Isolation and Identification of an acidoduric Streptomyces sp. from Forest Soil (산림토양으로 부터 내산성 streptomyces sp. 균주의 분리 및 동정)

  • In this study, an acidoduric Streptomyces strain was isolated and identified from acidic forest soil around Dankook University, Cheonan Campus. This isolated strain had rod-shaped, smooth, non-motile spore and the shape of spore chain was compact spiral. This structure appeared similar to the sporangium of the genus Streptosporangium but this strain proved to be a Streptomyces strain by an electron microscopic study and cell wall analysis. This strain showed a best growth on neutral medium, was also able to grow on the acidic media of pH 4.0 and pH 5.0. The color determination of this strain on various agar media and other physiological tests were carried out by ISP-methods. From these results, the isolated strain was considered to be Streptomyces mirabilis.

  • Seven white rot fungi (Irpex lactenis, Coriolus hirsutus, Lopharia mirabilis, Schizopora paradoxa, Ganoderma lucidum, Pleurotus ostreatus and Pycnoporus coccineus) native to Korea and two famous exotic lignin degradable white rot fungi (Coriolus versicolor and Phanerochaete chrysosporium) were investigated to clarify their physiological and physicochemical characteristics on white-rotted wood blocks. G. lucidum degraded wood blocks more seriously than those by exotic lignin-degrading fungi, C. versicolor and P. chrysosporium, but only slightly decreasecl the strength of wood which was compared to the weight loss, persumably on the account of its small use of cellulose when attacking wood. It is quite interesting to note that the holocellulose degradation rate of G. lucidum was also higher than any of the other tested fungi. The order of fungi, according to the lignin-decomposing rates, was G. lucidum>P. coccineus>C. versicolor>S. paradoxa>P. chrysosporium>L. mirabilis>P. ostreatus>C. hirsutus>I. lactenis. The lignin degradation of G. lucidum and P. coccineus which were collected in Korea was greater than that of C. versicolor and P. chrysosporium. If holocellulose degradation is not considered. G. lcidum has the merit of actual application in biomass conversion due to linin removal.

Purification and Properties of a Cysteinylglycinase from Proteus mirabilis (Proteus mirabilis가 생산하는 Cysteinylglycinase의 정제 및 성질)

  • Cysteinylglycinase was partially purified from Proteus mirabilis by consecutive procedure. The specific activity was increased about 16-fold to that of cell-free extract. The enzyme was found rather unstable on ammonium sulfate precipitation ann the precipitated enzyme protein became partially insoluble during dialysis. The precipitated enzyme was found to be solubilized by treatment of 4% Triton X-100 effectiviely, The optimum temperature and pH of the enzyme activity were 35$^{\circ}C$ and 7.3, respectively. After heat treatment of the enzyme at 5$0^{\circ}C$ for 30 min, it lost the activity to 70%. The enzyme was stable at pH 7.0-8.0. The molecular weight of the cysteinylglycinase was found to be about 190,000 by Sephadex G-150 gel filtration. The enzyme was activated by the addition of Mn$^{2+}$ and $Mg^{2+}$ ions. The maximal activation was obtained in preincubation with $Mg^{2+}$ ion for 30 min. The enzyme catalyzed the hydrolysis of various dipeptides and tripeptides. The Km and Vmax values for cysteinylglycine were 1.60 mM and 0.24 m unit/ mg, respectively.

  • 세균내 glutathione의 동태를 연구하기 위한 일환으로 P. mirabilis로부터 cysteinylglycine 분해효소를 정제 검토하였다. 본 균이 생산하는 cysteinylglycine 분해효소의 정제는 무세포추출액에 비해 비활성이 10배 증가하였고 0.68%의 낮은 수율을 나타내었다. 본 호소는 (NH$_4$)$_2$ SO$_4$ 침전과정에서 활성을 크게 손실하는 등 정제과정에서 불안정하였으며 투석 중에 형성하는 불용성 침전물은 4% Trition X-100 처리에 의해 효과적으로 용해되었다.

  • The nucleotide sequence of inducible chloramphenicol acetyl-transferase(CAT) gene isolated from a small plasmid pSBK203 of Staphylococcus aureus was determined. The base sequence shows that structural gene of pSBK203-CAT encodes a protein of 213 amino acids and has a leader region which encodes a short polypeptide of 9 amino-acids in its upstream. vertical bar /sup 35/S vertical bar-Methionine labelled CAT gene product in minicell showed almost same mobility with pC194-CAT of which molecular weight is 24Kdal on polyacrylamide gel electrophoresis. Predicted amino acid sequence of pSBK203-CAT has revealed a high degree of homology with the CATs of pC194 and pC221 than those of cat-86, Tn9 and proteus mirabilis PM13.

In-Vitro, Anti-Bacterial Activities of Aqueous Extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo Against Gram Positive and Gram Negative Bacteria

  • Objective: Evaluations of the in-vitro anti-bacterial activities of aqueous extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and Shilajita mumiyo against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) and gram-negative bacteria (Escherichia coli, klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa) are reasonable since these ethnomedicinal plants have been used in Persian folk medicine for treating skin diseases, venereal diseases, respiratory problems and nervous disorders for ages. Methods: The well diffusion method (KB testing) with a concentration of $250{\mu}g/disc$ was used for evaluating the minimal inhibitory concentrations (MIC). Maximum synergistic effects of different combinations of components were also observed. Results: A particular combination of Acacia catechu (L.F.) Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo extracts possesses an outstanding anti-bacterial activity. It's inhibiting effect on microorganisms is significant when compared to the control group (P<0.05). Staphylococcus aureus was the most sensitive microorganism. The highest anti-bacterial activity against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) or gram-negative bacteria (Escherichia coli, Klebsiella pneumonia, Proteus mirabilis, and Pseudomonas aeruginosa) was exerted by formula number 2 (table 1). Conclusion: The results reveal the presence of anti-bacterial activities of Acacia catechu, Castanea sativa husk, Ephedra sp. and Mumiyo against gram-positive and gram-negative bacteria. Synergistic effects in a combined formula, especially in formula number 2 (ASLAN$^{(R)}$) can lead to potential sources of new antiseptic agents for treatment of acute or chronic skin ulcers. These results considering the significant anti-bacterial effect of the present formulation, support ethnopharmacological uses against diarrheal and venereal diseases and demonstrate use of these plants to treat infectious diseases.

