Immunostimulating effects of lactic acid bacteria as biological response modifier is a subject of growing interest, but the knowledge of these focused on some bacteria as Lactobacillus and Bifidobacterium. In this study, we investigated the effects of Pediococcus pentosaceus EROM101 on the immunostimulating and anti-cancer activity in murine model. P. pentosaceus was mainly found in Kimchi and fermented sea food and is facultatively anaerobic, catalase-netative, gram-positive cocci arranged in pairs, tetrads and clusters. The immunostimulating effects of P. pentosaceus EROM101 were evaluated using IgA production assay of Peyer's patch and proliferation assay of exudated immune cells of Balb/C mice fed P. pentosaceus EROM101 for 3 weeks. The macrophage and splenocyte proliferation were enhanced by orally administrated of P. pentosaceus EROM101. Also, IgA production in Peyer's patch increased by P. pentosaceus EROM101. Anti-cancer activity of P. pentosaceus EROM101 was appeared in Sarcoma 180 tumor-bearing ICR mice. However, this bacterium lysate itself appeared to have noncytotoxic substance against Sarcoma 180 cell in vitro. These results suggested that P. pentosaceus EROM101 reinforce immune system and therefore was revealed to be anti-cancer activity in mice.
The purpose of this study is to examine the physiological characteristics and anti-diabetic effects of Pediococcus pentosaceus KI62. The α-amylase and α-glucosidase inhibitory activity of P. pentosaceus KI62 was 94.86±3.30% and 98.59±0.52%, respectively. In MRS broth containing 3% maltodextrin inoculated by P. pentosaceus KI62, the amounts of short chain fatty acids (SCFA) were propionic acid 18.05±1.85 mg/kg, acetic acid 1.12±0.07 g/100 mL, and butyric acid 2.19±0.061 g/kg, and those of medium chain fatty acids (MCFA) were C8 0.262±0.031 mg/kg, C10 0.279±0.021 mg/kg, and C12 0.203±0.009 mg/kg. Compared to sixteen antibiotics, P. pentosaceus KI62 had the highest sensitivity to penicillin-G and rifampicin, as well as the highest resistance to vancomycin and ampicillin. The strain also showed higher leucine arylamidase and valine arylamidase activities than other enzyme activities, but it did not produce β-glucuronidase which is carcinogenic enzymes. The survival rate of P. pentosaceus KI62 in 0.3% bile was 91.67%. Moreover, the strain showed a 98.63% survival rate in pH 2.0. P. pentosaceus KI62 exhibits resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus at rates of 29.41%, 38.10%, 51.72%, and 50.47%, respectively. P. pentosaceus (23.31%) showed a similar adhesion ability to L. rhamnosus GG, the positive control (24.49%). These results show that P. pentosaceus KI62 has possibility as a probiotic with anti-diabetic effects.
In this study, we investigated the mechanism of inhibition of pathogenic bacteria by Pediococcus pentosaceus strain SH-10 isolated from hard Clam Meretrix meretrix sikhae. When P. pentosaceus SH-10 was co-cultured in MRS broth with pathogenic bacteria, including Bacillus cereus, Listeria monocytogenes, Salmonella choleraesuis and Staphyloccus aureus, no viable pathogenic cells were detected after 18 h of incubation. However, pediocin or a pediocin-like bacteriocin was not detected in cultures of P. pentosaceus SH-10 by the agar diffusion method. Organic acids were produced in MRS broth in proportion to the incubation time of P. pentosaceus SH-10. These results indicate that P. pentosaceus SH-10 inhibited the growth of pathogenic bacteria by lowering the pH of the growth medium through the production of organic acids, including sodium lactate, sodium acetate, and sodium citrate.
Lactic acid bacteria are generally recognized as beneficial probiotic organisms. Recent studies revealed that the potential of probiotic strains was essentially dependent on the bacterial-binding and adhesion capabilities to gut epithelial cells and the hydrophobicity of the cell surface. In this study, we screened some indigenous lactic acid bacteria from Kimchi and selected one lactic acid bacterium as a potential probiotic based on its cell surface hydrophobicity. Analysis of the 16S rRNA gene sequences of probiotic isolates indicated that the selected isolate (BCNU 9070 strain) was a member of Pediococcus pentosaceus. P. pentosaceus BCNU 9070 showed resistance to bile acids and acidic pH. The P. pentosaceus BCNU 9070 strain also inhibited the cell growth of six food-borne pathogens including Listeria monocytogenes and Shigella sonnei. In addition, the P. pentosaceus BCNU 9070 strain expressed bile salt hydrolase activity and showed an ability to assimilate cholesterol in vitro. On the basis of these results, P. pentosaceus BCNU 9070 is considered to have probiotic potential for applications in functional foodstuffs.

The antifungal activity against cheese fungi by lactic acid bacteria isolated from kimchi was investigated. Eight fungi were isolated from cheese in the cheese ripening room. Two of them were identified as Penicillium and Cladosporium via ITS-5.8S rDNA analysis. Twenty-two species of lactic acid bacteria with antifungal activity were isolated from kimchi. Two of them were identified as Lactobacillus and Pediococcus via 16S rRNA sequence analysis. Of the 22 lactic acid bacteria species, six were selected (L. sakei subsp. ALJ011, L. sakei subsp. ALI033, L. sakei subsp. ALGy039, P. pentosaceus ALJ015, P. pentosaceus ALJ024 and P. pentosaceus ALJ026) due to their higher activity against the eight fungi isolated from cheese in the cheese ripening room; and among the six species, the P. pentosaceus ALJ015 and P. pentosaceus ALJ024 isolates from the Jeonju area kimchi and the L. sakei subsp. ALI033 isolate from the Iimsil area kimchi had higher antifungal activity than the other lactic acid bacteria. The minimum inhibitory concentration (MIC) of L. sakei subsp. ALI033 against the eight fungi isolated from cheese in the cheese ripening room was $62.5{\mu}g/mL$.
