Production of Lipase-catalyzed Structured Lipid from Olive Oil with Omega-3 Polyunsaturated Fatty Acids

  • Acidolysis of olive oil with omega-3 (n-3) polyunsaturated fatty acids (PUFAs) was carried out to produce a structured lipid. Novozym $435^{(R)}$ from Candida antarctica was used as the biocatalyst. Response surface methodology (RSM) was used to determine optimum conditions for lipase-catalyzed enrichment of olive oil. Three factors, 5 levels, central composite design was used. The effects of incubation time, temperature, and substrate mole ratio on incorporation ratio (n-3 fatty acids/total fatty acids, %) were investigated. From the evaluation of response surface graphs, the optimal conditions for incorporation of long chain n-3 PUFAs into olive oil were $40-60^{\circ}C$ for temperature, 30-45 hr for reaction time, and 3:1-5:1 (n-3 fatty acids/olive oil) for substrate mole ratio. Experiments conducted under optimized conditions predicted by the model equation obtained from RSM yielded structured lipids with 50.8% n-3 PUFAs. This value agreed well with that predicted by the model. Oxidative stability tests showed that the product was more susceptible to oxidation than unmodified olive oil. Antioxidant addition improved the oxidative stability of the product.

Evaluation of Major Nutrients of Domestic Farmed Eels Anguilla japonica (국내 양식 뱀장어 (Anguilla japonica)에서의 주요 영양학적 성분의 평가)

  • To evaluate the proximate composition and main nutritive ingredients (vitamin A, $V_A$; vitamin E, $V_E$; cholesterol; the composition of fatty acids) between eels cultured in different domestic eel farms, we analyzed edible parts of eels that were cultured in three "Water Recirculation System (WRS)" farms and three "Still-Water System (SWS)" farms. We found differences in crude protein, crude lipids and the ash content of eel meat between the farms, but water content showed no significant difference. $V_A$ and $V_E$ contents did not show meaningful differences between the two culture methods, but there was a noticeable distinction between the eel farms. In particular, $V_E$ content showed significant differences between the eels of each farm (highest farm, about 25 mg/100 g dry wt; lowest farm, about 1 mg/100 g dry wt). The fatty acids of cultured eels were composed of monoenes (MUFA, 55-60%), saturates (SFA, 30-33%), $\omega$-6 highly unsaturates (HUFA, 1.6-2%) and $\omega$-3 HUFA (8-9.2%). The MUFA content of WRS eels ($59.9{\pm}1.97%$) was higher than that of SWS eels ($55.8{\pm}0.48%$). The contents of other fatty acids were higher in SWS eels (SFA, $33.0{\pm}0.52%$; $\omega$-6FUFA, $1.98{\pm}0.2%$; $\omega$-3 HUFA, $9.2{\pm}0.67%$) than in WRS eels (SFA, $30.5{\pm}1.34%$; $\omega$-6 HUFA, $1.62{\pm}0.19%$; $\omega$-3 HUFA, $8.0{\pm}0.5%$).

Effects of Chronic Ethanol Consumption and Taurine Supplementation on Hepatic Total and Phospholipid Fatty Acid Compositions in Rats (만성적인 에탄올 섭취와 타우린보강이 흰쥐간의 총지방산 및 인지질지방산 조성에 미치는 영향)

