Production of Endoglucanase, Beta-glucosidase and Xylanase by Bacillus licheniformis Grown on Minimal Nutrient Medium Containing Agriculture Residues

  • Bacillus licheniformis was grown in minimal nutrient medium containing 1% (w/v) of distillers dried grain with soluble (DDGS), palm kernel meal (PKM), wheat bran (WB) or copra meal (CM), and the enzyme activity of endoglucanase, ${\beta}$-glucosidase, xylanase and reducing sugars was measured to investigate a possibility of using cost-effective agricultural residues in producing cellulolytic and hemicellulolytic enzymes. The CM gave the highest endoglucanase activity of 0.68 units/mL among added substrates at 48 h. CM yielded the highest titres of 0.58 units/ml of ${\beta}$-glucosidase, compared to 0.33, 0.23, and 0.16 units/mL by PKM, WB, and DDGS, respectively, at 72 h. Xylanase production was the highest (0.34 units/mL) when CM was added. The supernatant from fermentation of CM had the highest reducing sugars than other additional substrates at all intervals (0.10, 0.12, 0.10, and 0.11 mg/mL respectively). It is concluded that Bacillus licheniformis is capable of producing multiple cellulo- and hemicellololytic enzymes for bioethanol production using cost-effective agricultural residues, especially CM, as a sole nutrient source.

Estimating Nutrients Delivery Ratios at the Subwatershed Scale -A Case Study at the Bochung-A Watershed- (소유역 유달율 추정공식 개발 -보청A유역을 중심으로-)

  • The characteristics of delivered nutrient loads were analyzed and the regression equations to estimate delivery ratios of nutrients (TN and TP) were developed using HSPF simulation results at six subwatersheds within the Bochung A unit watershed during 1998-2007. TN delivery ratio was higher than TP delivery ratio because significant amounts of TP was considered to be attached at soil as ${PO_4}^-$ during delivery process from discharged point of nutrient source to main stream. As a results of correlation analysis, factors related to geomorphic characteristics had not statistical correlation with TN and TP delivery ratios. TN loading rate from living and specific stream flow had statistical negative and positive correlation, respectively, with TN delivery ratio. TP loading rates from all sources and from land cover and specific stream flow had statistical negative, negative and positive correlation, respectively. The specific stream flow represents the most strong correlation with nutrient delivery ratios. The regression equations to estimate delivery ratios for TN and TP were developed by including statistical correlated factors and showed high efficiency of 0.98 and 0.95 of coefficient of determination for TN and TP, respectively.

Effect of Supplementing Sheep with Sunflower Acid Oil or its Calcium Soap on Nutrient Utilization

  • Four adult rams ($22.25{\pm}0.90kg$) were used in a $4{\times}4$ latin square design to evaluate the rations without ($T_1$) or with supplementation of sunflower acid oil at 5 ($T_2$), 10 ($T_3$) or calcium soap at 10% of dietary DM ($T_4$) on nutrient digestibility and balances of nitrogen, calcium and phosphorus. The basal ration contained 60 parts Brazilian napier grass hay and 40 parts concentrate mixture. The DM, CF, NDF and ADF digestibilities and nitrogen retention (g/d) decreased (p<0.01) by inclusion of sunflower acid oil at 5% of dietary DM. In addition, depression (p<0.01) in digestibilities of CP, nitrogen free extract (NFE), cellulose, hemicellulose, retention of calcium and phosphorus (g/d) were also observed with increasing the level of sunflower acid oil to 10% of dietary DM. The EE digestibility, total digestible nutrients (TDN) content and calcium retention (g/d) were significantly higher (p<0.01) for ration supplemented with calcium soap. It is concluded that sunflower acid oil supplementation in free form as low as 5% of dietary DM is deleterious to fibre digestion in sheep while as calcium soap, it can be fed up to 10% of dietary DM as an energy source without any adverse effect.

  • The purpose of this study is to computerize all the necessary information on the daily food value and nutritional status for individuals and groups. In this research, a FOCUS-16jXT (16 bit personal computer ) compatible with IBM-PCjXT was used, and the database files and programs were created by using the dBASEIII package. The food life evaluation system consists of 3 subsystems of Reference, Nutrition Status Assessment and Food Source. The findings are summerized a8 follows: 1. Reference: This subsystem enables users to proceed to the next step, if necessary, by describing each subsystem. 2. Nutrition Status Assessment. 1) Food Habit Assessment: This subsystem determines whether the user has a good food habit or not, based on the answers for ten questions about daily food life. 2) Obesity Assessment: This subsystem calculates Broca index, which is used as a indicator of obesity. 3) Nutrient Intakes: When personal data such as age, sex, weight, height and food consumptions are input, it is possible to calculate the followings. i) Comparison between the amounts intaked and the recommended dietary daily allowances of various nutrients ii) Nutrient intakes from each food group and their composition rates for the nutrients iii) Nutrient intakes per unit body surface area iv) Composition of lipid intake 3. Food Sources: The appropriate food sources for the lacking nutrients will be recommended to the subjects.

