Vemurafenib has recently been used as drug for treatment of melanomas with $BRAF^{V600E}$ mutation. Unfortunately, treatment with only vemurafenib has not been sufficiently effective, with recurrence after a short period. In this study, three vemurafenib-resistant $BRAF^{V600E}$ melanoma cell lines, $A375P^R$, $A375M^R$ and SKMEL-$28^R$, were established from the original A375P, A375M and SKMEL-28 cell lines. Examination of the molecular mechanisms showed that the phosphorylation levels of MEK and ERK, which play key roles in the RAS/RAF/MEK/ERK signaling pathway, were reduced in these three cell lines, with increased phosphorylation levels of pAKTs limited to SKMEL-$28^R$ cells. Treatment of SKMEL-$28^R$ cells with 100 nM paclitaxel resulted in increased apoptosis and decreased cellular proliferation, invasion and colony formation via reduction of expression levels of EGFR and pAKTs. Moreover, vemurafenib-induced pAKTs in SKMEL-$28^R$ were decreased by treatment with an AKT inhibitor, MK-2206. Taken together, our results revealed that resistance mechanisms of $BRAF^{V600E}$-mutation melanoma cells to vemurafenib depended on the cell type. Our results suggested that paclitaxel should be considered as a drug in combination with vemurafenib to treat melanoma cells.

Disagreement among the various publications providing lists of Korean endemic plants makes confusion inevitable. We summarized the six previous reports providing comprehensive lists of endemic plants in Korea: 407 taxa in Lee (1982), 570 taxa in Paik (1994), 759 taxa in Kim (2004), 328 taxa in Korea National Arboretum (2005), 515 taxa in the Ministry of Environment (2005) and 289 taxa in Flora of Korea Editorial Committee (2007). The total number of endemic plants described in the previous reports was 970 taxa, including 89 families, 302 genera, 496 species, 3 subspecies, 218 varieties, and 253 formae. Endemic plants listed four times or more were collected to compare the data in terms of scientific names and synonyms (339 taxa in 59 families and 155 genera). If the varieties and formae were excluded, the resulting number of endemic plants was 252 taxa for the 339 purported taxa analyzed. Seven of the 155 genera analyzed were Korean endemic genera. Among the 339 taxa, the same scientific names were used in the original publications for 256 taxa (76%), while different scientific names were used for 83 taxa (24%). The four largest families were Compositae (42 taxa, 12.4%), Ranunculaceae (19 taxa, 5.6%), Rosaceae (19 taxa, 5.6%), and Scrophulariaceae (19 taxa, 5.6%). Saussurea (Compositae) had the highest number of taxa within one genus (17 taxa; 5% of total endemic taxa).
The chloroplast DNA sequences of Megaleranthis saniculifolia, an endemic and monotypic endangered plant species, were completed in this study (GenBank FJ597983). The genome is 159,924 bp in length. It harbors a pair of IR regions consisting of 26,608 bp each. The lengths of the LSC and SSC regions are 88,326 bp and 18,382 bp, respectively. The structural organizations, gene and intron contents, gene orders, AT contents, codon usages, and transcription units of the Megaleranthis chloroplast genome are similar to those of typical land plant cp DNAs. However, the detailed features of Megaleranthis chloroplast genomes are substantially different from that of Ranunculus, which belongs to the same family, the Ranunculaceae. First, the Megaleranthis cp DNA was 4,797 bp longer than that of Ranunculus due to an expanded IR region into the SSC region and duplicated sequence elements in several spacer regions of the Megaleranthis cp genome. Second, the chloroplast genomes of Megaleranthis and Ranunculus evidence 5.6% sequence divergence in the coding regions, 8.9% sequence divergence in the intron regions, and 18.7% sequence divergence in the intergenic spacer regions, respectively. In both the coding and noncoding regions, average nucleotide substitution rates differed markedly, depending on the genome position. Our data strongly implicate the positional effects of the evolutionary modes of chloroplast genes. The genes evidencing higher levels of base substitutions also have higher incidences of indel mutations and low Ka/Ks ratios. A total of 54 simple sequence repeat loci were identified from the Megaleranthis cp genome. The existence of rich cp SSR loci in the Megaleranthis cp genome provides a rare opportunity to study the population genetic structures of this endangered species. Our phylogenetic trees based on the two independent markers, the nuclear ITS and chloroplast MatK sequences, strongly support the inclusion of the Megaleranthis to the Trollius. Therefore, our molecular trees support Ohwi's original treatment of Megaleranthis saniculifolia to Trollius chosenensis Ohwi.
