Dyeing Property of Polyester in Byebath Containing Water and Water Miscible Organic Solvents (물/극성유기용매 혼합욕에서 폴리에스테르의 염색)

  • Polyester filaments were dyed with disperse dye in dyebath containing water and water miscible organic solvents . acetone, 1,4-dioxane, DMF. In case of Acetone and 1,4-dioxane, the equilibrium dyeuptake was maximun at the volume fraction 0.05. The equilibrium dye uptakes were decreased as volun~e fraction of organic solvents were increased. When the volume fractions of water miscible organic solvents were varied, dye uptake was increased constantly with dyeing time. In dyebath containing water and water miscible organicsolvent, the dyeuptake was increased quickly during initial 40∼ 60 min. and slowly increased there after. The slope of Ct/Coo to t was greater in dyebath containing water and water miscible organic solvents than dyebath containing water. The differences of the slope with volulne fractions of water miscible organic solvent were not shown big.

An Extract from Hydrolyzed Normal Human Urine which Induces Drug Binding Defects (정상인뇨의 가수분해에 의한 의약품결합 저해유도인자의 추출)

  • Uremia is associated with defective protein binding of weakly acidic drugs, whereas the protein binding of basic drugs tends to be normal. The exact chemical nature of compound(s) and mechanism for these changes as yet is unknown, and has not been defined. Organic solvent extraction of pooled normal human urine following hydrolysis by hydrochloric acid produced an extract, which when added to normal human serum, was capable of inducing binding defects similar to those in uremia. Binding defects were observed with the weakly acidic drugs such as nafcillin, salicylate, sulfamethoxazole and phenytoin while the binding of the basic drugs such as trimethoprim and quinidine were unaffected. The binding defects induced by the hydrolyzed urine extract could readily be corrected by same organic solvent extraction of acidified serum and the defects could be transferred to the normal human serum using the organic solvent layer at the physiologic pH (7.4). Followed by reacidification ind extraction of the binding defects induced serum with the same solvent, separated several fractions were obtained on thin-layer chromatography. One of these fractions could reinduce the binding defects and this factor(s) is apparently weakly acidic compounds and tightly bound to serum at physiologic pH, but extractable at acidic pH, and its molecular weight range is approximately 500 or less similar to those seen in uremia. These findings strongly support the hypothesis that the drug binding defect in uremia is due to the accumulation of endogenous metabolic products which arc normally excreted by the kidneys but accumulate in renal failure.

Effects of Vegetable Extracts by Solvent Separation on Alcohol Dehydrogenase Activity from Saccharomyces cerevisiae (채소의 용매분획 추출물들이 Saccharomyces cerevisiae의 alcohol dehydrogenase 활성에 미치는 영향)

  • The effects of extracts from bean sprout (Glycine max), dropwort (Oenathe javanica) and radish (Raphanus sativus Var. hortensis for. acanthiformis) by solvent separation on alcohol dehydrogenase (ADH) activity in vitro were investigated. The extracts were obtained from alcohol extracts of bean sprout, dropwort and radish, followed by solvent separation. Aqueous fractions facilitated much higher ADH activity than organic fractions. The facilitating rates of bean sprout, dropwort and radish in aqueous fractions were 125.75%, 104.94% and 87.63%, respectively. Basic fractions showed the highest facilitating rate with about 40%. Also other fractions showed below 20% facilitating rate and didn't show great difference from organic fractions. Phenolic fractions didn't show great effect on ADH activity.

Evaluation of the Antioxidant Properties of Pediastrum duplex and Dactylococcopsis fascicularis Microalgae

  • Interest in plant-derived food additives has increased recently, with efforts to identify natural antioxidant sources to replace synthetic antioxidants. This study evaluated the antioxidant effects of organic solvent fractions of 80% methanol extracts from Pediastrum duplex and Dactylococcopsis fascicularis microalgae. Among the solvent fractions tested, the ethyl acetate and n-hexane fractions from P duplex effectively scavenged 79.8% and 74.5% of DPPH free radicals, respectively. The chloroform fraction from D. fascicularis showed the strongest $H_2O_2$ scavenging ability (49.7%). The greatest scavenging of hydroxyl radicals (73.1%) was exhibited by the 80% methanol extract from P duplex. Aqueous residue and ethyl acetate fraction from P duplex provided the strongest nitric oxide scavenging (57.7%) and metal chelating effect (82.1%), respectively. Chloroform and ethyl acetate fractions from P duplex and n-hexane fraction from D. fascicularis exhibited significantly greater inhibition of lipid peroxidation than the commercial antioxidants. These results suggested that P duplex and D. fascicularis microalgae having potential anti oxidative compounds with various properties could be utilized in the food and pharmaceutical industries.

