성선자극 Hormone 수준이 Golder Hamster의 과배란 및 수정에 미치는 영향 (Effects of Superovulation and Fertilization on Gonadotrophic Hormone Levels in the Golden Hamster)

  • This study was carried out to investigate the effects of PMSG and/or HCG treatment on superovulation and fertilization in the golden hamster. The results obtained were summarized as follows: 1. The female groups treated with 30 IU PMSG or 30 IU PMSG+25IU HCG ovulated more eggs than those treated with 15 IU PMSG or 15 IU PMSG+25IU HCG(P<.01). All the PMSG treatment groups superovulated as compared with the untreated control group(P<.01). There were no differences on fertilization rate between the superovulated groups and the control group. 2. The fertilized ova were obtained only by the female group treated with 30 IU PMSG at 1000hr on day 1(morning of ovulation) of the estrous cycle. 3. The intervals between PMSG and HCG injection necessary to obtain the consistent superovulation and fertilized ova were 66hr and 72hr. 4. The superovulated ova were collected from oviduct 48hr, oviduct and uterus 72hr, and uterus 96 hr after mating. 80.3% of two cell, 75.8% of eight cell, and 73.7% of blastocyst of the ovulated ova occurred 48hr, 72hr, and 96hr after mating, respectively.

Propolis 투여가 Ovalbumin으로 유발된 마우스의 알러지성 천식에 미치는 영향 (Effects of Propolis on the Ovalbumin-Induced Allergic Asthma in Mice)

  • Objective : Propolis (PP) has been used in oriental medicine. PP has various biological activities. However, its immunoregulatory and anti-inflammatory activities have not been well studied. In this study, I investigated these activities of PP by using ovalbumin-induced allergic asthma in mice. Methods : To examine the effect of PP on allergic asthma, mice were sensitized with $100{\mu}g$ of OVA and 1mg of aluminum potasssium sulfate (Alum; Sigma) intraperitoneally on day 0 and 7. On day 14, mice were challenged on consecutive 3 days with 5% OVA and AHR was assessed 24 hours after the last challenge. To examine severity of AHR, I examined the population of eosinophiles and T cells in spleen and lung and cytokine production in T cells. Futhermore, I examined histological changes during the OVA-induced allergic asthma. Results and Conclusion : PP reduced the population of eosinophil and CD4+ T cells on the OVA-induced AHR mice model. PP also inhibited IL-4 production but increased INF-g production in T cells. These results suggest that PP may be beneficial material for allergic asthma.

加味柱枝湯이 알레르기 비염에 미치는 영향에 관한 실험적 연구 (The Experimental Stydy on the Anti-Allergic Rhinitis Effects of the Gamigyeji-tang)

  • Major symptoms of allergic rhinitis are nasal obstructions, sneezing and watery rhinorrhea. Gamigyeji-tang has been used to treat for watery rhinorrhea, which is one of the symptoms of allergic rhinitis. This experimental study was done to rescarch effects of Gamigyeji-tang. We have studied effect of mice on OVA-induced Production of IL-4, IL-5, IFN-${\gamma}$ by Murine Splenocytes, and effect of OVA-induced total IgE and OVA-Specific IgE. The results were as follows ; 1. In IL-4 study, Gamigyeji-tang treated group was proved significant inhibitory effect(p〈0.005) 2. In IL-5 study, Gamigyeji-tang treated group was proved significant inhibitory effect.(p〈0.05) 3. In IFN-${\gamma}$ study, Gamigyeji-tang treated group was proved significant inhibitory effect(p〈0.000001) 4. In Total IgE, Gamigyeji-tang treated group didn't showed significant inhibitory effect. 5. In OVA-specific IgE, Gamigyeji-tang treated group didn't showed significant inhibitory effect. According to this result, Gamigyejj-tang was concluded to be effective on anti-allergic action. More studies are required to investigate the mechanism of inhibition by herbal medicine in allergic rhinitis model.

