The Effect of Kamijoaguiem on Osteoporosis Induced by Ovariectomy in Rats (가미좌귀음(加味左歸飮)이 난소적출(卵巢摘出)로 유도(誘導)된 백서(白鼠)의 골다공증(骨多孔症) 미치는 영향(影響))

  • Purpose : This study was peformed to evaluate the effect of Kamijoaguiem(JGE) on the bone mass and its related factors. Methods : We used ovariectomized rat as an estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 7th day after operating ovariectomy, rats were administered with JGE per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of JGE. Results : Bone density, bone ash density, bone calcium, magnesium and phosphorus was decreased in osteoporotic rats. JGE improved the decreased bone density, bone ash density and the decreased bone magnesium, but JGE didn't improve the decreased bone calcium and phosphorus in osteoporotic rats. Osteocalcin in serum and hydroxy-proline excretion in urine were increased in osteoporotic rats. Their levels were decreased when JGE was administered. ALP activity in serum was increased in osteoporotic rats. JGE didn't induce any significant changes. JGE showed significant increase in serum calcium level, total protein level, albumin level, BUN level, serum LDH activity. JGE didn't show significant increase in serum T-cholesterol density, triglyceride density, HDL-cholesterol density. JGE didn't show significant increase in RBC number, hemoglobin level, platelet number, hematocrit level. JGE showed inhibitory effect on the degradation of bone-matrix in osteoporotic rats, in histological examination to Hematoxylin-eosin stain. Conclusion : JGE might improve bone density due to inhibition of bone resolution in osteoporotic rats. It suggest that JGE may be useful prescription in osteoporosis.

Effect of Excess Calcium and Iron Supplement on Bone Loss, Nephrocalcinosis and Renal Function in Osteoporotic Model Rats (골다공증 모델 흰쥐에서 칼슘과 철 보충제의 과다섭취가 골격손실과 신석회침착 및 신장기능에 미치는 영향)

  • This study examined the effects of excess intake of calcium (Ca) and iron (Fe) supplement on bone loss, nephrocalcinosis and renal function in osteoporotic model rats. Seven-week-old female rats were first fed a Ca-deficient diet for four weeks after ovariectomy operation, and then one of nine experimental diets for additional eight weeks, containing three levels of Ca, normal (0.5%) or high (1.5%) or excess (2.5%) and three levels of Fe, normal (35ppm) or high (210ppm) or excess (350ppm). The osteoporotic model rats showed a remarkable increase in body weight, serum alkaline phosphatase (ALP) and decrease in breaking force, Ca, P, Mg contents of femur. Serum Ca concentration was not significantly affected by dietary Ca and Fe levles. Liver Ca content increased in rats fed a high-and excess-Ca diet. Kidney Ca content and microscopic Ca deposition remarkably increased in osteoporotic model rats compared to control group, and showed a tendency to decrease in rats fed a excess-Ca diet. Breaking force of femur increased with increasing dietary Ca levels, but Ca, P contents of femur and serum ALP were not significantly affected by dietary Ca and Fe levels. Serum total protein decreased in rats fed a excess-Ca diet, BUN increased in rats fed a excess-Ca diet, while serum uric acid and creatinine were not significantly affected by dietary Ca levels. Urinary creatinine, GFR increased in rats fed a high-and excess-Ca, diet, and GFR was highest in rats fed a excess-Ca/excess-Fe diet. These results suggest that excess intake of Ca may increase breaking force of femur, but not increase mineral contents of femur, and decrease kidney function. Furthermore, excess intake of Fe and Ca concurrently may aggravate kidney function leading to potential health problems in ovariectomized osteoporotic model rats.

  • Osteoporosis is defined as a decrease in bone mass that leads to an increased risk of fracture. The therapeutic effect of $1{\alpha}$,25 dihydroxycholecalciferol, the hormonal form of vitamin $D_3$ that mediates calcium translation in intestine and bone, on the healing process of fracture has still been controversial. These studies were designed to understand the healing process of normal fibular fracture, the osteoporotic changes after ovariectomy, and the therapeutic effects of $1{\alpha}$,25 dihydroxycholecalciferol on the osteoporotic fracture in rats. The simple transverse fractures of rat fibulae were produced with a rotating diamond saw. The changes of the biochemical and mechanical indices of rats were investigated. The mechanical study based on bending test revealed the healing of the fibular fracture in the 5th week after simple transverse fracture. The osteoporosis impaired more the healing of osteoporotic fibular fracture than normal non-osteoporotic fibular fracture. The healing process of osteoporotic fracture was facilitated by the treatment with $1{\alpha}$,25 dihydroxycholecalciferol, however, was delayed more than the healing process of normal fracture. The bone strength based on the bending test also confirmed this tendency. The bone strengths in the 5th week after fracture of normal bone, osteoporotic bone, and $1{\alpha}$,25 dihydroxycholecalciferol-treated osteoporotic bone were 75%, 41%, and 67%, respectively, in comparison with those of intact bone. In conclusion, $1{\alpha}$,25 dihydroxycholecalciferol was effective in promoting the osteoporotic fracture healing.