Screening and identification of bioorganic light-emitting substances from marine macrophytes

  • Organic light-emitting materials in marine macrophytes from various coastal environments were identified. Twentyeight species from the solvent fractions were examined and identified as candidates for bioorganic light-emitting materials using photoluminescence (PL) spectra and gas chromatography-mass spectrometry. We selected 16 solvent fractions from a total of 1,221 prepared from Ishige okamurae, Sargassum confusum, Grateloupia elliptica, Chondracanthus intermedius, Porphyra yezoensis, Meristotheca papulosa, Gelidium amansii, and Scytosiphon lomentaria. The maximum light-emitting PL spectra appeared at various colors, mainly between blue and green, based on chromaticity coordinates, from solvent fractions of M. papulosa, G. amansii, G. elliptica, P. yezoensis, S. lomentaria, I. okamurae, and C. intermedius. These results will contribute to the development of novel organic light-emitting materials.

  • The solubility characteristics of organic compounds were studied in terms of the extraction efficiency as a function of the polarity of the organic solvent, and the acidity of water in urban aerosol samples collected in University of East Anglia (UEA), Norwich, England. The extraction efficiency of organic compounds were evaluated with respect to the organic carbon, -nitrogen and -hydrogen by means of a wide range of solvent which include polar and nonpolar organic solvents as well as acids and alkaline water. In addition, after being dissolved in aqueous solution, the aqueous chemistry of organic compounds were studied in terms of the organic metal complexes in aerosol, which were studied with oxalic acid, copper, and zinc. The results of this study indicate that solubility characteristics of organic compounds depend on the polarity of the solvents and the acidity of the solvents. In particular, some organic compounds are water soluble, even though they are much smaller than acetone soluble fractions. In the comparison between polar organic solvent extraction and non- polar organic solvent extraction, it can be thought that significant fraction of organic compounds analysed in the aerosol samples, are polar organic compounds because of the higher extraction efficiencies of organic compounds in polar organic solvent extraction than in nonpolar organic solvent extraction. Regarding the study of the oxalic -metal complexes, it can be thought that most oxalic acids are present in the form of oxalic -copper complexes in the aerosols collected at UEA.

Extracts from Pinus densiflora Siebold et Zuccarini Sawdust Inhibit the Mycelial Growth of Lentinus edodes (Berk.) Sing., Edible Mushroom

  • The extracts of Pinus densiflora sawdust by n hexane, ethyl acetate and methanol solvent were investigated to identify their mycelial growth inhibition against Lentinus edodes. The yields of n hexane soluble fraction, ethyl acetate-soluble fraction, and methanol soluble fraction from P. densiflora sawdust were obtained 1.36%, 2.21% and 4.03% using organic solvent, respectively. The mycelial growth inhibition of L. edodes was the greatest for n hexane extract, ranging from 36.5% to 47.6% at concentrations of 125 ppm to 1,000 ppm, with the values for all concentrations significantly different from one another. After direct extraction of P. densiflora sawdust using n hexane, ethyl acetate and methanol, each extract was separated into three fractions by silica gel column chromatography and then the fractions were isolated on the values of $R_f$ by thin layer chromatography. The mycelial growth inhibition against L. edodes was recognized in the fractions II (33.5%) and III (37.6%) of n hexane extract, the fraction II (21.4%) of ethyl acetate extract and the fraction II (26.4%) of methanol extract. The fractions III of n-hexane extract showed the highest growth inhibition among the nine fractions of the organic solvent extract.

Isolation and Concentration of Organic Components from a Complex Matrix into Three Fractions of Different Volatilities (복합 유기혼합물체로부터 휘발성이 서로 다른 세 유기화합물 그룹의 분리 농축방법의 연구)

  • A simple micro-sampling system is described which facilitates isolation and concentration of complex organic constituents into three fractions of different volatilities. The method involves the headspace trapping of very volatile components from a complex matrix onto a porous polymer, Tenax GC, followed by the solvent elution of the matrix and the subsequent fractions of the eluate into volatile and less-volatile fractions. The headspace and the volatile fractions are then analyzed by high-resolution capillary gas chromatography. The less-volatile fraction is analyzed by high-performance liquid chromatography. Experimental details and the results obtained using tobacco leaves as a complex organic matrix are presented.

Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

Antimicrobial and Antioxidative Activities of Solvent Fractions of Quercus mongolica Leaf (신갈나무 잎의 용매분획별 항균 및 항산화 효과)

  • The ethanol extract of Quercus mongolica leaf was fractionated by various organic solvents, and their antimicrobial and antioxidative activities were investigated against several microorganisms. The ethanol extract of Quercus mongolica leaf at two thousand $\mu\textrm{g}$ per disc showed 17~21mm inhibition zone against Gram postive and Gram negative bacteria. Among the various solvent fractions from ethanol extract of Quercus mongolica leaf, the hexane fraction showed the strongest antimicrobial activity. Minimal inhibitory concentration (MIC) of hexane fraction was 250$\mu\textrm{g}$/mL against Bacillus cereus, 250~500$\mu\textrm{g}$/mL against Listeria monocytogenes, 500$\mu\textrm{g}$/mL against Staphylococcus aureus, Salmonella typhimurium, Escherichia coli O157, and Pseudomonas aeruginosa. Also, the hexane and chloroform fraction had the similar antioxidative activity compared to that of butylated hydroxy toluene(BHT).