  • Out of 826 isolates of Streptomyces isolated from different soils, 65 isolates were selected and identified according to their morphological and physiological characters. A total of 46 species of Streptomyces were identified, and the following 38 species were newly recorded in Korea. 1) Melanoid pigments producing group; S. lavendulae, S. purpurascens, S. violarus, S. lateritius, S. olivochromogenes, S. purpeofuscus, S. fulvoviolaceus, S. gallilaeus, S. naganishii, S. hygroscopicus subsp. ossamyceticus, S. nevagawaensis, S. anandii, S. phaeopurpureus, S. mirabilis, S. arenae, S. massasporeus, S. eurythermus, S. tuirus. 2) Melanoid pigments non-producing group; S. fellus, S. flavidovirens, S. exfoliatus, S. fumanus, S. termitum, S. glomeroaurantiacus, S. luteofluorescens, S. prunicolor, S. fradiae, S. tauricus, S. griseolus, S. misakiensis, S. poonensis, S. antimycoticus, S. diastaticus subsp. ardesiacus, S. nigrifaciens, S. tendae, S. narbonensis subsp. josamvceticus, S. atroolivaceus, S. chrysomallus subsp. fumigatus.

Studies on Proteus Spp Isolated from Clinical Sources (임상검체에서 분리된 Proteus Spp에 관한 고찰)

  • Seven hundred and forty eight strains of proteus spp were isolated from clinical sources at National Medical Center during recent five years(1968 to 1972). 1. These strains identified as follows: P. mirabilis 357 strains(47.7%) P. vulgaris 140 strains(18.7%) P. morganii 134 strains(17.9%) P. rettgeri 63 strains(8.4%) P. inconstans 54 strains(7.2%) 2. About the incidence between both sexes, male is higher than female. 3. Total numbers of the age group from $20{\sim}29$ years, $30{\sim}39$ years, and $40{\sim}49$ years old respectively are higher than total numbers from other age groups. 4. Seasonal trend shows hot season e.g. August to September is more prevalent. 5. Correlation of strain source with clinical department shows the highest incidence of general surgery next to this, orthopedic surgery, chest surgery, internal medicine, neurosurgery, ENT and pediatrics etc. in descending order. 6. Correlation of strain source with clinical material shows the highest incidence of pus next to this urine, stool, sputum etc. in descending order. 7. The rate of susceptibility to several antibiotics is low in general except gentamicin(82.7%) and carbenicillin(72.9%). However, in case of gentamicin sensitive strains are 15(9.6%) and moderately sensitive strains are 114(73.1%).

Characterization of Proteus vulgaris K80 Lipase Immobilized on Amine-Terminated Magnetic Microparticles

  • Proteus vulgaris K80 lipase was expressed in Escherichia coli BL21 (DE3) cells and immobilized on amine-terminated magnetic microparticles (Mag-MPs). The immobilization yield and activity retention were 84.15% and 7.87%, respectively. A homology model of lipase K80 was constructed using P. mirabilis lipase as the template. Many lysine residues were located on the protein surface, remote from active sites. The biochemical characteristics of immobilized lipase K80 were compared with the soluble free form of lipase K80. The optimum temperature of K80-Mag-MPs was $60^{\circ}C$, which was $20^{\circ}C$ higher than that of the soluble form. K80-Mag-MPs also tended to be more stable than the soluble form at elevated temperatures and a broad range of pH. K80-Mag-MP maintained its stable form at up to $40^{\circ}C$ and in a pH range of 5.0-10.0, whereas soluble K80 maintained its activity up to $35^{\circ}C$ and pH 6.0-10.0. K80-Mag-MPs had broader substrate specificity compared with that of soluble K80. K80-Mag-MPs showed about 80% residual relative activity after five recovery trials. These results indicate the potential benefit of K80-Mag-MPs as a biocatalyst in various industries.

Evaluation of Urease Inhibition Activity of Zerumbone in vitro (제럼본의 요소가수분해효소 활성 억제 평가)

  • A key virulence factor for urinary tract pathogens is the enzyme urease, which catalyzes the hydrolysis of urea into ammonium ions and carbonic acid. Urease activity plays an important role in the pathogenesis of urinary tract infection. In this study, the inhibitory effect of zerumbone against six urease-producing bacteria (Klebsiella oxytoca, K. pneumoniae, Morganella morganii, Proteus mirabilis, P. vulgaris, and Staphylococcus saprophyticus) and their urease activities were evaluated. The results of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests showed that zerumbone had antibacterial effect against these six urease-producing bacteria. The MIC and MBC of zerumbone ranged from 0.5 to 2 mM and 1 to 4 mM, respectively. In the urease inhibitory assay, zerumbone showed better urease inhibition ($56.28{\pm}2.45-37.83{\pm}3.47%$) than the standard urease inhibitor, acetohydroxamic acid ($40.46{\pm}1.94-22.99{\pm}3.53%$). However, zerumbone did not affect the levels of the urease subunit. These results clearly indicated that zerumbone has antibacterial potential against urease-producing bacteria and possesses excellent bacterial urease inhibition properties.
