The current left-turn split model adopted in COSMOS has an inherent limitation when a loop detector in the left-turn lanes was disconnected for a period of time. In this instance, the current model always allocated minimum green time to the left-turn phase, thus optimal split and efficient signal operation for the intersection was not guaranteed. In this paper, four mathmatical models using detector information of the intersection and four empirical models using historical profiles were developed and investigated for different traffic conditions to improve the operational efficiency of the intersection. From the model evaluation test, the empirical model using a four-week historical profile produced the least error among the eight models investigated. NETSIM simulation test results also showed that the proposed model could give significantly reduced delay time as compared to the current model. From these results, the operational efficency of the signalized intersections under the real-time control can be greatly improved by using the model proposed in case of the left-turn detector failure.
Aflatoxin B1 (AFB1) is produced by Aspergillus flavus growing in feedstuffs. Early detection of maize contamination by aflatoxigenic fungi is advantageous since aflatoxins exert adverse health effects. In this study, we report the development of an optimized conventional PCR for AFB1 detection and a rapid, sensitive and simple screening Real-time PCR (qPCR) with SYBR Green and two pairs of primers targeting the aflR genes which involved aflatoxin biosynthesis. AFB1 contaminated maize samples were divided into three groups by the toxin concentration. Genomic DNA was extracted from those samples. The target genes for A. flavus were tested by conventional PCR and the PCR products were analyzed by electrophoresis. A conventional PCR was carried out as nested PCR to verify the gene amplicon sizes. PCR-RFLP patterns, obtained with Hinc II and Pvu II enzyme analysis showed the differences to distinguish aflatoxin-producing fungi. However, they are not quantitative and need a separation of the products on gel and their visualization under UV light. On the other hand, qPCR facilitates the monitoring of the reaction as it progresses. It does not require post-PCR handling, which reduces the risk of cross-contamination and handling errors. It results in a much faster throughout. We found that the optimal primer annealing temperature was $65^{\circ}C$. The optimized template and primer concentration were $1.5{\mu}L\;(50ng/{\mu}L)$ and $3{\mu}L\;(10{\mu}M/{\mu}L)$ respectively. SYBR Green qPCR of four genes demonstrated amplification curves and melting peaks for tub1, afIM, afIR, and afID genes are at $88.0^{\circ}C$, $87.5^{\circ}C$, $83.5^{\circ}C$, and $89.5^{\circ}C$ respectively. Consequently, it was found that the four primers had elevated annealing temperatures, nevertheless it is desirable since it enhances the DNA binding specificity of the dye. New qPCR protocol could be employed for the determination of aflatoxin content in feedstuff samples.
This study was performed to compare nutritional compounds and physiological functions of five domestic and imported green tea cultivars at three time points. The five cultivars were compared for theanine, γ-aminobutyric acid, and catechin content by LC-MS/MS and HPLC. Furthermore, the five tea cultivars were functionally characterized with respect to antioxidant activity, brain cell protective effect, and inhibitions of α-glucosidase and HMG-CoA reductase activities. Among green tea cultivars, Chamnok had the highest content of catechins (198 mg/g DW), theanine (11.89 mg/g DW), and tannin (23.6 mg/g DW). Considering functional properties, Chamnok treatment resulted in the maximum viability of brain cells and reduced the cortisol content of SH-SY5Y cells. The inhibition of α-glucosidase and HMG-CoA reductase was the strongest following Chamnok treatment (72.9% and 69.8%, respectively). These results indicate that Chamnok could be optimal for consumption or favorable processing owing to its high nutritional compounds, such as theanine and catechin, and remarkable brain cell protective effects.

Background: Ginseng berries (GBs) show temporal metabolic variations among different maturation stages, determining their organoleptic and functional properties. Methods: We analyzed metabolic variations concomitant to five different maturation stages of GBs including immature green (IG), mature green (MG), partially red (PR), fully red (FR), and overmature red (OR) using mass spectrometry (MS)-based metabolomic profiling and multivariate analyses. Results: The partial least squares discriminant analysis score plot based on gas chromatography-MS datasets highlighted metabolic disparity between preharvest (IG and MG) and harvest/postharvest (PR, FR, and OR) GB extracts along PLS1 (34.9%) with MG distinctly segregated across PLS2 (18.2%). Forty-three significantly discriminant primary metabolites were identified encompassing five developmental stages (variable importance in projection > 1.0, p < 0.05). Among them, most amino acids, organic acids, 5-C sugars, ethanolamines, purines, and palmitic acid were detected in preharvest GB extracts, whereas 6-C sugars, phenolic acid, and oleamide levels were distinctly higher during later maturation stages. Similarly, the partial least squares discriminant analysis based on liquid chromatography-MS datasets displayed preharvest and harvest/postharvest stages clustered across PLS1 (11.1 %); however, MG and PR were separated from IG, FR, and OR along PLS2 (5.6 %). Overall, 24 secondary metabolites were observed significantly discriminant (variable importance in projection > 1.0, p < 0.05), with most displaying higher relative abundance during preharvest stages excluding ginsenosides Rg1 and Re. Furthermore, we observed strong positive correlations between total flavonoid and phenolic metabolite contents in GB extracts and antioxidant activity. Conclusion: Comprehending the dynamic metabolic variations associated with GB maturation stages rationalize their optimal harvest time per se the related agroeconomic traits.
