Streptomyces 속 균주가 생성한 물질의 생물활성 (제삼보) 정제 및 영양요구성 (Biological Active Substance Produced by a Strain of Streptomyces sp. (Part.III) Purification and Nutritional Requirement.)

  • 공시균의 배양여액으로부터 독성물상을 chloroform 에 전용하여 농축한 후 avicel column chromatography 및 avicel TLC로 정제하였다. 독성물질 생성에 요구되는 영양원은 생육에 필요한 영양첨과 일치하지 않았으며 Valine, asparagine, arginine, 요소, D-glucose, D-mannose, L-rhamnose, D-ribose, D-xylose, D-arabitol, starch 등이 독성물질생성에 요구됨이 인정되었다. 또한 오탄당의 대사과정중 ribose, xylose, fructose, glucose가 대사되는 과정이 본 물질생성의 주된 경로로 추정된다. Vitamin류는 무관함에 비해 Mg가 필수적으로 요구되었다. 물질의 생성은 균의 증식에 비례되었으며 6일간 3$0^{\circ}C$에서 배양했을때 가장 많이 생성되었다. 독성물질 처리후의 잉어 조직은 아가미 신장 췌장등에서 핵진의 굴곡과 더불어 접의 심한 수축과 세포질의 괴사가 현저하였다. 본 물질의 화학식은 $C_{38}$ $H_{66}$ $NO_4$로 추정되며 UV조사시 회청색형광을 나타내었다.

Analysis and Evaluation of Lake Sediment

  • This study was performed to estimate interrelation between characteristics of sediment and nutrient releare from sediment in Dae-cheong lake. For the investigations, sediments were sampled in June and October 1997 at fish farms, embayment, and the main stream of Dae-cheong lake. Items for investigation are as follows; water content, weight loss on ignition(IG), porosity of sediment, Total Kjeldahl Nitrogen(TKN), content of element(H, N, C), nutrient release rate. Water content and porosity were measured to conjecture the physical trait and grain size. And weight loss on ignition was measured to determine the contents of organic substance. For the determination of nutrient release rate, $PO_4-P$ and $NH_4-N$ concentration of interstitial water and overlying water were measured. Release rate of nutrients which has direct influenced upon the water quality were 0.05-8.63mg-$P/m^2{\cdot}day$ and 4.99-36.56mg-$N/m^2{\cdot}day$. And it was found that release rate was measured higher in the 1st sampling than in the 2nd sampling. And for determination of the humus level of sediment, carbon and nitrogen content were measured by using elemental analyzer. Generally, C/N ratio is used to determine humus level of lake sediment. As a result of elemental analysis, C/N ratio was determined in the range of 7.64~11.55, so humus level of Dae-cheong lake sediment was estimated from mesohumic state to oligohumic state.

양액 처리에 따른 수경재배 새싹보리 수량과 폴리페놀 및 무기질 함량 비교분석 (Comparative analysis of hydroponically cultivated barley sprouts yield, polyphenol and mineral content by nutrient solution treatment)

  • 본 연구는 생육 기간에 따른 새싹보리 식물체의 길이, 무게 및 클로로필 함량 분석과 수확량을 통해 양액 처리에 따른 재배 효율성을 평가하고, 생산된 새싹보리의 무기성분 분석과 용매별 추출물의 총 폴리페놀 및 총플라보노이드 함량과 HPLC를 이용한 기능성 물질 분석을 통해 새싹보리의 산업적 이용 가능성을 높이고자 하였다. 새싹보리의 생육에서 무처리구보다 양액 처리구에서 생육속도가 빨랐으며 특히 지상부 길이가 크게 신장하였다. 지상부 길이가 더 증가한 만큼 생체중 또한 증가하였지만 수분 흡수량의 차이보다는 건물중의 증가로 이어졌다. 이는 수확량의 차이로 나타났다. 무처리구와 비교하였을 때 양액 처리구에서 생산량이 유의적으로 증가하였다. 클로로필 함량 분석을 통해 생육 9일차에서 광합성을 통한 독립영양 과정을 확인하였고, 생육 3일차와 6일차에서 양액 처리를 통한 무기성분 투입이 초기 생장에 보조적인 효과가 있다고 판단된다. 새싹보리의 폴리페놀 함량 분석 결과 총 폴리페놀 함량은 무처리구에서 높았지만 총플라보노이드 함량과 luteolin 함량은 양액 처리에 의해 증가하였다. 또한 추출 용매를 물로 사용하였을 때 폴리페놀 추출에 효과적이었다. 따라서 새싹보리 수경재배에서 양액의 투입이 높은 폴리페놀을 가진 새싹보리를 생산하고 생산량 증대에도 효과가 있음을 시사한다.

3가 크롬의 보조제로서의 역할 (The Role of Trivalent Chromium as a Supplement)

