센텔라아시아티카정량추출물의 리포좀 캡슐기술 개발과 피부개선효과 (Skin Improvement Effects and Development of Liposome Capsule Technology Using Centella Asiatica Extract Powder)

  • 본 연구에서는 의약품과 화장품에 사용되는 고순도의 센텔라아시아티카정량추출물을 리포좀화하는 방법과 이를 이용한 크림을 개발하여 주름개선에 대한 in-vivo 임상평가에 대하여 연구한 결과를 보고한다. 리포좀 라멜라 베지클에 센텔라아사아티카정량추출물을 안정화시킨 센텔라좀-10EX을 만들기 위하여, 5%의 하이드로제네이티드레시틴과 2%의 수크로오스다이스테아레이트를 사용하여 완성시킬 수 있었다. 센텔라좀-10EX의 외관은 저점성의 크림상, 색상은 엷은 황색, 냄새는 원료 본연의 고유취가 있었다. pH는 약 6.12 이었으며, 비중은 1.09, 산가는 약 0.35이었다. 리포좀 베지클에 함유된 센텔라아시아티카정량추출물의 주성분의 함량은 10,800ppm의 아시아틱애씨드, 10,900ppm의 아시아티코사이드, 6,000ppm의 마테카식애씨드, 1,600ppm의 마데카소사이드가 함유하고 있으며, 장기간 보관 시에도 변색이 없었고, 열역학적으로 주성분이 안정하게 유지되고 있음을 알 수 있었다. 센텔라좀-10EX의 리포좀 베지클의 구조를 메커니즘 적으로 분석하기 위하여, 투과전자현미경 (Cryo-TEM)으로 관찰한 결과 다중층막이 형성되어 수분이 채워져 있고, 그 주변에는 10~60여층의 다중층의 리포좀 라멜라 베지클 형성되었음을 확인하였다. 임상시험(in-vivo)으로써, 센텔라좀 크림의 5주동안 바른 후의 보습효과는 28.3%로 플라세보 보다 유의차 있게 증가되었다. 피부탄력효과는 13.6%로 플라세보 보다 유의차 있게 보습력이 증가되었다. 눈가의 피부잔주름 개선효과는 플라세보 크림보다 23.52%가 개선되었다. 이 연구 결과를 통하여 개발된 센텔라좀-10EX의 리포좀 베지클에 봉입 안정화하는 처방과 제조방법에 대하여 연구할 수 있었으며, 이를 이용한 크림에 대한 주름개선효과 등의 임상연구를 통하여 얻어진 결과들이 화장품 산업에 있어서 피부과학을 연구하고, 고 효능을 가진 고품격 화장품을 개발하는데 폭넓게 활용될 수 있을 것으로 기대한다.

Overexpression of Anti-apoptotic Molecules and Sax Translocation to Mitochondria by Pharbitis Nil Extracts in AGS

  • Conventional medicines have usually sorted to a number of treatments such asoperation, radiotherapy, and chemotherapy. The existing anti-cancer agents, designed to eradicate cancer cells, have strong toxicities, also with leading to harmful side effects. Recently, a number of researches on natural products have been actively carried out in efforts to develop new treatments that can decrease side effects or increase anti-cancer effects. We performed this study to understand the molecular basis underlying the antitumor effects of Pharbitis nil, and Plantago asiatica, which have been used for herbal medicinal treatments against cancers in East Asia. We analyzed the effects of these medicinal herbs on proliferation and on expression of cell growth/apoptosis related molecules, with using an AGS gastric cancer cell line. The treatment of Pharbitis nil dramatically reduced cell viabilities in a dose and time-dependent manner, but Plantago asiatica didn't. FACS analysis and Annexin V staining assay also showed that Pharbitis nil induce apoptotic cell death of AGS. Expression analyses via RT-PCR and Western blots revealed that Pharbitis nil didn't increase expression of the p53 and its downstream effector p21/sup wafl/, and that the both increased expression of apoptosis related Sax and cleavage of active caspase-3 protein. We also confirmed the translocation of Sax to mitochondria. Collectively, our data demonstrate that Pharbitis nilinduce growth inhibition and apoptosis of human gastric cancer cells, and these effects are correlated with down- and up-regulation of growth-regulating apoptotic and tumor suppressor genes, respectively.

