Optimization of Shark (Squatina oculata) Cartilage Hydrolysis for the Preparation of Chondroitin Sulfate

  • Enzymatic hydrolysis of shark (Squatina oculata) cartilage (SC) was optimized by response surface methodology (RSM) for chondroitin sulfate (CS) preparation. Among 11 commercial proteases, Maxazyme NNP showed highest productivity (CS yield per enzyme cost) of CS. Optimal hydrolysis conditions determined by RSM were 1.63% and 2.87 hr for enzyme concentration and hydrolysis time ($r^2\;=\;0.9527$, p<0.0l), respectively and highest yield of hydrolysate under the conditions was 42.3%. The yield ($43.1{\pm}2.1%$) and CS content ($24.8{\pm}0.1%$) of hydrolysate at optimal condition verified statistical optimization of SC enzymatic hydrolysis was valid.

  • CIGS thin films have received a great attention as a promising material for solar cells due to their high absorption coefficient, appropriate bandgap, long-term stability, and low cost production. CIGS thin films have been deposited by various methods such as co-evaporation, sputtering, spray pyrolysis and electro-deposition. In this study, Cu(In,Ga)Se2(CIGS) thin films were prepared using a single quaternary target by rf magnetron sputtering. The effect of sulfurization on the structural, compositional and electrical properties of the films was examined in order to develop the deposition process. An optimal sulfurization process will be selected for the preparation of CIGS thin films with good structural, optical and electrical properties by applying various sulfurization processes. In addition, the electrical properties of CIGS thin films were investigated by post-deposition annealing process. The carrier concentration of CIG(SSe) thin films after sulfurization was increased from $10^{14}cm^{-3}$ to $10^{16}cm^{-3}$ and the resistivity was increased from 10 ${\Omega}cm$ to $10^3$ ${\Omega}cm$. It is confirmed that CIG(SSe) thin films prepared at optimal deposition condition have similar atomic ratio to the target value after sulfurization.

Processing Optimization and Quality Characteristics of Low-Fat Yogurt Prepared with Roselle (로젤 첨가 저지방 요구르트의 제조 조건 최적화 및 품질특성)

  • The purpose of this study was to determine the optimal amounts and mixing condition of skim milk powder (A), and roselle (B) for the production of yogurt prepared with roselle. The experiment was designed according to the central composite design for estimating the response surface method, which yielded ten experimental points, including two replicates for the skim milk powder and roselle. The physicochemical and mechanical analysis of each sample, including pH (P<0.001), titratable acidity (P<0.001), color (P<0.05), viscosity (P<0.001), showed significant differences. Antioxidant properties (total phenolic content, DPPH free radical scavenging activity) and viable cell counts of lactic acid were significantly different (P<0.05). The sensory measurements were significantly different in color, flavor, sourness, texture, and overall quality (P<0.05). The optimal formulation, calculated by numerical and graphical methods, was 7.82 g of skim milk powder and 2.09 g of roselle. From findings of this study, the roselle may be used in yogurt and can be applied for other food industries.

Optimizing Steeping Conditions of Waxy Rice Based on the Sensory Properties of Gangjung (a Traditional Korean Oil-Puffed Snack) (강정의 관능적 특성에 의한 찹쌀의 수침조건 최적화)

  • This study was conducted to determine the optimal steeping period and temperature for Gangjung production, using response surface methodology based on the previously reported sensory characteristics. Five sensory attributes ('degree of expansion', 'sourness', 'butyric acid flavor', 'hardness' and 'degree of melting'), which showed high variability explained ($R^2$) and recognized to be important for the quality of Gangjung were selected for optimization. As a result, the optimal steeping temperature and period of waxy rice were determined to be $31.5^{\circ}C$ and 9 days, respectively.

  • Topology optimization is applied for the optimal design of various products to ensure weight reduction and productivity improvement. Reducing the weight of the mold while maintaining its rigidity can ensure shortening of the production cycle, stabilization of the mold temperature, and reduction of mold material costs. In this study, a topology optimization technique was applied to the optimal design of the injection mold, and a topology-optimized model of the mold was obtained. First, the injection mold for the square specimens was modeled. Subsequently, a structural analysis was performed by implementing a load condition generated during the injection molding process. Topology optimization was performed based on the structural analysis results, and the models of the initial and topology-optimized designs were manufactured at 1/4 magnification using a 3D printer. Consequently, compared with the existing model, the weight of the topology-optimized model decreased by 9.8%, and the manufacturing time decreased by 7.61%.

Investigation of 180W separation by transient single withdrawal cascade using Salp Swarm optimization algorithm

  • The 180W is the lightest isotope of Tungsten with small abundance ratio. It is slightly radioactive (α decay), with an extremely long half-life. Its separation is possible by non-conventional single withdrawal cascades. The 180W is used in radioisotopes production and study of metals through gamma-ray spectroscopy. In this paper, single withdrawal cascade model is developed to evaluate multicomponent separation in non-conventional transient cascades, and available experimental results are used for validation. Numerical studies for separation of 180W in a transient single withdrawal cascade are performed. Parameters affecting the separation and equilibrium time of cascade such as number of stages, cascade arrangements, feed location and flow rate for a fixed number of gas centrifuges (GC) are investigated. The Salp Swarm Algorithm (SSA) as a bio-inspired optimization algorithm is applied as a novel method to minimize the feed consumption to obtain desired concentration in the collection tank. Examining different cascade arrangements, it is observed in arrangements with more stages, the separation is further efficient. Based on the obtained results, with increasing feed flow rate, for fixed product concentration, the cascade equilibrium time decreases. Also, it is shown while the feed location is the farthest stage from the collection tank, the separation and cascade equilibrium time are well-organized. Finally, using SSA optimal parameters of the cascade is calculated, and optimal arrangement to produce 5 gr of 180W with 90% concentration in the tank, is proposed.

