Development of Terra MODIS data pre-processing system on WWW

  • Terra MODIS is one of the few space-borne sensors currently capable of acquiring radiometric data over the range of view angles. Institute of Industrial Science, University of Tokyo, has been receiving Terra MODIS data at Tokyo since May 2001 and Asian Institute of Technology at Bangkok since May 2001. They can cover whole East Asia and is expected to monitor environmental changes regularly such as deforestation, forest fires, floods and typhoon. Over eight hundred scenes have been archived in the storage system and they occupy 2 TB of disk space so far. In this study, MODIS data processing system on WWW is developed including following functions: spectral subset (250m, 500m, 1000m channels), radiometric correction to radiance, spatial subset of geocoded data as a rectangular area with latitude-longitude grid system in HDF format, generation of a quick look file in JPEG format. Users will be notified just after all the process have finished via e-mail. Using this system enables us to process MODIS data on WWW with a few input parameters and download the processed data by FTP access. An easy to use interface is expected to promote the use of MODIS data. This system is available via the Internet on the following URL from September 1 2002, "http : //webmodis.iis.u-tokyo.ac.jp/".

이용도가 낮은 어패류의 가수분해물을 이용한 속성액젓의 제조 (Rapid Processing of Hydrolyzed Sauce Using Low-Usefulness Fish and Shellfish)

  • A rapid processing method for fermented sauce of favorable flavor was investigated with low-usefulness marine resources. Hydrolyzed at optimal conditions for 6 hours usuing alcalase, and separated by molecularporous membrane. It's very effective for remove bitter taste at below M.W. 100 dalton, and effective at below M.W.500 dalton. Added 2% invert sugar in fermented sauce at below M.W.500 dalton, and thermal treatment at 100$^{\circ}C$ for 30 minutes were improved flavor. Chemical composition of fermented sauce using hair tail were 80.7% of moisture, 2.2% of carbohydrate, 1.8% of total nitrogen, 1.6% of amino nitrogen and pH was 6.5. The ratio of amino nitrogen in total nitrogen was 77.8%. And chemical composition of fermented sauce used gizzard shad, kangdale, pen shell and oyster were similar to fermented sauce used hair tail. Total nitrogen were above 1.8% and the ratio of amino nitrogen in total nitrogen was 77.7∼84.2% in all fermented sauce. Amino acid contents in fermented sauce used hair tail, gizzard shad, kangdale, pen shell and oyster were 4,318.1mg%, 4,681.3mg%, 3,156.2mg% and 4,175.0mg%, respectively. And the predominant free amino acid were glutamic acid, lysine and glycine in all fermented sauce.

  • The AOPs research defined by creating a sufficient amount of OH radicals from the dissolution of organic materials through photoxidation and research for a complete elimination of residual organic materials by membrane are actively ongoing. This research focuses on the hybrid processing of AOPs and M/F membrane to dissolve and eliminate organic chemicals in drinking water which are suspected of carcinogens. For this purpose, underground water was used as a source of drinking water for the hybrid processing of AOPs oxidation and M/F membrane, and a pilot plant test device was installed indoor. Carcinogenic chemicals of VOCs and pesticide were artificially mixed with the drinking water, which was then diluted close to natural water in order to examine treatment efficiency and draw optimal operation conditions. The samples used for this experiment include four chemicals phenol, chloroform, in VOCs and parathion, carbaryl in pesticide. As a result of the experiments conducted with simple, and compound solutions, the conditions to sufficiently dissolve and eliminate carcinogenic chemicals from the hybrid processing of where carcinogens were artificially added are : (hydrogen peroxide) prescribed solution 100 mg/L under pH 5.5~6.0, and the temperature $12{\sim}16^{\circ}C$, at the normal temperature and pressure. $d-O_3$ volume of 5.0 ppm and above and 30-40 minutes of reaction time are most appropriate and using MF/UF for membrane was ideal.

