ITS에 의한 한국 내 Pelvetia속 분류군의 계통학적 연구 (Phylogeny Study of Genus Pelvetia in Korea by Internal Transcribed Spacer Sequence (ITS))

  • 갈조류 모자반과 Pelvetia속은 다세포 조류로 많은 해조류를 포함하고 있으며 북반구 태평양과 대서양에 분포되어 있다. Pelvetia속에 속하는 우리나라의 동종 및 근연한 같은 속내 종에 대해 유전자은행을 통해 밝혀진 ITS에 의한 서열을 이용하여 계통관계를 조사하였다. 한국의 P. babingtonii은 북미의 P. babingtonii AF102957과 유사한 핵산서열로 계통도 분석에서 같은 분지군을 형성하였다. 한국의 P. siliquosa은 역시 북미의 P. siliquosa AF102958과 유사한 핵산서열로 계통도 분석에서 같은 분지군을 형성하였다. Pelvetia 속 내 여러 종간은 결실이나 삽입에 의한 indel이 많은 반면 같은 종내 계통은 치환에 의한 차이가 현저하였다. 이 속은 NJ분석에서 크게 두 분지군으로 나뉘는데 한 그룹은 P. canaliculata와 P. limitata이며 나머지 그룹은 P. siliquosa, P. compressa, P. babingtonii을 포함하고 있었다. ITS 서열로 한국 내 분류군과 북미 간 분류군이 잘 구분되었다. MP 분석에서도 높은 지지도로 잘 구분되었다. 따라서 ITS 서열로 종 동정에 이용할 수 있었으며, 종의 보전이나 생식질 보전에 기초로 이용될 수 있을 것으로 사료된다.

  • Fucosterol isolated from Pelvetia siliquosa was tested for its anti-diabetic activity in vivo. Fucosterol, when administered orally at 30 mg/kg in streptozotocin-induced diabetic rats, was caused a significant decrease in serum glucose concentrations, and exhibited an inhibition of sorbitol accumulations in the lenses. Fucosterol, when administered orally at 300 mg/kg in epinephrine-induced diabetic rats, was also caused an inhibition of blood glucose level and glycogen degradation. These results demonstrated that fucosterol is a main anti-diabetic principle from the marine algae P. siliquosa.

  • A new method for the quantitative determination of fucosterol in the marine algae Pelvetia siliquosa by gas chromatography was established. A HP-1 column programmed as $200^{\circ}C\;(5\;min)\;{\rightarrow}\;5/min\;{\rightarrow}\;280^{\circ}C\;(10\;min)$ was developed. The analysis of fucosterol in P. siliquosa was successfully carried out within 30 min.

Isolation of Fucosterol from Pelvetia siliquosa by High-speed Countercurrent Chromatography

  • We report here the use of high-speed countercurrent chromatography (HSCCC) in the preparative isolation and purification of the bioactive component, fucosterol, from Pelvetia siliquosa. A crude extract was obtained by ultrasonic extraction of powdered P. siliquosa using methylene chloride and was then subjected to separation and purification by HSCCC, coupled with evaporative light-scattering detection. Preparative HSCCC was performed successfully using a two-phase solvent system, n-heptane:methanol (3:2, v/v), to obtain 10.96 mg fucosterol with 96.8% purity from 50 mg of crude extract; the recovery rate was approximately 90.5%.

국내 자생 해조류 추출물의 항산화능 비교분석 연구 (Comparative Analysis of Antioxidant Activity of Korean Seaweeds Extracts)

  • Seaweed-derived foods have long been popular in Korea because of their high content of nutrients that are beneficial to the human body. Recently, Korean seaweeds have been used as raw materials to produce new natural products with health benefits. Herein, we compared the antioxidant activity of 16 Korean seaweed extracts to explore their potential utility as health foods. The total phenolic content (TPC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of seaweed extracts were determined. We also investigated their ability to protect human diploid fibroblast (HDF) cells against hydrogen peroxide. The results showed that seaweed extracts at a concentration of 100 ㎍/mL did not cause any cell toxicity. Sargassum thunbergii (Jichung-i) had the highest TPC and radical scavenging effects, followed by Porphyra tenera (Gim), Silvetia siliquosa (Tteumbugi), and Sargassum fusiforme (Tot). Hydrogen peroxide increased the production of intracellular reactive oxygen species, while P. tenera (Gim), Saccharina japonica (Dasima), and S. thunbergii (Jichung-i) extracts significantly decreased it. The effect was highest in the S. thunbergii (Jichung-i)-treated HDF cells. These findings indicate that S. thunbergii (Jichung-i) shows promise as a potential antioxidant raw material.
