골다공증 모델 흰쥐에서 고수준의 단백질의 칼슘 섭취가 칼슘대사 및 신장기능에 미치는 영향 (Effects of High Protein and Calcium Intakes on Calcium Metabolism and Renal Function in Ovariectomized Osteporosis Rat Model)

  • This study was to investigate interaction between dietary protein and Ca levels in Ca metabolism and renal function in osteporosis rats. Five week-old female rats were fed a low Ca diet for 4 weeks after ovariectomy operation to establish rat models of osteoporosis. The ovariectomized osteoporosis rats were divided into six groups and were fed experimental diets which contained two levels of protein, normal (20%) and high(40%) , and three levels of Ca, low (0.06%), normal (0.47%) and high(0.94%) for 4 weeks , respectively. The ovaricetmized rat model of osteoporosis showed a remarkable decrease in serum Ca concentration, fresh weight and breaking force of femur, Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca and P contents of femur, and apparent absorption and retention of Ca. The supplementations of Ca at the dietary levels of normal and high levels significantly enhanced Ca bioavailability shown in the above experimental rat models of osteoporosis, regardless of dietary protein levels ; whereas the rats which were fed the low Ca diet demonstrated rather a decrease in its bioavailability. Irrespectively of the dietary Ca levels, the rats which were fed high protein diet exhibited an increase in kidney weight, urinary Ca, volume and hydroxyproline, and glomerular filtration ratio(GFR). The results show that dietary protein and calcium levels affect the renal function and Ca metabolism independently, while the interaction between protein and calcium have not been shown.

연교의 파골세포 분화 및 골 흡수 억제 기전 연구 (Forsythiae Fructus Extract Inhibits RANKL-Induced Osteoclast Differentiation and Prevent Bone Loss in OVX-Induced Osteoporosis Rat)

  • Objectives : Osteoporosis is a condition characterized by low bone mass and increased bone fragility. It has become a major problem of senior citizens. The purpose of this study is to experiment the effect of water extract of Forsythiae Fructus (wFF) on osteoclast differentiation; and the other purpose is to examine the effect of wFF on osteoporosis in ovariectomized rat. Methods : To investigate the effect of wFF on osteoclast differentiation and activity, RAW 264.7 cells were used. The number of TRAP positive cell, TRAP activity, pit area, mRNA expression of makers (RANK, TRAP, CA II, CTK, MMP-9, NFATc1, c-Fos), protein expression of makers (NFATc1, c-Fos) were investigated. For in vivo study, 40 female Sprague-Dawley (SD) rats were induced osteoporosis by ovariectomy (OVX) and then tested for anti-osteoporosis effect by administration of wFF. Results : wFF suppressed osteoclatogenesis, TRAP activity and pit area formation. Moreover, wFF decreased the expression of master differentiation factors (NFATc1, c-Fos) and also reduced the osteoclastogenesis-related markers (TRAP, CA II, CTK, MMP-9). These suggest that wFF inhibit osteoclasts differentiation and bone resorption. In the OVX rat model, wFF inhibited decreasing of BMD and trabecular area. Conclusions : Forsythiae Fructus should be effective for osteoporosis prevention and treatment.

  • Objectives: In this study, the author aimed to evaluate the effect of BuOH fraction(YK) from Cinnamomum loureirii on osteoblast proliferation in murine rat calvarial cells. Methods: The osteoblast separated from murine calvariae was cultivated for 10 days and evaluated the cell function. After the addition of YK on the culture medium, we determined the effect of YK on the cell proliferation, alkaline phosphatase activity, apoptosis of the cultivated osteoblast, protein synthesis and collagen synthesis. Results: YK increased the proliferation of rat calvarial osteoblast. YK increased ALP activity of rat calvarial osteoblast. YK did not change the survival rate of rat calvarial osteoblast. YK increased protein synthesis of rat calvarial osteoblast. YK increased collagen synthesis of rat calvarial osteoblast. Conclusions: This study suggests that YK might improve the osteoporosis resulted from augumentation of osteoblast proliferation.

녹용 추출물 투여가 콜다공증 유발 Rat 에 미치는 효과에 관한 연구 II. 녹용 추출물 투여가 골다공증 유발 Rat 의 체중, 골회분량, 대퇴 및 장기중량 및 조직상의 변화에 관한 연구 (Studies on the Effects of Antler Extract in Osteoporosis-Induced Rats II. Effect of Antler Extract on Body Weight It, Femur Weight It, Bone Ash Quantity, Organ Weight and Histological in Osteoporosis-Induced Rats)