기능성 어육단백질의 젤화 특성과 산업적 응용-1. 가열변성 중 화학결합에 미치는 pH의 영향 (Gelation Properties and Industrial Application of Functional Protein from Fish Muscle-1. Effect of pH on Chemical Bonds during Thermal Denaturation)

  • 근원섬유단백질과 알칼리 용액으로 처리하여 회수한 단백질의 가열 중 ANS 소수성의 변화, IR스펙트럼의 변화, SH기의 변화, 전기영동상의 변화 및 엔탈피의 변화로 비교하여 회수 단백질의 가열 변성기구를 조사하였다. 갈고등어와 냉동 꼬마민어의 근원섬유단백질은 가열 온도의 상승과 더불어 소수성 잔기가 외부로 노출되고 소수성 상호작용은 6$0^{\circ}C$ 부근에서 최대로 일어나지만, 근형질 단백질을 포함하는 회수단백질은 근원섬유 단백질과 ANS 소수성에서 차이를 보였다. pH가 증가함에 따라 1636 $cm^{-1}$ /에 해당하는 peak가 증가하였다. 반응성 SH기와 총 SH기 수의 차이는 pH 7.0과 pH 10에서 비교적 큰 것으로 나타났다. SDS-PAGE 상에서 pH가 상승함에 따라 myosin heavy chain의 중합체가 확인되었다. 알칼리 공정으로 제조한 회수 단백질을 시차주사열량계로 분석한 결과, 회수 단백질은 33.1,44.3 및 65.5$^{\circ}C$에서 전이 온도가 나타나며, myosin의 변성 에 해당하는 온도인 51.7$^{\circ}C$의 peak는 보이지 않았다. 회수 단백질의 가열 젤은 극단적 인 pH 처리에 의한 $\alpha$-helix 구조의 $\beta$-sheet 구조 전환, 가열에 의한 S-S 결합의 형성과, myosin heavy chain 중합체 형성에 의한 것으로 보인다.

미생물을 이용한 신예미 자철광으로부터 철 침출에 관한 연구 (Microbial Leaching of Iron from Shinyemi Magnetite Ore)

  • 갯벌 퇴적물에서 분리한 미생물(Haejae-1)을 이용하여 혐기성 및 호기성 조건 하에서 신예미 자철광으로부터 철 침출 및 침출된 철의 생광화작용에 따른 2차 광물의 형성을 연구하였다. 침출에 사용한 미생물은 Haejae-1 (Shewanella sp.)을 이용하였으며 자철광(자철광 : 미생물 성장 배지 = 100 : 1)과 글루코스(10 mM)를 주입한 후 호기성과 혐기성 조건하에서 1개월간 상온에서 실험을 실시하였다. 미생물의 성장 동안 자철광으로부터 침출된 철과 망간의 농도를 ICP-AES를 이용하여 분석하였다. 미생물의 성장과 철의 침출기간 동안 미생물 성장 배지의 열역학적 조건 변화를 알아보기 위하여 미생물 성장 배지의 Eh와 pH를 혐기성 조건하에서 측정하였다. 자철광으로부터 미생물의 성장과 철의 침출, 그리고 자철광의 광물조성 변화 및 2차 광물의 형성을 알아보기 위하여 핀-선 회절분석(XRD), 그리고 SEM-EDX 분석을 실시하였다. XRD 분석결과에 따르면 신예미 자철광의 주 구성광물은 자철석으로 이루어져 있었다. 자철석이 주 구성광물인 신예미 자철광을 이용하여 1개월 동안 미생물에 의한 금속이온의 침출 실험을 실시한 후 호기성에서 15 mg/L의 철(total Fe)과 3.41 mg/L의 망간이 침출 되었으며, 혐기성에서는 32.8 mg/L의 철(total Fe)과 5.23 mg/L의 망간이 침출되었다. 이는 철 환원 미생물이 활동하였기 때문으로 사료된다. 호기성과 혐기성에서의 미생물의 활동에 따른 pH와 Eh의 변화를 측정한 결과, 호기성 조건 하에서는 Eh는 +144.9 mV에서 -331.7 mV로 변화되었으며 pH는 8.3 에서 7.2로 감소하였다. 혐기성 조건하에서 Eh는 -2.3 mV에서 -494.6 mV로 변화되었으며 pH는 8.2에서 7.0으로 감소하였다. Eh의 변화는 미생물에 위한 글루코스의 산화에 따른 전자의 방출에 따른 결과로 사료되며, pH의 감소는 글루코스의 산화에 따른 유기산의 생성에 기인한 것으로 사료된다.