  • This study evaluated the effects of chronic ethanol consumption and/or taurine supplementation on hepatic total, phospholipid fatty acid composition and the metabolism of rats fed one of three purified liquid diets for 8 weeks. the rats followed either the control diet (CD, ethanol-free and taurine-free diet); ethanol diet (ED, CD+ 50g ethanol/L) or ethanol-taurine diet (ETD, ED+3.75g taurne/L). Chronic ethanol consumption and/or dietary taurine supplementation were associated with altered hepatic total and phospholipid fatty acid composition. compared to the values for the control rats, ED or ETD significantly decreased the percentage of total monounsaturated fatty acids ($\Sigma$MUFA), and increased the percentage of total polyunsaturated fatty acids ($\Sigma$PUFA) of hepatic total lipids(p〈0.01). Percentages of 14:0(P〈0.01) and 16:0(p〈0.001) were sigificantly lower, and those of 18:0(p〈0.01), 20:0(p〈0.001), 20:3$\omega$6(p〈0.01) and 22:4$\omega$6(p〈0.01) in hepatic total fatty acid compositions were oserved in rats fed ETD versus those fed ED or ETD. No significant differences in hepatic total fatty acid compositions were observed in rats fed ETD versus those fed ED. Percentages of 24:0(p〈0.01), 16:1(p〈0.05), 20:1(p〈0.01), 18:2$\omega$6(p〈0.01) and 18:3$\omega$3(p〈0.05) in hepati phospholipids were significantly higher, and those of 14:0(p〈0.01), 16:0(p〈0.001), 20:3$\omega$3(p〈0.05) in hepatic phospholipids were significantly higher, and those of 14:0(p〈0.01), 16:0(p〈0.001), 20:3$\omega$3(p〈0.001), 22:6$\omega$3(p〈0.001) and $\Sigma$$\omega$3(P〈0.001) were significantly lower in rats fed ED or ETD compared to the values for the control rats. The Δ5 desaturation index(20:3$\omega$6⇒20:4$\omega$6) and elongation index (20:5$\omega$3⇒22:5$\omega$3) of hepatic phospholipid index (20:3$\omega$6⇒20:4$\omega$6) and decreased Δ4 desaturation index (22:5$\omega$3⇒22:6$\omega$3) compared to the values for the ED rats. These changes in hepatic fatty acid composition induced by chronic ethanol consumption and/or taurine supplementation might be associated with the modulations of physical properties of the hepatic cell membrane and its sensitivity to peroxidation damage.

Relationships between Fatty Acid Intake and Serum Lipids in Postmenopausal Women (폐경 후 여성의 지방산 섭취실태와 혈청 지질농도와의 관련성)

  • The intake of fatty acids, and the effects of fatty acid intake patterns on serum lipid concentrations was investigated in 112 postmenopausal women. Dietary intakes of fatty acids were assessed by means of a semi-quantitative food questionnaire with food models and other measuring tools. The intakes of saturated fatty acid(SFA), monounsaturated fatty acid(MFA) and polyunsaturated fatty acid(PUFA) were 10.85g, 11.89g and 12.09g, respectively. The ratios of P/M/S and $\omega$6/$\omega$3 fatty acid were 1.19 : 1.10 : 1 and 6.65 ; 1, respectively. Serum lipid concentrations were adjusted for age, menopausal period, BMI, stress scores, exercise habit and energy intake and the relationship between fatty acid intake and serum lipids was measured using Pearson's correlation coefficient. Serum cholesterol concentration was positively correlated with dietary intake of SFA (p<0.10), MUFA(p<0.05), palmitic acid(p<0.10), stearic acid(p<0.05), oleic aicd(p<0.05), and arachidonic acid(p<0.10). Serumk LDL-cholesterol concentration was positively correlated with MUFA(p<0.10), oleic acid(p<0.10), oleic acid(p<0.10), and arachidonic acid (p<0.10). Serum triglyceride concentration was positively correlated with arachidonic acid(p<0.10). However, serum HDL-cholesterol concentration was not found to be significantly correlated with any fatty acids.

The Relationship between Serum Cholesterol Levels and Dietary Fatty Acid Patterns, Plasma Fatty Acids, and Other Lipid Profile among Korean Adults (성인 남녀의 혈청 콜레스테롤의 수준과 지방산 섭취 양상, 혈중 지질 및 지방산 조성의 상관 관계 연구)

