Optimal Conditions for the Production of Immunostimulating Polysaccharides from the Suspension Culture of Angelica gigas Cells. (면역증강성 다당 생산을 위한 참당귀 세포배양의 최적조건)

  • An Immunostimulating polysaccharide was produced from the suspension culture of Angelica gigas H4, plant cells. In order to enhance the polysaccharide production by the A. gigas cell culture, medium composition and physical conditions were optimized. Schenk and Hildebrandt (SH) medium was selected as an optimal basal medium for the growth of A. gigas. The maximum cell and polysaccharide concentration obtained in SH medium were 15.8 g DCW/l and 0.85 g polysaccharide/l, respectively, at $25^{\circ}C$ under dark condition. For the enhanced polysaccharide production, a polysaccharide production medium (PPM) was established by modifying Gamborg B5 medium with optimized carbon sources, growth regulators, organic and inorganic elements. Optimal initial pH and temperature were 6.0-6.6 and $20^{\circ}C$, respectively, and the dark condition was better than the light condition. The maximum polysaccharide concentration of 1.5 g/l could be obtained through the optimization of the medium composition and physical conditions.

  • The purpose of the present study is to find the optimal growth condition of Crypthecodinium cohnii. Using a design of experiments, it was found that the yeast extract was the most significant factor for cell growth. The effect of glucose was less than that of yeast extract and the effect of NaCl was negligible. The highest cell density, 1O.96g/L was obtained at the concentration of yeast extract 7.5g/L.

  • Chronic kidney disease-mineral bone disorder (CKD-MBD) is a systemic disorder of mineral and bone metabolism caused by CKD. Patients with early-stage CKD who present with disordered regulation of bone and mineral metabolism may be asymptomatic. However, if untreated, the condition can be a significant barrier in achieving optimal bone strength, linear growth, and cardiovascular health in pediatric patients with CKD. Thus, the current study evaluated the definition, pathogenesis, diagnosis, and management of pediatric CKD-MBD.

Optimal Culture Condition and Extracellular Enzyme Activity of Metarhizium anisopliae (녹강균(Metarhizium anisopliae)의 최적 배양조건 및 효소활성)

  • In order to use Metarhizizmn~ anisopliae as a biological pesticide, effect of envrionmental factors on nlycelial growth, spore formation, and extracellular enzyme activity in culture broth of M. anisopliae DGUM 35001 was determined. Optimal temperature was 26^{\circ}C.$ and optimal pH ranged from 5 to 9. Among the complex media tested, MCM and SDPY media were the most favorable for mycelial growth. When Czapek-Dox agar was used as a mnimal medium, glucose and sucrose among the saccharides were very excellent source of carbohydrate. Among the biopolyners tested. chitin was the most favorable source for mycelial growth and produced high aerial inycelia. Urea and ammonium phosphate as an inorganic nitrogen source and bacto-peptone and soytone as an organic nitrogen source enhanced the mycelial growth When serine as a source of amino acid was supplemented, excellent mycelial growth was shown. Large amount of spores could be obtained from the aerial mycelia of starch medium. When the culture broth was filtrated and then the concentrate with ammonium sulfate was used as a crnde enzyme solution, high enzyme activities of amylase and protease were shown. However, lipase and chitinase activities were comparatively low.

Selection of Optimal Application Condition of Corn Waste Biochar for Improvement of Corn Growth and Soil Fertility

  • This study was conducted to select an optimal approach to corn waste biochar (BC) application, and to evaluate the effects of combined application of BC and inorganic fertilizer (IF) on corn growth and soil chemical properties in a pot experiment. Corn growth differed with BC application timing and BC application levels. Based on the characteristics of corn growth in pot-based experiments, the selected optimal application conditions of BC were application of $500kg\;10a^{-1}$ at 20 days before sowing. Also, the chemical properties of the tested soil with BC after corn harvesting were significantly improved than those in the other treatments. In particular, soil pH and CEC regardless of application conditions were markedly increased by 0.04~0.19 units and $0.08{\sim}2.58coml_c\;kg^{-1}$ in BC treatments than without BC treatments. Additionally, combined application of BC and IF had greater effects on corn growth than single application of BC. Therefore, the results suggest using properly BC application conditions and a combination of BC and IF for effective corn cultivation in an upland field.

Characterization of Single-walled Carbon Nanotubes Synthesized by Water-assisted Catalytic Chemical Vapor Deposition

  • The influence of the water vapor on the growth of single-walled carbon nanotubes (SWCNTs) was investigated. SWCNTs were synthesized by catalytic chemical vapor deposition of acetylene over Fe-Mo/MgO catalyst with injection of water vapor. The morphologies and structures of the water-assisted SWCNTs were investigated according to the growth conditions such as water vapor concentrations, flow rate of the gas, furnace temperature, and growth time. Water-assisted SWCNTs exhibited large bundle morphological features with well-alignment of each CNT, while SWCNTs synthesized in the absence of water vapor showed entangled CNT with the random orientation. We also found that the diameter of the SWCNT bundle could be controlled by the growth condition. In our optimal growth condition, the product yield and the purity were 300 wt. % and 75%, which were 7.5 and 2.5 times higher than those of SWCNTs synthesized without water vapor, respectively. A Simulation Study on the Improvement of Lighting Condition on Sidewalks Considering the Type and Growth of Roadside Trees (가로수의 유형 및 성장을 고려한 보행로 조명환경 개선에 관한 시뮬레이션 연구)

  • In recently, a growing concern for the health of urban residents increased interests in a variety of outdoor activities simply be done in terms of cost and time. They are specially interested in low-impact and safe exercises around residential or working area. Walking is the one of easily doing exercise in daytime or nighttime near residential area. The sidewalks of boulevard near the residential area is the best place for exercise because of easy access and the green space with roadside trees. However, if the nighttime is not guaranteed the proper lighting condition, the possibility of exposure to crime and the threat to pedestrian safety can be increased. Because roadside trees are one of the potential obstacle for lighting condition, supplementary lightings are important to mitigate interruption for safety. To meet such a need, the purpose of this study is to propose a simulation approach which improves lighting condition on sidewalks of boulevard with variety of roadside trees. To do so, the simulation approach is applied for analyzing the interrupted condition by classified five standard types of roadside trees considering the growth of them and finding optimal layout of supplementary luminaires by lighting types. The results of this approach shows that it is useful for assessing the safety of pedestrian in nighttime.

A Study on High Temperature Low Cycle Fatigue Crack Growth Modelling by Neural Networks (신경회로망을 이용한 고온 저사이클 피로균열성장 모델링에 관한 연구)

  • This paper presents crack growth analysis approach on the basis of neural networks, a branch of cognitive science to high temperature low cycle fatigue that shows strong nonlinearity in material behavior. As the number of data patterns on crack growth increase, pattern classification occurs well and two point representation scheme with gradient of crack growth curve simulates crack growth rate better than one point representation scheme. Optimal number of learning data exists and excessive number of learning data increases estimated mean error with remarkable learning time J-da/dt relation predicted by neural networks shows that test condition with unlearned data is simulated well within estimated mean error(5%).
