  • This study aims to detect the change of marine aquaculture farm within the boundary of Hallyeo Marine National Park. Comparison has been made on the Landsat images taken in 1984 and 2002 respectively by using feature extraction methods and other image analysis techniques. During the 18 year period between 1984 and 2002, total area of the aquaculture farms has been decreased in 63 percent. The reason for the change seems to be that aquaculture farms became concentrated only around the Geoje Islands due to the growth of the labor- and capital-intensive cage aquaculture for the expensive fish species instead of traditional oyster farming. Authors suggest the monitoring using remotely-sensed data as the best tool for the management of marine aquaculture farms on the basis of accuracy of analysis and relatively cheap cost. Management strategies of salmon farms in Tasmania, Australia has been analyzed to find the field techniques necessary for the management of aquaculture.

어패류에서의 Vibrio vulnificus 분리 (Isolation of Vibrio vulnificus from Shellfish)

  • Vibrio vulnificus is an organism capable of causing septicemia and wound infection in compromised patients. The source of infection is known to be raw oysters and others. The prevalence of the organism in Korean sea water and shellfish is not known. The authors surveyed shellfish and other specimens obtained mainly from a market in Seoul and from ones in Inchon. Five cultures of V. vulnificus were isolated from oyster and clam samples. Two isolates had typical characteristics of the strains isolated from patients, i.e., definite hemolysis and typical biochemical reactions. However, other 2 isolates were sucrose positive, although the identity were confirmed by Center for Disease Control. We do not know wether such strains are pathogenic or not. For the isolation of V. vulnificus from environmental samples, TCBS agar and VV agar were not very selective or differential. We isolated our strains with the use of OF-lactose agar and SPS agar. However OF-lactose agar did not support good growth of V. vulnificus, while SPS agar did not suppress other vibrios.

Effect of Sodium Hypochlorite for Controlling Bacterial Blotch on Pleurotus ostreatus

  • Sodium hypochlorite alkaline was tested against Pseudomonas tolaasii causing bacterial blotch on cultivated oyster mushroom (Pleurotus ostreatus). The minimum inhibitory concentration of sodium hypochlorite against P. tolaasii contained active chlorine (AC) at 1.4 mg/l on plate assay. The highest cultivation yield was obtained from the treatment of AC 5.7 mg/l. Treatment of sodium hypochlorite at the rate of higher than AC 11.4 mg/l resulted in reduced yields at the harvest. However, the population of total bacteria on the bed surface treated with AC 5.7 mg/l of sodium hypochlorite was maintained to some extent. Inhibitory concentration against total bacteria on the bed surface was over AC 22.8 mg/l. Mushroom mycelium was damaged and its growth strongly inhibited at the concentration of AC 200 mg/l. Mushroom caps showed yellowish symptom by chemical injury by treatments of AC 74.1 mg/l or higher. Sporocarps infected by P. tolaasii were irrevocable at any concentration of sodium hypochlorite. Routine watering with AC 5.7 mg/l from mushroom initiation to the end of picking resulted in reduced bacterial blotch incidence of 40% and 86% at two mushroom farms. The treatment resulted in higher quality mushroom production compared to that conventionally watered with tap water alone.

Physiological Characteristics of Green Mold(Trichoderma spp.) Isolated from Oyster Mushroom(Pleurotus spp.)

  • This study was conducted to investigate physiological characteristics of Trichoderma spp. isolated from Pleurotus spp. Damage tests of Pleurotus spp. and mycotoxins tests of Trichoderma spp. were also done. The optimal growth temperature of Trichoderma spp. was $27{\sim}30^{\circ}C$. Although, T. longibrachiatum was able to grow at $37^{\circ}C$ and grew $30{\sim}40$ times faster than Pleurotus. The colony colour on PDA medium of T. cf. virens was yellowish green, T. longibrachiatum was yellow, and T. harzianum was turning to bright green. In damage tests of Pleurotus by Trichoderma, T. cf. virens caused the most severe damage to Pleurotus. T. longibrachiatum and T. harzianum caused less damage on Pleurotus but were able to cause greater damage to P. eryngii. One of the mushroom cultivars, P. ostreatus 8 was the most resistant to all Trichoderma spp.. Chitinolytic mycotoxin released by Trichoderma spp. caused 52.7% damage to Pleurotus. Mycotoxins released by T. longibrachiatum caused the greatest damaged(78.6%) on P. eryngii.

