Comparison of QuEChERS and Solid Phase Extraction for Accurate Determination of Pesticide Residues in Kimchi Cabbage and Strawberry using Isotope Dilution Mass Spectrometry

  • QuEChERS is used worldwide as a universal sample preparation method with many benefits, such as being quick, easy, cheap, effective, rugged and safe. This study examined whether QuEChERS can be employed in isotope dilution mass spectrometry (ID-MS) for accurate analysis of pesticides in food. The ratios of fortified values and measured values of malathion and fenitrothion using the QuEChERS method were compared with those using the solid phase extract (SPE) method which was previously used in this laboratory. The separations of the two pesticides on DB-5MS and VF-1701MS columns were compared. Malathion and fenitrothion were fortified into kimchi cabbage and pretreated with the QuEChERS method and the SPE method. The results obtained using the DB-5MS column varied according to the sample preparation method, column and pesticide level. Using the VF-1701 column, ratios were 98-102% by both QuEChERS and Carb/NH2 SPE method for all fortification level. Malathion and fenitrothion were fortified into strawberry samples for comparison with kimchi cabbage. The results for the strawberry samples indicated that the ratios were not influenced by the sample preparation methods or GC column. The QuEChERS method could be acceptable in the ID-MS method for pesticide residue analysis in food, however other conditions should be carefully considered for accurate determination, such as the column, amount of analyte and food matrix.

  • New ${\beta}$-methoxyacrylate derivatives 1-4 having thio-enol side chain were prepared and subjected to in vivo screening for fungicidal activity against phytopathogenic fungi and many of them showed good fungicidal activities against especially rice blast and wheat leaf rust at 100 ppm.

  • Objectives : This study was designed to evaluate the pesticide residue of herb medicine through high density ozone water cleaning system. Materials and Methods : We purchased ginseng(Panax ginseng) on market and sprayed pesticides (Diazinon) on ginseng. We analyzed pesticide residue according to washing methods(untreated, ozone water cleaning, ultrasonic cleaning, Water cleaning). As a result of each washing methods, in ozone water cleaning method pesticide residue was much less remained than in Ultrasonic Cleaning method and water cleaning method. Conclusions : High density ozone water cleaning is the best method for removing pesticides of herbal medicine.

Monitoring of Pesticide Residues in Commercial Environment-friendly Stalk and Stem Vegetables and Leafy Vegetables and Risk Assessment (유통 친환경 엽채류와 엽경채류 중 잔류농약 실태조사 및 안전성 평가)

  • In order to monitor the levels of pesticides in environment-friendly stalk and stem vegetables and leafy vegetables selling at markets in Korea. A total of 637 (395 organic agricultural products, 242 pesticide-free agricultural products) samples of 21 stalk and stem vegetables and leafy vegetables were collected twice from markets in July and August 2010 in Korea. Pesticide residues in samples were analyzed by multiresidue method for 240 pesticides using GC-ECD/NPD and HPLC-DAD/FLD. The suspected-pesticides were confirmed with a GC-MSD. As a result of analysis, six pesticides were detected from eight samples, representing a detection rate of 1.3% and amounts of pesticide residues in samples were as follows: alachlor 0.043, chlorfenapyr 0.022-0.324, diazinon 0.024, dicofol 0.009-0.138, dithiopyr 0.008, metolachlor 0.025 mg/kg. Their residue levels were below the MRLs. Estimated daily intakes (EDIs) of the pesticides detected from stalk and stem vegetables and leafy vegetables were less than 25% of their acceptable daily intakes (ADIs), representing that residue levels of the pesticides detected were evaluated as safe for consumption. However, five samples containing pesticide residues were unsuitable environment-friendly products because of pesticides detected more than their 10% MRLs in organic and pesticide-free agricultural products.

  • Since the chemical pesticides have been played a major role in crop protection practices during last 5 to 7 decades, social concerns on the pesticide residues in and on food commodities as well as environmental compartments have also growing with endless demands and interests. Most national regulation authorities over the world have paid a special attention on the data requirements for pesticide registration. In addition, even the registered pesticides also should follow the re-registration process, which meets today's guidelines and regulatory triggers and safety profiles. More recently, a defined interest in the international bodies has given to the global conservation program from the environmental contamination; these involves persistent organic pollutants (POPs), endocrine disrupting chemicals (EDs), biocides, etc.. In order to secure the food safety and keep our circumference sound, in-depth efforts getting information from global networks have perpetually to be given under relevant national agencies. At the same time, a nation-wide survey of the residues has also to be in operation to monitor the tendency of the toxicant in/on foods, feeds, and environmental segments. In final, the scientifically assessed results on safety should be opened to the public to provide the right-to-know for the consumers.

