레스테롤혈증 환자의 지질 성상에 미치는 영향)

  • It is known that onion increases antioxidative and antibiotic capacity, and decreases blood pressure and cholesterol levels. The study was carried out to investigate whether ethanol extract of onion can enhance blood lipid profiles in hypercholesterolemic patients. Foully three hypercholesterolemic patients were randomly divided into 2 groups; 25 in controls and 18 in experimental group. The control group was offered to take placebo for 4 weeks (200 ml/day), and experimental group was offered to consume ethanol extract of onion for 8 weeks (200 ml/day that corresponds to 500 g of onion). There were no differences in sex, age, body mass index (BMI) and educational levels between groups before treatment. After 8 weeks of onion extract consumption, plasma triglycerides level was significantly decreased in experimental group compared to control (p<0.05). Total cholesterol level was tended to decrease in experimental group, but there was no significant difference of change between control and experimental group. The levels of plasma glucose and LDL-cholesterol were not changed significantly after onion extract consumption. In summary, alcohol extract of onion may have a positive effect on the lipid profiles of hypercholesterolemia patients through decreasing plasma triglyceride concentration.

Identification of Single Nucleotide Polymorphisms (SNPs) of the Bovine Growth Hormone (bGH) Gene Associated with Growth and Carcass Traits in Hanwoo

  • The purpose of this study was to find any association of the bovine growth hormone (bGH) gene with growth and carcass quality traits in Korean native cattle, Hanwoo. Genomic DNA was extracted from 21 Hanwoo individuals, and the 47 to 2,528 bp region of the bGH 2,856 bp (GenBank accession number M57764) including the promoter and the five exons was sequenced. A total of ten bGH SNPs were confirmed, including four (253 C>T, 303 C>T, 502 C>T, and 559 G>A) in the promoter, one (679 C>T) in exon 1, one (1,692 T>C) in intron 3, and four (2141 C>G, 2258 C>T, 2277 C>T, and 2291 A>C) in exon 5. The ten bGH SNPs were genotyped for a sample of 242 Hanwoo steers and association tests were performed to find any significant SNP that was correlated with growth and carcass quality. Of the SNPs, the 303 C>T SNP in the promoter region was significantly associated with 6-month-old weight, the 559 G>A SNP with longissimus dorsi muscle area, the 2141 C>G SNP in exon 5 with daily weight gain, and the 2258 C>T SNP with daily weight gain and carcass weight (p<0.05). The significant SNPs need to be verified in other Hanwoo populations before considering implementation of marker-assisted selection for genetic improvement of growth and carcass quality in Hanwoo.

Measuring Stages of Change, Perceived Barriers and Self efficacy for Physical Activity in Saudi Arabia

  • Background: The benefits of physical activity are well established and recognized to prevent adults from many chronic diseases and particularly some forms of cancers. Objectives: The present study was conducted to investigate the present status of physical activity among Saudi adults in Al-Ahsa, and to examine the association between the stages of change for physical activity and perceived barriers, and self efficacy. Methods: A cross-sectional study of 242 subjects (118 males and 124 females) attending health centers aged between 20-56 years, were personally interviewed for demographic data, anthropometric measurement, physical activity level, stages of change for physical activity, self efficacy and perceived barriers. Results: Forty eight percent of the females were overweight and 16.9% of the males were obese with no significant difference between the genders for BMI categories. More than half of the females were inactive and 39% of the males were physically active with a significant difference (P=0.007). Twenty percent of the males were in maintenance stage, while similar percentage of the females were in contemplation stage. However the majority of the subjects were in pre-contemplation stage with a significant difference across the stages. Males had a higher mean score of self efficacy and less external barriers of physical activity. The major barrier among the females was lack of time ($7.2{\pm}1.4$) and in the males, lack of motivation ($7.7{\pm}1.4$). The females had less internal ($21.2{\pm}3.8$) barriers comparable to the males ($23.08{\pm}4.7$). Both genders had a significant relationship between stages of changes of physical activity and perceived barriers (internal and external), but in the females no significant difference across the stages was observed for self efficacy unlike the males who had a significant difference for self efficacy and self efficacy categories. Conclusion: The present study provided useful data on stages of change for physical activity and some psychosocial factors (self efficacy and perceived barriers) that can help to tailor strategies aiming at increasing physical activity level according to self efficacy and to the barriers detected, to prevent many chronic diseases including certain types of cancer in Saudi Arabia.

The Relationship between Lipid Peroxidation of Red Blood Cell Membrane, and Mean Corpuscular Volume and Liver Enzyme Markers in Alcohol Dependence Patients (알코올 의존 환자의 적혈구막 지질과산화 정도와 적혈구 평균 용적 및 간 효소 지표와의 관련성)

  • Objectives : Alcohol-induced oxidative stress has been known to injure various tissues or organs. This stress is related with free radicals which are produced as the result of long-term alcohol consumption. Malonyldialdehyde(MDA) is produced by the interaction of free radicals and cell membrane lipids, and indicates the degree of lipid peroxidation indirectly. The purpose of this study was to investigate the relationship between red blood cell(RBC) membrane lipid peroxidation by free radicals, and associated hepatic injuries and hematologic changes. Methods : Thirty-three subjects diagnosed as alcohol dependence according to DSM-IV diagnostic criteria were evaluated within 72 hours after discontinuing alcohol drinking. Clinical characteristics were evaluated by CAGE questionnaire and Korean Michigan Alcoholism Screening Test(MAST). RBC membrane MDA level was measured as the marker of RBC membrane lipid peroxidation. Aspartate aminotransferase(AST), alanine aminotransferase(ALT) and gamma-glutamyltransferase(GGT) were used as the biochemical markers of liver damage due to alcohol ingestion. The alcohol-induced hematologic change was assessed by mean corpuscular volume(MCV). Results : The results were as follows. Clinical characteristics were not different between two groups having normal and abnormal levels of AST, ALT, GGT or MCV. The levels of MDA were not correlated with the clinical characteristics and serum levels of AST, ALT and GGT. However, there was a significant correlation between the levels of MDA and the value of MCV(p=0.017). Conclusions : These findings suggest that oxidative stress in alcohol dependence may not be reflected in liver enzyme markers such as AST, ALT and GGT, but may be reflected in MCV.
