Effect of Several Species of the Family Rubiacea on Cytotoxicity and Apoptosis in HL-60 cells

  • Herbal medicines have been utilized to treat a variety of diseases, including cancer. Several species of the family rubiaceae have been reported to have antitumor activity. In this study, we report the cytotoxicity and antitumor activity exhibited dy the methanol extracts prepared from Rubia radix (RRME), Uncaria gambir (UGME) and Oldenlandia diffusa (ODME) (family: Rubiaceae) against human promyleloid leukemia cell line, HL-60. The cytotoxicity of RRME (2~20 ${\mu}g/ml$), UGME (20~200 ${\mu}g/ml$) and ODME (20~200 ${\mu}g/ml$) were assessed dy the MTT reduction assay. IC50 values for RRME, UGME and ODME were 11.0, 99.5 and 106.1 ${\mu}g/ml$, respectively. When the HL-60 cells were treated with RRME (10 ${\mu}g/ml$), UGME (120 ${\mu}g/ml$) and ODME (140 ${\mu}g/ml$) for 24 h, several apoptotic characteristics such as DNA fragmentation and morphologic changes were observed. Furthermore, flow cytometric analysis was peformed to determine the percent of apoptotic cells. The poupulation of sub-G1 hypodiploid cells was increased 37.49% in RRME treatment, 12.49% in UGME treatment and 7.21% in ODME treatment compared with untreated control cells (2.64%). To further confirm apoptotic cell death, we assayed caspase-3, -8 and -9 activities in RRME, UGME and ODME-treated cells. After treatment of RRME, UGME and ODME for 12 h, caspase-3, -8 and -9 activities significantly increased.compared to untreated control cells. These results show that RRME, UGME and ODME induced apoptotic cell death in HL-60 cells and may have a possibility of potential antitumor activities.

백화사설초(白花蛇舌草), 산자고(山慈姑), 절패모(浙貝母)에 의한 MDA-MB-231 인체 유방암 세포에서의 항암 효과 (Anti-cancer Effects of Oldenlandia diffusa, Cremastra appendiculata and Fritillaria thunbergii on MDA-MB-231 Human Breast Cancer Cells)

  • O. diffusa, C. appendiculata and F. thunbergii are reported to possess many pharmacological activities including anti-oxidant, anti-inflammatory, anti-hypertension, anti-diabetic and anti-cancer effects. However, their anti-cancer activities in human breast cancer have not been clearly elucidated yet. Objectives: In the present study, we compared the in vitro cytotoxic effects of single and complex treatment of O. diffusa, C. appendiculata and F. thunbergii in human breast cancer MDA-MB-231 cells. Methods: After we treated human breast cancer MDA-MB-231 cells with O. diffusa, C. appendiculata and F. thunbergii. we evaluated viability, growth inhibition, morphological changes, apoptotic body formation, measurement of the cell cycle and formation of DNA fragmentation of these cells. Results: We found that single treatment of O. diffusa and F. thunbergii could inhibit cell proliferation in human breast cancer MDA-MB-231 cells. However, complex treatment of O. diffusa, C. appendiculata and F. thunbergii had weak or no effect on the cell proliferation of MDA-MB-231 cells. The first, anti-proliferative effects of O. diffusa in MDA-MB-231 cells was associated with G2/M arrest of cell cycle and apoptotic cell death. The second, anti-proliferative effect of F. thunbergii in MDA-MB-231 cells was associated with apoptotic cell death. Conclusions: Taken together, these findings suggest that O. diffusa and F. thunbergii may be a potential chemotherapeutic agent for the control of human breast cancer cells, further studies will be needed to identify the molecular mechanisms.

산화적 상해로 인한 상피세포암 세포(KB) 억제에 미치는 천연약용식물 추출물의 상승효과 (Synergistic Effects of Natural Medicinal Plant Extracts on Growth Inhibition of Carcinoma (KB) Cells under Oxidative Stress)

  • 본 연구에서는 천연 약용작물 추출물을 대상으로 하여 산화적 스트레스 하에서 상피세포암 세포 생장억제의 상승효과를 검색하였다. 한국인 호발암인 간암, 위암 및 대장암 등에 효과가 있다고 보고된 총 51 종의 천연 약용작물을 선정하여 메탄올 추출물 시료를 제조하고 , 과산화수소에 의해 유도된 산화적 스트레스 하에서 이 추출물의 암세포생장억제상승효과를 검색하였다. 사용된 51종의 시료 중 27 시료가 다양한 정도의 활성을 나타내었다. 활성이 나타난 27 시료 중 고련피, 곽향, 노나무, 도인, 방기, 백두웅, 백화사설초, 전호, 오미자, 조각자, 천궁의 11시료에서 산화적 스트레스 부가시 $IC_{50}$ 값이 50% 이상 감소되었으며, 다른 16 시료엣 50-25%의 $IC_{50}$ 값의 감소가 나타났다. 우수한 상승효과를 나타낸 11 시료는 산화적 스트레스에 용량 의존적인 상승효과를 나타내었다. 그 중 가장 우수한 상승효과를 나타낸 시료는 노나무(Catalpa ovata)로서 과산화수소수 75 및 100 uM을 가하여 산화적 스트레스를 부가하였을 때 각각 61 및 28%의 $IC_{50}$ 값의 감소를 초래하였다. 그 외 고련피, 곽향, 도인, 백두옹, 오미자 등이 우수한 효과를 나타내었다.