Optimization of Cellulase Production from Paenibacillus jamilae BRC 15-1 (Paenibacillus jamilae BRC15-1의 Cellulase 생산 최적화)

  • In this study was selected the cellulolytic microorganism and investigated optimum condition of cellulase production for the cellulosic bioethanol production. A bacterial strain Paenibacillus jamilae BRC15-1, was isolated from soil of domestic reclaimed land. For optimizing cellulase production from the selected strain, various culture parameters were investigated such as culture medium, pH (pH 4~10), temperature ($25{\sim}50^{\circ}C$) and culture time (2~72 h). As a result, P. jamilae BRC15-1 efficiently produced cellulase from cellulosic biomass under following conditions: 24 h of culture time (pH 7, $40^{\circ}C$) in manufactured media of CMC (carboxymethyl cellulose) with peptone. Optimum saccharifying condition of crude enzyme produced from P. jamilae BRC15-1 was identified on pH 6 and $40^{\circ}C$ of reaction temperature, respectively. This crude enzyme from P. jamilae BRC15-1 was used for saccharification of pretreated sweet sorghum (Sorghum bicolor var. dulciusculum Ohwi) bagasse under the optimal condition. Finally, pretreated sweet sorghum bagasse including 0.1 g of glucan was saccharified by crude enzyme of P. jamilae BRC15-1 into 2.75 mg glucose, 0.79 mg xylose and 1.12 mg arabinose.

Continuous Mesophilic-Dry Anaerobic Digestion of Organic Solid Waste (유기성고형폐기물의 연속 중온 건식혐기성소화)

  • Continuous dry anaerobic digestion of organic solid wastes (30% TS, Total Solids) comprised of food waste and paper was performed under mesophilic condition. During the operation, hydraulic retention time (HRT) was decreased as follows: 150 d, 100 d, 60 d, and 40 d, which corresponded to the solid loading rate of 2.0, 3.0, 5.0, and 7.5 kg TS/$m^3$/d, respectively. Volumetric biogas production rate ($m^3$/$m^3$/d) increased as HRT decreased, and the highest biogas production rate of 3.49${\pm}$0.31 $m^3$/$m^3$/d was achieved at 40 d of HRT. At this HRT, high volatile solids (VS) reduction of 76% was maintained, and methane production yield of 0.25 $m^3$/kg $TS_{added}$ was achieved, indicating 67.4% conversion of organic solid waste to bioenergy. The highest biogas production yield of 0.52 $m^3$/kg $TS_{added}$ was achieved at 100 d of HRT, but it did not change much with respect to HRT. For the ease feed pumping, some amount of digester sludge was recycled and mixed with fresh feed to decrease the solid content. Recirculation volume of 5Q was found to be the optimal in this experimental condition. Specific methanogenic activity (SMA) of microorganisms at mesophilic-dry condition was 2.66, 1.94, and 1.20 mL $CH_4$/g VS/d using acetate, butyrate, and propionate as a substrate, respectively.

Characteristics of Growth and Germination of Salicornia herbacea L. for the Soil salinity and Manure Condition (토양염분.시비 조건에 따른 퉁퉁마디 생장 및 발아 특성)

  • This experiment was carried out to investigate the cultivation condition of soil salinity, manure and to find out the cultivation capability of Salicornia herbacea. The optimal growth condition of soil salinity was between 1% and 4%. The growth of groups for fertilization was significantly better than control group. Salicornia herbacea grew very up from 7 kg/l0a to 9 kg/l0a for N, 12 kg/l0a for P and the K-fertilizer group was better than control group but there was not significantly different among the conditions of K-concentration. The germination was good from $0{\textperthousand}\;to\;5{\textperthousand}$ for salinity, from $20^{\circ}C\;to\;30^{\circ}C$ for temperature. On the experiment cultivation, the production by hill seeding was $5.40{\sim}5.90 kg/m^2$ and was significantly higher than the yield by broadcast sowing which was $4.01{\sim}4.20 kg/m^2$. The production by hill seeding was 2.7 times than natural production and the production by broadcast sowing 1.9 times.

SynGas Production from Propane using GlidArc Plasma Reforming (부채꼴방전 플라즈마 개질을 이용한 프로판으로부터의 합성가스 생산)

  • The purpose of this paper was to investigate the reforming characteristics and optimum operating condition of the GlidArc-assisted $C_3H_8$ reforming reaction for the synthesis gas(SynGas) production without formation of carbon black from propane using GildArc plasma reforming. Also, in order to increase the hydrogen production and the propane conversion rate, 13 wt % nickel catalyst was filled into the catalytic reactor and parametric screening studies were conducted, in which there were the variations of vapor mole ratio$(H_2O/C_3H_8),\;CO_2$ mole ratio($CO_2/C_3H_8$), input power and injection flow rate. When the variations of vapor mole ratio, $CO_2$ mole ratio, input power and injection flow rate were 1.86, 0.48, 1.37 kW and 14 L/min, respectively, the conversion rate of the propane reached its most optimal condition, or 62.6%. Under the condition mentioned above, the dry basic concentrations of the SynGas were $H_2\;44.4%,\;CO\;18.2%,\;CH_4\;11.2%,\;C_2H_2\;2.0%,\;C_3H_6\;1.6%,\;C_2H_4\;0.6%\;and\;C_3H_4$ 0.4%. The conversion rate of carbon dioxide was 29.2% and the concentration ratio of hydrogen to carbon monoxide($H_2/CO$) in the SynGas was 2.4.
