Changes in Quality Attributes of Sigumjang with Fermentation (시금장 발효기간에 따른 품질 변화)

  • The various components of sigumjang were analyzed according to fermentation time. Aerobic bacteria were increased with fermentation time. Yeasts and molds were increased until 3 days after fermentation and then not changed. Six free sugars, 3 volatile organic acids and 6 non-volatile organic acids were detected. The content of free amino acids was $1,407.9{\sim}3,053.9\;mg%$. Glutamic acid was most abundant component among the amino acids, followed by phenylalanine, arginine and valine. The ratio of essential amino acid was $34.4{\sim}37.0\;mg%$. The content of mineral was $12,966.7{\sim}13,864.5\;mg%$. Potassium was the most abundant in quantity among the minerals in sigumjang except sodium which was added artificially. The principal fatty acids were linoleic acid, palmitic acid and oleic acid. The ratio of unsaturated fatty acids was $76.3{\pm}1.1%$. As a result of sensory evaluation, the highest score was obtained 3 to 5 days after fermentation.

Changes in Aflatoxin and Flavor Components of Traditional Sigumjang (전통 시금장 발효기간별 Aflatoxin 및 휘발성 향기성분의 변화)

  • This study was conducted to investigate various components and aflatoxin of sigumjang during fermentation time. The contents of moisture were decreased during fermentation time. The contents of protein, fat and ash were $19.8{\sim}20.8%,\;1.3{\sim}1.6%$ and $3.1{\sim}3.2%$, respectively. The pH was gradually acidized. The result of measurement of surface color showed: L-value, $29.45{\sim}30.94\;a-value,\;+3.01{\sim}+3.63;\;b-value,\;+5.39{\sim}+6.34$. Aflatoxin was not detected during fermentation. Among 81 kinds of flavor components identified in sigumjang, esters was most in number followed by acids, aldehydes, alcohols and phenols.

Cellular Iron Uptake from Aqueous Solutions depending on Reaction Conditions by genetically engineered Saccharomyces cerevisiae (재조합 Saccharomyces cerevisiae에 있어서 반응조건에 따른 수용성 철의 생체 흡수)

  • Cellular iron uptake was performed in the yeast Saccharomyces cerevisiae that transformed with human ferritin H- and L-chain genes. The recombinant yeasts were enriched in YEP medium supplemented with $2\%$ galactose for 3 days and the iron uptake was followed by incubating the cells with iron in 20 mM MOPS buffer (pH 6.5). The reactions were examined under different conditions including the iron compounds of Fe(II) and Fe(III), the concentration of iron, the concentration of cells and the reaction time. From our results, the recombinant yeast YGH2 producing H-chain ferritin showed higher cellular iron concentration at the cell concentration of 100 mg/ml than 200 mg/ml. Iron presented as Fe(II) rather than Fe(III) was taken up more efficiently. Iron uptake increased slightly when iron was added up to 14.3 mM Fe(II) and then its cellular iron concentration was $16.7{\pm}0.7\;{\mu}mol/g$ cell wet wt. In addition, the iron uptake reaction reached to maximum at about 2 hr incubation.

In-vivo Studies on Effect of Lipo-PGE1 on Neoangiogenesis of Composite Graft in a Rabbit Model (가토모델에서 Lipo-PGE1이 복합조직이식편의 미세혈관신생에 미치는 영향)