The Standardized Recipe and Nutrient Analysis of Stir-fried Whip-arm Octopus in Busan (부산지역 낙지볶음의 표준조리법 개발 및 영양소 분석)

  • The purpose of this study was to develop the standardized recipe and to analyze the nutrients of stir-fried whip-arm octopus as one of the kind of traditional local foods in Busan. The standardized recipe was developed by using cookbooks, home recipes, and the recipes by commercial food restaurants. The nutrient was analyzed by using an AOAC method. Sensory evaluations were made on nine sensory attributes by a 12-member panel. As the results, the whip-arm octopus was recorded as a food source for health, rejuvenation in Jasanobo. Since early 1900, the stir-fried method has been used for cooking with the ship-arm octopus. The main ingredients were whip-arm octopus, onion, large green onion as the local stir-fried whip-arm octopus in Busan. The ingredients of soup were shrimp, little neck clam, and water. Seasoning sauce was mixed with red pepper powder, soy sauce, sesame oil, chopped garlic, chopped onion, and sugar. The seasoning sauce was fermented for three days. The results of sensory scores were salty seasoning 3.66/5.00, taste 3.75/5.00, thickness 3.84/5.00, and smell 4.09. Nutrient retention per 100g of the stir-fried whip-arm octopus was 67.54kcal, protein 6.43g, fat 1.66g, Ca 28.06mg, Fe 2.56mg, cholesterol 68mg, and taurin 51mg. Fatty acid consists of various unsaturated fatty acids such as linoleic acid(46.24%) and oleic acid(33.67%).

Response of growing pig in dietary sucrose supplementation on growth performance, nutrient digestibility, fecal score, and serum cortisol

  • Sucrose is a common disaccharide sugar that is used in pig diet mainly as an energy source as well as to improve the palatability of diet. This study investigated the effects of dietary sucrose supplementation on the growth performance, nutrient digestibility, fecal score, and serum cortisol of growing pigs. A total of 96 growing pigs were randomly allocated into three treatment groups (8 repetitions per treatment, 4 pigs per pen). Dietary treatments included: control (CON), basal diets; treatment 1 (TRT1), basal diet with 0.75% sucrose; and treatment 2 (TRT2), basal diet with 1% sucrose. Dietary sucrose supplementation tended to improve (p < 0.10) the average daily gain compared to CON group. Moreover, increased (p < 0.05) feed intake was found in the sucrose supplemented group compared to the CON group. In comparison to the CON diet the nutrient digestibility of energy tended to increase (p < 0.10) by sucrose supplemented diet. Fecal score was not altered through dietary sucrose supplementation. Decreased (p < 0.05) serum cortisol was found in both the 0.75% sucrose and 1% sucrose supplemented diet than the CON diet. In conclusion, sucrose can be a suitable feed ingredient for growing pigs as it can improve the palatability of diet as well as feed intake, energy digestibility and reduce stress through reducing serum cortisol.

Pollutant Load Characteristics of a Rural Watershed of Juam Lake (주암호 농촌 소유역 오염부하특성)

  • A monitoring study has been conducted to identify hydrologic conditions, water quality and nutrient loading characteristics of small watershed in Juam Lake. Climate data of the watershed were collected; flow rate was measured and water quality sampling was conducted at the watershed outlet for this study. Water quality data revealed that T-P concentrations meet I grade of lake water quality standard during non-storm period, but degraded up to II-III grade of lake water quality standard during storm period. The observed T-N concentrations always exceeded lake water quality standard. Therefore, T-P was identified as limiting chemical constituent for eutrophication of Juam Lake. T-P concentration of non-storm period also revealed that point source pollution is not serious in the watershed. Three year monitoring results showed that the observed T-N losses were $10.85\~18.88$ kg/ha and T-P losses were $0.028\~0.323$ kg/ha during six month (Mar. - Oct.), respectively. Major portion of runoff amount discharged by a few storm events a year and nutrient load showed apparent seasonal variation. Huge runoff amounts were generated by intense storms, which make application of water treatment or detention facilities ineffective. Monitoring results confirmed that water quality improvement by abating nonpoint source pollution in rural watershed of monsoon climate should be focused on source control. T-P losses from paddy field seemed to consist of significant amount of total load from study watershed. Therefore, management of drainage from paddy field is considered to be important for preventing algal blooming problem in Juam Lake.