This study examined the suitability of characteristics of potential strains of probiotic bacteria. Among 25 lactic acid bacteria isolated from Korean traditional fermented food, the Hard Clam Meretrix meretrix Shikhae, the SH-10 strain, which exhibited superior resistance to low pH and bile salts, was selected as a potential probiotic bacteria. By examining carbohydrate utilization, morphological properties, and the 16S rRNA gene sequence, the SH-10 strain was identified as Pediococcus pentosaceus (hereafter, P. pentosaceus SH-10). P. pentosaceus SH-10 was resistant to amikacin, cefotetan, ciprofloxacin, gentamicin, kanamycin, nalidixic acid, streptomycin, and vancomycin. Tests of antimicrobial activities against pathogens such as Bacillus cereus, Listeria monocytogenes, Salmonella choleraesuis, and Staphylococcus aureus, indicated that P. pentosaceus SH-10 inhibited the growth of pathogenic bacteria. These results suggest that P. pentosaceus SH-10 can be developed as a probiotic bacteria.
Two lactic strains, Leuconostoc mesenteroides Lu5 and Pediococcus pentosaceus P1 isolated from Kimchi, were used to determine the optimum conditions for protoplast formation and regeneration. The maximum protoplast formation rate was obtained with both strains at early exponential growth phase and decreased rapidly during growth phase. For P. pentosaceus P1, 30 $\mu$g/ml of lysozyme treatment was sufficient to obtain over 90% of protoplast formation and 300 $\mu$g/ml for L. mesenteroides Lu5, showing great difference in sensitivity of these strains to lysozyme. For both strains, best results were obtained at pH 7, using 0.5 M sucrose as osmotic stabilizer. For regeneration of protoplast, the highest regeneration rate was obtained after 15 minutes of lysozyme treatment and declined drastically with prolonged digestion.
Pediococcus pentosaceus ID-7 was isolated from kimchi, a Korean fermented food, and it showed high activity for lactose hydrolysis. The ${\beta}$-galactosidase of P. pentosaceus ID-7 belongs to the GH2 group, which is composed of two distinct proteins. The heterodimeric LacLM type of ${\beta}$-galactosidase found in P. pentosaceus ID-7 consists of two genes partially overlapped, lacL and lacM encoding LacL (72.2 kDa) and LacM (35.4 kDa). In this study, Escherichia coli MM294 was used for the production of LacL, LacM, and LacLM. These three types of recombinant proteins were expressed, purified, and characterized. The specific activities of LacLM and LacL were 339 and 31 U/mg, respectively. However, activity was not detected with LacM alone. The optimal pH of LacLM and LacL was pH 7.5 and pH 7.0, and the optimal temperature of LacLM and LacL was $40^{\circ}C$ and $50^{\circ}C$, respectively. The optimal temperature changes indicate that LacLM is able to achieve higher activity at a relatively lower temperature. LacLM was strongly activated by $Mg^{2+}$, $Mn^{2+}$, and $Zn^{2+}$, which was not true for LacL. Consistent with this, EDTA strongly inactivated LacLM and LacL, but the presence of reducing agents did not dramatically alter the activity. Taken together, multiple alignment of amino acid sequences and phylogenetic analysis results of LacL and LacM of P. pentosaceus ID-7 suggest the evolution of LacL into LacLM and that the use of divalent metal ions results in higher activity.
A lactic acid bacterial strain resistant to several antibiotics such as oxytetracycline, tylosin, neomycin and sulfathiazole, which have been often used as a therapeutic agent in livestock, was isolated from the porcine gastrointestinal tract. The isolate YB-55 was identified as belonging to the genus Pediococcus with the highest similarity to P. pentosaceus on the basis of its 16S rRNA sequence and biochemical properties. The isolated strain showed viability of over 85% at pH 3.0 and was resistant to bile salt. The strain produced lactic acid of 12.3 g/L by jar fermentation and maintained its viability in the presence of antibiotics at dosage related therapeutic effect, suggesting P. pentosaceus YB-55 may of ffr potential as a probiotics for livestock.
Three lactic acid bacteria (C-1 K-3 and T-1 strain) were isolated from Nuruk and characterized subsequently. They were useful strains for production of lactic acid and their growth was inhibited at 10% ethanol pH 4 These strains were identified as lactococcus lactis subsp. lactis NR C-1 Leuconostoc mesenteroides subsp. mesenterides NR K-3 and pediococcus pentosaceus NR T-1 respectively by morphological physiological and biochemical characterization Lac lactis subsp lactis NR C-1 showed the highest lactic acid productivity. Leu measenteroides subsp mesenteroides NR K-3 showed stable lactic acid productivity and its growth was inhibited at pH 4. P pentosaceus NR T-1 had lower lactic acid productivity than the other two bacteria but it could not grow at 10% ethanol pH 4 The lactic acid productivity of these three strains in MRS broth were higher than that in Skim milk media the optimum pH and temperature for the lactic acid production of the three strains were 30-32$^{\circ}C$ and pH 6.0∼6.8 Glucose was the optimal carbon souorce for the lactic acid production. In terms of antagonism lac lactis subsp lactis NR c-1 showed somewhat inhibitory efects against some Gram positive rod and cocci such as Lactobacillus brevis and Streptococcus mitis. And Leu mesenteroides subsp mesenteroides NR K-3 showed the inhibitory effects against Streptococcus mitis but P. pentosaceus NR T-1 didn't show any inhibitory effects against tested strains.
