Potential Control of Foodborne Pathogenic Bacteria by Pediococcus pentosaceus and Lactobacillus graminis Isolated from Fresh Vegetables

  • The consumption of fresh vegetables has been related to recurrent outbreaks of foodborne diseases (FBD) worldwide. Therefore, the development of effective alternative technologies is necessary to improve the safety of these products. This study aimed to isolate and identify epiphytic lactic acid bacteria (LAB) from fresh fruits and leafy vegetables and characterize their antagonistic capacity due to their ability to produce bacteriocins or antibacterial compounds. For this, 92 LAB isolates from fruits and leafy vegetables were screened for antagonistic activity. Two strains with the highest and broadest antagonistic activities were selected for further characterization; one from cantaloupe melon (strain CM175) and one from cilantro leaves (strain C15). The cell-free supernatants (CFS) of CM175 and C15 were found to exhibit antagonistic activity against FBD-causing pathogens. The CM175 and C15 strains were identified as Pediococcus pentosaceus and Lactobacillus graminis, respectively. Notably, the P. pentosaceus CM175 CFS stopped the growth of Salmonella Typhimurium, Salmonella Saintpaul, Staphylococcus aureus, and Listeria monocytogenes, and delayed Escherichia coli O157:H7 growth. Moreover, L. graminis C15 CFS delayed the growth of all indicator pathogens, but did not completely stop it. Organic acids and bacteriocin-like molecules were determined to be possibly exerting the observed antagonistic activity of the identified LAB strains. Thus, application of the antagonistic compounds produced by Pediococcus pentosaceus and Lactobacillus graminis could be a novel and ecological strategy in developing antimicrobial biopreservatives for the food industry and mitigating FBD by reducing the biological contamination in fruit and vegetable orchards, mainly via their potential in controlling both gram-negative and gram-positive pathogenic bacteria.

Assessment of Pre-Harvest Environmental Factors in Domestic Production of Organic Lettuce (국내 유기상추의 생산환경 조사분석)

  • Among pre-harvest environmental factors, increasing attention has been paid to the effects of chemical and microbiological factors on fresh produce. The occurrence and prevalence of these factors have been usually studied with regard to the final products at the post-harvesting stage and/or when they are sold in the market. However, the origin and routes of transmission of both factors remain to be clarified. In the present study, we examined the contamination levels of food-borne pathogens and chemical factors such as pesticide residues and heavy metals in 83 and 43 samples, respectively, including various soil, water, and fertilizer samples, as well as post-harvested and processed samples. Among the organic farming samples, only one pesticide, dimethomorph, was detected in the soil sample, however no pesticides were observed from any other samples in organic farming system. Thus, it was thought that might be contaminated from conventional farm land in the vicinity. Whereas many pesticide residues were detected in conventional farming systems such as soil, fertilizer, water, and fresh produce as expected. Furthermore, heavy metals detected from all tested samples did not shown contamination levels higher than the standard limit. We comparatively assessed the levels of contamination by food-borne pathogens on the samples from organic and conventional farming systems, and found aerobic bacteria at approximately 7 log CFU/g, with no significant differences observed between the two systems. Coliforms were present at lower levels than aerobic bacteria. No human pathogens were present among the coliforms detected, indicating that these bacteria are saprophytes without the ability to cause food-borne illnesses. In contrast, among the high-risk food-borne pathogens, only sporadic cells of Bacillus cereus were found on samples of organic farming system. These data extend previous findings that the most prevalent food-borne pathogen is B. cereus and demonstrate that it spreads to whole living plants via soil.

Investigation of Chemical Properties of the Jujube Orchard Soils at Boeun Region in Chungbuk (충북 보은지역 대추재배 토양의 화학적 특성 조사)

  • BACKGROUND: Recently, as the consumption of fresh jujube is increased, fertilizer in jujube cultivation is excessively used to supply nutrient for large fruit produce. This study was conducted to obtain the useful data related to optimum nutrient management technique for fresh jujube cultivation. METHODS AND RESULTS: Nutrient contents of the jujube orchard soils were investigated at 30 different jujube orchards in Boeun, Chungbuk. Soil samples were collected from the different orchards in June, both 2012 and 2013. Soil chemical properties such as pH, organic matter, available phosphate, and exchangeable potassium, calcium, and magnesium were analyzed. Soil available phosphates in optimum level for jujube cultivation were 7% and 13% of total samples in the 2011 and 2012 respectively, and 73% and 57% were higher than optimum level. In Exchangeable K, 37%, 30% were optimum level, 63%, 67% were higher in the year 2011 and 2012 respectively. CONCLUSION: These results showed that nutrient contents of soils were accumulated in jujube orchard of Boeun area. Especially, available phosphate and exchangeable potassium were greatly higher than their optimum level for jujube cultivation respectively.

  • The purpose of this article is twofold: 1) to provide nutrition professionals with the history of how the concept of sustainable diets was introduced to the nutrition profession and 2) to describe how different sustainable agricultural practices offer potential for improving human nutrition and health. The idea of connecting sustainable agriculture With the promotion of human nutrition and health is not new. It is a concept that was introduced to the nutrition profession more than 20 years ago. To foster healthful, sustainable diets, consumers need to choose more whole foods. From the standpoint of nutrition and health, whole foods are naturally higher in fiber and lower in fat, sodium, sugar and additives compared to highly processed foods. From the standpoint of agricultural sustainability, whole foods bypass the high-energy costs of food processing and transportation. Organic farming systems offer potential benefits to human health through reducing farmers' exposure to pesticides and by increasing the total phenolic content in selected food crops. Participation in farmers' market nutrition programs and gardening has been associated with increased fruit and vegetable intake. Gardening may also be a way to increase access to fresh produce in low-income populations that do not have access to nutritious food outlets and to increase physical activity. Further research is needed to assess the human nutrition and health benefits of other types of sustainable agriculture strategies and to investigate the links among different agricultural practices with nutrient and total phenolic content in a wider variety of important food crops.

