Determination of optimum gamma ray range for radiation mutagenesis and hormesis in quinoa (Chenopodium quinoa Willd.)

  • Quinoa (Chenopodium quinoa Willd.) is one of the ancient crops cultivated in the Andes region at an altitude of 3,500-4000m in Chile and Bolivia from 5000 BC. It contains a large amount of protein, minerals and vitamins in comparison with other crops. The cultivation area has been increasing worldwide because of its excellent resistance to various abiotic stress such as salinity, drought and low temperature. ${\gamma}$-Ray radiation of high dose is often used as a tool to induce mutations in plant breeding, but it has a deleterious effect on organisms. However, the radiation may have a positive stimulatory effect of 'hormesis' in the low dose range. This experiment was carried out to investigate the optimum dose range for creating the quinoa genetic resources and to investigate the hormesis effect at low dose on the quinoa. This experiment was performed for 120 days from November, 2016 to February, 2017 in the greenhouse of Gyeongsang National University. ${\gamma}$-Ray radiation was irradiated to seeds at 0 Gy, 50 Gy, 100 Gy, 200 Gy, 300 Gy, 400 Gy, 600 Gy, 800 Gy and 1000 Gy for 8 hours. (50 Gy) using the low level radiation facility ($Co^{60}$) of Cooperative Research Institute of Radiation Research Institute, KAERI. Fifty seeds were placed on each petri dish lined with wet filter paper and germination rate was measured at a time interval of 2 hours for 40 hrs. The length of the root length was measured one week after germination. Each treatment was carried out in 3 replicates. The growth of seedlings were investigated for 10 days after transplanting of 30 day-old seedlings. The plant height, NDVI, SPAD, Fv/Fm, and panicle weight were measured. The germination rate was highest at 50Gy and 0Gy and the rate of seeds treated with 400Gy or higher rate decreased to 25% of the seeds treated with 50Gy. The emergence rate of seedling in pot experiment was higher at the dose of 200 Gy, 300 Gy and 400 Gy than at 0 and 50Gy. However, the rate was lower at strong radiation higher than 600Gy at which $1^{st}$ leaf was not expanded fully and dead due to extreme overgrowth at 44 days after treatment (DAT). The highest value of panicle weight was observed at 50Gy (6.15g) and 100Gy (5.57g). On the other hand, the weight at high irradiated dose of 300Gy and 400Gy was decreased by about 55% compared to low dose (50 Gy). NDVI measurement also showed the highest value at 50 Gy as the growth progressed. SPAD was the highest at 400 Gy and showed positive correlation with irradiation dose except 0 Gy. Fv/Fm was high at 50 Gy up to 30 DAT and no difference between treatments was observed except for 400 Gy from 44 DAT. The plant height was the highest in 50Gy during the growing period and was higher in the order of 50Dy, 100Gy, 0Gy, 200Gy, 300Gy and 400Gy in 88 DAT. In this experiment, the optimal radiation dose for hormesis was 50Gy and 100Gy, and the optimal radiation dose for mutagenesis seems to be 400 Gy.

Effect of Lime on Growth of Rice and Changes in pH, Eh, Fe2+ and Al in an Acid Sulfate Soil (특이산성토양에서 석회시용이 벼의 생육과 토양의 pH, Eh, Fe2+, Al 변화에 미치는 영향)

