Economic Analysis based on Estimation of Optimal Facilities of Oyster Aquaculture in Jaran Bay (자란만 해역의 굴 양식어업 적정시설량 평가에 기초한 경제성 분석)

  • Maintaining a certain level of fisheries production is due to the increase in aquaculture production. In recent years, aquaculture plays an important role in fishery production in Korea. Nevertheless, aquaculture should be carefully managed by regulations because it also causes environmental load which can threat sustainability of aquaculture. For this reason, Korean government has regulated density of culturing facilities with the Fisheries Law and its adjective decrees. The regulatory compliance of fisheries businesses is very low because the criteria for the regulation of the density were not enacted with scientific research. Thus, this research was aimed to obtain scientific criteria for the regulation of oyster aquaculture in Jaran Bay with economic assessment. For this research, we collected the data necessary for the assessment on five investigation points in the bay for two years. With the data, simulation for the growth rate of oyster was performed and the result showed that at least 25.5% of facilities should be reduced. Also, it was revealed that removing 2 long lines would be most beneficial. The NPV of the best measurement was 35,120,300 won and IRR was 11.7%. With this research, the government will gain more accurate regulatory compliance due to the scientific approach. Moreover, fisheries businesses in oyster aquaculture can obtain flexibility to cope with market fluctuation.

Effect of Pasteurized Substrates in Various Condition on the Mycelial Growth of Oyster Mushroom, Pleurotus spp. (볏짚배지의 살균조건이 느타리버섯균의 균사생장에 미치는 영향)

  • Various conditions for mushroom were investigated to study their effects on the mycelial growth of oyster mushroom and pathogens. Our results show that the optimal pasteurization temperatures for mycelial growth and pathogens are $60^{\circ}C\;and\;121^{\circ}C$ respectively. Both were then decreased by heating over $60^{\circ}C$ and getting down temperature under $121^{\circ}C$, respectively. In the experiment for pasteurization time, it was showed that both 8 and 12 hours pasteurization were greater than 24 hours treatment for the mycelial growth. In contrast, we didn't find any particular effect by different soaking time of substrates. The pH In substrates was leveled well by pasteurization and after-fermentation even though the acidity was lowered by extending the soaking time. Our results also show that the mycelial growth is greatest at $25{\sim}30^{\circ}C$ regardless of pasteurization time for substrates, Through the experiment with paddy straw as a substrate, it was showed that two time pasteurization was greater than one time for mycelium growth and pro-fermentation of paddy straw before pasteurization have resulted in worth mycelial growth compared with that of normal condition.

Studies on the Culture of Oyster, Crassostrea gigas (Thunberg) in Eastern Coast of Korean (동해안 참굴 양식에 관한 연구)

  • For improvement of fishermen's income by oyster cultivation in eastern coast of Korea, the spats of oyster (Crassostrea gigas) collected from Hadong in southern coast of Korea were transplanted to Chumunjin and cultured by hanging long line method from August 1994 to July 1995 for examination of their growth and ground environmental conditions. The environmental conditions show the annual range of water temperature as 8.33~$25.62^{\circ}C$, salinity as 32.84~$34.28\textperthousand$, PO4-P as 0.09~0.40$\mu$g-at/1, dissolived inorganic nitrogen (DIN) as 0.32~3.21$\mu$g-at/1. The growth of oyster in hanging culture was as follows : 88.7mm (shell hight) in upper layer (7m), 84.9mm in middle layer (10m) and 78.0mm in lower layer (13m) after one-year cultivation. Thus, the oysters in upper layer had grown relatively faster than those in lower layer. The relationships between shell hight (SH) and shell length (SL) in each culturing depth as follows : SL=0.5403 SH+8.5486 (r=0.9959) in upper layer, SL=0.5813 SH+3.7775 (r=0.9869) in middle layer, and SL=0.5159 SH+6.8736 (r=0.9961) in lower layer. The meat weight of oyster was the highest value as 13.24g in upper layer, and 12.68g in middle layer, and lowest as 10.96g in lower layer. The relationships between total wight (TW) and meat weight (MW) with culturing water layer were as follows : MW=0.1933 TW+0.1051 (r=0.0073) in upper layer, MW=0.1915 TW+0.1894 (r=0.9984) in middle layer, and MW=0.1650 TW+0.0558 (r=0.9983) in lower layer.

