Novel non-apoptotic cell death: ferroptosis (새로운 non-apoptotic 세포사멸: ferroptosis)

  • Ferroptosis is a newly recognized type of cell death that results from iron-dependent lipid peroxidation and is different from other types of cell death, such as apoptosis, necrosis, and autophagic cell death. This type of cell death is characterized by mitochondrial shrinkage with an increased mitochondrial membrane density and outer mitochondrial membrane rupture. Ferroptosis can be induced by a loss of activity of system $X_c{^-}$ and the inhibition of glutathione peroxidase 4, followed by the accumulation of lipid reactive oxygen species (ROS). In addition, inactivation of the mevalonate and transsulfuration pathways is involved in the induction of ferroptosis. Moreover, nicotinamide adenine dinucleotide phosphate oxidase and p53 promote ferroptosis by increasing ROS production, while heat shock protein beta-1 and nuclear factor erythroid 2-related factor 2 inhibit ferroptosis by reducing iron uptake. This article outlines the molecular mechanisms and signaling pathways of ferroptosis regulation, and explains the roles of ferroptosis in human disease.

Secretion of the cloned serratia marcescens nuclease in escherichia coli (Serratia marcescens nuclease의 escherichia coli에서의 분비)

  • Secretion of Serratia marcescens nuclease by E. coli harboring pNUC4 was investigated. 29.2, 54.2 and 16.6% of total nuclease were observed in culture medium, periplasm, and cytoplasm of E. coli, respectively. To investigate the secretion mechanism of Serratia nuclease by E. coli, secretion kinetics of nuclease was examined in the presences of sodium azide, and energy metabolism inhibitor; procaine, an exoprotein processing inhibitor; and chloramphenicol, a protein synthesis inhibitor. In the presence of sodium azide, periplasmic unclease was gradually decreased and the extracellular nyclease was linearly increased according to the incubation time. Similar results were obtained in presences of procaine and chloramphenicol. From these results, we concluded that two transport processes are involved in nuclease secretion: secretion of nuclease through the inner membrane is occurred by an energy-dependent process and probably requiring precusor processing: secretion of nuclease through outer membrane does not require energy, de novo protein synthesis, and precursor processing.

Scanning Electron Microscopic Study on the Tissue Mast Cells of Mammals (포유류 조직비만세포의 주사전자현미경적 연구)

  • A Scanning electron microscope which can obtain additional information not readily available with either the light or transmission electron microscope was used to study the mast cell shape and its granules in normal mammal tissue(rat mesentery, stomach and mouse stomach) by fretting cut using liquid nitrogen. The results showed that rat mesentery and mouse stomach mast cell surfaces had no ridges and microvilli, but revealed several microvilli projecting into the surrounding connective tissue in the rat stomach mast cell. The shape of the mast cell varied from discoid(in the rat mesenteric mast cell) to ellipsoid (rat and mouse stomach), ranging from 7.5 to $10{\mu}m$ in diameter. The shape of the nucleus was ellipsoid and nucleic membrane was adherent to the outer surface of the granules. The granules, approximately 0.2 to $0.9{\mu}m$ in diameter, were various shapes. Frequently, rounded protrusions of cytoplasmic granules could be discerned under the cell membrane. Many small granules were seen in the cytoplasm.

  • The pro-apoptotic BCL-2 family protein BID activates BAK and/or BAX, which form oligomeric pores in the mitochondrial outer membrane. This results in the release of cytochrome c into the cytoplasm, initiating the apoptotic cascade. Here, we utilized liposomes encapsulating sulfo-rhodamine at a controlled temperature to improve upon a previously reported assay system with enhanced sensitivity and specificity for measuring membrane permeabilization by BID-dependent BAK activation. BAK activation was inhibited by BCL-$X_L$ protein but not by a mutant protein with impaired anti-apoptotic activity. With the assay system, we screened a chemical library and identified several compounds including trifluoperazine, a mitochondrial apoptosis-induced channel blocker. It inhibited BAK activation by direct binding to BAK and blocking the oligomerization of BAK.

  • The effects of calcium channel blockers (CAB's) verapamil, diltiazem and nicardipine, on erythrocyte ghost membranes have been studied. Using the fluorospectroscopic method, it was observed that the fluidity of the inner layer of ghost membranes was increased with an increase of drug concentrations but did not any changes in the fluidity of the outer layer. These drugs showed protectuve effect against hypotonic hemolysis of erythrocytes. Thus, the expansion of surface area in response to corpuscular volume of erythrocytes in the presence of CAB's is seemed to play an important role in protecting hypotonic hemolysis of erythrocytes.

Effects of n-Alkanols on the Lateral Diffusion of Total Phospholipid Fraction Extracted from Brain Membranes

  • We investigated the effects of n-alkanols on the range and rate of the lateral diffusion of 1, 3-di(1-pyrenyl)propane in the model membranes of total phospholipid fraction extracted from synaptosomal plasma membrane vesicles. n-Akanols increased the range and rate of the lateral diffusion of 1, 3-di(1-pyrenyl)propane in the bulk model membrane structures (inner + outer monolayers) and the potencies of n-alknols up to 1-nonanol increased by 1 order of magnitude as the carbon chain length increases by two carbon atoms. The cut-off phenomenon was reached at 1-decanol, where further icnrease in hydrocarbon length resulted in a decrease in the lateral diffusion. However, significant changes in the 1'/1 value were not observed by methanol (from 100 to 2500 mM), ethanol (from 25 to 800 mM), and 1-propanol (from 10 to 250 mM) over entire concentration.

  • The glandular secretory system of the capitate gandular trichomes in leaf of Thymus quinquecostatus Celakovsky was examined by transmission electron microscope. The glandular trichome was consisted of three cell layers; an basal cell layer, a stalk cell with single-celled intermediate layer and a discoid secretory layer with thickened cuticle. The secretory cell was dense, rich in mitochondria, rER, plastds, Golgi complex and had many vesicular structure. Typical plastids with reticulate body and plastoglobule were present in glandular trichome. The tytoplasm of secretory cell was filled with osmiophilic secretory materials. The secretory vesicles, originated from Golgi complex, appeared as membrane bounded vesicles and secreted to the outer wall surface. The presences of well developed rER, mitochondria, Golgi complex, and membrane-bounded vesicles fused with plasmalemma in the secreting cells indicate that the granulocrine mechanism of secretion was occurring in T. quinquecostatus. Subcuticular cavity was developed between the cuticular layer and the secretory cell wall, and it formed above the secretory cell upon separation of cuticle-wall.

Extraction of Chromium (Vl) Son in Waste Water through the Liquid Surfactant Membrane with Open-Type Perforated Reciprocating-Plate Column (개방형 왕복 다공판 추출탑을 이용한 액막법에 의한 폐수중 Cr(Vl)의 추출)

  • The objective of this study was to investigate the extraction of chromium(Vl) ion in waste water through the liquid surfactant membrane with open-type Perforated Reciprocating-Plate Column. Extraction experiments were conducted to measure the effect of flow characteristics of continuous and dispersed phase and stroke velocity, sodium hydroxide concentraction in internal aqueous phase, sulfuric acid concentraction in outer phase, and residence time distribution and measured extraction velocity. The result of experiments showed that extraction velocity of chromium ion was maximum when stroke velocity was 180 1/min and dispersion phase velocity was 30m11min, continuous phase velocity was 20m1/min. Extraction velocity of chromium ion increased with increasing difference of hydrogen ion concentraction of dispersion and continuous phase and column stage decreased.