국내산 향신료로 제조한 발효소시지의 특성 (Characteristics of Fermented Sausages with korean Native Spices)

  • the objective of this study was to develop the Korean style fermented sausage by addition of Korean native spices(Mugwort Artemisaia asiatica ;pine needles pinus thunbergii ; Japanese pepper Zanth-oxylum prperitum ; Arrowroot pueraria hirsuta) instead of foreign-original spices(white pepper corian-der)pH of all sausages was lowered rapidly during ripening. pH reductio of sausages with foreign spices was mere rapid than that with Korean native spices except for the sausages with all mixtured Korean native spices. Aw was not affected by differentiated spices. The "a" value among Hunter Color Value of sausages that produced with mixed Korean native spices was higher than sausages with foreign spices Lactic acid bacteria rapidly increased and reached to 108cfu/g after 2 days of ripening. Korean native spices used to this work did not inhibit the inhibit of lactic acid bacteria in fermented sausages but enterobacteria did not detected after 10 days of fermentation in the sausages with mixed Korean native spices.

  • Effects of plantago-mucilage A (P-MA) on the immune responses were studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and P-MA at doses of 7, 21 and 63 mg/kg were orally administered to mice once a day for 21 consecutive days. Mice were immunized and challenged with sheep red blood cells (SRBC). P-MA at 63 mg/kg/day significantly increased the body weight gain and the relative weights of spleen and thymus, as compared with those in controls. However, there were no significant effects on liver weight due to P-MA treatment. Plaque forming cells (PFC) and hemagglutination (HA) titers to SRBC were significantly enhanced in mice dosed at 21 and 63 mg/kg/day P-MA, as compared with those in controls. Delayed-type hypersensitivity (DTH) reaction to SRBC, phagocyte activity and circulating leukocyte were also significantly increased in mice dosed at 63 mg/kg/day P-MA. These results demonstrate that P-MA markedly enhances both humoral immune and allergic reaction to SRBC at concentrations which don't act on the relative weight of liver.

Screening for Herbs Having Inhibitory Effect on Egg Hatching of Strongyloides venezuelensis

  • In vitro bioassays of aqueous extracts of 25 herbal species were performed by ovicidal activity against Strongvloides venuelensis. 500 mg of feces from Mongorian gerbil infected with S. venuelensis were placed into polyvinyl culture with extracts of herb and anthelmintic drugs in triplicates. Larval activity inside the polyvinyl bag was recorded microscopically at 12 hours, and every 24 hours thereafter for up to 72 hrs incubation. Eggs were not hatched within 72 hrs and degeneration was evident in the presence of extracts. The ovicidal activity characterized by the absence of L$_3$ in fecal cultures inside the polyvinyl bag. 1. Among 25 herbs, 12 herbs had ovicidal effects and caused degeneration of larvae in egg. S. angustifolia, I. helenium, A. korenum, P. tenuifolia, F. virridissima, A. asiatica, S. chinensis, A. tataricus, S. baicalensis, M. rimosa, S. glabra and Z. piperitum were the most ovicidal effect. However, P. thunbergiana, T. chinensis, C. flos, G. uralensis, C. obutusifolia, C. fortunei, A. koreana, P. grandiflorum, P. tenuifolia (Radix), P. mume and A. asphodelodes showed no ovicidal effects in vinyl bag culture. 2. Of 25 hrebs, 4 herbs had ovicidal and wormicidal effects, P. thunbergiana, C. flos, B. Striata and T. chinensis were characterized by degeneration of larvae.

  • Allelopathy 작용성(作用性)을 나타내는 4종(種)의 잡초(雜草) 중에 존재(存在)하는 휘발성(揮發性) terpene 물질(物質)을 GC 및 GC-MS로 동정(同定)하였다. 쑥에서는 26종(種)의 휘발성(揮發性) terpene 류(類)를 분리(分離) 동정(同定)하였는데 이들은 monoterpene 16종(種)과 sesquiterpene 10종(種)이었다. 또한 토끼풀에서는 4종(種), 쇠비름과 냉이에서 각(各) 3종(種)이 확인(確認)되었다. 동정(同定)된 terpene 중 allelopathy 작용성(作用性)을 나타내는 물질(物質)로는 쑥에서 ${\alpha}$-phellandrene, 1,8-cineole, limonene, ${\beta}$-pinene, borneol, selinene 및 caryophyllene, 토끼풀에서 ${\alpha}$-pinene과 ${\beta}$-caryophyllene, 그리고 냉이에서 ${\alpha}$-pinene 등이었고, 쇠비름 중에는 allelopathy 작용성(作用性)을 나타내는 휘발성(揮發性) terpene 류(類)는 없는 것으로 나타났다.