  • Among pigment used at work of Dan-Chung, Emerald Green is specific illuminating fluorescent light of green. It is very difficult to change other organic or inorganic pigment. All of the internal high class pigment has rare light. But Emerald Green is superior to fresh color and stability out of industrial chemical products. It forms over 50% of quantity and importance of a pattern painting. Emerald Green prohibited to produce because of its toxicpollutants, so required to changing pigment development.   • This study aims to find the taste compounds of wild loach, Misgurnis mizolepis, which is important fresh-water fish in this country. In order to elucidate the origin of the taste of wild loach meat, free amino acids, organic bases, nucleotides and their related compounds, organic acids, sugars and minerals were analyzed, and then followed by organoleptic test of synthetic extract prepared on the basis of the analytical data. Sensory evaluations of synthetic extracts prepared by omitting each extractive components were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, histidine, threonine, glycine, lysine, alanine were abundant. IMP and AMP were dominant in the wild loach in content. Total creatinine and butyric acid were abundant. As for the minerals, $Na^+,\;K^+,\;Cl^-\;and\;PO_4^{3-}$ were found to be the major ions. From the results of omission test of synthetic extracts the major components which contribute to produce the taste were arginine, glutamic acid, valine, aspartic acid, IMP, succinic acid, $Na^+,\;K^+,\;Ca^{2+}\;Mg^{2+}\;Cl^-\;and\;PO_4^{3-}$.

Monitoring of Pathogenic Bacteria in Organic Vegetables from Korean Market (국내 유통중인 유기농 채소류의 미생물 분포도 분석)

  • This study was undertaken to assess the microbiological quality and prevalence of pathogens in organic vegetables produced in Korea. A total of 189 organically grown vegetable samples (perilla leaf 50, lettuce 50, tomato 39, cucumber 50) were analyzed for the presence of aerobic plate count, Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, and Yersinia enterocolitica. The total aerobic plate counts were in the range of 4.2 to $7.7log\;CFU\;g^{-1}$ for perilla leaf, 5.0 to $8.0log\;CFU\;g^{-1}$ for lettuce, 4.0 to $7.5log\;CFU\;g^{-1}$ for tomato, and 6.6 to $8.6log\;CFU\;g^{-1}$ for cucumber. The highest counts were found in cucumber. E. coli O157:H7, Salmonella spp., S. aureus, L. monocytogenes, and Y. enterocolitica were not detected from any organically grown vegetable samples. This research suggests that continuous monitoring in organic vegetables is required to improve fresh produce safety.

  • This study was directed to define the taste compounds of crucian carp, Carassius caressius, free amino acids, nucleotides and their related compounds, organic bases, sugars, organic acids and minerals in the extracts of crucian carp were analyzed, and then followed by sensory evaluation of synthetic extracts prepared from 44 pure chemicals on the basis of the analytical data. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, histidine was dominant occupying $46\%$ of the total free amino acids. The other abundant free amino acids were glycine, lysine, alanine and taurine. As for the nucleotides, IMP was dominant showing about $80\%$ of the total of nucleotides. The most abundant organic base was total creatinine. The content of betaine was poor and TMAO were trace in content. The main organic acids were succinic, propionic, butyric and valeric acid. Small amount of glucose, fructose and inositol were detected and ribose and arabinose were trace in content $K^+,\;Na^+,\;PO_4^{3-}\;and\;Cl^-$ were found to be the major ions and small amount of $Ca^{++}\;and\;Mg^{2+}$ were deleted. Judging from the results of omission test, the major components which contribute to produce the taste were serine, glutamic acid, lysine, arginine, tyrosine, phenylalanine, IMP, $Na^+,\;K^+\;and\;PO_4^{3-}$.

Among organic bases total creatinine was abundant. The amount of betaine was 24 mg/100 g. The main organic acid were butyric acid, valeric acid and succinic acid. As for the sugars, glucose and inositol were 1-2 mg/ 100g in content. $K^+,\;Na^+,\;PO_{4}^{3-}\;and\;Cl^-$ were found to be the major ions. From the results of omission test the major components which contribute to produce the taste were glycine, serine, glutamic acid, IMP, $Na^+,\;K^+,\;Cl^-,\;PO_{4}^{3-}$, lysine, alanine, isoleucine, aspartic acid and creatinine.

Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, glycine was dominant occupying $25\%$ of total free amino acids. The other abundant free amino acids were lysine, taurine and alanine. Judging from the result of analysis of nucleotides, IMP was dominant showing about $70\%$ of total nucleotides while ATP, ADP, AMP, inosine and hypoxanthine were low in content. Among organic bases, total creatinine was abundant and its nitrogen content occupied more than $50\%$ of the total extractive nitrogen. The amount of betaine was 18mg/100g and trace amount of trimethylamine and trimethylamine oxide were detected. The main organic acids were succinic, butyric, propionic and valeric acid. On the other hand, oxalic, fumaric, maleic, tartaric and citric acids were analyzed in trace. As for the sugars, glucose was found to be the most abundant monosaccharide. Extremely small amounts of fructoae, inositol were also detected and ribose and arabinose were trace in content. $K^+,\;Na^+,\;PO_{4}^{3-}$ and $Cl^-$ were found to be the major ions and small amount of $Ca^{2+}$ were defected. The synthetic extract, prepared with about 40 pure chemicals based on the analytical data satisfactorily revealed the natural taste of the original extract except slight difference in meaty taste and mildness. From the results of omission test the major components which contribute to produce the taste were serine, IMP, succinic acid and $PO_{4}^{3-}$.