  • 본 연구는 골다공증유발 rat에 녹용추출물의 투여가 예방 및 치료효과에 미치는 영향을 구명하고자, 양측 난소를 적출한 후 골다공증이 유발된 rat에 암, 수녹용 추출물 525, 1,250 mg/kg을 투여했을 때 체중, 대퇴골의 중량, 장기중량, 골회분량의 측정 및 조직학적, 전자현미경적 검사 등을 실시하였다. 1. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 체중은 정상대조군에 비하여 유의한 증가를 나타냈다 (p<0.05). 2. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 우측, 좌측의 femur 중량은 정상대조군에 비하여 감소하였다. 3. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 골회분량은 정상대조군에 비하여 약간 감소된 치를 나타냈으나 유의성은 인정되지 않았다. 4. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 간과 비장 및 신장의 각각 정상대조군에 비하여 감소된 치를 나타냈다. 5. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 조직학적 및 전자현미경적 소견은 정상대조군에서는 소주골간의 연결이 비교적 잘 유지되어 있고 골소실을 발견할 수 없었으나 난소적출군은 정상군에 비하여 소주골이 가늘어졌거나 소주골간 연결부분의 대다수가 끊어져 골양이 많이 소실되었음이 관찰되었으며, 녹용처리군에서는 소주골의 형태가 굵고 연결부분이 잘 유지되어 정상군과 유사한 소견이 관찰되었다. 위의 결과들에서 암, 수녹용 추출액은 난소제거후의 골소설을 억제하므로 골다공증의 예방과 치료에 효과가 있는 것으로 판단된다.

  • Objectives: In this study, the author aimed to evaluate the effect of EtOH extract of Ganoderma lucidum (GLE) on osteoblast proliferation in rat fetus calvarial cells. Methods: The osteoblast separated from rat fetus calvariae was cultivated for 6~21 days and evaluated the cell function. After the addition of GLE on the culture medium, we determined the effect of GLE on the cell viability, cell proliferation, bone matrix protein synthesis, alkaline phosphatase (ALP) activity, collagen synthesis and calcified nodule formation of the cultivated osteoblast. Results: GLE did not change the survival rate of rat calvarial osteoblast. GLE increased the proliferation of rat calvarial osteoblast. GLE increased ALP activity of rat calvarial osteoblast. GLE increased bone matrix protein synthesis of rat calvarial osteoblast. GLE increased collagen synthesis of rat calvarial osteoblast. GLE slightly affected calcified nodule formation of rat calvarial osteoblast. Conclusions: This study suggests that Ganoderma lucidum might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

건지황(乾地黃) 추출물이 Rat fetus 두개골로부터 분리한 조골세포에 미치는 영향 (The Effect of Dried Roots of Rehmannia glutinosa Extract on Osteoblast in Rat Fetus Calvarial Cells)

  • Objectives: Osteoporosis is characterized by bone loss and morbidity with osteoporotic fracture. In this study, the author aimed to evaluate the effect of dried roots of Rehmannia glutinosa extract (RGE) on osteoblast proliferation in murine calvarial cells. Methods: The osteoblast separated from murine calvariae was cultivated for 6 days and evaluated the cell function. After the addition of RGE on the culture medium, we determined the effect of RGE on the cell viability, cell proliferation, protein synthesis, alkaline phosphatase activity, collagen synthesis and calcified nodule formation of the cultivated osteoblast. Results: The results were summarized as follows. 1. RGE did not change the survival rate of rat calvarial osteoblast. 2. RGE increased the proliferation of rat calvarial osteoblast. 3. RGE increased ALP activity of rat calvarial osteoblast., 4. RGE slightly affected protein synthesis of rat calvarial osteoblast. 5. RGE increased collagen synthesis of rat calvarial osteoblast. 6. RGE slightly affected calcified nodule formation of rat calvarial osteoblast. Conclusions: From these results, it is concluded that RG might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

골다공증 동물모델에서 약침치료에 대한 국내 연구보고 고찰 (Effects of the Pharmacopuncture in Animal Models for Treatment of Osteoporosis: A Review of Animal Study Reports Published in Korea)

  • Objectives This study is to review the effect of pharmacopuncture on treatment of osteoporosis in animal models reported in korean domestic journals. Methods The databases (Koreantk, KISS, NDSL) were searched with term as osteoporosis, and animal study reports on osteoporosis with pharmacopuncture were reviewed. Animal model, intervention, and osteoporosis indicator were extracted. Results 22 articles were reviewed. 11 studies used ddy mouse and 9 studies used SD rat. 20 studies used ovariectomy to induce osteoporosis. 21 studies used simple pharmacopuncture. Cervi pantotrichum cornu was most frequently used pharmacopuncture and Umgok (KI10) was most frequently used acupuncture point. Each study shows significant changes of osteoporosis indicators. Conclusions Pharmacopuncture is expected to be a positive effect on osteoporosis.