  • Purpose: The survival of composite graft is dependent on three steps, (1) plasmatic imbibitions, (2) inosculation, and (3) neovascularization. Among the many trials to increase the survival rate of composite graft, prostaglandin E1 (PGE1) has beneficial effects on the microcirculatory level with vasodilating, antithrombotic, anti-inflammatory and neoangiogenic properties. Lipo-PGE1 which is lipid microspheres containing PGE1 had developed to compensate the systemic and local side effects of PGE1. This study was proposed to determine whether Lipo-PGE1 administration enhanced the survival of composite graft through neovascularization quantitatively in a rabbit ear model. Methods: Fourteen New Zealand White Rabbits each weighing 3~4 kg were divided in two groups: (1) intravenous Lipo-PGE1 injection group and (2) control group. A $2{\times}1\;cm$ sized, full-thickness rectangular composite graft was harvested in each auricle. Then, the graft was reaaproximated in situ using a 5-0 nylon suture. For the experimental group, $3{\mu}g$/kg/day of Lipo-PGE1 ($5{\mu}g$/mL) was administered intravenously through the marginal vein of the ear for 14 days. The control group was received no pharmacologic treatment. On the 14th postoperative day, composite graft of the ear was harvested and immunochemistry staining used Monoclonal mouse anti-CD 31 antibody was performed. Neoangiogenesis was quantified by counting the vessels that showed luminal structures surrounded by the brown color-stained epithelium and counted from 10 random high-power fields (400x) by independent blinded observer. Statistical analysis (Wilcoxon Signed Ranks test for nonparametric data) was performed using SPSS v12.0, with values of p<0.05 considered significant. Results: The mean number of the microvessels was $15.48{\pm}8.65$ in the experimental group and $9.82{\pm}7.25$ in the control group (p=0.028). Conclusion: The use of Lipo-PGE1 facilitated the neoangiogenesis, resulted in the improvement of the survival rate of graft. On the basis of this results, we could support wider application of Lipo-PGE1 for more effective therapeutic angiogenesis and successful survival in various cases of composite graft in the human.

Studies on the Physicochemical Characteristics of Traditional Kochujang (전통 고추장의 품질특성)

  • Physicochemical and microbial characteristics of traditional kochujang (fermented hot pepper-soybean paste) collected from 55 households at different regions were investigated. The traditional kochujang contained $46.71{\pm}5.98%$ moisture, $46.87{\pm}8.83%$ total sugar, $11.77{\pm}3.90%$ crude protein, $15.01{\pm}6.48%$ salt, $27.52{\pm}7.32%$ reducing sugar, $0.26{\pm}0.15%$ amino nitrogen and $2.69{\pm}2.35%$ ethanol. The pH and titrable acidity were $4.60{\pm}0.23$ and $27.26{\pm}10.98\;ml/10\;g$, respectively. The average water activity of traditional kochujang were $0.79{\pm}0.04$. The Hunter L, a, and b values of kochujang were $16.03{\pm}2.89$, $20.42{\pm}4.37$, and $9.71{\pm}1.92$, respectively. The viable cell counts of aerobic, anaerobic bacteria and yeasts in the traditional kochujang were $1.02{\times}10^8{\pm}1.29{\times}10^8\;CFU/g$, $2.24{\times}10^7{\pm}3.90{\times}10^7\;CFU/g$ and $5.90{\times}10^5{\pm}2.25{\times}10^6\;CFU/g$, respectively. The kochujang collected from various regions showed quite strong liquefying and saccharogenic amylase and protease at different level by samples.

Preparation and Its Taste-Active Components of Grass puffer (Takifugu niphobles) Extracts (복섬 엑스분의 추출 및 정미발현성분의 조성)

  • To develop natural intermediate flavoring substances, optimal hydrolysis conditions and taste compounds for two step enzyme hydrolysate(TSEH) using Grass puffer(Takifugu niphobles) were investigated. The optimal conditions for TSEH method were revealed in temperature at $55^{\circ}C$ 3 hour digestion with Alcalase (pH 7.5) at the 1st step and 2 hour at $45^{\circ}C$ digestion with Flavourzyme(pH 6.0~6.5) at the 2nd step. TSEH method was superior to hot-water extraction on the aspect of yield, nitrogen contents and organoleptic taste quality such as umami and control of bitter taste formation. In taste active-components in Glass puffer TSEH, total free amino acid content was 4,502 mg%, major free amino acids were Pro, Leu, Lys, Hypro, Tau, Arg, Phe, Ala, Glu and Val in ordor. As for nucleotides, IMP(372 mg%) was the principal component and also contents of TMAO, total creatinine, and betaine were 43, 278 and 41 mg% in Glass puffer TSEH, respectively. The major inorganic ions in TSEH were Na(949 mg%), K(222 mg%), Cl(1,180 mg%) and $PO_4$(1,081 mg%).