  • The purposes of this research were to assess dietary fatty acid patterns and to elucidate the relationship between the serum cholesterol levels and dietary fatty acid patterns, plasma fatty acid compositions, BMI (body mass index), and other lipid profile. The subjects were 151 adults aged 23 to 80 years, selected from the Outpatient Clinic and Cardiova-scular Department of the Seoul Municipal Hospital. Dietary data were obtained using three day food records. Sixteen dietary fatty acids were analyzed using Korean and US nutrient databases. The subjects were divided into three serum cholesterol levels: desirable (< 200 mg/dl, N = 44), borderline-risk ($\geq$ 200 - < 240 mg/dl, N = 35), and high-risk ($\geq$ 240 mg/dl, N = 72) groups. The high-risk group had higher BMI, waist, and waist to hip ratio (WHR) than the desirable and borderline-risk groups. Serum concentrations of triglyceride, LDL cholesterol and LDL/HDL cholesterol ratio were significantly higher in the high-risk group as compared to those in the other two groups. The serum cholesterol levels were highly correlated with BMI (r = 0.435), triglyceride (r = 0.425) and LDL/HDL cholesterol (r = 0.870) ratio. The highest fatty acid intake was from oleic acid (33 - 34% of total fatty acid intakes), which was followed by linoleic acid (27%), palmitic acid (19%), and stearic acid (7%). There was no correlation between the serum cholesterol levels and the dietary fatty acid intakes, polyunsaturateumonounsaturateusaturated fatty acids (P/M/S) and $\omega$6/$\omega$3 ratios. The correlation between plasma fatty acids such as myristic acid, oleic acid, linoleic acid, and docosahexaenoic acid and serum cholesterol levels was also weak. Molecular Dynamic Simulations of the Fatty Acid Bilayer Containing Very Long Chain Transmembrane Dicarboxylic Acids

  • Recent research results regarding the very long chain transmembrane ${\alpha},{\omega}-dicarboxylic$ components in the membrane of extremophilic eubacteria, such as Sarcina ventriculi, Thennotoga maritima, and Thermoanaerobacter ethanolicus have raised interesting questions concerning the physical and biochemical function on these components in the membrane. In order to understand the dynamic characteristics of these acids which reside in the bilayer membrane, 580 ps molecular dynamic simulations at 300 K were performed for two model systems. These systems were the bilayer with regular chain (C16:0 or C18:1) fatty acid methyl esters and the fatty acid bilayer containing very long chain transmembrane dicarboxylic acid methyl esters (${\alpha},{\omega}-15,16-dimethyltriacotane-dioate$ dimethyl ester; C32:0). Our analyses indicate that very long chain transmembrane dicarboxylic acids have a noticeable influence on the bilayer dynamics at a sub-nanosecond time scale. The center-ofmass mean-squared-displacement (MSD) of regular chain fatty acids adjacent to the very long chain transmembrane dicarboxylic acids decreased, the long-axis order parameter increased, and the reorientational motions of methylene groups were slowed along the hydrocarbon chains. These results indicate that the very long chain transmembrane dicarboxylic acids reduce the molecular order of the whole bilayer membrane.

A Comparative Study on the Dietary Attitudes and Nutritional Status of Preschoolers in Different Income Levels in Seoul and Kyunggi-Do: 2. Focusing on Preschoolers' Nutrients and Fatty Acid Intakes (서울과 경기 일부지역의 소득수준별 미취학 아동의 식생활 태도 및 영양상태에 관한 비교 연구: 2. 영양소 및 지방산 섭취실태를 중심으로)