Sisyrinchium micranthum Cav., a newly naturalized species, was found in Seogwipo, Jeju-do. This species is native to Central and South America and is reported as a widely invasive species around the world. Sisyrinchium micranthum is a sister species of S. rosulatum E. P. Bicknell (Deung-Sim-But-Kkot in Korean), reported as an invasive species earlier in Korea, and both species are members of the subgenus Echthronema. Sisyrinchium micranthum is distinguished from S. rosulatum by the urceolate shape of its flowers and by the large size of its fruits and seeds. Its local name is "Yeon-Deung-Sim-But-Kkot" based on the light blue corolla. We also found that Sisyrinchium angustifolium Mill., previously used for Deung-Sim-But-Kkot, was misapplied. It belongs to the subgenus Sisyrinchium and is morphologically very different from S. rosulatum. We corrected the scientific name of Deung-Sim-But-Kkot by changing it from S. angustifolium to S. rosulatum based on the type specimens, original descriptions, and recent studies.

A Heat Recovery Steam Generator(HRSG) is the main component of a Combined Cycle Power Plant(CCPP). It is a very large structure that is made from relatively thin metal sheets. Therefore, the structural integrity of an HRSG is very important to ensure safe operation during plant lifetime. In particular, thermal deformation and thermal fatigue have been revealed as the main causes of the mechanical degradation of an HRSG. In order to prevent unexpected damage, safety evaluation based on a large-scale analysis is necessary. Therefore, this study aims to improve the safety of HRSG by using Finite Element Analysis(FEA) results derived from large-scale analysis. Furthermore, the modified design is verified by comparing it with the original one. This result will be used as basic data for improving the safety of a vertical-type HRSG.

The insecticidal toxin gene of Bacillus thuringiensis (Bt) is one of the most commonly used in the development of genetically modified (GM) crops. In this research, we analyzed Bt rice showing lepidopteran pest-resistance. The Bt gene is a synthetic Cry1Ac composed of optimal codons for plants, and the Bt protein is targeted to the chloroplast by a transit peptide. Three Cry1Ac rice events (C103-3, C127-1, and C7-1) were analyzed for molecular characterization. C103-3 contains two copies of T-DNA where the left border (LB) region is truncated. Both C7-1 and C127-1 have a single copy of T-DNA, but a part of the vector backbone DNA is inserted into the genome of C127-1; thus, only C7-1 had intact T-DNA. Progenies of C7-1 crossed with the original cultivar, Nakdong, and double-haploid lines from anther culture of lines crossed with the elite cultivar, Dongjin, were analyzed for T-DNA flanking genomic DNA and genotyping. Results showed that an intact T-DNA region without the vector backbone was inserted into the genome and was stably inherited through generations. The C7-1 homozygous event could be used as breeding material to develop GM rice with pest resistance.
This study was conducted to establish the most appropriate nitrogen levels and planting densities which can attribute to increase of aromatic substances in tobacco variety, Hyangcho, maintaining its original aroma. An increase of nitrogen fertilization from 3 to 12 kg per 10a increased total alkaloid content 2.0% to 3.3%, while an increased nitrogen fertilization decreased the contents of total sugar, starch, fatty acid etc., Which seem to be the positive components relating to tobacco aroma the contents of alkaloids were decreased and total fatty acids were increased as the planting densities increased. The higher contents of 2. 3-methylbutanoic and 3-methylpentanoic acid was obtained at the density of 3 plant per pit (12,498 plant/10a). The components determined in the neutural fraction such as solanone, furfural, furfurylalcohol and benzyl alcohol were presented in higher amount at the higher densities. The volatile acids such as 2. 3-methylbutanoic acid 3-methylpentanoic acid decreased and also the contents of butanol, furfutal, furfuryl alcohol, hexanol, cinnamil, acetophenone, benzyl acetate and solanone etc., were decreased as the nitrogen level increased from 3 to 12 kg per l0a.
