  • Reactive oxygen species are highly reactive oxidant molecules and react with cellular components, causing oxidative damages. These damages may play a significant role in the causation of several chronic diseases. Antioxidative defence systems are present in the body to protect cells from oxidative damages. The first line of defence include dietary antioxidants and enzymes such as superoxide dismutase. Cynanchum wilfordii Hemeseley has been used for centuries as a tonic nutraceutical in China and Korea.   • [$SiO_2$] film coated as a passivation layer for YBCO based electronic devices is investigated by measuring the micro wave properties of micro strip line resonators. The $SiO_2$ film coated resonators are compared with coated resonators for two degradation conditions, a $200^{\circ}C$ annealing in air and an exposure to air at $85^{\circ}C\;85\%$ relative humidity. The $SiO_2$ film reduces the YBCO thin film degradation caused by oxygen stoichiometry change and reaction with water.

Dust particles-induced intracellular Ca2+ signaling and reactive oxygen species in lung fibroblast cell line MRC5

  • Epidemiologic interest in particulate matter (PM) is growing particularly because of its impact of respiratory health. It has been elucidated that PM evoked inflammatory signal in pulmonary epithelia. However, it has not been established $Ca^{2+}$ signaling mechanisms involved in acute PM-derived signaling in pulmonary fibroblasts. In the present study, we explored dust particles PM modulated intracellular $Ca^{2+}$ signaling and sought to provide a therapeutic strategy by antagonizing PM-induced intracellular $Ca^{2+}$ signaling in human lung fibroblasts MRC5 cells. We demonstrated that PM10, less than $10{\mu}m$, induced intracellular $Ca^{2+}$ signaling, which was mediated by extracellular $Ca^{2+}$. The PM10-mediated intracellular $Ca^{2+}$ signaling was attenuated by antioxidants, phospholipase blockers, polyADPR polymerase 1 inhibitor, and transient receptor potential melastatin 2 (TRPM2) inhibitors. In addition, PM-mediated increases in reactive oxygen species were attenuated by TRPM2 blockers, clotrimazole (CLZ) and N-(p-amylcinnamoyl) anthranilic acid (ACA). Our results showed that PM10 enhanced reactive oxygen species signal by measuring DCF fluorescence and the DCF signal attenuated by both TRPM2 blockers CLZ and ACA. Here, we suggest functional inhibition of TRPM2 channels as a potential therapeutic strategy for modulation of dust particle-mediated signaling and oxidative stress accompanying lung diseases.

75톤급 액체로켓엔진 연소시험에서의 액체산소 공급부 예냉특성 고찰 (Investigation of Chill Down Characteristics of Liquid Oxygen Feeding System in 75 Tonf-class Liquid Rocket Engine Firing Test)

  • 한국형발사체의 1단 및 2단 엔진으로 사용될 75톤급 액체로켓엔진의 연소시험이 엔진 연소시험설비에서 수행되었다. 이 엔진은 산화제로 극저온 유체인 액체산소를 사용하므로, 연소시험을 위해서는 필수적으로 설비의 공급 배관 및 엔진의 예냉과정이 진행되어 주어진 엔진 입구 온도 및 압력 요구조건이 만족되어야 한다. 따라서 향후 효율적인 시험 운용을 위해서는 시험 시 설비와 엔진에서의 예냉특성 및 예냉과정에서 소모되는 액체산소의 양을 파악하는 것이 중요하다. 이 논문에서는 75톤급 액체로켓엔진의 연소시험에서 예냉과정의 고찰을 통하여 런탱크 가압 전, 후 각 단계에서의 설비배관 및 엔진의 예냉특성을 평가하였으며, 시험 시 소모되는 액체산소의 양을 평가하였다.

Aloe 세포 배양계에서 $CO_2,\;O_2,\;C_2H_4$ gas가 세포 생장 및 다당체 생산에 미치는 영향

  • 본 연구에서는, gas 성분의 조성을 on-line으로 자동제어할 수 있는 시스템을 확립하고, 이를 이용하여 각 성분들이 식물세포에 미치는 영향에 대해 조사하였다. 실험결과, $CO_2$가 첨가되었을 때, 세포의 생장에는 큰 차이를 볼 수 없었으나, 2차 대사물 및 polysaccharide의 생산성을 증대시켰다. 특히, polysaccharide의 경우, $CO_2$가 참가되었을 때 1.8배의 생산성 증대를 나타내었다.

  • The first line of antioxidant defense against reactive oxygen species includes the enzymatic activity of the superoxide dismutase (SOD) that catalyzes the disproportionation of superoxide to hydrogen peroxide and water. The SOD mainly removes highly toxic $O_2$$^{[-10]}$ and also prevents $O_2$$^{[-10]}$ mediated reduction of iron and subsequent OH$^{[-10]}$ generation. Along with an interest in SOD as a first line of defense against damage mediated by the superoxide anion, the SOD1 enzyme has been subjected to investigation in the molecular and cellular level.   • Bioreactor을 좀더 효율적으로 운전하기 위하여 여러 가지 gas 성분의 조성을 on-line으로 자동제어 및 분석할 수 있는 시스템을 연구하였으며, 이 시스템을 이용하여 각 gas 성분이 식물세포에 미치는 영향을 여러 관점에서 비교 ${\cdot}$ 분석하였다. 실험결과, 산소는 exponential phase을 증가시켰으며, 이산화탄소와 ethylene은 lag phase을 줄여주는 결과를 보였다. 이산화탄소는 2차대사물질인 aloin의 생산량을 증대시켰으며, 다당의 경우를 보면 현저한 생산량의 차이(약 1.8배)를 나타내었다. Ethylene을 주입한 샘플에서는 glucose 소비속도가 매우 빨랐으며, aloin의 생산량을 높여주는 결과를 보였다.

  • Upon a turbopump's running, cavitation may occur at the inlet of the LOx pump by pressure drop and heat transfer along the LOx feeding line. Since the cavitation can cause serious damage to the pump or to stop running, the absence of the cavitation at the inlet of a turbopump should be confirmed before the using the turbopump. In the present study, the calculation of the volume fraction of LOx gas phase at the inlet of the pump are performed with different temperatures of LOx in the tank, pressure drops and heat transfers along the feeding line. This calculation method can be applied to define the limits of thermal and hydraulic characteristics during the design of a LOx feeding system.
