Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.
일부 그람양성세균들은 고온, 저온, 염, 에탄올, 산소와 영양분 고갈과 같은 다양한 스트레스 상태에 노출되면, 일반 스트레스반응(general stress response)에 의해서 일련의 스트레스 단백질군을 발현시켜 외부 스트레스를 극복하고 세균의 생존력을 증가시킨다. 비병원성균인 Bacillus subtilis의 일반 스트레스반응에 관해서는 많은 연구가 이루어져 있으므로 다른 균의 연구모델로 이용이 가능하다. 본 총설에서는 B. subtilis와 병원성균인 Listeria monocytogenes의 일반 스트레스반응의 유사성과 차이점을 B. subtilis를 모델로 하여 비교하였다. 두 균의 일반 스트레스반응은 대체 전사 인자인 ${\sigma}^B$ (alternative transcription factor sigma B)에 의해서 조절되고 신호전달 네트워크 또한 매우 유사하며, ${\sigma}^B$ 의존성 유전자들에 의해 150여 개의 스트레스 단백질들이 발현된다. 그러나 L. monocytogenes는 B. subtilis의 에너지 스트레스 신호 경로를 가지고 있지 않은 점과, 일반 스트레스반응에 의해 병독 유전자들(virulence genes)이 조절되는 것이 가장 큰 차이점이다. 그러므로 L. monocytogenes의 생리 및 병원성 규명을 위해서는 일반 스트레스반응에 관한 이해가 매우 중요하다.
The nutritional worth of Sarson Saag Waste (SSW), a cannery waste, was assessed in comparison with conventional complete diet as a total mixed ration (TMR), and a conventional green fodder, Avena sativa. Each diet was offered ad libitum, supplemented with mineral mixture and common salt, to 4 male murrah buffaloes. The control TMR was made iso-nitrogenous to SSW. Simultaneously, each diet was offered to 3 rumen fistulated male buffaloes for assessing the biochemical changes in the rumen. The nutrient digestibility of unconventional SSW was comparable to that of conventional green fodder-A. sativa but significantly (p<0.05) higher than that of control TMR. The tri-chloro acetic acid (TCA) precipitable-N in the strained rumen liquor of animals fed SSW was considerably higher than that of animals fed A.sativa. The urinary excretion of total purine derivatives was comparable in animals fed SSW and conventional green fodder but significantly (p<0.05) higher than those fed conventional control TMR. The significantly (p<0.05) lower purine nitrogen index (PNI) in animals fed control TMR resulted in significantly (p<0.05) lower microbial protein synthesis than that in animals fed SSW and conventional green fodder. The N-excretion as per cent of nitrogen intake was significantly (p<0.05) lower in animals fed SSW as compared to either of the conventional feeds tested, resulting in significantly (p<0.05) higher Nretention and apparent biological value. SSW supplemented with mineral mixture could serve as an excellent source of nutrients for ruminants.
This study investigated the influence of anthropometric data and nutrient intake on bone mineral density(BMD) and biochemical markers of bone metabolism The mean age of 21 premenopausal women were 47.0 years and that of 41 postmenopausal women whose menopausal age was 49.46 years were 60.56 years. The waist and WHR of postmenopausal women were significantly higher than those of premenopausal ones. The animal protein intake of premenopausal and postmenopausal women were 38.5 and 21.03 g which comprised 54.35 and $31.84\%$ of total protein intake, respectively. The calcium intake of premenopausal and postmenopausal women were 446.45 and 546.97mg which was 63.78 and $78.14\%$ of Korean RDA, respectively. The ALP(Alkaline phosphatase) of premenopausal women was 65.81 U/L, which was significantly lower than that(90.24 U/L) of postmenopausal women (p<0.01). BMD of lumbar spine of premenopausal women was correlated significantly with body weight(r=0.690, p<0.01), waist(r=0.682, p<0.01), WHR(r=0.672, p<0.01), BMI(r=0.559, p<0.01), and body fat(r=0.457, p<0.01). Urinary Ca/creatinine ratio of the premenopausal women was negatively correlated with plant protein(r=-0.529, p<0.05) and plant calcium(r=-0.579, p<0.05). BMD of lumbar spine of postmenopausal women showed positive correlation with lean body mass(r=0.469, p<0.01) and body weight(r=0.383, p<0.05). Urinary Ca/creatinine ratio for the postmenopausal women was positively correlated with ALP(r=0.404, p<0.01) and urinary Na/creatinine ratio(r=0.389, p<0.05). In conclusion, it is necessary to maintain adequate body weight and to increase calcium intake for the premenopausal women. It is also important to increase muscle mass and reduce salt intake for the postmenopausal women.
Livestock manure is generally beneficial to soil and crop production when appropriate amount is applied, but excessive application may be detrimental to soil and water environments. A proper protocol of livestock waste treatment is required to manage the quality of soil and water. A trickling filter system using rice straw media was employed to treat piggery wastewater from small-scaled livestock farms as an alternative to the currently available methods. Batches of piggery wastewater were treated with this system, and the byproducts of rice straw media and trickling filtrate were applied to the soil with cultivating rye (Secale cereale L.). Objective of this research was to characterize these byproducts for the sustainable soil amendments and rye production. Both the treated straw medium and filtrate were proven to be effective organic fertilizers for rye plant development, with the enhanced but balanced absorption of nutrients. The synergistic effects of filtrate in addition to straw application did not show, but the filtrate appeared to lead to a higher water content of the plant. No specific nutrient deficiency or toxicity symptom was shown due to the salts derived from the byproducts applied. Chemical parameters of the soil quality were significantly improved with the application of straw medium either with or without the filtrate. Judging from parameters relating to the salt accumulations, such as sodium adsorption ratio (SAR), electrical conductivity (EC), exchangeable sodium percentage (ESP), potassium adsorption ratio (KAR), and residual P concentrations, the byproducts from piggery wastewater exhibited no detrimental effects on soil quality within the ranges of treatments used. In addition to the effectiveness of the rice straw trickling filter system for the small-scaled swine farms, both rice straw medium and filtrate could be recycled for the sustainable soil amendment and plant nutrition.