Physiology characteristics of genus Hypocrea isolated at oyster mushrooms of the shelf cultivation (균상재배 느타리버섯에서 분리한 Hypocrea속 균의 생리적 특성)

  • Hypocrea disease is the most severe disease of oyster mushroom cultivation in Korea. Physiological and ecological studies were performed on the pathogens (Hypocrea spp.) to obtain basic information for developing the integrated disease management system. Fourteen isolates of Hypocrea were collected from oyster mushroom house in five areas. Pathogenic fungi causing disease of oyster mushroom were identified as Hypocrea sp. based on morphological characteristics and pathogenicity. Two isolates (H-1, H-12) showed the fastest growth at $15^{\circ}C$ but four isolates (H-8, H-9, H-13, H-14) showed slower growth than those of other isolates at $20^{\circ}C$ and $25^{\circ}C$. Stroma with ascocarps and ascospore were produced on PDA under fluorescent light. The five isolates produced stroma with ascocarps and ascospores. Formation of fruiting body of strains H-14 of Hypocrea were the best out of all the strains on the potato dextrose agar (PDA). Also, fruiting bodies and ascospores were completely produced under fluorescent light. The growth of the isolates was correlated with total carbon content. The stroma of the isolates was formed mainly in histidine and asparagine treatment and especially in histidine-70 and asparagine-100 treatment. In the test of pathogenicity, after and before spawning showed very fast incidence of disease.

Breeding and characterization of a new white cultivar of Pleurotus ostreatus, 'Sena' (갓이 백색인 느타리 신품종 '세나'의 육성 및 특성)

  • The development of automated bottle cultivation systems has facilitated the large-scale production of Pleurotus ostreatus, a commonly cultivated oyster mushroom species in South Korea. However, as the consumption of this product is decreasing and production quantities are exceeding demand, farmers are seeking various other mushroom types and cultivars. In response to this, we have developed a new oyster mushroom cultivar named 'Sena'. This high-yielding cultivar has a white pileus and excellent quality. The white oyster mushroom cultivars 'Goni' and 'Miso' were selected as parental strains from the genetic resources of the National Institute of Horticultural and Herbal Science's Mushroom Division. By crossing their monokaryons, hybrids were developed and subjected to cultivation trials and characteristic evaluations to select the superior cultivar. The optimal temperature for 'Sena' mycelial growth is 25-30℃, with inhibition occurring at temperatures above 30℃, whereas the temperature for mushroom growth is 14-18℃. The mushrooms grow in clusters, with the white pileus having a shallow funnel shape. Optimal mycelial growth occurs in malt extract agar medium. When cultivated in 1,100 cc bottles, the 'Sena' cultivar had 35 available individuals, surpassing the number 16 available from the control cultivar 'Goni'. The yield per bottle also increased by approximately 157 g, a 24% increase over the control cultivar amount. When 300 g samples of harvested mushrooms were packed and stored at 4℃ in a cold storage facility for 28 days, the weight loss rate of 'Sena' was approximately 4.22%, lower than that of 'Goni'. Moreover, the changes in pileus and stipe whiteness (measuring 6.99 and 8.33, respectively) were also lower than those of the control cultivar. Since the appearance of a white cap is crucial for quality assessment, the 'Sena' cultivar is superior to the 'Goni' cultivar in terms of both weight and quality after undergoing low-temperature storage.