  • A pot experiment with an acid sulfate soil from Kimhae was carried out to find out the cause of toxicity in rice plant. The effect of liming on changes in pH, Eh, Al, and $Fe^{2+}$ in soil and leachate was examined at two-week interval during the growth of rice. Also, total $P_2O_5$, $SiO_2$, Fe and Al contents in plants at harvesting stage were determined. In the early stage, the rice plant in the check soil showed the same healthy growth as did in limed soil even at high Al in soil and leachate. Around panicle forming stage, reddish brown mottlings suddenly infested all over the plants when accompanied with strong reduction, and afterward growth was severely retarded, and finally caused the significant difference in yield. During the strong reduction, significant amount of sulfide was formed only in check soils, but no free $H_2S$ was detected. Appreciable Al was still present in soil and leachate, and $Fe^{2+}$ in check soil was lower than that in limed soil, but $Fe^{2+}$ in leachate was slightly higher. Limed soils were more reduced and produced more $Fe^{2+}$ due to increased microorganism activity in the neutral pH. In the leachate, the check showed slightly higher $Fe^{2+}$ concentration but considerably higher than limed one at later stage. Appreciable amount of Al was detected only in check soil and leachate from transplanting to panicle formation stage. Plant tissues at harvesting stage contained very low P regardless of liming. Uptake of Si was markedly increased by liming. Contents of Fe an Al was markedly higher in check than limed one, but difference in Fe content was more drastic possibly due to more Fe uptake in presence of markedly higher $Fe^{2+}$ in soil and leachate at later growing stage. In conclusion toxic symptom in this acid sulfate soil seems to be primarily due to Al when accompanied with low pH and strong reduction. But association with $Fe^{2+}$ toxicity is not completely excluded. In order to differentiate the effect of $Fe^{2+}$ from that of Al more detailed plant analysis at different stage is required.

Effects of Simulated Acid Rain on Nutrient Contents of Pinus densiflora S. et Z. and Forsythia koreana Nak. Seedlings (인공산성우(人工酸性雨)가 소나무 및 개나리묘(苗)의 식물체내(植物體內) 함유성분(含有成分)에 미치는 영향(影響))

  • For the purpose of examining the effects of simulated acid rain on nutrient contents of plant tissues in Pintos densiflora seedlings and Forsythia koreana rooted cuttings, the experimental design of randomized block arrangement with three replications was implemented in the experimental field of Yesan National Agricultural Junior College. One-year-old Pinus densiflora seedlings and Forsythia koreana cuttings were planted in the pots filled the mixed soils(nursery soil : forest soil of siliceous sandy loam=1 : 1 v/v) in the early spring of 1986. The regime of artificial acid rain, in terms of spray frequency per month and spray amount at single treatment per plot, was simulated on the basis of climatological data averaged from 30 years records. Simulated acid rain(pH 2.0, pH 4.0, and pH 5.5 as control) containing sulfuric and nutric acid in the ratio of 3 : 2(chemical equivalant basis) diluted with ground water were treated on the experimental plants under condition of cutting off the natural precipitation with vinyl tunnel, during the growing season(May 1 to August 31) in 1986. The results obtained in this study were as follow : (1) As for the nitrogen contents in plant tissues, P. densiflora increased significantly in one-year-old stembranch and root tissues, and F. koreana increased significantly in leaf and root tissues, as the pH levels of acid rain decreased. (2) The available phosphate contents in root tissues of P. densiflora, and in leaf and root tissues of F. koreana were significantly decreased, as the pH levels of acid rain decreased. (3) $K_2O$, CaO and MgO contents in plant tissues were significantly decreased in the both species as the pH levels decreased. And the effects of acid rain on F. koreana were higher than those of P. densiflora. (4) Sulfur contents of plant tissues in the both species were increased at pH 2.0 treatment. There were significant differences among three acid rain treatments in leaf and root tissues of P. densiflora, and in all parts of F. koreana. (5) In the effects of simulated acid rain on the both species and the tested soils, in general, F. koreana revealed higher sensitiveness than P. densiflora, and the lower pH levels of simulated acid rain were treated, the more sharp reaction was showed.

Expression of Organogenesis-related Genes and Analysis of Genetic Stability by ISSR Markers of Regenerants Derived from the Process of in vitro Organogenesis in Japanese Blood Grass (Imperata cylindrica 'Rubra') (기내배양 홍띠 단계별 재분화체의 기관분화 관련 유전자 발현과 ISSR에 기반한 유전적 안정성 분석)