A single process using hot water (0% green liquor) and green liquor (GL) was investigated for pre-pulping extraction on two types of raw material. The GL was applied at different alkali charges of 0-5% on a dry wood weight basis. The extractions were performed at an H-factor 900 at $180^{\circ}C$. The 0% and 3% GL extraction detected acetic acid (AA) at 10.02 and $9.94g/{\ell}$, extracts derived from hardwood, 2.46 and $3.76g/{\ell}$, extracts derived from softwood, respectively. The single liquid-liquid extraction (LLE) was studied using tri-n-alkylphosphine oxide (TAPO). Response surface methodology (RSM) was employed as an efficient approach for predictive model building and optimization of AA recovery conditions. The extraction of AA was evaluated with a three-level factorial design. Three independent variables, pH (0.5-3.5), temperature ($25-65^{\circ}C$), and residence time (24-48 min) were investigated. Applying the RSM models obtained, the optimal conditions selected of extracts derived from hard- and softwood with a 3% GL were approximately pH 1.4, $26.6^{\circ}C$, 43.8 min and approximately pH 0.7, $25.2^{\circ}C$, 24.6 min, respectively. The predicted and experimental values of AA recovery yield were similar whilst sugar retention was 100%.

The grain complex of Saemangeum is created for promoting the foundation of agriculture combined the global competitiveness. However, the masterplan is being also revised with changing of local conditions and social needs. Thus, the dynamic masterplan is needed to consider the change of time for Saemangeum project. The present study was made to set up the dynamic masterplan of Saemangeum grain complex for handling the change such as project progress, local environment, and project conditions flexibly. In this study, the dynamic masterplan for the progressive development of water supply, farmland composition, and introduction facilities is presented to the 6-2 zone in three stages. We believed that the water supply would be possible through the pumping and desalination facilities with the development stages. The farmland composition proceeded for each complex with reclamation, soil preparation, and soft soil processing. And it is planned to carry out crop cultivation from the complex where the construction is completed first. The introduction facilities were analyzed focusing on the silos and forage loading facilities, and the optimal location of them was selected using road and accessibility. The concept of dynamic masterplan may provide the direction for the planning and progress of reclamation project.
Corn cobs were fermented with Aspergillus niger to produce soluble dietary fiber (SDF) of high quality and excellent food safety. In this work, the fermentation process was optimized by single-factor test and response surface methodology (RSM). The optimal fermentation conditions were determined to be a material-liquid ratio of 1:30, an inoculum concentration of 11%, a temperature of 32℃, a time of 6 days, and a shaking speed of 200 r/min. Under these conditions, the SDF yield of corn cob increased from 2.34% to 11.92%, and the ratio of soluble dietary fiber to total dietary fiber (SDF/TDF) reached 19.08%, meeting the requirements for high-quality dietary fiber (SDF/TDF of more than 10%). Scanning electron microscopy (SEM) and Fourier-transformed infrared spectroscopy (FT-IR) analysis revealed that the fermentation effectively degraded part of cellulose and hemicellulose, resulting in the formation of a loose and porous structure. After fermentation the water swelling capacity, water-holding capacity, and oil-holding capacity of the corn cob SDF were significantly improved and the adsorption capacity of glucose, cholesterol, and nitrite ions all increased by more than 20%. Moreover, the total phenolic content increased by 20.96%, which correlated with the higher antioxidant activity of SDF. Overall, the fermentation of corn cobs by A. niger increased the yield and enhanced the functional properties of dietary fiber (DF) as well.

