Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.
Growth and flowering of Cymbidium 'Red Fire' and 'Yokihi' plants were examined in a greenhouse with cooling systems in summer, and with night interruption (NI) lighting in winter as a forcing culture system. The greenhouse was divided into two sections with separate cooling controls during the summer season. One section was cooled by a mist system (mist), while the other section was cooled by a shade screen (shade). During the winter, the greenhouse was redivided into three sections within each cooling system. Plants were grown with NI either at a low light intensity of $3-7{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(LNI) or a high l ight intensity of $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(HNI) u sing h igh-pressure sodium l amps during the 22:00-02:00 HR. The control plants were grown under 9 h short-day condition. NI for 16 weeks and cooling for 9 weeks were employed twice during the 2 years of the experimental period. The air temperature was approximately $2^{\circ}C$ lower in the mist than in the shade and the relative humidity was 80 ${\pm}5%$ in the mist compared to $55{\pm}5%$ in the shade. The daily light integral in the mist section was 48% higher than in the shade section. The time from initial planting to flowering pseudobulb emergence decreased with both LNI and HNI for both cultivars, regardless of the cooling treatments. Under NI conditions, however, between 60% and 1 00% of plants of both cultivars flowered in the mist, whereas no or 20% of 'Red Fire' or 'Yokihi' plants, respectively, flowered in the shade treatment over 2 years. Plants grown under the mist had bigger pseudobulbs than those grown in the shade under both NI treatments. These results show that commercial use of NI in winter and a mist cooling system in summer would decrease crop production time to 2 years and increase profits in Cymbidium forcing culture.
Cymbidium mosaic virus (CymMV) is one of economically important viruses that cause significant losses of orchids in the world. In the present study, a reverse transcription recombinase polymerase amplification (RT-RPA) assay combined with a lateral flow immunostrip (LFI) assay was developed for the detection of CymMV in orchid plants. A pair of primers containing fluorescent probes at each terminus that amplifies highly specifically a part of the coat protein gene of CymMV was determined for RT-RPA assay. The RT-RPA assay involved incubation at an isothermal temperature (39℃) and could be performed rapidly within 30 min. In addition, no cross-reactivity was observed to occur with odontoglossum ringspot virus and cymbidium chlorotic mosaic virus. The RT-RPA with LFI assay (RT-RPA-LFI) for CymMV showed 100 times more sensitivity than conventional reverse transcription polymerase chain reaction (RT-PCR). Furthermore, the RT-PCR-LFI assay demonstrated the simplicity and the rapidity of CymMV detection since the assay did not require any equipment, by comparing results with those of conventional RT-PCR. On-site application of the RT-RPA-LFI assay was validated for the detection of CymMV in field-collected orchids, indicating a simple, rapid, sensitive, and reliable method for detecting CymMV in orchids.
In vitro mass multiplication of Vanilla planifolia was investigated using node as explant. Multiple shoots were developed in MS medium supplemented with $2.0mgl^{-1}$ 6-benzylaminopurine and $1.0mgl^{-1}$$\alpha$-naphthalene acetic acid. Multiple shoots were maintained for 6-T weeks with regular subculturing at the end of $3^{rd}$ week onto fresh medium. The maximum number of shoots at the rate of 12.8 per node segment was achieved over a period of four weeks. The elongated shoots were separated from the shoot clusters and were transferred onto half strength MS medium supplemented with indole-3-acetic acid ($1.0mgl^{-1}$) over a period of 28 days for induction of roots. The development of roots was observed on $7^{th}$ day of incubation. The in vitro raised plantlets were transferred to poly-cups, covered with polyethylene sheets and maintained under shade net for 25 days for hardening. Finally these plants were transferred to field and recorded that 85 % of tissue cultured plants were survived. From the present study, a simple and efficient micropropagation protocol was developed for Vanilla planifolia using single node segments as explants.