Two new species of Trichoderma isolated from commercially grown oyster mushroom, Pleurotus ostreatus (oral)

  • We describe two new Trichoderma species associated with oyster mushroom in Korea. Trichoderma green mould has been one of the most serious diseases of oyster mushroom in Korea. Of these the predominant species are two unrecorded species. We designed as Trichoderma sp. Korean type 1 (Th K1) and Trichoderma sp. Korean type 2 (Th K2), respectively. Th K1 and Th K2 can be distinguished from previously reported Trichoderma species as well as each other in morphological characteristics including growth rate at 35$^{\circ}C$, colony morphology, conidia shape and branch pattern of phialides. Sequence of the ITS region of rDNA, the protein coding translation elongation factor gene(EF-1${\alpha}$), and RNA polymeraseII (RPB2) not only clearly separated Trichoderma sp. Korean types from their closely related T. harzianum biotype but also distinguished them from each other. Analyses of the EF-1${\alpha}$ and RPB2 sequences were found to be more useful for establishing systematic relationships among Trichoderma isolates than those of the ITS sequence.   • This study aimed to investigate possible cause of slow and abnormal growth of oyster mushrooms, Pleurotus florida and P. ostreatus collected from bad crop farms. Spherical virus particles of 30 nm in diameter from P. florida, 23 nm particles from P. ostreatus, and both 23 and 30 nm particles were also found from interspecies mated culture between P. florida and P. osreatus. The virus particles might be associated with the bad crop of Pleurotus species.

  • In order to understand the effect of introduction of oyster rack culture on natural short-necked clam, Ruditapes philippinarum, stock, we investigated the status of natural clam stock prior to introduction of oyster rack culture at Yeongheung Coast of Wando Island. The growth equation of the clam was estimated as: $L_t=61.46{\cdot}(1-e^{-0.172(t+0.155)})$ from ring radius composition of shell. Instantaneous coefficient of total mortality and natural mortality were calculated as: 2.4087/year and 0.478/year, respectively. The age at first capture was estimated to be 2.55 year. The total biomass was estimated to 3.23 ton in the bed (0.8 ha). Applied by these parameters, the annual recruit biomass and the current yield per recruit (Y/R) was corresponded to $114.7individuals/m^2$ and $92.0g/m^2$, respectively. Maximum sustainable yield (MSY) was estimated 0.77 ton/year which was close to annual catches, 0.74 ton/year, in the area.

Judgement of Resistant Cultivar by Screening method for Resistance of Oyster Mushroom to Trichoderma disease in vitro (푸른곰팡이균의 저항성 품종 검정방법에 의한 느타리버섯 균주의 저항성 판별)

  • In coculturing with strains of Trichoderma and oyster mushroom, we could detect the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We selected the isolates ASI 2183, ASI2504 and ASI 2477 as varieties that showed the resistance. The isolates ASI 2240, ASI 2479 and ASI 2181 were the best in their resistance against Trichoderma in the method using culture filtrate. In common, the isolates ASI 2479 and ASI 2240 were selected in both methods. In post-inoculation method, the isolates ASI 2479, ASI 2333 and ASI 2181 were selected and ASI 2302 was susceptible. For the same isolate of Trichoderma, the resistance varied depending on the isolates of oyster mushroom used in the experiments. Because we could detect the interactions between Trichoderma and oyster mushroom, it is possible to detect the level of the resistance that differs in the varieties. However, there were the cases of detecting the level of the resistance in repetitions with the same isolate, which may be caused by the vitality of isolates of Trichoderma and oyster mushroom. It is efficient to test the resistance with the resistant isolate of Pleurotus salmoneostramine and the susceptible isolate of ASI 2302.

Characteristics of a new oyster mushroom variety 『Jinmi』 for the bottle cultivation (느타리버섯 신품종 육성 연구(I) -병재배용 신품종 『진미』 느타리버섯의 특성-)

  • Jinmi oyster mushroom(Pleurotus ostreatus) were bred in Mushroom Research Institute, Gyonggi Province A.R.E.S in 2003. This oyster mushroom were bred and cultivated one after mating single spores collected from K3-2 and ASI2018-249. The major characteristics of the mushroom are showing a lot of pinheadings, the gray-colored and infundibuliform pileus. The optimum temperature for the mycelial growth was around $25{\sim}30^{\circ}C$ and that for the pinheading and growth of fruitbody was around $15{\pm}1^{\circ}C$. Around 22 days at $20^{\circ}C$ were required for incubation of Jinmi and the yield was shown high by 145.9g/bottle.
