  • This paper investigated environmental effects for passive, air-breathing, and vapor-feeding direct methanol fuel cells. In these experiments, experimental parameters are temperature($30^{\circ}C$, $40^{\circ}C$ and relative humidity(25%, 50%, 75%). From these experimental results, the water contents play a key role in terms of optimal ionic conductivity at the cathode catalyst layer. In case of pure methanol feeding, the performance is inversely proportional to the relative humidity. The water generation resulting from methanol crossover maintains ionic conductivity at the cathode. On the contrary, diluted methanol solution (50wt.%) lowers methanol crossover to the cathode. In order to increase ionic conductivity, the relatively high humidity is required to the cathode catalyst layer for the water generation. The relative humidity scales with the performance.

  • This study was intended to develop the automatic feeding depth control system of the head-feed combine which could feed the rice head into threshing unit at the optimal depth regardless of plant height and uneven ground surface. In the control system, one-board microcomputer was used for the controller instead of conventional electric circuits. Field test of the combine equipped with the control system was conducted to evaluate its overall performance. It was also investigated how the location and time delay of rice head sensor affect the system performance.

Red Pigment Overproduction by Fed-Batch Culture of Monascus anka (Monascus anka로부터 유기배양에 의한 적색소의 대량생산)

  • The production of red pigment from glucose by fed-batch culture of Monascus anka was investigated. In batch culture using fermentor, 200 rpm of agitation speed, 1vvm of aeration volume, and 10% (v/v) of inoculum size were optimal, respectively. The red pigment production was increased by removal of wall-attached mycelium. In an intermittent feeding fed-batch culture, dry cell weight increased to 30 g/l, adn the red pigment content reached 350 of absorbance at 495nm. In a continuous feeding fed-batch culture, dry cell weight increased to 22g/l, and the red pigment content reached 190 of absorbance at 495nm.

  • Breastfeeding is the best source of nutrition for every infant, and exclusive breastfeeding for 6 months is usually optimal in the common clinical situation. However, inappropriate complementary feeding could lead to a nutrient-deficient status, such as iron deficiency anemia, vitamin D deficiency, and growth faltering. The recent epidemic outbreak of obesity in Korean children emphasizes the need for us to control children's daily sedentary life style and their intakes of high caloric foods in order to prevent obesity. Recent assessment of breastfeeding in Korea has shown that the rate is between 63% and 89%; thus, up-to-dated evidence-based nutritional management of breastfeeding infants to prevent common nutrient deficiencies or excesses should be taught to all clinicians and health care providers.

  • A high speed feeding type laser cutting M/C is developed for precise cutting of sheet metal. This paper has been carried out to obtain cutting characteristics and optimal cutting conditions of the developed high speed feeding type laser cutting M/C by a design of experiments. Cutting speed. laser power. laser duty and gas pressure are control factors for the surface roughness. The major factors affecting the surface roughness and the optimum cutting conditions are studied with minimum experiments using Taguchi method.

The Optimum Feeding Frequency in Growing Korean Rockfish (Sebastes schlegeli) Rearing at the Temperature of 15℃ and 19℃

  • Two feeding trials were conducted to determine the optimum feeding frequency in growing Korean rockfish, (Sebastes schlegeli) reared at the temperatures of $15^{\circ}C$ and $19^{\circ}C$. Fish averaging $92.2{\pm}0.7$ g (mean${\pm}$standard deviation [SD]) at $15.0{\pm}0.5^{\circ}C$ and $100.2{\pm}0.4g$ ($mean{\pm}SD$) at $19.0{\pm}0.5^{\circ}C$ water temperature were randomly distributed into each of 15 indoor tanks containing 250-L sea water from a semi-recirculation system. A total of five feeding frequency groups were set up in three replicates as follows: one meal in a day at 08:00 hour, two meals a day at 08:00 and 17:00 hours, three meals a day at 08:00, 14:00, and 20:00 hours, four meals a day at 08:00, 12:00, 16:00, and 20:00 hours, and one meal every 2 days at 08:00 hour. Fish were fed at the rate of 1.2% body weight (BW)/d at $15^{\circ}C$ and 1.5% BW/d at $19^{\circ}C$. At the end of 8 wks of feeding trial weight gain and specific growth rate were significantly higher at the fish fed groups of one meal a day and two meals a day at $15^{\circ}C$ and fish fed groups of 1 meal every 2 days at $19^{\circ}C$ were significantly lower than those of all other fish fed groups. Glutamic oxaloacetic transaminase and glutamic pyruvic transaminase of fish fed group at 1 meal every 2 days was significantly higher than those of all other fish fed groups in both experiments. Weight gain, specific growth rate and condition factor were gradually decreased as the feeding frequency increased. The results indicate that growing Korean rockfish 92 and 100 g perform better at $15^{\circ}C$ than $19^{\circ}C$ water temperature. As we expected, current results have indicated that a feeding frequency of 1 meal a day is optimal for the improvement of weight gain in growing Korean rockfish grown from 92 g to 133 g at $15^{\circ}C$ and 100 g to 132 g at $19^{\circ}C$ water temperature.

