The effects of Mn on growth and nutrient status of Pinus densiflora seedlings grown in a nutrient culture solution were investigated. Mn concentrations was added as manganese chloride at 0, 30 and 60ppm to the nutrient culture solution. The 2-year-old seedlings were transplanted into the solution maintained at pH 4.0, and grown for 90 days in a greenhouse. The Mn treatment induced a significant reduction in the dry weight growth of the seedlings. The relative growth rate(RGR) and net assimilation rate(NAR) of the seedlings decreased with increasing Mn concentrations in the nutrient culture solutions. For the nutrient status of the seedlings, Ca and Mg content in trunk and root was least in 60ppm Mn treatment, and Mn content in needle was about 3 times more than in root. Also the net photosynthetic rate of the seedlings was significantly lower both in 30ppm and 60ppm Mn treatment compared to them in 0ppm. This result suggests that the reductions in the RGR and NAR of the seedlings may be resulted from the inhibition of net photosynthesis by the mixed effect of lower nutrient uptake of roots and excess accumulation of Mn in needle.
This study was carried out to investigate the effects of changes of ionic strength according to growth stage on growth and flowering of Dendranthema grandiflorum (Ramat.) Kitamura 'Seiun' grown hydroponically in perlite. The stage I, II, and III covered early vegetative growth (27-40 days after planting), latter vegetative growth (41-54 days), and reproductive growth (55-80 days), respectively. The 2 strength (1S and 2S) of nutrient solution were treated in stage I, whereas 3 strengths (1S, 1-2S, and 2S) were treated in stage II. Then, total 9 treatments in stage III were designated by 3 treatments (tap water, 1S, and 2S) for each 3 strengths in stage II. Each nutrient solution was applied 8 times per day. At vegetative growth stage (54 days after planting), stem length was highest when irrigated 8 times a day with 1S nutrient solution. Both photosynthesis and transpiration rate were higher in 1S than those in other treatments (1-2S, 2S), whereas leaf chlorophyll content was highest in 2S treatment. Ion content of plant treated with 2S was higher than other treatments. Growth (plant height, leaf area, stem length), fresh weight, and dry weight of each plant organ after flower bud formation were better in tap water treatment (1-1-0) than other 1S treatments (1-1-1, 1-1-2). Regarding the number of days to flowering, tap water treatment was the most effective. Thus, after flower bud formation supplying tap water or lower concentration of nutrient solution than those used during the vegetative growth stage was economical in saving chemical fertilizers, shortening the number of days to flowering, reducing salt accumulation in media, saving efforts of leaching, and reducing ground water contamination.
The production of poly-$\beta$-hydroxybutyrate(PHB) from methanol by batch and fed-batch cultivations of Methylobacterium sp. GL-10 was studied. PHB accumulation was stimulated by the nutrients deficiency including, NH4+, SO42-, and K+. The nitrogen deficiency was the most critical factor for PHB accumulation. In batch cultivation, the maximum cell concentration and PHB content were 1.86g/l and 0.62g/l, respectively, with 1.0%(v/v) of methanol and 0.5g/1 of ammonium sulfate. The mass doubling time of Methylobacterum sp. GL-10 was in the range of 4-5 hrs. The cell growth and PHB accumulation were severely inhibited at the methanol concentration over than 2% (v/v). To overcome methanol Inhibition, constant feeding and intermittent feedillg fed-batch cultivations were adopted, using C/N molar ratio as a control factor. In constant feeding fed-batch process, cell concentration was increased up to 2.67g/1, and PHB yield was enhanced from 0.33 of batch culture to 0.53. The relatively low cell concentration was caused by methanol accumulated in culture broth at late growth phase. To prevent methanol accumulation and to maximize PHB production, DO-state intermittent fed-batch cultivation was attempted. The cell and PHB concentration was reached up to 4.55g/1 and 1.80g/1, respectively. It was possible to maintain methanol concentration low and also to feed nutrient of desired C/N molar ratio.