  • According to economical status, under-nutrition and over-nutrition coexist in preschool children in Korea. Subjects consumed more than RDA in most nutrients, and children in of the upper income class tended to have more nutrients than those of the low income class. Especially in case of energy, protein, fat, vitamin $B_2$, and P there were significant differences among the different classes. The mean calorie compositions of carbohydrates, protein and fat were $57{\sim}59:\;16{\sim}17:\;26{\sim}27$ and fat calorie percentage was higher than quantity recommended for Korean adults. Energy, protein, Fe, vitamin A, $B_1$ and C intakes were taken mostly from plant food sources. In the upper income class group, intakes of protein and vitamin $B_2$ from animal food sources were higher than in any other income classes. Intakes of iron, vitamin A, $B_1$ and C were largely provided by plant food group. Especially 44-45% of vitamin A were taken from vegetable and fruits group, which indicated that about 50% of vitamin A intake was the form of ${\beta}-carotene$. Also compared with other groups, in the upper income class group, the intakes of energy, protein, Ca, p, vitamin $B_1$ and vitamin $B_2$ from cereal and potatoes were significantly lower, and those of Ca and P from milks and meats, fishes & eggs were significantly higher(p<0.05). The mean fat intake in all subjects was $38{\sim}43g/day$. Regardless of income class, oleic acid($6.6{\sim}8.4g$) and linoleic acid($6.7{\sim}8.1g$) were the most abundant fatty acids contained in the diet. The upper class group consumed significantly more total saturated fatty acids and total monounsaturated fatty acids(p<0.05). In polyunsaturated fatty acids, there were no differences between 3 different income classes, but intakes of total ${\omega}6$ fatty acids in the upper class tended to be higher and those of total o3 fatty acids in low class tended to be higher. Therefore, ${\omega}6/{\omega}3$ tended to be higher in the upper class group. Regardless of income classes, P/M/S and ${\omega}6/{\omega}3$ ratios in all subjects were $0.9{\sim}l.3/1.0{\sim}1.1/1.0$ and $5.2{\sim}7.4/1.0$, respectively and were in a desirable range. Cholesterol intake of subject was 184-218 mg/day, which was comparable to the value of Korean adult intake.

Oil Contents and Fatty Acid Composition of Korean Perilla (Perilla ocimoides L.) Collections

  • This study was carried out to obtain informations on the variations of oil content and fatty acid composition among 90 Korean perilla collections. Average oil content of 90 perilla collections was 44.2% with a range from 29.7% to 61.9%. Perilla collections with late-maturing, super-large seed and gray seed coat showed higher oil content than other types in general. Average saturated fatty acid content in perilla oil was 9.0% with a range from 8.2% to 10.7%, while average unsaturated fatty acid content varied from 89.3% to 91.8% with a mean of 91.0%. Contents of saturated and unsaturated fatty acids were not related to maturity. There were no differences in the contents of saturated and unsaturated fatty acids according to maturity. Linoleic acid and linolenic acid contents were the highest in the super large-sized group(15.5%) and in the large-sized seed group(61.4%), respectively, while contents of fatty acids among the perilla collections were variable with different seed coat colors. Most of the traits studied were not significantly correlated with oil content, but linoleic acid($\omega$-6) content was negatively correlated (r=-0.217*) with linolenic acid($\omega$-3) content.

Monitoring of compositions of gamma-linolenic and omega-3 fatty acids in some functional foods consumed in market (유통중인 건강기능식품의 감마-리놀렌산 및 오메가-3 지방산 함량 모니터링)

  • The compositions of health functional food products (HFFP; 18 products) containing gamma linolenic acid (GLA; $C_{18:3}$, n-6) and omega-3 fatty acids (EPA and DHA) were investigated. The contents of index components (especially, GLA and omega-3) in HFFPs were monitored by GC-FID analysis. Among the GLA products (sample No. 1~8), the content of GLA in most samples (except sample No.6) ranged from 8.04 to 9.98 g/100 g. These results were suitable for the reference standard (more 7.0 g/100 g) of HFF. In the omega-3 products (sample No. 10 and 15) derived from harp seal oil (HSO), the total contents of EPA and DHA were 14.21-15.98 g/100 g, respectively. These values were suitable for the reference standard (more 12.0 g/100 g) of HFF. Besides, among the omega-3 products (sample No.9, 11~14, 16~18) derived from fish oil, the total content of EPA and DHA ranged from 24.11 to 31.20 g/100 g. These results were suitable for the reference standard (more 18.0 g/100 g) of HFF. In the result of TLC analysis, the HFFPs of 18 were mainly composed of triacylglycerols (TAGs). The content of trans fatty acid in 18 HFFPs was detected in less than 0.30 g/100 g. For the detection of trans fatty acid, $^1H$-NMR (600 MHz) can be used because chemical shift of trans fatty acid was observed at 5.3 ppm in this study.