Effects of different sulfur ion concentration in nutrient solution and light source on glucosinolate contents in kale sprouts (Brassica oleracea var. acephala)

  • The aim of this study was to investigate the amount of glucosinolates (GSLs) in kale sprouts (Brassica oleracea L. var. acephala) ('TBC') according to different concentrations of sulfur ions in sprout's nutrient solutions (0.0, 0.5, 1.0, and 2.0 mM) and to different light sources [Fluorescent lamp, Red, Blue, and Mix (R+B) LED]. Kale sprouts were cultivated in a growth chamber for 13 days in sulfur solutions. Kale sprouts were treated with fluorescent lamp and LED light sources for 5 days, from eight days after sowing to harvest. Amount of seven types of GSLs (progoitrin, sinigrin, 4-hydroxyglucobrassicin, glucobrassicin, 4-methoxyglucobrassicin, gluconasturtiin, and neoglucobrassicin) were measured in kale sprouts after harvest. The total GSL content was influenced by different sulfur solution concentration, and it was the highest at S 0.5 mM ($172.54{\mu}mol{\cdot}g^{-1}DW$) and the lowest at S 2.0 mM ($163.09{\mu}mol{\cdot}g^{-1}DW$). The GSL content was influenced by different light source, and it was the highest with Red LED ($159.23{\mu}mol{\cdot}g^{-1}DW$) and the lowest with Blue LED ($147.57{\mu}mol{\cdot}g^{-1}DW$). As the sulfur solution concentration increased under all light source, progoitrin and sinigrin contents tended to decrease while glucobrassicin content showed an upward tendency for all of the light sources. The content of glucobrassicin was higher than that of progitrin when treated with sulfur solutions for all LED light sources. Sinigrin, which has excellent anti-cancer effects, showed the highest rate (92.2%) among all the GSLs, under all of the light sources.

A Study on Autocontrolled SBR for Biological Nutrient Removal with External Carbon Sources (외부탄소원 주입시 영양염류의 생물학적 제거를 위한 자동제어 SBR 공정에 관한 연구)

  • The purpose of this study is to develop effective operating process in order to achieve more suitable conditions of Anoxic-Oxic-Anoxic-Stripper(AOAS) SBR through real-time control. To improve the removal efficiency, glucose, methanol and synthetic food waste acid fermentant were added as an external carbon source, In the case of glucose and synthetic food waste acid fermentant, TN, TP were removed to average 86.9%, 73.0% respectively. Methanol was removed to average 64.6%, 55.4% respectively. The synthetic food waste acid fermentant proved to be the most efficient and allowed for the substitution of an external carbon source. The removal rate of $COD_{Cr}$, was approximately 90% at all cases. The results of the study that a correlation between ORP (Oxidation-Reduction Potential), pH and DO and nitrification or denitrification when an external carbon source is added and when it isn't was showed that ${\Delta}ORP$ is suitable parameter. ORP reacted properly to denitrification (${\Delta}ORP<-10$) and nitrification (${\Delta}ORP<0$). The use of real-time control saved anywhere between 61 and 67 minutes at the anoxic(1) stage and 26 to 52 minutes at the oxic(1) stage. When the time saved from the anoxic(1) and oxic(1) was added to the anoxic(2) stage for the removal efficiency of TN and TP increased from 0.7 to 13.9% and 12 to 35 % respectively.

Evaluation of COD Solubilization and Reduction of Waste Activated Sludge by pH Control (pH 조절을 통한 폐활성 슬러지의 COD 가용화 및 감량화 평가)

  • From the view point of biological wastewater treatment, C/N ratio is one of the most important factor in biological nutrient removal process. However, municipal sewage in Korea is characterized by extremely low content of carbon source and relatively higher portion of N source. Accordingly, it is necessary to dose external carbon source in order to obtain higher degree of carbon source within the process. In this study, the effects of pH pretreatment as an alternative plan for increasing carbon source on the cell disruption and COD solubility of waste activated sludge were conducted under well defined experimental conditions. During 5 hours, the value of COD solubilization rate ($S_R$) at pH 11.5 is approximately 4.4 times higher than the value of $S_R$ at pH 9.5. It is expected that the level of SCOD increased due to the result from cell disruption. However, VSS/TSS ratio was not significantly changed after 5 hours. As Alkalinity changes gradually from less than 15, 30 and 60 meq NaOH/L, average RBCOD/SCOD fraction showed 34, 36 and 45%,respectively.