  • The in vitro organogenesis is one of important issues in plant embryology, and somaclonal variations are existing in calli and/or regenerants induced from a process of the organogenesis with in vitro circumstances. In this study, expressions of organogenesis-related genes were evaluated and genetic stability of regenerants derived from the process of in vitro organogenesis were measured using ISSR markers in Imperata cylindrica 'Rubra', Poaceae. The expressions of organogenesis-related genes were detected all of regenerants at the process of the organogenesis. All ISSR markers produced with an average of 71 bands per in vitro-cultured regenerants, and the scorable bands were varied from two to eight with an average of 5.14 bands per a primer. The polymorphism rates of the in vitro regenerants were higher than that of mother plants (1.4%), showing 4.1% (pot-cultured regenerants), 4.3% (field-cultured regenerants), 4.2% (in vitro-cultured regenerants), 5.6% (calli with green shoots) and 1.4% (calli), respectively. The genetic similarity matrix (GSM) among all accessions ranged from 0.747 to 1.0 with a mean of 0.868. GSM of the regenerants showed differences (from 0.972 to 1.00) compared with that of mother plants (0.991). According to the clustering analysis, two independent groups were divided into; the one is mother plants and regenerants cultured at room and open field, the other is regenerants cultured in vitro. The results give a new insight for understanding the dynamics of organogenesis in monocot plant.

The Effects of Nitrogen Type and Fertilized Depth on Leaching and Absorption of Nitrogen in Paddy Soil and Growth and Yields of Rice (질소비료(窒素肥料)의 형태(形態)와 시용심도(施用深度)가 질소(窒素)의 용탈흡수(溶脫吸收) 및 수도(水滔) 생육수량(生育收量)에 미치는 영향(影響))

  • In order to find the effects of nitrogen type (ammonium sulfate and urea fertilizer) and fertilized depth, (0~10cm, 0cm, 5~10cm, 10~15cm, 15~20cm, and 20cm below) on leaching and absorption of nitrogen in paddy soil, and growth and yields of rice, the pot culture experiment was carried out, using the variety Jaekun, one of the Korean leading variety. Experimental results were Summarized as follows: 1. No variations of the pH of percolating water were induced by the differences of nitrogen types and their fertilized depth (Table. 2). 2. The leaching of nitrogen was less in ammonium sulfate and top soil fertilizing plots than in urea and subsoil fertilizing plot, and the growth of rice in early stage was more promoted in ammonium sulfate and topsoil fertilizing plots (Table. 1, 7 and 8). 3. Leachng of nitrogen through the percolating water almost came to an end at the most numerous tiller stage (Table 1). 4. The absorption of nitrogen of each part of the rice plant in the harvesting stage correlated closely with the yields of each part (Table 5, 6, 9 and 10) and the leaching of nitrogen in the early stage was inversely proportion to the absorption of nitrogen of rice plant in the harvesting time (Table 1, 5, 6, 9 and 10). 5. The number of spikes was more numerous in ammonium sulfate plots than in urea plots on an average, so that the yields were higher in the ammonium sulfate plots than in urea plots although no differences in the grain number per spike were found in above two plots. The number of spikes was more numerous in topsoil fertilizing plots than in subsoil fertilizing plots, but the grain number per spike was less in former than in latter, so that no difference in yields was found. The absorption of nitrogen correlated closely with the yields in complete paddy grains (Table 5, 9, and 10). 6. At the ammonium sulfate fertilizing plots, the number of spikes was more numerous in topsoil fertilizing plots than in subsoil fertilizing plots, (among the each of the topsoil plots, 0~10cm and 5~10cm fertilizing plots kept more spikes than the 0cm fertilizing plots), but the grain number per spike was less in former than in latter (among the each of topsoil plots, no differences were found), so that no significant difference in yields was showed between the topsoil and subsoil fertilizing plots, but the results showed the tendency that the yields were highest in 0~10cm plots and the lowest in 20cm below plots. At the urea fertilizing plots, the number of spikes decreased in proportion to the increasing of fertilized depth, but no variations were found in the grain number per spike, so that the yields decreased in proportion to the increasing of fertilized depth. The absorption of nitrogen correlated closely with the yields in complete paddy grains (Table 5, 6, 9, and 10). 7. When fertilized in topsoil, the number of spikes was more numerous in ammonium sulfate plot than in urea plot, but the grain number per spike variated reversely, so that no differences were found in the yields between the ammonium sulfate and the urea plots, when fertilized in subsoil, both the number of spikes and the grain number per spike were larger in ammonium sulfate than in urea plot, so that the yields were also higher in ammonium sulfate plots (Table 5, 6, 9 and 10). 8. The weight of straw and its nitrogen absorption were higher in ammonium sulfate plot than in urea plot and decreased in proportion to the increasing of fertilized depth. Among the each of topsoil fertilizing plots, the 0~10cm and the 5~10cm fertilizing plots excelled the 0cm plot (Table 5, 6, 9 and 10). 9. No significant variations in the fertilizer treatments were found in the characters of heading date, maturing date, length of culm, length of spike, weight of empty grain, 1,000 grain weight, and one liter weight.