EXPERIMENTAL STUDY OF PERI-IMPLANT TISSUE REACTION IN OVARIECTOMIZED OSTEOPOROTIC RATS (난소절제로 유도된 골다공증 흰쥐에서 implant 주위 조직 반응에 관한 실험적 연구)

  • This study was designed to investigate the peri-implant tissue reaction in ovariectomized osteoporotic female rats, and to evaluate effects of estrogen, calcitonin, parathyroid hormone on the bone - implant interface in osteoporotic rats. 120 Sprague - Dawley rats were used in this experiments. Osteoporosis was induced by bilateral ovariectomy. They were divided 5 groups : sham-operated control group(Sham), ovariectomized group (OVX), OVX and estrogen treated group (OVX+E), OVX and PTH treated group (OVX+PTH), and OVX and calcitonin treated group (OVX+CT). Eight weeks after ovariectomy, two titanium screw implants were inserted into the left tibia of each rat. Eight weeks after the insertion of the implants, the periotest values (PTV) of implant were examined, and the rats were sacrificed, and examined the reaction of bone tissue surrounding the implant both histologically and histomorphometrically. The bone density and ash weight of opposite right tibia were examined. Over 40 rats were fractured on left tibia that was implant inserted. On histologically finding, all groups were osseointegrated well, especially in OVX+PTH group. In OVX group, tibial cortical bone showed many large harversian canal and microfracture lines. The OVX+PTH group showed the lowest mean PTV (-2.33) (p<0.05), and the hightest mean bone - implant contact percentage (89%) (p>0.05). But the OVX+CT group showed the highest mean bone density ($5.45mg/cm^3$) and ash weight (56.12%) (p<0.05). The results indicate that PTH treatment enhances osseointegration of implant in OVX rats, and CT treatment depresses bone turnover and prevent the development of osteopenia in OVX rats.

Effects of $1\alpha$, 25 Dihydroxycholecalciferol on Osteoporotic Fracture : Light Microscopic and Scanning Electron Microscopic Observation ($1\alpha$, Dihydroxycholecalciferol의 골다공증성 골절 치유효과 : 광학현미경 및 주사전자현미경적 관찰)

  • Vitamin D is one of important factors involved in the regulation of bone metabolism. In osteoporosis, the therapeutic effect of vitamin D on the healing process of fracture has still been controversial. These studies were designed to understand the healing process of normal fibular fracture and the therapeutic effects of $1\alpha$, 25 dihydroxycholecalciferol on the osteoporotic fracture in rats. The histological and ultrastructural studies revealed the healing of the fibular fracture in the 5th week after simple transverse fracture. The osteoporosis impaired more the healing of osteoporotic fibular fracture than normal non-osteoporotic fibular fracture. The healing process of osteoporotic fracture was facilitated by the treatment with $1\alpha$, 25 dihydroxycholecalciferol, however, was delayed more than the healing process of normal fracture. These results suggest that $1\alpha$, 25 dihydroxycholecalciferol was effective for reducing the deleterious effects of osteoporosis in fracture healing.

  • Objective & Methods : The purpose of this study is to observe the effects of Cyperi Rhizoma Herbal-acupuncture infusion solution at Umgok($KI_{10}$) on Osteoporotic Rats Induced by Ovariectomy. The author performed several experimental items to analyze cytotoxic, osteoporosis evaluation, change of ALP, osteocalcin, Ca and histological change of tibia. Result : 1. BMD were increased non-meaningly in CR-HA than control group. 2. ALP was not inhibited by CR-HA, osteocalcin was decreased meaningly in CR-HA than control group. 3. Osteoclast like cell in tibia was increased meaningly in control group, decreased meaningly in CR-HA. 4. In the histological study in tibia, TBV was significantly increased, GPL was significantly decreased in the CR-HA than control group. Conclusions : From the result above, the results suggest that CR-HA at $KI_{10}$ has significant effect on osteoporosis, and to be put to practical use in the future osteoporosis clinic.