Orchidaceous plants have symbiotic relationships with endophytic fungi, including mycorrhizal fungi, which play important roles in the seed germination and growth of the host plants. In this study, endophytic fungal communities isolated from the roots of Cephalanthera longibracteata collected from three different sites in Korea were analyzed, and it was determined whether fungal communities were preferentially correlated with the sites. The fungal isolates were identified by sequence analysis of the internal transcribed spacer regions of rDNA. In total, 30 species of endophytic fungi, including two species of mycorrhizal fungi belonging to the genus Tulasnella, were identified. Leptodontidium orchidicola showed the highest frequency and was isolated from all root samples. Species diversity and richness were not significantly different among sites. However, the community structure of the endophytic fungi significantly differed among sites, suggesting that the site characteristics affected the community composition of the endophytic fungi colonizing the roots of C. longibracteata. Our findings will aid in developing methods involving the use of symbiotic fungi for orchid conservation and restoration in native habitats.

Sclerotium rot was found on Cymbidium orchids at Seosan-si, Chungcheongnam-do, Korea, in July, 2010. Symptoms occurred on low leaves, which turned yellowish, after which the entire plant wilted. Severely infected plants were blighted and eventually died. White mycelial mats and sclerotia appeared on pseudobulbs. Based on the mycological characteristics and pathogenicity, the causal fungus was identified as Sclerotium rolfsii. This is the first report of new Sclerotium rot on Cymbidium spp. caused by S. rolfsii in Korea.
Phytophthora crown rot of cymbidium was observed in Cheju island since June of 1996. The disease initiated at the basal portion of infected plant progressed upward to lower leaves. Soon after distinct water-soaking lesions appeared on lower leaves, the plant was wilted, blighted and died. Four orchid farms at Sogwipo out of 16 surveyed in the island were infected by the disease estimating 5~20% infection rates. The causal fungus was identified as P. palmivora based on following distinguishing characteristics. All isolates were heterothallic as A1 types and readily produced chlamydospores with cultural age. Sporangia were conspicuous papillate, ellipsoidal to ovoid, highly deciduous with short pedicels ca. 3~4 ${\mu}{\textrm}{m}$. Koch's rules were satisfied by a pathogenicity test and re-isolation of the fungus from inoculated plants. The pathogen has never been reported in Cheju island previously and its firstly recorded as the cause of Phytophthora crown rot of cymbidium in Korea.
난과식물의 형질전환 유도의 기초연구로서 자란(Bletilla striata)의 미성숙 종자로 부터 embryogenic callus를 유도하고, 이 배발생 캘러스의 현탁배양을 하여 이로부터 분리한 원형질체에 여러가지 조건하에서 electroporation을 행하고 원형질체의 생존율(viability)을 조사하였다. Voltage와 capacitancy의 증가에 의해 생존율은 감소를 가져 왔으며, HBM buffer에서 $4^{\circ}C$로 electroporation시 원형질체의 생존율에 있어서 안정성을 높일 수 있었다.
신품종 Phalaenopsis 'SM 333'은 2002년 상미원에서 Phalaenopsis 'Odoriko'와 Phalaenopsis 'Be Tris'를 모부본으로 인공교배하여 육성한 $F_1$ 개체 중에서 선발한 핑크 소형 다화 분지성이다. 2003년부터 2004년까지 2년에 걸쳐 실생 300개체를 양성하여 이들 중에서 영양생장과 개화특성이 우수한 개체 02-03-33을 선발하였다. 2004년과 2005년에 1차, 2차 특성검정을 통하여 품종의 안정성과 균일성을 확인하고 'SM 333'으로 명명하였다. 'SM 333'은 화색이 화사하고 선명한 핑크(RP69D)이며, 화형은 앉아피기로 안정되어 있다. 꽃의 길이와 폭은 각각 5.0, 5.8cm로 소형이다. 복총상화서로 볼륨이 있으며 소형 캐주얼분화에 적합하다. 잎의 길이와 폭은 각각 20.8, 6.5cm이며 엽형은 수평이다. 기내증식율이 높고 변이가 거의 없으며 영양생장 우수하여 재배관리가 용이하다. 2009년 12월 1일 품종등록(등록번호 제2916호)하여 종자산업법에 의해 보호받고 있다.
