Postprandial Ammonia Excretion and Oxygen Consumption Rates in Olive Flounder Paralichthys olivaceus Fed Two Different Feed Types According to Water Temperature Change

  • Postprandial ammonia excretion and oxygen consumption in olive flounder Paralichthys olivaceus fed two different feed types, moist pellet (MP) and expanded pellet (EP) diets, to satiation were determined at $12^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, and $25^{\circ}C$ for 48 h. The ammonia excretion and oxygen consumption rates increased with increasing water temperature. However, the postprandial times for the maximum rates of ammonia excretion and oxygen consumption were shortened from 12 h to 6 h after feeding with increasing water temperature. The ammonia excretion and oxygen consumption rates of the fish fed EP were significantly higher (P < 0.05) than those fed MP at 12 h post-feeding both for $12^{\circ}C$ and $15^{\circ}C$. The highest (P < 0.05) weight-specific ammonia excretion rates at $12^{\circ}C$ were observed in the fish fed EP and MP at $12.1mg\;NH_3-N\;kg^{-1}h^{-1}$ and $8.7mg\;NH_3-N\;kg^{-1}h^{-1}$, respectively, for 12 h and 9 h after feeding. The highest (P < 0.05) weight-specific oxygen consumption rates at $12^{\circ}C$ were observed in fish fed EP and MP at $116.4mg\;kg^{-1}h^{-1}$ and $101.0mg\;kg^{-1}h^{-1}$, respectively, for 12 h after feeding. The highest ammonia excretion rates at $25^{\circ}C$ in the fish fed EP and MP increased to $16.9mg\;NH_3-N\;kg^{-1}h^{-1}$ and $18.3mg\;NH_3-N\;kg^{-1}h^{-1}$, respectively, for 6 h after feeding. The highest (P < 0.05) weight-specific oxygen consumption rates at $25^{\circ}C$ were observed in fish fed EP and MP at $184.3mg\;O_2kg^{-1}h^{-1}$ and $197.3mg\;O_2kg^{-1}h^{-1}$, respectively. These data are valuable for the design of biofilters and development of effluent treatment technologies for the land-based flounder farms.

Influence of Oxygen Consumption on Pregnancy Rates of Hanwoo Calves following Embryo Transfer

  • Recently, several approaches have been used to measure the oxygen consumption rates of individual embryos, but relationship between oxygen consumption and pregnancy rates of Hanwoo following embryo transfer has not yet been reported. In this study, we investigated the correlation between oxygen consumption rate and pregnancy rates of Hanwoo embryo using a SECM. In addition to, the expression of apoptosis-related genes was determined using real-time PCR by extracting RNA according to the oxygen consumption of in vivo embryo. First, we found that the oxygen consumption significantly increased in blastocyst-stage embryos (blastocyst) compared to early blastocyst stage embryos, indicating that oxygen consumption reflects the embryo quality (Grade I). The oxygen consumption or GI blastocysts were significantly higher than those of GII blastocysts ($10.2{\times}10^{14}/mol\;s^{-1}$ versus $6.4{\times}10^{14}/mol\;s^{-1}$, p<0.05). Pregnant rate in recipient cow was 0, 60 and 80% in the transplantation of embryo with the oxygen consumption of below 10.0, 10.0~12.0 and over $12.0{\times}10^{14}/mol\;s^{-1}$, respectively. Apoptosis regulatory genes, Hsp-70.1 were significantly increased in over-10.0 group than below 10.0 group but in Caspase-3, Bax and P53 gene, there was no significant difference. In conclusion, These results suggest that measurement of oxygen consumption maybe help increase the pregnant rate of Hanwoo embryos.

Estimation of Oxygen Consumption Rates by Using an Oxygen Microelectrode in Ganghwa Intertidal Flat (강화도 남부 갯벌퇴적물에서 산소 미세전극을 이용한 산소소모율 추정)

  • We measured oxygen microprofiles using an oxygen microelectrode in Ganghwa intertidal flat in April and September. Oxygen consumption rate was calculated by using three different methods based on the oxygen microprofiles. The method using the PROFILE software was thought to be the most reliable among the three methods. The oxygen consumption rates calculated at station D1 by using the PROFILE software were 10.5 and 6.27mmol $m^{-2}d^{-1}$ in April and September, respectively. At station D2, they were 10.9mmol $m^{-2}d^{-1}$ in April and 5.39 mmol $m^{-2}d^{-1}$ in September. There was little spatial variation, but large seasonal variation, with almost two times larger values in April than in September. The higher rate in April is ascribed to higher oxygen concentration in the seawater and higher organic carbon content in the surface sediments, which probably accelerate oxygen consumption for organic matter decomposition in the sediments. Aerobic remineralization rates estimated from the oxygen consumption rates ranged from 4.14 to 8.07 mmol C $m^{-2}d^{-1}$ in Ganghwa intertidal flat, which were much lower than the anaerobic remineralization rate.