  • Objectives : The purpose of this study is to observe the effects of RUBI FRUCTUS phamacopuncture infusion solution at $KI_{10}$ on osteoporotic Rats Induced by ovariectomy. Methods : The author performed several experimental items to analyze cytotoxic, osteoporosis evaluation, change of ALP, creatinine, Ca, P, osteocalcin, Ca/P ratio and histological change of serum and tibia. Results : 1. Creatinine and phosphorus in serum was decreased significantly in RF-HA than control group. 2. Calcium and phosphorus in tibia was increased significantly in RF-HA than control group. 3. Osteoclast like cell in tibia was increased significantly in control group, decreased significantly in RF-HA. 4. In the histological study in tibia, TBT was significantly increased, GPL was significantly decreased in the RF-HA than control group. Conclusions : This study suggested that RF-HA at KI10 has significant effect on osteoporosis.

Histological and Immunohistological Studies of the Thyroid Glands of the Osteoporotic Rats Induced by Ovariectomy (난소절제로 유발된 골다공증 시 흰쥐 갑상샘에 대한 조직학적 및 면역조직화학적 연구)

  • In order to investigate the histological and immunohistochemical changes of thyroid glands, and serum osteocalcini estradiol, pmgesterone, follicular-stimulating hormone (FSH), and calcium levels in the osteoporotic rats induced by ovariectomy, normal(sham-operated group) and experimental groups(ovariectomized group) were divided. The experimental groups were subdivide6 to 5, 10, 15, and 20 weeks after ovariectomized groups. The histological structure of the thyroid gland were observed in normal structure throughout all experimental periods but the calcitonin-immunoreartive cells in the experimental groups were significantly decreased compared with normal groups and degranulations of calcitonin-immunoreactive cells were observed. Theses numbers were decreased with time after ovariectomy. However, the serum osteocalcin and FSH levels of experimental groups were significantly increased compared with normal groups and these levels were increased along with time after ovariectomy, but serum estradiol, progestemn and calcium levels of experimental group were significantly decreased compared with normal groups and these levels were decreased with time after ovariectomy.

Effects of the Gamidokwalgisaeng-tang(GDS) Oral Administration and Electro-acupuncture at Chishil$(BL_{52})$, Hyonjong$(GB_{39})$ on Osteoporotic Rats Induced by Ovariectomy (가미독활기생탕(加味獨活寄生湯)의 경구투여(經口投與)와 지실(志室).현종(懸鍾) 전침(電鍼) 병용(倂用) 시술(施術)이 흰 쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objectives : This study was carried out to determine the effects of Gamidokwalgisaeng-tang(GDS) oral administration and electro-acupuncture at $BL_{52}$ & $GB_{39}$ on the postmenopausal osteoporosis. Methods: In order to study the effects of the Gamidokwalgisaeng-tang(GDS) oral administration and electro-acupuncture at $BL_{52}$, $GB_{39}$ on osteoporotic rats induced by ovariectomy, body weight, femur weight, uterine weight, serum level of osteocalcin, alkaline phosphatase(ALP), cholesterol, triglyceride, estradiol, androstenedione, testosterone and urinary excretions of calcium & phosphorus were measured. Results : 1. Body weight in all experimental group decreased significantly in comparison with ovariectomized group(OVX). 2. The weight of femur compared with body increased significantly in GDS oral administration and combination treatment group in comparison with ovariectomized group(OVX). 3. Uterine endometrial morphology is caused by ovariectomy and the weight of uterus decreased significantly in control and all experimental group in comparison with sham. 4. The serum level of osteocalcin decreased in all experimental group in comparison with OVX, but significantly in electro-acupuncture and combination treatment group. 5. The serum alkaline phosphatase(ALP) activity decreased in electro-acupuncture and combination treatment group in comparison with OVX, but only significantly in electro-acupuncture group. 6. The serum level of cholesterol increased significantly in OVX in comparison with sham and decreased in all experimental group in comparison with OVX, not insignificantly. The serum level of triglyceride increased in all experimental group in comparison with OVX, not insignificantly. 7. Urinary excretions of calcium and phosphorus decreased significantly in all experimental group in comparison with OVX. Conclusion : We suggest that GDS oral administration and electro-acupuncture at $BL_{52}$ & $GB_{39}$ are available for prevention and curing about the postmenopausal osteoporosis.