Near future dynamics of water qualities (nutrient concentration) of the Han River was predicted, based on a mathematical model representing the relationship between the nutrient concentration in th river wagter and environmental factors (population density, land-use types, rock compositions and nutrient accumulation) in the basin. The population density and land-use types were forecasted to change distinctly in the downstream area, especially in Seoul City area in 1985~1990 whereas any environmental factor was not expected to change its level significantly in both upstream and middle reaches areas. It was indicated by the model that the nutrients concentration in the up- and mid-streams would keep its level in future as it was, but it would increase drastially in the downstream area. For the preservation of the water qualities in the downstream at least to keep its level as it was in 1980, practical countermeasures were proposed, based on the assessment of the contribution of each environmental factor to the water qualities.
The freshwater green alga Chlorococcum sp. grew on NH_4^{+},{\;}NO_3^{-}$, urea, yeast extract, and peptone as the nitrogen source showing similar pattens of growth and secondary carotenoid (SC) production. However, the most suitable nitrogen source for the induction fo SC was urea. The dffects of nutrient levels (urea, phosphate, sulfate, ferrous iron, and salt) on growth and SC production were stydied by varying the concentration of each nutrient in batch cultures. High biomass production was achieved in cultures containing 20-28 mM urea, 4.8-10 mM phosphate, 1.6 mM sulfate, 70 mM phosphate, 1.6 mM sulfate, 170 mM NACl, and $50{\;}\mu\textrm{M}$ iron. The optimum concentrations of nutrients for biomass and for the SC accumulation in biomass were evaluated and the two media for achieving high biomass production and SC production were thus developed. The extent to which each parameter to stimulate the formation of SC in the alga were varied and the potentially improned SC prodution by manipulating the nutrient levels in the modified media were descussed.
BACKGROUND/OBJECTIVES: Epigenetic regulation by nutrients can influence the development of specific diseases. This study sought to examine the effect of individual nutrients and nutrient families in the context of preventing chronic metabolic diseases via epigenetic regulation. The inhibition of lipid accumulation and inflammation by nutrients including proteins, lipids, vitamins, and minerals were observed, and histone acetylation by histone acetyltransferase (HAT) was measured. Correlative analyses were also performed. MATERIALS/METHODS: Nutrients were selected according to information from the Korean Ministry of Food and Drug Safety. Selected nutrient functionalities, including the attenuation of fatty acid-induced lipid accumulation and lipopolysaccharide-mediated acute inflammation were evaluated in mouse macrophage Raw264.7 and mouse hepatocyte AML-12 cells. Effects of the selected nutrients on in vitro HAT inhibition were also evaluated. RESULTS: Nitric oxide (NO) production correlated with HAT activity, which was regulated by the amino acids group, suggesting that amino acids potentially contribute to the attenuation of NO production via the inhibition of HAT activity. Unsaturated fatty acids tended to attenuate inflammation by inhibiting NO production, which may be attributable to the inhibition of in vitro HAT activity. In contrast to water-soluble vitamins, the lipid-soluble vitamins significantly decreased NO production. Water- and lipid-soluble vitamins both exhibited significant inhibitory activities against HAT. In addition, calcium and manganese significantly inhibited lipid accumulation, NO production, and HAT activity. CONCLUSIONS: Several candidate nutrients and their family members may have roles in the prevention of diseases, including hepatic steatosis and inflammation-related diseases (i.e., nonalcoholic steatohepatitis) via epigenetic regulation. Further studies are warranted to determine which specific amino acids, unsaturated fatty acids and lipid-soluble vitamins or specific minerals influence the development of steatosis and inflammatory-related diseases.

The objective of this study was to produce a nutrient delivery system (NDS) using sialic acid extracted from edible bird's nest (EBN), which improves brain function in patients with Alzheimer's disease and Parkinson's disease, by affinity bead technology (ABT). The inhibitory activity of acetylcholinesterase (AChE) and pyramidal cells in the dentate gyrus of the hippocampus were analyzed to investigate the effect of a sialic acid NDS on Alzheimer's disease. Also, the effect of a sialic acid NDS on Parkinson's disease was evaluated by rota-rod test and pole test in an animal model. Among the groups treated with donepezil, EBN, and sialic acid NDS, the AChE activity was the lowest in the sialic acid NDS-treated group. The results of the hippocampus analysis of the rat model confirmed that the sialic acid NDS inhibited amyloid-beta accumulation depending upon the concentration. Also, the sialic acid NDS group showed more improvement in motor deterioration than the1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced group in both the rota-rod test and pole test. Therefore, the sialic acid NDS had an effect of protecting not only Alzheimer's disease by inhibiting AChE and amyloid-beta accumulation, but Parkinson's disease by preventing neurotoxicity induced by MPTP.