  • Changes in Oxygen consumption and filtration rates were investigated to understand physiological rhythms for 24 hours of the Manila clam, Ruditapes philippinarum. physiological rhythms in the oxygen consumption and filtration rates at 15$^{\circ}C$ and 25$^{\circ}C$ were showed diurnal tidal rhythms, appearing two peaks for 24 hours: maximum at night-high tide and minimum at day-low tide. No rapid variations in oxygen consumption and filtration rates for 24 hours appeared at two different water temperatures.

  • As a prioiminary study on usage of metabolic charateristics of the indicator species for indirect estimation of environmental water quality, effects of environmental pollutants on survival, filtration and oxygen consumption rates in Corbicula aeana were investigated at 17$^{\circ}C$ and $25^{\circ}C$ in 10 day afrer treatmint of pollutants. In case of glucose and complex fertilizer, the survival rates of the clams were 100% without any relation to individual sizes and water temperatures. In small sizes at $25^{\circ}C$, the survival rates of the clams by NH$_{4}$CI concentration were shown 95% at 10 mg/1 and 15mg/1, and 90% at 20mg/1, respectively. But the survival rate was 95%at 20 mg/1 of NH$_{4}$CI concentration in small size at 17$^{\circ}C$. The higher filtration and oxygin consumption rates were shown in small size at higher water timperatures(over $25^{\circ}C$), and generally filtration and oxygen consumption rates decreased with increase of glucose, complex fertilizer and NH$_{4}$CI concentrations, respectively. In general, effects of filtration and oxygen consumption rates at NH$_{4}$CI concentrations were shown slightly larger than those of glucose and complex fertilizer.

Effects of Heavy Metals on Clearance and Oxygen Consumption Rates of the Sea Squirt Halocynthia roretzi According to Various Body Sizes

  • To evaluate the biological response of sea squirt Halocynthia roretzi with different body size to heavy metals and its suitability for ecotoxicity assays, the effects of Cr, Cu and Zn on its clearance and oxygen consumption rates were investigated. Clearance and oxygen consumption rates of H. roretzi with various body sizes were calculated at different metal concentrations. Both clearance and oxygen consumption rate were negatively correlated with body sizes. Clearance rate of H. roretzi decreased gradually with increasing concentration of heavy metal, the decreasing rate was in an order of Cr>Cu>Zn. The oxygen consumption rate first increased at low metal concentration (below $100{\mu}g\;L^{-1}$) and then decreased rapidly with increasing metal concentrations. The decreasing rate was in an order of Cu>Cr=Zn. There was a trend that the clearance rate and oxygen consumption rate decreased drastically under a concentration of $400{\mu}g\;L^{-1}$, and then decreased smoothly when the metal ion concentration increased continually. So the oxygen consumption and clearance rate at a concentration of $400{\mu}g\;L^{-1}$ Cu could be thought as a suitable biological tool for exotoxicology analysis.

Physiological viewpoint of the recommended safe weights of load for manual materials handling tasks (인력물자취급의 권장안전하중에 대한 생리학적 고찰)

  • The objective of this study was to make a comparison of the oxygen consumption rates during the lifting activities and the physiological criteria of the recommended weights of RWL, AL, and MPL by NIOSH Guideline. The physical Work Capacity (PWC) based on the bicycle ergometer was 2562.71ml/min, and the one based on the treadmill was 2874.89ml/min for the college male students of Korea. Lifting activities with four different lifting frequencies(2, 5, 8, 11 lifts/min) for one lifting range from floor to 76cm height were studied. The oxygen consumption rates and the heart rates were measured or recorded while subjects were lifting the weight of RWL, AL, and MPL. The heart rates and the oxygen consumption rates increased as the frequency increased from 2 to lifts/min. However, those slightly decreased at the frequency of 11 lifts/ min. The measured oxygen consumption rates were ranging from 2.3% to 29.6% higher than the physiological criteria 620, 700, and 1000ml/min, respectively, of the RWL, AL, and MPL for all the lifting frequencies (5, 8, 11 lift/min) except 2 lifts/min. It si suggested that the physiological criteria of NIOSH Guideline should be based on the lifting PWC, which can consider the type of lifting activity and the frequency of the task, rather than using the PWC by ergometer or treadmill. The measured oxygen consumption rates were ranging from 13.26% to 40.11% higher than the values estimated using the models by Garg and Kim. From these findings it is suggested that the NIOSH Equation should not be directly applied to Korean without resonable modifications.