This study examined the decomposition of blades and culms of aquatic emergent plant species, Zizania latifolia, Phragmites communis and Typha angustata, and changes in nutrient contents during decomposition. Z. latifolia, P. communis and T. angustata were the most frequently occurring species in Lake Paldang of Han River, Korea. Experiments were carried out from July 27 to December 14, 2005 in Lake Paldang using the litter bag method. The remaining masses of blade litter of each species at the end of experimental period were 21.2% of initial dry weight in Z. latifolia, 32.5% in P. communis, and 44.7% in T. angustata. In addition, the remaining mass of culm was 22.6% of initial dry mass in Z. latifolia, 56.4% in P. communis, and 38.1% in T. angustata. During the litter decomposition period, P, K, Na, and Mg concentration decreased rapidly within 10 days, but Ca and Mg concentration declined slowly. K contents remained below 10% of initial values in all litter samples retrieved during decomposition, whereas Ca and Mg concentration remained above 40% and 50% during decomposition in all three species. Na, P and Mn contents in litter varied among species and plant parts. P concentration in culms of P. communis remained at about 60% of initial concentration throughout the study, but the remaining P content in culms of Z. latifolia was only 10% of the original value at the end of the study period. The Mn concentration in blades of P. communis increased about 15-fold relative to the initial content by the end of experiment.
Background: Nutrient release during litter decomposition was investigated in Vitex doniana, Terminalia avecinioides, Sarcocephallus latifolius, and Parinari curatellifolius in Makurdi, Benue State Nigeria (January 10 to March 10 and from June 10 to August 10, 2016). Leaf decomposition was measured as loss in mass of litter over time using the decay model Wt/W0 = e-kd t, while $Kd=-{\frac{1}{t}}In({\frac{Wt}{W0}})$ was used to evaluate decomposition rate. Time taken for half of litter to decompose was measured using T50 = ln 2/k; while nutrient accumulation index was evaluated as $NAI=(\frac{{\omega}t\;Xt}{{\omega}oXo})$. Results: Average mass of litter remaining after exposure ranged from 96.15 g, (V. doniana) to 78.11 g, (S. lafolius) in dry (November to March) and wet (April to October) seasons. Decomposition rate was averagely faster in the wet season (0.0030) than in the dry season (0.0022) with P. curatellifolius (0.0028) and T. avecinioides (0.0039) having the fastest decomposition rates in dry and wet seasons. Mean residence time (days) ranged from 929 to 356, while the time (days) for half the original mass to decompose ranged from 622 to 201 (dry and wet seasons). ANOVA revealed highly significant differences (p < 0.01) in decomposition rates and exposure time (days) and a significant interaction (p < 0.05) between species and exposure time in both seasons. Conclusion: Slow decomposition in the plant leaves implied carbon retention in the ecosystem and slow release of CO2 back to the atmosphere, while nitrogen was mineralized in both seasons. The plants therefore showed effectiveness in nutrient cycling and support productivity in the ecosystem.
The aims of the study were to analyze (1) the effects of Prosopis juliflora (Prosopis) on the spatial distribution and soil seed banks (SSB) diversity and density, (2) the effects of environmental factors on SSB diversity and density (number of seeds in the soil per unit area), and (3) the effects of animal fecal droppings on SSB diversity, density, and dispersal. Aboveground vegetation data were collected from different Prosopis-infested habitats from quadrats (20 × 20 m) in Prosopis thickets, Prosopis + native species stand, non-invaded woodlands, and open grazing lands. In each Prosopis-infested habitats, soil samples were collected from the litter layer and three successive soil layer, i.e., 0-3 cm, 3-6 cm, and 6-9 cm. Seeds from soil samples and animal fecal matter were separated in the green house using the seedling emergence technique. Invasion of Prosopis had significant effects on the soil seed bank diversity. Results revealed that the mean value of the Shannon diversity of non-invaded woodlands was being higher by 19.2%, 18.5%, and 11.0% than Prosopis thickets; Prosopis + native species stand and open grazing lands, respectively. The seed diversity and richness, recovered from 6-9-cm-deep layer were the highest. On the other hand, the density of Prosopis seeds was the highest in the litter layer. About 156 of seeds/kg (92.9%) of seeds were germinated from cattle fecal matter. However, in a small proportion of seedlings, 12 of seeds/kg (7.1%) were germinated from shot fecal matter. Thus, as the seeds in the soil were low in the study areas, in situ and ex situ conservation of original plants and reseeding of persistent grass species such as Cynodon dactylon, Cenchrus ciliaris, Chrysopogon plumulosus, and Brachiaria ramosa are recommended.