Purification of a Pore-forming Peptide Toxin, Tolaasin, Produced by Pseudomonas tolaasii 6264

  • Tolaasin, a pore-forming peptide toxin, is produced by Pseudomonas tolaasii and causes brown blotch disease of the cultivated mushrooms. P. tolaasii 6264 was isolated from the oyster mushroom damaged by the disease in Korean. In order to isolate tolaasin molecules, the supernatant of bacterial culture was harvested at the stationary phase of growth. Tolaasin was prepared by ammonium sulfate precipitation and three steps of chromatograpies, including a gel permeation and two ion exchange chromatographies. Specific hemolytic activity of tolaasin was increased from 1.7 to 162.0 HU $mg^{-1}$ protein, a 98-fold increase, and the purification yield was 16.3%. Tolaasin preparation obtained at each purification step was analyzed by HPLC and SDS-PAGE. Two major peptides were detected from all chromatographic preparations. Their molecular masses were analyzed by MALDI-TOF mass spectrometry and they were identified as tolaasin I and tolaasin II. These results demonstrate that the method used in this study is simple, time-saving, and successful for the preparation of tolaasin.

오미자박과 미강 첨가배지가 느타리버섯 자실체의 γ-aminobutyric acid(GABA) 함량에 미치는 효과 (Effect of schizandra berry dregs and rice bran treatment on γ-aminobutyric acid (GABA) content enhancement in Pleurotus ostreatus)

  • 느타리버섯(Pleurotus ostreatus)'흑타리'의 기본배지 조성에 농산물 부산물 유래 첨가용 배지를 추가하였을 경우 기억력 증진물질 중인 하나인 GABA(Gamma-amino butyric acid)함량을 증가시킬 수 있는 배지재료와 첨가수준을 구명한 결과는 다음과 같다. '흑타리'는 5% 녹차가루 처리에서 발이율과 수량이 현저히 떨어졌고, 다시마가루는 첨가된 전체 처리구에서 생육이 불량하였으며, 10% 녹차박과 오미자박 처리구에서는 생육이 가장 양호하였으나 녹차박 처리구보다는 오미자박 처리구에서 기능성물질 함량이 더 우수한 것으로 분석되었다. 또한, 첨가 수준에 따른 자실체의 GABA 함량은 무처리 대비 녹차가루는 2%, 녹차박은 10%, 다시마가루는 1% 정도 증가되었으나, 오미자박은 5%, 10%, 15%, 미강은 1%처리구에서 1.2~2.1배 수준까지 증가되는 양상을 보였다. 결론적으로, 느타리버섯 '흑타리'의 발이율과 생육 뿐만 아니라 GABA 함량 증가를 고려한 적합 첨가 배지로는 오미자박과 미강처리시 가장 양호했으며, 자실체 GABA 함량은 오미자박 10% 처리구에서 2.1배, 미강 1% 처리구에서 12%정도가 증가됨을 알 수 있었다.

  • 본 시험은 일본 Miyagi Ken 굴 연구소에서 1969년 채묘된 O. lurida 및 C. angulata를 항공편으로 우송하여 1970년 1월 20일부터 12월까지 경남 거제군 하청면 연구리 옥계지선에서 수하식으로 성육시켜 조사한 것이며 그 결과는 다음과 같다. 1. O. lurida는 평균 각고 36.4mm 각장 29,5mm인 둥근모양으로서 소형굴로 자랐고, C. angulata는 각고 87.4mm, 각장 52.3mm인 타원형으로서 대형굴 형태로 성장했다. 2. O. lurida, C. angulata는 다같이 수온이 $16^{\circ}C$ 이상일 때 6, 7월시에 성장도가 좋았으며, 이때의 비중은 $1.01717\~1.02368$ 사이였다. 3. 각고 각장간의 상대적인 변화는 O. lurida는 년간 큰 차가 없었으나, C. angulata는 7월에서부터 현저한 차를 이루어 11월에 35.8mm로 그 차가 가장 컸다. 4. 치사율은 O. lurida는 $35.2\%$였으며, C. angulata는 $21.7\%$를 나타냈다.