When methanol was the sole carbon source, Methylobacterium organophilum NCIB 11278, a facultative methylotroph, accumulated Poly-$\beta$-hydroxybutyrate (PHB) as 59% (w/w) of dry cell weight under potassium limitation, 37% under sulfate limitation, and 33% under nitrogen limitation. Based on a stoichiometric analysis of PHB synthesis from methanol, it was suspected that PHB synthesis is accompanied by the overproduction of energy, either 6-10 ATP and 1 $FADH_2$ or 6 ATP and 3 NADPH to balance the NADH requirement, per PHB monomer. This was confirmed by observation of increased intracellular ATP levels during PHB accumulation. The intracellular ATP with limited potassium, sulfate, and ammonium increased to 0.185, 0.452, and 0.390 $\mu$moles ATP/g Xr (residual cell mass) during PHB accumulation, respectively. The intracellular ATP level under potassium limitation was similar to that when there was no nutrient limitation and no PHB accumulation, 0.152- 0.186 $\mu$moles ATP/g Xr. We propose that the maximum PHB accumulation observed when potassium was limited is a result of the energy balance during PHB accumulation. Microorganisms have high energy requirements under potassium limitation. Enhanced PHB accumulation, in ammonium and sulfate limited conditions with the addition of 2,4-dinitrophenol, which dissipates surplus energy, proves this assumption. With the addition of 1 mM of 2,4-dinitrophenol, the PHB content increased from 32.4% to 58.5% of dry cell weight when nitrogen limited and from 15.1 % to 31.0% of dry cell weight when sulfate limited.

A process for the production of bioplastics from wastewater with an open microbial culture was developed and evaluated. The process consists of a selection reactor to select bacteria in feast/famine regime and an accumulation reactor to produce PHA using selected bacteria. Polyhydroxyalkanoate(PHAs) accumulating bacteria could be efficiently grown in a sequencing batch reactor(SBR) without any growth limitation. For the high production of PHA limitation such as oxygen and nutrients seemed to be needed. Accumulation experiments were performed to evaluate the level of accumulation of PHA. Limited aeration had no effect, but nutrients limitation showed high accumulation. Bacteria which were selected in the SBR could accumulate PHA till 60% of cellular dry weight in accumulation experiments under nitrogen limitation. PHA accumulation rate decreased with increasing PHA content in the cells. Clearly, PHA accumulation rate has a strong correlation with the PHA content of the cells.
Microorganisms capable of accumulating poly-p-hydroxybutyric acid(PHB) were isolated from soil by enrichment culture technique. Among them, the strain designated as FL-027 had high PHB productivity and was identified as Alcaligenes. The optimal medium composition for cell growth was 8.0 $g/\ell$ of fructose and 3.0 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 5.04 at pH 7.0 and $30^{\circ}C$. To investigate the optimal conditions for the PHB accumulation, we divided the process into two stages; the first stage for the growth of the cell in nutrient-rich medium and the second stage for the PHB accumulation in nutrient-deficiency medium. The optimal conditions for PHB accumulation were 8.0 $g/\ell$ of fructose and 0.25 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 60 at pH 6.5 and $30^{\circ}C$. PHB accumulation was stimulated by deficiency of nutrients such as $NH_4^+$, $Ca^{2+}$, $SO_4^{2+}$ in medium. Among them. $NH_4^+$ deficiency was chosen because of its effectiveness. We found the inhibition of cell growth by fructose in batch culture. In order to keep the fructose concentration at an optimal level, intermittent feeding fed-batch culture was employed, and the cell concentration as high as 10.83 $g/\ell$ whose PHB content was responsible for 43% of the dry cell weight. The purified PHB was identified as homopolymer of 3-hydroxybutyric acid by using IR and $^1H-NMR$.