금수육군전(金水六君煎) extract 투여가 Ovalbumin으로 유발된 마우스의 알레르기성 천식에 미치는 영향 (Effects of Kumsooyukkun-jeon extract on the Ovalbumin-Induced Allergic Asthma in Mice)

  • Objective : Kumsoouyukkun-joen extract(KSE) has been used treating asthma for a long time in korea. In the present study, I examined the effects of KSE on the ovalbumin(OVA)-induced airway hyper-reactivity(AHR). Methods : To examine the effects of KSE on asthma, mice were sensitized with $100{\mu}g$ of OVA and 1 mg of aluminum potassium sulfate(Alum; Sigma)intraperitoneally on day 0 and 7. On day 14, mice were challenged on 3 consecutive days with 5% OVA and AHR was assessed 24 hrs after the last challenge. To examine severity of AHR, I examined eosinophil population and cytokine production in bronchoaveloar lavage fluid(BALF) and stained with hematoxylin and eosin in lung and also measured enhanced pause(Penh) in AHR from mice. Results and Conclusion : KSE potently inhibited the development of Penh and also reduced the number of eosinophil during OVA-induced AHR. KSE also inhibited cytokines production such as IL-4, IL-5 in BALF. Furthermore, KSE inhibited percentage of eosinophil in BALF. These results suggest that KSE may be beneficial oriental medicine for AHR.

Metformin Suppresses MHC-Restricted Antigen Presentation by Inhibiting Co-Stimulatory Factors and MHC Molecules in APCs

  • Metformin is widely used for T2D therapy but its cellular mechanism of action is undefined. Recent studies on the mechanism of metformin in T2D have demonstrated involvement of the immune system. Current immunotherapies focus on the potential of immunomodulatory strategies for the treatment of T2D. In this study, we examined the effects of metformin on the antigen-presenting function of antigen-presenting cells (APCs). Metformin decreased both MHC class I and class II-restricted presentation of OVA and suppressed the expression of both MHC molecules and co-stimulatory factors such as CD54, CD80, and CD86 in DCs, but did not affect the phagocytic activity toward exogenous OVA. The class II-restricted OVA presentation-regulating activity of metformin was also confirmed using mice that had been injected with metformin followed by soluble OVA. These results provide an understanding of the mechanisms of the T cell response-regulating activity of metformin through the inhibition of MHC-restricted antigen presentation in relation to its actions on APCs.

Inhallation of e-Cigarette Cartridge Solution Aggravates Allergen-induced Airway Inflammation and Hyper-responsiveness in Mice

  • Electronic cigarettes (e-cigarettes) are becoming increasingly popular worldwide and their cellular effects warrant further evaluation. In this study, we investigated the effects of an e-cigarette cartridge solution on allergen related asthmatic airway inflammation (AI) and airway hyperresponsiveness (AHR), when it is delivered by intratracheal route in mice. Asthmatic AI and AHR were induced by systemic sensitization to ovalbumin (OVA) followed by intratracheal, intraperitoneal, and aerosol allergen challenges in BALB/c mice. The cartridge solution of e-cigarette (containing 16 mg/ml nicotine) was diluted 50 times and $100{\mu}l$ of the diluted solution was intratracheally instilled to OVA-sensitized (OVA-S) mice two times a week for 10 weeks. Long-term e-cigarette inhalation elicited no remarkable changes in the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase enzymes in serum, however, increased infiltration of inflammatory cells including eosinophils, into airways from blood, aggravated the asthmatic AI and AHR, and stimulated the production of cytokines such as interleukin (IL)-4, IL-5 and IL-13, and OVA-specific IgE production. Our data suggest that the inhalation of e-cigarette solutions can function as an important factor to exacerbate the allergy-induced asthma symptoms. Further studies are needed to address the effects of e-cigarette solutions on human health.