무에서 추출한 myrosinase의 정제 및 효소학적 특성 (Purification and enzymatic characteristics of myrosinase from radish)

  • 무에서 DEAE Bio-Gel, Con-A 및 Superose-6 칼럼을 이용하여 myrosinase를 정제하고 그 효소학적 특성을 검토한 결과 myrosinase(II)는 2개의 subunits를 가졌으며, 이들의 분자량은 SDS-GAGE상에서 각각 53 및 39 KD였다. 정제된 무효소의 비활성도는 37,500 units/mg이었으며, 정제도는 44배였다. 최적 활성 pH는 phosphate 및 Tris-HCl 완충액에서 $6.5{\sim}7.0$이었으며, pH7.0에서 그 효소활성이 가장 안정하였다. 활성 최적온도는 $37{\sim}38^{\circ}C$였고, 열안정성은 $30^{\circ}C$ 이하였다. 무 myrosinase에 대한 무기염의 영향은 구리 및 수은 이온은 효소 활성을 매우 저해하며, ascorbic acid의 농도별 영향은 1 mM일 때 최대활성을 나타내었다. Ascorbic acid analogue에 대한 활성은 dehydroascorbic acid에 대해서는 거의 없었으며, 나머지 analogue들도 ascorbic acid보다 상당히 활성이 낮았다. 무 myrosinase에 대한 환원제의 영향은 2-mercaptoethanol과 dithiothreitol에 의해서는 활성을 나타내지 않았으나, 이들을 ascorbic acid 등 2-mercaptoethanol과 함께 첨가하면 활성을 나타내었다.

  • 젓갈에서 분리한 단백질분해균을 이용하여 속성 저식염 멸치젓의 제조조건과 저장중의 품질 안정성에 대하여 실험한 결과 저식염 멸치젓의 제조조건은 생멸치 100g에 대해 식염 1%, B. licheniformis p-5균 배양액 20m1($3.2{\times}10^{4}cells/ml$), sodium erythorbate 1%를 첨가한 후 $40^{\circ}C$, pH7.0에서 15시간 진탕배양(45strokes/min)시킨 후 저장성과 풍미를 고려하여 NaCl 3%, KCl 4%, 에탄을 4%(w/v), 마늘 및 생강가루 각각 0.5%씩 첨가하는 것이며, 저장중 휘발성 염기질소는 서서히 증가하였고, 히스타민 함량은 제조 직후 17.6mg/100g으로 위생상 안정하였다. 그리고 첨가된 Bacillus 속이 저장기간 동안 총균수의 상당량을 지배하였으며, 유리 아미노산 함량이 분석된 정미성분의 대부분을 차지하였다. 휘발성 성분중 휘발성산이 젓갈의 냄새에 기여도가 켰으며 다음으로 카르보닐 화합물, 염기의 순이었다.