  • This research was carried as a part of the basic study, in which the aptitude of theKorean oak varieties as barrels for aging apple fine spirits was investigated, and thefollowing results were obtained. 1. Following was the result of the chemical analysis of the fruits which are now mass-produced and can be used as a substitute for raw materials for wine production. Apple (Malus pumila Miller var. domestica Schneider) : Total sugar. total acid, volatile acid and pectin of Jonathan (Hong-og) were 13.95%, 0.46%, 0.012%, 0.20% respectively. Total sugar, total acid, volatile acid and pectin of Ralls (Koog-kwang) were 13.35%, 0.43%, 0.011%, 0.45% respectively. 2. Because of low yield of apple juice due to cellulose, pectin, hemicellulose which are present besides sugars, acids in apples, the apple juice were treated with xylanase of Aspergillus niger SUAFM-430, cellulase and pectinase of Aspergillus niger SUAFM-6. This treatment increased the yield of apple juice. And the apple juice was sterilized by adding potassium metabisulfite $(K_2S_20_5)$ and Saccharomyces cerevisae var. ellipsoideus Rasse Johannisberg II (SUAFM-1018) as a cultivation yeast, which has a strong fermentation power was used to ferment. The yield of apple wine based on raw material was 86-87%. The amount of ethanol, extract and methanol obtained from Jonathan and Ralls were 13.5%, 5.4%, 0.04-0.05% respectively. 3. Wines were distilled for two times by the pot still method to make fine spirits. The yield of fine spirits from apple wine mash was 86.6%, and the pH of fine spirits from Jonathan and Ralls were 4.1, 4.2 respectively. 4. The oak chips made of inner part or outer part of 24 Korean oak varieties were used to select the barrel for aging fine spirits. Two oak chips (one oak chip: $1{\times}1{\times}5cm$) of the inner part or of the outer part of each oak variety were dipped into 300 ml of fine spirits, which was bottled in 640ml beer bottle, and followed aging. The colors, flavors and tastes of the fine spirits were checked during 6 months. A. As a criterion for the first screening of oak barrels for aging fine spirits, the rate five of color extraction was determined. The oak chips showed good results in their order as follows and the best 5 varieties were selected. Gal-cham: Quercus aliena Blume (Inner part), Gul-cham: Quercus variabilis Blume (Outer part), Gal-chain: Quercus aliena Blume (Outer part), Jol-cham: Quercus serrata Thumb (Inner and Outer part). Sin-gal-cham: Quercus mongolica Fisher (Outer and Inner part) Sang-su-ri: Quercus acutissima Carruthers (Outer and Inner part) B. To find out the influence of aging temperature on aging, apple fine spirits were aged by dipping each oak chip at room temperature $(24-25^{\circ}C)$) and $45^{\circ}C$. Aging at $45^{\circ}C$ gave the best result followed aging at $30^{\circ}C$ and then at room temperature. C. Apple fine spirits was aged for six months by dipping oak chips in Erlenmeyer flasks and was irradiated with U.V light. The U.V irradiation enhanced the aging effect by nearly two times, compared with the aging without U.V irradiation. D. In aging apple fine spirits by dipping two oak chips, it was observed that the extent of the extraction of most components of oak chips were strongly dependent upon the pH of fine spirits. E. Oak chips of five selected oak varieties and a Limousin white oak from France as a control were used. Each apple fine spitits was dipped by two oak chips, and was aged at room temperature $(24-25^{\circ}C)$, $30^{\circ}C$, $45^{\circ}C$, and with the U.V irradiation at room temperature shaking every week. After six months of aging, the panel test of these aged fine spirits (Young Brandy) showed the following result. Young brandy of apples aged at $45^{\circ}C$ by dipping oak chips of Gal-chain was almost as the fine spirits which were aged at room temperature by dipping Limousin white oak chips from France. Young brandy of with U.V. irradiation at room temperature which were aged by dipping oak chips of Gal-chain was a little worse than that from the fine spirits aged at room temperature by dipping Limousin white oak chips from France. And so, Korean oak varieties are thought to be able to be used for aging every apple fine spirit which was here investigated.