Apoptosis and Anti-proliferaction by Saussurea lappa and Pharbitis nil in AGS Human Gastric Cancer Cell Line

  • Objectives : We performed this study to understand the molecular basis of the antitumor effect of Saussurea lappa, Pharbitis nil, Plantago asiatica and Taraxacum mongolicum, which have been used for cancer treatment in Korean traditional medicine. Design: We analyzed, the effect of these medicinal herbs on proliferation and apoptosis of tumor cells and its association with gene expression, We performed semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) analysis of cell cycle- and apoptosis-related genes using a gastric cancer cell line AGS. Results : Cell counting assay and $[^3H]thymidine$ uptake analysis showed that Saussurea lappa and Pharbitis nil strongly inhibit cell proliferation of AGS in a dose-dependent manner. Interestingly, gene espression assay revealed that mRNA espression levels of c-Jun, c-Fos, c-Myc, and Cyclin D1 were markedly decreased by Saussurea lappa and Pharbitis nil. Furthermore, Saussurea lappa was identified to activate expression of the p53 tumor suppressor and its downstream effector $p21^{Wafl}$, which leads to $G_1$ cell cycle arrest and apoptosis. These observations suggest that the anticancer effect of Saussurea lappa and Pharbitis nil might be associated with their regulatory capability of tumor-related gene expression.

Bioceramic Effects to Enhance Secondary Metabolites Production in Tissue Culture of Some Medicinal Plants

  • We have investigated that a couple of soft ferrite ceramic powders having a spinal structure have shown the effect on growth and secondary metabolites production of some medicinal plants cultured in vitro. The addition of the ceramic powders as bare state to culture medium has stimulated the growth of Achyranthes japonica callus and plantlet, adventitious root of Hyoscyamus niger and Platycodon grandiflorum hairy root about 65, 75, 150 and 50%, respectively. Whereas Centella asiatica callus and plantlet, Scopolia parviflora hairy root, and Hyoscyamus albus adventitious root were not affected markedly. Moreover, the ceramic powder has enhanced the growth of H. niger adventitious roots even under conditions of irradiating alone without any direct contact between ceramic powder and media. Based on growth stimulation effect, the ceramic powders have enhanced the gross production of tropane alkaloid in H. niger adventitious root, and polyacetylene in P. grandiflorum hairy root about 35 and 30%, respectively.

  • Flora and distribution of vascular hygro- and hydrophytes were investigated from April to June, 1983 on the seven sampling sites along the estuary of Nagdong River. As the result, total 60 kinds of vascular plants are investigated, among them 13 kinds are aquatics; Rotala indica var. uliginosa, Myriophyllum spicatum, Allisma canalicultum, Vallisneria asiatica, Hydrilla verticillata, Potamogeton berchtoldii, P. crispus, Ruppia maritima, Phyllospadix iwatensis, Zostera marina, Juncus effusus, Phragmites communis, and Scirpus triqueter. Present hydrophytes are largely confined to the main-stream zone. The transition zone is poorly habitated by hydrophytes and considered for the distributconfined to the estuary and coastal zone. However, long-term conditions of salinity gradient appear to be primarily responsible for the present flora and distribution.

생약제(Miracle® 첨가가 포유모돈의 생산성, 자돈의 성장 및 혈액성상 변화에 미치는 영향 (Effects of Herbal Plant Mixture(Miracle®) Supplementation on the Productivities of Lactating Sows and Growth Performance and Hematological Characteristics of Piglets)

  • This study was conducted to evaluate effects of dietary herbal extracts mixturetMiracle" included Angelicae Gigantis Radix, Rehmanniae Radix, Cnidii Rhizoma, Glycyrrhizae Radix, Schizandrae Fructus, Plantago asiatica and Dioscoreae Radix) on the productivities of lactating sowsand growth performance and hematological characteristics of piglets. Twenty seven sows (Yorkshire ${\times}$ Landrace ${\times}$ Duroc, 1 to 3 parities) were used in a 21 day old trial. Dietary treatments included 1) CON (control; basal diet), 2) HPMO.l (basal diet + herbal plant mixture 0.1 %) and 3) HPMO.2 (basal diet + herbal plant mixture 0.2 %). Backfat thickness losses from farrowing to weaning was decreased in HPM treatmentscompared with control(linear effect, P $\leq$ 0.003). ADFI was decreased in sows fed herbal plant mixture(linear effect, P < 0.05). Also, treatments of dietary herbal plant mixture reduced day of return to estrus compared with control(linear effect, P < 0.05). Energy digestibility in HPM treatments were higher than control(linear effect, P $\leq$ 0.05). In piglets, weight gain(P > 0.05) and survivability(P> 0.05) after weaning werenot affected by treatments. No statistical differences were found(P> 0.05) forhematological characteristics in piglets. In conclusion, dietary herbal plant mixture reduces backfat thickness losses and day of return to estrus and improves energy digestibility in lactating sows.