To analyze the influence of various groundwater flow rates (specific discharge) on BHE system with balanced and unbalanced energy loads under assuming same initial temperature (15℃) of ground and groundwater, numerical modeling using FEFLOW was used for this study. When groundwater flow is increased from 1 × 10−7 to 4 × 10−7m/s under balanced energy load, the performance of BHE system is improved about 26.7% in summer and 22.7% at winter time in a single BHE case as well as about 12.0~18.6% in summer and 7.6~8.7% in winter time depending on the number of boreholes in the grid, their array type, and bore hole separation in multiple BHE system case. In other words, the performance of BHE system is improved due to lower avT in summer and higher avT in winter time when groundwater flow becomes larger. On the contrary it is decreased owing to higher avT in summer and lower avT in winter time when the numbers of BHEs in an array are increased, Geothermal plume created at down-gradient area by groundwater flow is relatively small in balanced load condition while quite large in unbalanced load condition. Groundwater flow enhances in general the thermal efficiency by transferring heat away from the BHEs. Therefore it is highly required to obtain and to use adequate informations on hydrogeologic characterristics (K, S, hydraulic gradient, seasonal variation of groundwater temperature and water level) along with integrating groundwater flow and also hydrogeothermal properties (thermal conductivity, seasonal variation of ground temperatures etc.) of the relevant area for achieving the optimal design of BHE system.
This study was carried out to investigate the optimal application rate and time of mixed expeller cake (MEC) for the replacement of chemical fertilizer. Dongjin-1, as cultivated rice was used at Fluvio-marine deposit in Honam plain paddy field. Soil chemical properties were improved by the application of MEC. Contents of total nitrogen and organic matter were higher in 70%, 100% plots of basal dressing than standard fertilizer application (SFA) plot. Cation exchangeable capacity was highly increased in 70% plot of basal dressing. Also, the content of organic matter in soil was increased with MEC application. Cation exchangeable capacity, total nitrogen and available phosphate were decreased according to late application time. The content of inorganic nitrogen in soil showed high tendency at more application rate of MEC, and nitrogen mineralization at harvest season have finished in 50%, 70% plots of basal dressing. The content of inorganic nitrogen in soil was increased according to late application time, however it was decreased in the late period of growth. Leaf color value became darker with increased application rate of MEC. Leaf color was dark green in MEC application plots at panicle formation stage, on the other hand, it was light green in 50%, 70% plots of basal dressing at heading stage. SPAD reading value of leaf-color was high during the whole growth stage in MEC application plots. More application rate of MEC showed higher tendency of fertilizer nitrogen absorption. Nitrogen use efficiency was the highest in 70% plot of basal dressing. Absorbed amount of fertilized nitrogen was increased in 10~15days before transplanting and nitrogen use efficiency was high according to the late application time. The ratio of perfect kernel and the content of protein on hulled rice showed high tendency at the less application rate of MEC. The ratio of head rice on milled rice showed high tendency at the less application rate of MEC. Rice yield increased 4% in 100% and 70% plots of basal dressing compare with SFA ($5.18Mg\;ha^{-1}$) plot respectively. Ear and culm length of rice were long according to the late application time, while the numbers of spikelet and ear were increased and the percentage of ripened grain was decreased. Rice yield was increased 2~5% in all MEC application plots compared to SFA plot and especially, increased 10~15days before transplanting in application plots. Italian ryegrass is generally cultivated by a winter forage in a paddy field, the harvest season overlaps with rice transplantion in some cases. In addition, farmers can miss the optimal seedtime for Italian ryegrass because of the rainy season. Therefore, this research was conducted between October 2012 and June 2013 in Kwangjin, Korea to examine the effect of spring-seeding on the yields and quality of Italian ryegrass for the determination of an alternative if the optimal seedtime is missed. This trial consists of four different seeding dates (15 February, 25 February, 5 March, and 15 March) and three varieties (Kowinmaster, Kowinearly, and Green farm). The Italian ryegrass varieties "Kowinmaster," "Kowinearly," and "Green farm" were sown on February 15. The heading date of each variety showed from May 12 to May 9 and interval time of five days but the Treatment of March 15 planting date showed three days interval. The Kowinmaster variety, planted on February 15, was 93.0 cm taller on March 15 with a height of 105.7 cm. The Kowinearly and Green farm varieties planted on February 15 were of a similar height on February 25 and received a poor lodging score when planted quickly. The dry matter (DM) yield of the Kowinearly planted on February 15 was high but that of Green farm was low at 6,609 kg/ha and a difference was not shown between the varieties. The DM yield was different for those varieties planted on February 15 and March 15 (P<0.05). The crude protein content (CPC) of those varieties planted on February 15 showed an average of 8.8%, while those that were planted on March 15 showed an average that was about 2.3% higher at 11.1%; furthermore, the latter also showed lower acid detergent fiber (ADF) and total digestible nutrient (TDN) contents, by 2.1% and 1.5%, respectively. The relative feed value was about 5.9% higher, depending on the seeding date, as well as being slower and showing less difference between the varieties. According to this study, Italian ryegrass planted in a paddy field of the southern region of Korea on February 25 will produce a high-quantity yield with nutritive value.