Generation of Reactive Oxygen Species by Nonenzymatic Reaction of Menadione with Protein Thiols in Plasma (Menadione과 Plasma내의 Protein Thiol의 비효소적인 화학반응에 의한 활성산소 생성)

  • Quinones have been reported to undergo nonenzymatic reaction with thiols to generate reactive oxygens. It is therefore possible that the nonenzymatic reaction of quinones with thiols in plasma could lead to potentJared cellular toxicity or disease. When 1 mM menadione was added in plasma under pH 11.2, 7.4 and 5.0, the increase in oxygen consumption rate was the order of pH 11.2 > pH 7.4 > pH 5.0. In addition, oxygen consumption rates under plasma anticoagulated with trisodium citrate solution (pH 7.85) was significantly higher than those with acid-citrate-dextrose solution (pH 6.87). SOD and catalase reduced the rate of oxygen consumption induced by menadione in plasma. Taken together, these results suggest that the menadione-induced increased oxygen consumption was due to nonenzymatic reaction of menadione with thiols in the plasma. The presence of plasma has an additive effect on the increased oxygen consumption rates induced by the menadione treatments on our model tissue, platelets, as compared between washed platelet (WP) and platelet rich plasma (PRP). Cytotoxicity, as determined by LDH release, are well correlated with the oxygen consumption rates observed in each system and strongly suggest that menadione-induced cytotoxicity can be increased with the presence of blood plasma.

Respiration Rate and Oxygen Intake by Change of Wheelchair Backrest Angle

  • This study was purposed to provide basic information on the correct application of a wheelchair's backrest angle by investigating the change in cardiopulmonary function according to backrest angle during propulsion. This study examined the effects of the wheelchair's backrest angle on the cardiopulmonary function by varying the angle to $0^{\circ}$, $10^{\circ}$ and $20^{\circ}$ with a propulsion velocity of 60 m/min. The experimental parameters were respiration rate, oxygen consumption rate and oxygen consumption rate/kg which were measured by a portable wireless oxygen consumption meter (COSMED, $K4b^2$). The results of the study were as follows: 1) There were no statistically significant differences in respiration rates due to changes in the wheelchair backrest angle (p>.05). 2) There were statistically significant differences in oxygen consumption rates due to changes in the wheelchair backrest angle (p<.05). 3) There were also statistically significant differences in the oxygen consumption rate/kg due to changes in the wheelchair backrest angle (p<.05). In conclusion, changes in the backrest angle of wheelchairs during propulsion influences oxygen consumption rates and heart rates, while respiration rates are not affected. Therefore, a training program for good seating and posture needs to be provided, and the wheelchair seating system should be equipped with the unadjustable-angle wheelchair to reduce the functional load on the cardiopulmonary system.

Respiration Rates of Individual Bovine In Vivo-Produced Embryos Measured with a Novel, Scanning Electrochemical Microscopy (Scanning Electrochemical Microscopy를 이용한 한우 체내 수정란의 호흡률 조사)

  • Oxygen consumption is a useful parameter for evaluating mammalian embryo quality, since individual bovine embryos was noninvasively quantified by scanning electrochemical microscopy (SECM). Recently, several approaches have been used to measure the oxygen consumption rates of individual embryos, but relationship between oxygen consumption and pregnancy rates of Hanwoo following embryo transfer has not yet been reported. In this study, we measured to investigate the correlation between oxygen consumption rate and pregnancy rates of Hanwoo embryo using a SECM. In addition to, the expression of pluripotent gene and anti-oxidant enzyme was determined using real-time PCR by extracting RNA according to the oxygen consumption of in vivo embryo. First, we found that the oxygen consumption significantly increased in blastocyst-stage embryos (blastocyst) compared to early blastocyst stage embryos, indicating that oxygen consumption reflects the embryo quality (Grade I). Oxygen consumption of blastocyst was measured using a SECM and total cell number of in vitro blastocyst was enumerated by counting cells stained by propidium iodide. The oxygen consumption or GI blastocysts were significantly higher than those of GII blastocysts ($10.2{\times}10^{15}/mols^{-1}$ versus $6.4{\times}10^{15}/mols^{-1}$, p<0.05). Total cell numbers of in vitro blastocysts were 74.8, 90.7 and 110.2 in the oxygen consumption of below 10.0, 10.0~12.0 and over $12.0{\sim}10^{15}/mols^{-1}$, respectively. Pregnant rate in recipient cow was 0, 60 and 80% in the transplantation of embryo with the oxygen consumption of below 10.0, 10.0~12.0 and over $12.0{\times}10^{15}/mols^{-1}$, respectively. GPX1 and SOD1 were significantly increased in over -10.0 group than below 10.0 groups but in catalase gene, there was no significant difference. On the other hand, In OCT-4 and Sox2, pluripotent gene, there was a significant difference (p<0.05) between the below-10.0 ($0.98{\pm}0.1$) and over 10.0 ($1.79{\pm}0.2$). In conclusion, these results suggest that measurement of oxygen consumption maybe help increase the pregnant rate of Hanwoo embryos.