Image contrast of whole bacteria was compared in Staphylococcus aureus and Escherichia coli depending on pre-stain suspension liquids by energy-filtering transmission electron microscopy. The two bacterial strains were suspended in three most commonly used liquids for negative staining (triple distilled water [DW], phosphate-buffered saline [PBS], and nutrient broth [NB]) and directly observed without staining or stained with neutralized potassium phosphotungstate (PTA), respectively. Even though in low contrast, unstained bacteria were observed owing to their inherent electron density and cell shape in zero-loss (elastic scattering) images. After being suspended in PBS, unstained bacteria appeared to have higher contrast and more refined periphery than DW-suspended ones, and extracellular appendage structures such as fimbriae and flagella could be discerned. The unstained bacteria appeared to be invariably surrounded with electron-lucent precipitates, possibly from PBS. As far as delineation of the structures, the combination of DW or PBS suspension with subsequent staining provided the most satisfactory results, as evidenced by the high contrast of bacterial morphology and appendage structures. However, after being suspended in NB and stained with PTA, bacteria often had too high contrast or poor staining, with electron-dense aggregates around the bacteria. These results suggest that suspension with concentrated organic aliquots including broth media before PTA staining could deteriorate image contrast, and should be used only in dilute form for visualizing bacterial morphology and appendage structures. Moreover the contrast enhancement of unstained bacteria by salt granules would be advantageous in demonstrating bacterial sorption of environmental particles like heavy metals, maintaining minimal contrast for cell imaging.
Vibrio vulnificus is a gram-negative, halophilic, oxidase-positive, lactose-positive, motile, rod shaped bacterium that has been associated with primary septicemia and wound infection. Elucidating the growth and survival of V. vulnificus in ecological conditions is of great importance to develop sanitary measure against this microorganism. Thus we simulated the ecological conditions and evaluated the effect. About $10^5\;CFU/ml$ of V. vulnificus was inoculated to fresh water, brackish water $(1\%\;NaCl)$, sea water $(3\%\;NaCl)$, and bottom deposit solution. The same concentration of V. vulnificus was also inoculated to distilled water, $1\%\;NaCl$ solution and $3\%\;NaCl$ solution as controls. These were stored at 4, 15 and $25^{\circ}C$, respectively and were used to assess the effects of temperature and salinity on the survival of V. vulnificus. In fresh water V. vulnificus could not survive regardless of storage temperature. In case of brackish water and sea water survival time of V. vulnificus was the longest at $25^{\circ}C$, and the number of V. vulnificus was decreased most rapidly at $4^{\circ}C$. V. vulnificus survived longer in brackish water than in any other conditions. In bottom deposit solution containing brackish water, the survival time of V. vulnificus was longer and the rate of decline was slower than that in brackish water. These results indicate that both biological and physicochemical factors such as temperature and salinity could affect survival of V. vulnificus. V. vulnificus, damaged in normal fresh water, did not grow on TCBS agar of selective plating medium but grew on BHI agar plate; However, V. vulnificus was recovered by addition of salt and nutrient materials.

A more efficient use of nutrients from organic sources can benefit both farmers and water quality in agriculture. In this paper we reviewed information related to the regulations and guidelines of a swine liquid fertilizer to propose an establishment scheme for the official standards of swine manure slurry as a liquid fertilizer for organic farming. According to the law of establishment and designation of official standard of fertilizers, the liquid fertilizer made with pig liquid manure belongs to the byproduct fertilizer as of one of organic fertilizer. However, the official standards for byproduct fertilizers including the liquid fertilizer set by the Rural Development Administration (RDA), especially for a liquid swine manure fertilizer, should be revised to meet the goals of laws pursuing the environmentally sound agriculture, especially for organic farming because the terms and relevant standards need additional specific standards alike the standards such as impurities and parameter for declaration in EU. Therefore, it is suggested that official standards for byproduct fertilizers should be revised with respect to nutrient, salt content as of SAR and Na content instead of NaCl. Also we need to develop the maximum limit of heavy metals in soil to which the liquid fertilizer can be applied, as well as application rate depending on EC of the liquid fertilizer.
본 연구에서는C. indica 자실체의 안정적인 대량생산에 기반이 되는 필수적인 균사체 생산을 위한 최적 배양조건을 구명 하고자하였다. C. indica 균사체의 효율적 배양을 위한 조건은 최적 배양 온도 30-32℃, pH 6.0, 탄소원 maltose 2%, 유기질소원 yeast extract 1%, 무기질소원 NaNO3 0.1%, 아미노산 asparagine 0.7%, 유기산 acetic acid 0.1%, 무기염류 MnSO4 0.7 mM가 최적 배양조건 이었다. 새로이 MYNA로 명명한 C. indica의 최적 배지는 기존의 PDA 배지보다 균사의 생장이 균일하고, PDA 배지의 배양기간과 비교하였을 때 10.5% 단축하는 효과가 있었다. MYNA 배지는 추후 C. indica의 재배에 널리 활용될 수 있을 것으로 판단된다.