Biological Control of Tobacco Cutworm, Spodoptera litura (Lepidoptera: Noctuidae) by Steinernematid and Heterorhabditid Entomopathogenic Nematodes (Steinernematid와 Heterorhabditid 곤충병원성 선충을 이용한 담배거세미나방 (Spodoptera litura)의 생물적 방제)

  • Five effective strains against tobacco cutworm, Spodoptera litura (Lepidoptera: Noctuidae), Steinernema carpocapsae (GSN1), Steinernema sp. (GSNUS-10), Steinernema sp. (GSNUS-14), Heterorhabditis bacteriophora Hamyang (HbH), and Heterorhabditis sp. (GSNUH-1) were selected among 14 isolates of Korean entomopathogenic nematode in laboratory tests. $LC_{50}$ values of above five strains against tobacco cutworm were various by different nematode strains and developmental stages of tobacco cutworm. $LC_{50}$ value of S. carpocapsae (GSN1) was the lowest by $4.0{\sim}8.3$ infective juveniles (Ijs) and 2nd instars of tobacco cutworm was most susceptible. Pathogenicity of five effective strains against tobacco cutworm depends on nematode strain, concentration, and application times. The most effective strain was determined as S. carpocapsae (GSN1). Two or three times of applications were effective regardless of nematode strain, or concentration. Efficacy of S. carpocapsae (GSN1), Steinernema (GSNUS-10), Steinernema (GSNUS-14), and Heterorhabditis (GSNUH-1) was variable depending on nematode strain, concentration, application times, and host variety. S. carpocapsae (GSN1) was the most effective and inoculation of 100,000 infective juveniles per m2 (720,000 Ijs/7.2 $m^2=1{\times}10^9$ Ijs/ha) resulted in higher efficacy. Three times of application of nematodes led to higher control efficacy than one or two applications. Efficacy of nematodes was higher on Chinese cabbage than cabbage or kale.

Synthesis of Artificial Zeolite from Fly Ash for Preparing Nursery Bed Soils and the Effects on the Growth of Chinese Cabbage (석탄회(石炭灰)를 이용한 육묘(育苗) 상토용(床土用) 인공(人工) 제올라이트의 제조와 배추 생육에 미치는 효과(效果))

  • To reduce the environmental contamination and to utilize fly ash massively produced from the coal power plant every year, we synthesized the artificial zeolite using fly ash treated with alkaline, and then analyzed the mineralogical and morphological properties by X-ray, IR, and SEM. The amount of $NH_4{^+}$, $K^+$, and $H_2PO_4{^-}$ adsorbed by the fly ash and the artificial zeolite were determined with reaction time, amount of adsorbate used, ion concentrations. The results obtained from the pot experiments packed with the top soil, amended with granulated artificial zeolite which was made by treatment of 4% polyvinylalcohol, showed that CEC of the artificial zeolite was $257.7cmol^+kg^{-1}$, that was almost 36 times greater than that of fly ash. The ratio of $SiO_2/Al_2O_3$ decreased but the amount of Na increased. The physico-chemical properties analyzed by X-ray, IT, and SEM represented that the artificial zeolite synthesized had a similar morphological structure to that of the natural zeolite. The structures of the artificial zeolite had a significantly enlarged surface having a lot of pores, while the fly ash looked like spherical smooth shape with having not pores on the surface. Thus, the artificial zeolite was successfully synthesized. The results of adsorption isotherms of fly ash and artificial zeolite showed that the amount of $NH_4{^+}$, $K^+$, and $H_2PO_4{^-}$ adsorbed increased as the equilibrium concentration increased, while $NH_4{^+}$ was strongly adsorbed on the surface of fly ash and artificial zeolite than that of $K^+$. The most distinctive growth of Chinese cabbage was found from the top soil(NPK + soils + 20% of granulated artificial zeolite + 5% of compost). Therefore, we concluded that one of the most effective methods to effectively recycle a fly ash was to make the artificial zeolite as we did in this experiment.