디지탈영상에서 골판톰의 프랙탈분석 (A fractal analysis of bone phantoms from digital images)

  • Purpose : (1) To analyse the effect of exposure time, ROI size and one impact factor in the image processing procedure on estimates of fractal dimension; and (2) to analyse the correlated relationship between the fractal dimension and the Cu-Eq value (bone density). Materials and Methods : The cylindric bone phantoms of 6 large and 5 small diameter having different bone densities respectively and human dry mandible segment with copper step wedge were radiographed at 1.0 and 1.2 sec esposure (70 kVp, 7 mA) using one occlusal film and digitized. Eleven rectangular ROIs from 11 cylindric bone phantoms and 4 rectan-gular ROIs from cortical, middle, periodontal regions, and socket of bone were selected. Gaussian blurred Image was subtracted from original image of each ROI and multiplied respectively by 1, 0.8, and 0.5, and then the image was made binary, eroded and dilated once, and skeletonized. The fractal dimension was calculated by means of a box counting method in the software ImageJ. Results : The fractal dimension was decreased gradually with continued bone density decrease showing strong correlations (bone phantom; r> 0.87, bone; r> 0.68) under 70 kVp 1.0 sec M = 0.8. Fractal dimensions showed the significant differerence (p < 0.05) between two different exposure times on the same small ROI of bone phantom. Fractal dimensions between two different sizes of ROI on bone phantom showed the significant differerence (p < 0.05) under 1.2 sec exposure, but did not show it (p > 0.05) under 1.0 sec exposure. Conclusions : Exposure time, ROI size, and modifying factor during subtracting could become impacting on the results of fractal dimension. Fractal analysis with thoroughly evaluated method considering the various impacting factors on the results could be useful in assessing the bone density in dental radiography.

Expression of manB Gene from Escherichia coli in Lactococcus lactis and Characterization of Its Bifunctional Enzyme, Phosphomannomutase

  • Phosphomannomutase (ManB) converts mannose-6-phosphate (M-6-P) to mannose-1-phosphate (M-1-P), which is a key metabolic precursor for the production of GDP-D-mannose used for production of glycoconjugates and post-translational modification of proteins. The aim of this study was to express the manB gene from Escherichia coli in Lactococcus lactis subsp. cremoris NZ9000 and to characterize the encoded enzyme. The manB gene from E. coli K12, of 1,371 bp and encoding 457 amino acids (52 kDa), was cloned and overexpressed in L. lactis NZ9000 using the nisin-controlled expression system. The enzyme was purified by Ni-NTA column chromatography and exhibited a specific activity of 5.34 units/mg, significantly higher than that of other previously reported ManB enzymes. The pH and temperature optima were 8.0 and $50^{\circ}C$, respectively. Interestingly, the ManB used in this study had two substrate specificity for both mannose-1-phosphate and glucose-1-phosphate, and the specific activity for glucose-1-phosphate was 3.76 units/mg showing 70% relative activity to that of mannose-1-phosphate. This is the first study on heterologous expression and characterization of ManB in lactic acid bacteria. The ManB expression system constructed in this study canbe used to synthesize rare sugars or glycoconjugates.

Production of bioactive ginsenoside Rg3(S) and compound K using recombinant Lactococcus lactis

  • Background: Ginsenoside Rg3(S) and compound K (C-K) are pharmacologically active components of ginseng that promote human health and improve quality of life. The aim of this study was to produce Rg3(S) and C-K from ginseng extract using recombinant Lactococcus lactis. Methods: L. lactis subsp. cremoris NZ9000 (L. lactis NZ9000), which harbors ${\beta}$-glucosidase genes (BglPm and BglBX10) from Paenibacillus mucilaginosus and Flavobacterium johnsoniae, respectively, was reacted with ginseng extract (protopanaxadiol-type ginsenoside mixture). Results: Crude enzyme activity of BglBX10 values comprised 0.001 unit/mL and 0.003 unit/mL in uninduced and induced preparations, respectively. When whole cells of L. lactis harboring pNZBglBX10 were treated with ginseng extract, after permeabilization of cells by xylene, Rb1 and Rd were converted into Rg3(S) with a conversion yield of 61%. C-K was also produced by sequential reactions of the permeabilized cells harboring each pNZBgl and pNZBglBX10, resulting in a 70% maximum conversion yield. Conclusion: This study demonstrates that the lactic acid bacteria having specific ${\beta}$-glucosidase activity can be used to enhance the health benefits of Panax ginseng in either fermented foods or bioconversion processes.