오이의 수경재배시 생육과 몇 가지 품질에 미치는 재활용 배지의 영향 (Effect of Recycled Substrates Culture on the Growth and Some quality Components of Hydroponically Grown Cucumber)

  • 오이의 생육과 괴실의 품질에 미치는 폐암면, 몇가지 폐버섯 배지, 밤나무 파쇄입자, 질석, 펄라이트, 상용암면의 효과를 실험한 바 다음과 같은 결괴를 얻었다. 야마자키 조성 오이 배양액을 이용한 배지경에서 식물체 생육은 폐암면내피트모스 혼용구와 입상 UR 암면에서 좋았다. 그러나 오이 총수량은 폐팽이버섯 배지에서 가장 많았다. 다음으로 펄라이트, 폐애느타리버섯 배지, 폐암면+피트모스(1:2 v/v) 순서였는데 이들 세 처리 간에는 유의차가 없었다. 당도와 경도는 펄라이트와 질석구에서 높았으며 그 이외는 처리구 상호간에 큰 차이가 없었다. 비타민 C 함량은 배지구간에 일정한 경향이 없었다. 이상의 결과로 미루어 폐배지도 오이의 수경재배 배지로 이용 가능성이 매우 크다는 것을 알 수 있었다.

Isolation of Bacteria Associated with the King Oyster Mushroom, Pleurotus eryngii

  • Eight distinct bacteria were isolated form diseased mycelia of the edible mushroom, Pleurotus eryngii. 16S rDNA sequence analysis showed that the isolates belonged to a variety of bacterial genera including Bacillus (LBS5), Enterobacter (LBS1), Sphingomonas (LBS8 and LBS10), Staphylococcus (LBS3, LBS4 and LBS9) and Moraxella (LBS6). Among them, 4 bacterial isolates including LBS1, LBS4, LBS5, and LBS9 evidenced growth inhibitory activity on the mushroom mycelia. The inhibitory activity on the growth of the mushroom fruiting bodies was evaluated by the treatment of the bacterial culture broth or the heat-treated cell-free supernatant of the broth. The treatment of the culture broths or the cell-free supernatants of LBS4 or LBS9 completely inhibited the formation of the fruiting body, thereby suggesting that the inhibitory agent is a heat-stable compound. In the case of LBS5, only the bacterial cell-containing culture broth was capable of inhibiting the formation of the fruiting body, whereas the cell-free supernatant did not, which suggests that an inhibitory agent generated by LBS5 is a protein or a heat-labile chemical compound, potentially a fungal cell wall-degrading enzyme. The culture broth of LBS1 was not inhibitory. However, its cell-free supernatant was capable of inhibiting the formation of fruiting bodies. This indicates that LBS1 may produce an inhibitory heat-stable chemical compound which is readily degraded by its own secreted enzyme.

신품종 느타리버섯 '화성2호'의 특성 (Characteristics of a new cultivar Pleurotus ostreatus, Hwaseong #2)

  • 다양한 느타리버섯 품종간의 교잡을 하였으며 균상재배용으로서 유용한 특성을 갖는 느타리버섯 품종을 육종코자하였다. 그 중 화성1호에서 단핵균을 분리한 후 수한1호 이핵균과 di-mono교배법으로 육성된 MST247ns가, 세균성 갈변병에 약하고 저온기 재배시 어려움이 많은 수한1호의 단점을 보완하면서 우수한 특성을 가지므로 최종 선발하여 "화성2호"로 명명하였다. 수한1호와 비교하여 대가 길고 조직이 무르지 않고 단단하였다. 화성2호는 중고온성인 수한1호와 비교하여 낮은 온도에서도 갓이 작고 대가 길며 저장성이 좋은 특성을 더욱 잘 나타내었다. 이러한 특성은 시장에서 선호하는 버섯으로서, 낮은 온도에서의 재배관리가 비교적 용이한 가을철에서 이른봄철까지 농가에서의 난방비 부담을 줄여주며 양질의 버섯을 생산할 수 있어 농가의 소득증대에 많은 도움을 줄 것으로 기대된다. '화성2호'의 주요특성으로는 가. 적정 균사배양온도는 $25-30^{\circ}C$이다. 나. 버섯발생 온도는 $8-15^{\circ}C$, 자실체의 적정 생육온도는 $9-14^{\circ}C$으로 가을철부터 봄철까지의 기간에 재배하기에 좋은 품종이다. 다. 수한1호와 비교하여 대가 길고, 갓은 얕은 깔때기형이다. 라. 낮은 온도에서 재배시에도 대가 길고 곧으며 대의 표면이 백색이며 조직이 단단하면서 탄력이 있으므로 저온기의 생육관리가 비교적 쉽고 저장성이 우수하다.