  • A pot experiment was conducted to study the effect of two methods of applying potassium fertilizer to rice. One basic application was compared with the split of same total quantity into four applications as follows: 15 days after transplanting (40%), Ear formation stage (30%), 13 days before heading (20%) and 7 days after heading (10%) Each of these two treatments was carried out on both untreated soil and soil to which wollastonite and lime material had been added. The number of ripened grains or the 1,000 grain weight was increased by application of potassium to untreated soil. However, on soil treated with lime and wollastonite only the number of total grains was increased by potassium application. In both cases, split application of the potassium was more affective than a single basic application. No significant increase in yield was obtained from a single basic application of potassium. However, split application of the same total quantity of potassium did give a significant increase in yield. A negative correlation was found between the content of $K_2O$ and that of other nutrients in the rice plant at two stages of growth. Significant negative correlation was obtained between the content of $K_2O$ and magnesium, phosphorus and nitrogen at ear formation stage, and between the content of $K_2O$ and calcium and silicate including magnesium, phosphorus and nitrogen at heading stage. This result also indicated that the depression of uptake of phosphorus and magnesium at ear formation stage and that of calcium and silicate at heading stage were decreased by potassium split application. However, the degression of uptake of nitrogen at ear formation stage and that of magnesium at heading stage were increased by potassium split application.

  • This investigation includes the sick soil phenomenon caused by the self-poisoning of Setaria italica, Sorghum nervosum, Zea mays and Miscanthus sinensis among Poaceae. It elucidates whether the poison is directly excreted from the root or the secondary product resulting from the decomposition in the soil; the effect of Miscantus sinensis on the germination and growth of other plants, and the effect of Zea mays grown between furrows to shade Angelica gigas on its growth. The results obtained are as follows; Supplied with the leakage water from the pots, in which the same plants as the test ones were grown, in anticipation of the poison to be directly excreted from their roots, Sataria itlaica and Zea mays exhibited the growth inhibition more than 30%, whereas Sorghum nervosum and Miscanthus sisnensis were not effected in growth at all. When cultivated in the soils mixed with the roots of the some plants as the test ones, in anticipation of the poison to be the secondary product resulting from the decomposition in soil, Setaria italica and Zea mays showd growth inhibition of more than 50%, which is greater than that of the case of the leakage water, and Miscanthus sinensis exhibited no inhibition either, whereas Sorghum nervosum in the 50% plot showed heavy growth inhibition of more than 80% to the case of the leakage water. The common or uncommon plants found easily in the group of Misscanthus scinenis were not affected by the extracts of the steam and leaves of Miscanthus sinensis in germination and growth. Supplied with the leakage water from the pots in which Miscanthus sinensis was grown, among Lespedeza crytobotrya, Oenothera odorata, Raphanus sativus val'. acarlthiformis, Zoysia japonica, Patrinia scabiosaefolia. which are easily found in the group of Miscanthus sinensis, only Patrinia scabiosaefolia was slightly inhibited in growth in the 100% plot, whereas the others did not show any inhibition at all. Mean while, Amaranthus patulus. Solanum nigrum, Capsella bursa-pastoris val'. triangularis, Alopecurus amurensis, Chenopodium album val'. centrorubrum, which could not be found in the group of Miscanthus sinensis, were all distinctly inhibited. In the experiment on the effect of Zea mays on the growth of Angelica gigas, its growth was severely inhibited by one-half to two thirds with the increased concentration in both the cases of growing in the mixture of the soil and the powdered root of Zea mays and being supplied with the leakage water from the pot in which Zea mays was grown.