Fed-Batch Culture for Polyhydroxyalkanoate Overproduction by Pseudomonas sp. HJ (Pseudomonas sp. HJ로부터 Polyhydroxyalkanoate 대량생산을 위한 유가식 배양)

  • The production of polyhydroxyalkanoate(PHA) from glucose by batch and fed-batch culture of Pseudomonas sp. HJ was studied. In batch culture using fermentor, 400 rpm of agitalion speed, 2 vvm of aeration rate, 18 hours of inoculum age, and 5% (vlv) of inoculum size were optimal. PHA production was not increased by deficiency of oxygen. In a batch culture, the final call mass was $6.251g/\ell$, and PHA content was 20% of dry cell weight. In a constant feeding fed-batch culture, cell mass increased to $33.24g/\ell$, and PHA content reached 48.9% of dry cell weight. In an intermittent feeding fed-batch culture, cell mass increased to $37.89g/\ell$, and PHA content reached 53.5% of dty cell weight.

Effects of Feeding Rice Protein Concentrate on Growth Performance and Ileal Digestibility in Early-weaned Pigs

  • These experiments were conducted to evaluate the feeding value of rice protein concentrate (RPC) in weaning pigs. In expt. I, a 5-week feeding trial was conducted with 126 pigs (L${\times}$Y${\times}$D; 21 d-old; 5.32${\pm}$0.34 kg). Treatments were spray-dried plasma protein (SDPP; control), soy protein concentrate (SPC) and RPC (phase 1), and dried porcine soluble (DPS; control), SPC and RPC (phase 2). An ileal digestibility trial was also conducted to compare digestibility of amino acids in the tested protein sources. In expt. II, 160 weaning pigs (L${\times}$Y${\times}$D; 21 d-old; 5.65${\pm}$0.35 kg) were used in a 5-week feeding trial to determine the optimal inclusion level of RPC in the diet. Treatments were control (9% SPC), and three levels of RPC instead of SPC in the diets (3, 6 and 9%). During phase 1, pigs fed SDPP showed better (p<0.05) ADG and FCR compared with those fed SPC or RPC, while there was no difference in ADFI among treatments. During phase 2, however, pigs fed DPS showed lower (p<0.05) ADG than those fed SPC or RPC. During the total period, there were no significant differences in ADG, ADFI and FCR among treatments. The apparent ileal digestibilities of his, lys, phe, thr and met were not different among the tested protein sources. The apparent ileal digestibilities of arg, ile, leu and val were lower (p<0.05) in RPC than SDPP. The true ileal digestibilities of arg and leu were lower (p<0.05) in RPC than SDPP and SPC. However, that of met was higher (p<0.05) in RPC than SDPP. In expt. II, there were no significant differences in ADG and FCR when SPC was substituted with RPC up to 9% during the total period. In conclusion, based on our experimental results, RPC would replace SPC in the complex prestarter diet, which is somewhat cheaper than SPC.

Efficient Production of ε-Poly-L-Lysine by Streptomyces ahygroscopicus Using One-Stage pH Control Fed-Batch Fermentation Coupled with Nutrient Feeding

  • ε-Poly-L-lysine (ε-PL) is a homopolymer of L-lysine molecules connected between the epsilon amino and alpha carboxyl groups. This polymer is currently used as a natural preservative in food. Insufficient biomass is a major problem in ε-PL fermentation. Here, to improve cell growth and ε-PL productivity, various nitrogen-rich nutrients were supplemented into flask cultures after 16 h cultivation, marking the onset of ε-PL biosynthesis. Yeast extract, soybean powder, corn powder, and beef extract significantly improved cell growth. In terms of ε-PL productivity, yeast extract at 0.5% (w/v) gave the maximum yield (2.24 g/l), 115.4% higher than the control (1.04 g/l), followed by soybean powder (1.86 g/l) at 1% (w/v) and corn powder (1.72 g/l) at 1% (w/v). However, supplementation with beef extract inhibited ε-PL production. The optimal time for supplementation for all nutrients examined was at 16 h cultivation. The kinetics of yeast-extract-supplemented cultures showed enhanced cell growth and production duration. Thus, the most commonly used two-stage pH control fed-batch fermentation method was modified by omitting the pH 5.0-controlled period, and coupling the procedure with nutrient feeding in the pH 3.9-controlled phase. Using this process, by continuously feeding 0.5 g/h of yeast extract, soybean powder, or corn powder into cultures in a 30 L fermenter, the final ε-PL titer reached 28.2 g/l, 23.7 g/l, and 21.4 g/l, respectively, 91.8%, 61.2%, and 45.6% higher than that of the control (14.7 g/l). This describes a promising option for the mass production of ε-PL.

High Production of L-Threonine using Controlled Feeding of L-Methionine and Phosphate by Escherichia coli Mutant (L-Methionine과 Phosphate의 제한 공급에 의한 Escherichia coli MT201로부터의 고농도 L-Threonine 생산)

  • L-Threonine fermentation process was constructed on batch and fed-batch culture by using Escherichia coli MT201. The production type of L-threonine was observed as growth-associated production in batch culture. In fed-batch culture studying optimal concentration of yeast extract in feeding media, when 600 g/l of glucose and 60 g/l of yeast extract were added in feeding media, 87 g/$\ell$ of L-threonine was produced. To improve cell growth and L-threonine production, the culture of high cell density was performed in fed-batch culture with oxygen enriched air and feeding media containing L-methionine and phosphate. Under the conditions, we could achieve the highest L-threonine production of98 g/$\ell$ at 60 h. The highest productivity of L-threonine was about 3.85 g/$\ell$/h.