Estimating Stem Volume Table of Quercus Acutissima in South Korea using Variable Exponent Equation (변량지수식을 이용한 전국 상수리나무의 입목수간재적표 추정)

  • This study was conducted to develop a stem volume table for Quercus acutissima in Korea by using Kozak's stem taper equation. In total, 2700 tree samples were collected around the country, and growth performance was investigated through compiling data on diameters by stem height and stem analysis. In order to test the stem taper equation's fitness, the fitness index (FI), bias, and mean absolute deviation (MAD) were analyzed. The fitness of the equation was estimated at 97%, bias as 0.017, and MAD turned out to be 1.118, respectively. Furthermore, there was a statistically significant volume difference between the current volume table and the new volume table (p = 0.0008, <0.005). The result indicates that using the new volume table that reflects the actual forest will reduce the loss when assessing wood resources and will improve the accuracy of forest statistics for national and local governments. A stem volume table, the main result of this research, which is utilized in the estimated stem taper equation, will provide growth information for Quercus acutissima, one of the main broadleaf species in Korea, and will function as a management indicator for rational forest management.

Exploration of β-Glucuronidase Activity of Lactic Acid Bacteria Isolated from Kimchi (김치에서 분리된 젖산균의 β-glucuronidase 활성 탐색)

  • Lactic acid bacteria (LAB) isolated from kimchi were studied for their ${\beta}$-glucuronidase activity. Among the 156 strains tested, 52 strains utilized glucuronic acid as a carbon source and their intracellular ${\beta}$-glucuronidase activities were significantly higher than their extracellular activities. Leuconostoc mesenteroides KFRI 73007 isolated from turnip kimchi exhibited the highest intracellular ${\beta}$-glucuronidase activity of $0.77{\pm}0.01U/mg$ protein, which was further increased to $1.14{\pm}0.01U/mg$ protein under optimized reaction conditions (pH 7, $37^{\circ}C$). The activity of ${\beta}$-glucuronidase was notably decreased by the addition of divalent cations, and glucuronic acid was the best carbon source to produce ${\beta}$-glucuronidase in Leu. mesenteroides KFRI 73007.

Development of a Stem Taper Equation and a Stem Table for Criptomeria japonica Stands in South Korea (삼나무의 수간곡선식 및 입목수간재적표 개발)

  • The aim of this study was to utilize Kozak's stem taper model to develop both a stem taper equation and a stem volume table for Criptomeria japonica, a tree species distributed across Korea. A total of 1,000 sample trees were cut and collected across the country to measure their diameters by stem height. The equation was then used to estimate examine their stem shapes. Our results show that the Fitness Index for the equation was 98.7%, the Mean Absolute Deviation (MAD) was -0.0142, and the MAD was 1.1640, thus indicating a high level of fitness. A statistically significant difference (p < 0.05) was also found from the analysis of discrepancies between a current table and the new table used in this study. It is therefore suggested that the new table-with data from actual stands-will contribute to enhancing the accuracy of national and municipal forest statistics and reducing losses caused by imprecise data on available forest resources.

Clinical and Angiographic Features of Secondary Postpartum Hemorrhage and the Outcomes of Transcatheter Arterial Embolization (속발성 분만 후 출혈의 임상 및 혈관 조영술의 특징과 경도관 동맥 색전술의 결과)

  • Purpose: To analyze the clinical and angiographic features with outcome of transcatheter arterial embolization in patients with secondary postpartum hemorrhage. Materials and Methods: Clinical details and angiographic features with assessment of arterial embolization were reviewed in total 38 patients underwent arterial embolization at single tertiary referral center. Results: Twenty patients (53%) had Cesarean section. The major causes of bleeding were iatrogenic vascular injury, and retained placenta (55%). The patterns of vaginal bleeding were recorded as intermittent (50%), or as persistent (50%). Seven patients (18%) were hemodynamically unstable at presentation. Positive angiographic findings appeared in eighteen patients (47.3%). The frequency of pseudoaneurysm was statistically high in the Cesarean section (p < 0.001). The used embolic agents except gelfoam were N-butyl cyanoacrylate (n = 7), and microcoil (n = 7). Unilateral selective embolization (26.3%) was shown effective in superselective embolization of bleeding focus. Technical and clinical success rate were 100% and 97.4%, respectively with no complication. Sixteen resumed regular menstruation, and one pregnancy were observed in patients with available follow-up of over 6 months. Conclusion: Considerable rate of hemodynamically unstable patients was observed with high rate of positive angiography findings. Given high successful rate and few complications, early angiographic assessment with embolization should be considered.