  • The AOPs research defined by creating a sufficient amount of OH radicals from the dissolution of organic materials through photoxidation and research for a complete elimination of residual organic materials by membrane are actively ongoing. This research focuses on the hybrid processing of AOPs and M/F membrane to dissolve and eliminate organic chemicals in drinking water which are suspected of carcinogens. For this purpose, underground water was used as a source of drinking water for the hybrid processing of AOPs oxidation and M/F membrane, and a pilot plant test device was installed indoor. Carcinogenic chemicals of VOCs and pesticide were artificially mixed with the drinking water, which was then diluted close to natural water in order to examine treatment efficiency and draw optimal operation conditions. The samples used for this experiment include four chemicals phenol, chloroform, in VOCs and parathion, carbaryl in pesticide. As a result of the experiments conducted with simple, and compound solutions, the conditions to sufficiently dissolve and eliminate carcinogenic chemicals from the hybrid processing of where carcinogens were artificially added are : (hydrogen peroxide) prescribed solution 100 mg/L under pH 5.5~6.0, and the temperature $12{\sim}16^{\circ}C$, at the normal temperature and pressure. $d-O_3$ volume of 5.0 ppm and above and 30-40 minutes of reaction time are most appropriate and using MF/UF for membrane was ideal.

Control of Diseases and Insects for Pesticide-free Cultivation of Leafy Vegetables (엽채류 유기재배의 병해충 관리)

  • This study was conducted to establish agricultural practices to control diseases and insects for chemical pesticide-free cultivation of leafy vegetables. Two diseases, gray mold(Botrytis cinerea) and soft rot(Erwinia carotovora), on lettuce were reduced by controlling temperature and humidity using air-circulation fan. The aphidophagous lady beetle(Harmonia axyridis) and primary parasitoids(Aphidius colemani) showed activity to control aphids density on Chinese cabbage. Co-application of cooking oil and yolk mixture (COY) and BT(Bacillus thuringiensis) decreased diseases including soft rot(Erwinia carotovora), downy mildew(Peronospora brassicae Gaumann), and powdery mildew(Eryslphe polygoni), and insects such as diamondback moth(Plutella xylostella) and beet armyworm(Spodoptera exigua Hubner). Neem extract treatment reduced downy mildew(Peronospora destructor) on Welsh onion.

Adsorption and residues of EPN in the soil of Cheju Island (제주도 토양에서 EPN의 용탈과 잔류)

  • The adsorption and leaching of organophosphorus pesticide, EPN (O-ethyl-O-4 -nitrophenyl phenyl phosphonothioate) were investigated in Namwon soil(black volcanic soil), Aewol soil(very dark brown volcanic soil) and Mureung soil(dark brown nonvolcanic soil) in Cheju Island. The residue of EPN was surveyed on coastal environment of Cheju in Aug. 1996. The organic matter of Namwon soil, Aewol soil and Mureung soil was 19.8, 6.2 and 2.4%, respectively. The cation exchange capacity of Namwon soil, Aewol soil and Mureung soil was 24.8, 13.0 and 9.5 meq/100 g, respectively. The Freundlich constant, k value, was 89.4, 26.9 and 9.25 for Namwon soil, Aewol soil and Mureung soil, respectively. The k value of Namwon soil with very high organic matter content and cation exchange capacity was the highest for Aewol soil and Mureung soil. The Freundlich constant, 1/n, show a high correlation with organic matter content, i.e. it is less than unity for organic matter rich soil of Namwon soil and greater than unity for organic matter poor soil of Mureung soil. The leaching of EPN was slower for Namwon soil with high k values, and faster for Mureung soil with low k values. The results of the study was demonstrated the potential of pollution for EPN have little leached into soil environment. EPN was not detected in seawater and sediment in the coastal environment in Cheju Island. EPN used in the farm on tile Cheju island were not residued in the coastal environment.

  • A method for the determination of 16 residual pesticides in crude deugs was examined. Crude drug was extracted with acetonitrile/water solution and the extract was partitioned between hexane/CH$_{2}$Cl$_{2}$ and NaCl saturated water. The organic layer was passed through cleanup column and subjected to the GC/ECD or GC/NPD analysis. Essential oil components in crude drug interfere with the analysis, and sulfuric acid treatment was adapted to overcome this interference.

  • As an ongoing research program for the development of environmentally friendly new herbicide, several hydantoin derivatives 5a - 5i possessing amide subgroup were synthesized and shown to have interesting herbicidal activities exhibiting symptoms as Protoporphyrinogen IX oxidase inhibitor under postemergence upland greenhouse screening. Among derivatives tested, compound 5h showed superior herbicidal activity against upland problem weed, digitaria sanguinalis and aeschynomene indica to reference compound fluthiacet-methyl.