방풍(防風)의 난알부민 유도 알레르기 비염 마우스에서의 항알레르기 효능 연구 (Effects of Saposhnikoviae Radix on allergic responses in OVA-induced Allergic rhinitis mice)

  • Objectives : In this study, we investigated the effect of Saposhnikoviae Radix on allergic responses in ovalbumin(OVA)-induced Allergic rhinitis(AR) mice. Methods : BALB/c mice were orally administrated with Saposhnikoviae Radix water extract (SRW, 50 mg/kg) or anti-histamine drug, Ketotifen (10 mg/kg) as a reference drug, followed by sensitization and challenge of OVA. Mice were measured clinical symptoms and the serum levels of histamine, IgE, IL-4, and IFN-${\gamma}$, and observed histopathological changes of nasal mucosa H&E staining. Results : SRW significantly decreased rubs and the serum levels of histamine, IgE, and IL-4, and then increased the serum levels of IFN-${\gamma}$ in OVA-induced AR mice, and inhibited histopathological changes of nasal mucosa with inflammation and the eosinophils infilteration. Conclusions : These data suggest that SRW has anti-allergic effect through the inhibitory property of Saposhnikoviae Radix against allergic responses in allergic rhinitis.

천식 모델 마우스에서 골쇄보의 항천식 효과 (The therapeutic effect of Drynariae Rhizoma in a mouse model of allergic asthma)

  • Objective : Allergic asthma is a chronic airway disease that affects millions of people in the developed world. The disease is characterized by concurring airway inflammation, Th2 cytokine production, increased mucus secretion, airway hyperresponsiveness (AHR) to inhaled antigen, and pulmonary fibrosis. To investigate the therapeutic and anti-asthmatic effects of Drynariae Rhizoma (DR), we examined the influence of DR on the development of pulmonary eosinophilic inflammation and airway hyperresponsiveness in a mouse model of allergic asthma. Methods : In this study, BALB/c mice were systemically sensitized to ovalbumin (OVA) followed intratracheally, intraperitoneally, and by aerosol allergen challenges. We investigated the effect of DR on airway hyperresponsiveness, pulmonary eosinophilic infiltration, various immune cell phenotypes, Th2 cytokine production and OVA specific IgE production in a mouse model of asthma. Results : In asthmatic mice, we found that DR.treated groups had suppressed eosinophil infiltration, allergic airway inflammation and AHR by suppressing the production of IL-5, IL-13 and OVA specific IgE. Conclusions : Our data suggest that the therapeutic mechanism by which DR effectively treats asthma is based on reductions of Th2 cytokines (IL-5), eotaxin, OVA-specific IgE production and eosinophil infiltration.

신비전(新鼻煎)의 항알레르기 효과 (Anti-allergic Effects of Shinbijen)

  • The discovery of drugs on the treatment of rhinitis and allergic disease is a very important subject in human heath. The Shinbijen has been used for centruries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from Shinbijen on OVA-mediated allergic reaction and studied its possible mechanism of action using fluorescenc microscopy and RT-PCR analysis. Shinbijen inhibited OVA-induced rhinitis, total cells in BFA and lymphocyte related to inflammation in mice. Shinbijen decreased immuno response, which activated by IL-4, COX-2 and iNOS expression. in tissue. Shinbijen reduced inflammatory molecule release from mice lung tissue and CD4/CD8 cells activated by cardiac blood. Shinbijen decreased OVA-induced IL-4 and iNOS levels in pulmonary alveoli. Our findings provide evidence that Shinbijen inhibits OVA-induced allergic reactions, and also demonstrate the involvement of inflammation and allergic disease in these effects.

마황(麻黃)의 기도과민반응의 효과 (Effect of Herba Ephedrae on Airway hyperreactivity)

In the present study, we examined the effect HE on the ovalbumin (OVA)-induced airway hyper-reactivity (AHR). Methods : To examine the effect of HE on AHR, mice were sensitized with 100 mg of OVA and 1mg of alum intraperitoneally on day 0 and 14. On day 28, mice were challenged on 3 consecutive days with 3% OVA and AHR was assessed 24 h after the last challenge. To examine severity of airway hyper-reactivity, we examined eosinophil population and cytokine production in bronchoaveloar lavage fluid(BALF) and stained with hematoxylin and eosin in lung. Results : HE potently inhibited the development of airway hyper-reactivity and also reduced the number of eosinophil during OVA-induced airway hyper-reactivity. HE also inhibited cytokines production such as IL-4, IL-5, IL-13 in BALF. Furthermore, HE inhibited proliferation of eosinophil in a dose dependent manner. Conclusions : These results suggest that HE may be beneficial oriental medicine for AHR.