  • 본 연구는 골다공증유발 rat에 녹용추출물의 투여가 예방 및 치료효과에 미치는 영향을 구명하고자, 양측 난소를 적출한 후 골다공증이 유발된 rat에 암, 수녹용 추출물 625, 1,250 mg/kg을 5 주간 투여하면서 혈청중 estradiol, progesterone, calcitonin, osteocalcin 농도와 Ca, P 및 ALP 함량변화등을 조사하였다. 1. 난소적출 후 골다공증이 유발된 rat에 암, 수녹용 추출물 625mg/kg, 1,250 mg/kg을 각각 투여했을 때 혈청내 estradiol 수준은 난소적출군이 20.80$\pm$1.86~40.50$\pm$pg/$m\ell$로서 정상대조군의 49.50 :t2. 70-50.80 :t3.13 pg/$m\ell$에 비하여 유의한 감소를 나타냈다 (p<0.05). 2. 골다공증 유발 rat 에 암 수녹용 추출물 625 mg/kg, 1,250 mg/kg을 각각 투여했을 때 혈청중 progesterone 수준은 정상대조군의 50.90$\pm$3.63~53.40$\pm$2.74 ng/$m\ell$에 비하여 큰 차이를 나타내지 않았다. 3. 골다공증 유발 rat 에 암, 수녹용 추출물 625 mg/kg, 1,250 mg/kg을 투여했을 때 혈청중 calcitonin 수준은 정상대조군의 0.64$\pm$0.03~0.68$\pm$0.04 pg/$m\ell$에 비 하여 약간 높은 증가를 나타냈으며 특히, 난소적출군은 대조군에 비해 현저한 감소를 나타냈다. 4. 골다공증 유발 rat 에 암, 수녹용 추출물 625 mg/kg, 1,250 mg/kg을 각각 투여했을 때 혈청중 osteocalcin 수준은 정상대 조군의 0.28$\pm$0.02 ~0.31$\pm$0.02 ng/$m\ell$에 비하여 약간 높은치를 나타냈다. 한편, 암, 수녹용 추출물의 처리군의 osteocalcin 치는 암컷녹용 추출물처리군이 약간 높았으나 유의성은 인정되지 않았다. 5. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 혈청중 Ca, P 함량은 각각 정상대조군에 비하여 약간 높은 치를 나타냈으며, 난소적출 무처치군은 대조군에 비해 유의한 증가를 나타냈다 (p<0.05). 6. 골다공증유발 rat에 암, 수녹용 추출물을 투여했을 때 혈청중 ALP 함량은 정상대조군에 비하여 증가치를 나타냈으며, 암, 수사슴녹용 추출물의 처리군중 대조군과 FA1,250 처리군과 MA625 처리군간에 유의한 변화가 인정되었다 (p<0.05). 위의 결과들에서 암, 수녹용 추출액은 난소제거후의 estrogen의 감소를 억제시키고 골소실을 억제하므로 골다공증의 예방과 치료에 효과가 있는 것으로 판단된다.

가미소요산(加味逍遙散)이 난소적출로 유발된 흰 쥐의 골다공증에 미치는 영향 (A Study on the Effects of Gamisoyosan on Ovariectomized Osteoporosis in Rats)

  • Purpose: Osteoporosis that occurred after menopause is one of the most common metabolic disease. and it is that bone mass significant decrease in comparison with normal people. Its symptoms are fracture, decrease of height, etc. The purpose of this study is to examine what are the effects of the Gamisoyosan on the aged ovariectomized rat model of postmenopausal osteoporosis. Methods: 24 female rats were ovariectomized and divided into normal group(normal), ovariectomized group(control), and treated with Gamisoyosan group(treated). During 8 weeks we dosed D.W to sham and control group, and dosed Gamisoyosan(105 mg/100 g) to experimental group. After 8 weeks we sacrificed rats and measured the body weight, albumin, P, Calcium, ALP, total cholesterol, triglyceride, AST, ALT, T4, estradiol, the femur weight, the tibia ash weight, trabecular bone area, trabecular thickness, osteoclast number. Results: The level of serum albumin, calcium, AST, ALT, estradiol in Gamisoyosan group showed significant increase in comparison with control group. The level of serum triglyceride in Gamisoyosan group showed significant decrease in comparison with control group. Trabecular bone area and osteoclast number in Gamisoyosan group showed significant increase in comparison with control group. Conclusions: Reviewing these experimental results, Gamisoyosan has shown to activate osteogenesis and prevent bone resorption, and it should show efficacy on prevention and improvement of osteoporosis after menopause.

어성초 추출물 부탄올 분획이 Bone Marrow Stromal Cell 분열능 및 골다공증 랫드에 미치는 영향 (Effects of Houyttnia cordata on Bone Marrow Stromal Cell and Osteoporetic Rat)

  • This study was performed to evaluate the effect of HCB on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with HCB, extract of Houyttnia cordata, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of HCB. Osteoporotic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns.