  • 인슐린 작용을 강화시키는 LMWCr의 발견과 크롬 결핍과 관련 있는 포도당 불내성(glucose intolerance)이 크롬 투여 시 반응을 보여준 임상적 증명 이후, 크롬은 확실한 필수 영양소로 인식되고 보조제로 사용되고 있다. 시험관 실험에서 고농도의 3가 크롬을 배양 세포에 노출 시, 일부 연구 결과에서 염색체이상 유발과 변이원성 효과가 발견되긴 하였으나 이는 비타민 A, D, 니코틴산, 셀레늄 등과 같은 많은 다른 영양소의 과잉 섭취 시 독성 효과를 나타내는 정도로 해석되며, 시험관 실험 결과들이 발암성의 증거로 해석되지는 않는다. 3가 크롬을 경구 보조제로 투여된 임상 치료에서 환자들은 독성효과를 보이지 않았으며, 혈장 내 크롬 수치가 비 투여된 사람들에 비하여 조금 높았다. 크롬을 TPN 형태로 투여받은 환자들에게서도 우려했던 신체ㆍ정신적 효과와의 상관 관계가 없었으며 신기능부전을 나타낸 경우는 신장 독성을 주는 약들을 함께 복용한 경우로서, 우려되는 부작용은 없는 것으로 인정된다. 임상학적, 실험적으로 사용된 3가 크롬의 용량은 ESADDI 규정된 범위 50∼200 $\mu$g/day에서 전혀 독성을 나타내지 않았으며, 일반적인 사용에 기준이 될 것이다. EPA는 모든 관련 자료들을 검토한 후에 크롬의 참고용량 (RfD) 수치를 70 mg/day로 결론지었으며, 다른 영양소들의 용량과 비교했을 때 대단히 넓은 수치이다. 3가 크롬은 당뇨병, 임신성 당뇨병, 인슐린 내성, 비정상적 지방대사를 개선시키기 위한 영양소로서 가능성을 가진다. 또한 크롬의 보충은 제II형 당뇨병과 다양한 심혈관계 질환을 예방하거나 치료하는데 가장 유용한 것으로 밝혀졌다. 앞으로는 크롬 보조제 사용 시, 일부 연구자들이 3가 크롬 1000 $\mu$g/day의 안전성을 입증한 것과 같이 용량을 늘려 임상실험을 시도한 후 안전과 효능을 증명하는 것이 필요하다.

변형된 활성슬러지공법의 폐수처리에 관한 실험적 연구 (An Experimental Study on Wastewater Treatment by Modified Activated Sludge Process)

  • 본 연구는 활성슬러지공법을 변형시킨 무산소일염기일호기반응조를 사용해서 질산화일탈질소반응 및 인의 방출인초과축적반응을 유도하여 합성페수의 유기물, 질소 및 인을 동시에 제거하였다. 무질소조, 무염기 및 호기호 각각에 대한 반응특성을 조사하기 위해 실험실규모의 반응조가 운영되었으며 MLSS농도를 5000, 4200, 3300, 2600, 1900mg/l로, 변화시켜 COD제거율에 대한 생물학적 동력학계수를 결정하였다. 연구결과, 유기물 및 영양염이 변형된 슬러지공법에 의해 동시에 제거될 수 있음을 알았고, ortho-p 제거효율은 66%∼99%로 높게 나타났다.

Wetting Agent and Phosphorus for Quick Establishment of Kentucky Bluegrass

  • Wetting agent is a substance that reduces the soil water repellency and causes a liquid to spread more easily into the soil. Wetting agents are well known as to improve irrigation efficiency. Phosphorus is one of the 17 essential nutrient elements and promote faster seed germination. The objective of this study was to evaluate wetting agent and P rates for fast germination and establishment of Kentucky bluegrass. Three levels of wetting agent which were 0.23, 0.46, and $0.92ml\;m^{-2}$ were applied and the P treatments were 5, 10, and $15g\;m^{-2}$. The medium and high rate of wetting agent at the high P rate had the greatest turfgrass coverage and took 28 days to reach 50% turfgrass coverage regardless of P application. Based on this study, wetting agent is effective for fast germination and establishment of Kentucky bluegrass if sufficient phosphorus is applied.

모유의 무기질과 면역물질함량에 영향을 미치는 요인에 관한 연구 (Effect of Maternal Factors on the Concentrations of Minerals and Immunological Substance in Breast Milk)

  • The purpose of this study was to evaluate the effect of maternal nutritional status and health behaviors on the concentrations of minerals (Zn, Fe, Ca) and the immunological substances (lactoferrin, sIgA, Iysozyme) in breast milk. Breast milk was collected from 193 healthy Korean women from obstetric clinics and postpartum care centers in Seoul. : 99 colostrum (1 - 5 days postpartum), 33 transitional milk (6 - 10 days postpartum), 61 mature milk (11 - 50 days postpartum). The concentrations of minerals and immunological substance were highest in colostrum and decreased with lactational period. Concentrations of Zn and Fe reduced significantly from colostrum to mature milk, however, Ca concentration stayed constant throughout the lactational period. Contents of lactoferrin, sIgA, and lysozyme were significantly lower in mature milk than in colostrum. Mother's nutritional status, assessed by prepregnancy BMI, had an effect only on colostrum, but not on transition and mature milk. Fe concentration of colostrum was significantly lower in underweight (prepregnancy BMI < 18.5) than in overweight mothers (prepregnancy BMI $\geq$ 23.0). Also lower tendency was observed for sIgA and lysozyme contents, even though the difference was not statistically significant. Pregnancy weight gain had no effect on the breast milk component. Since nutritional factors had some effect on colostrum, the health behaviors of mothers providing colostrum were assessed. The mother's behavior of smoking, drinking, morning sickness, parity, disease, nutrient supplement use had no significant effect on the breast milk component, however, Zn, sIgA, and lysozyme were the somewhat affected components by maternal health behavior.

Purification of Antioxidant substance from the stem bark of Rhus verniciflua

  • The Rhus verniciflua contains alkly(en)-catechol type allergens with a saiurated or unsaturated alkly chain of 15 or 17 carbon atoms. It has been recognized as an extremely active allergen causing skin reactions similar In poison ivy. The allergic contact dermatitis induced by the urushiol is known to be mediated be T lymphocytes whicht specifically recognize the hepten urushiol. Therefore. direct use of this plant as a medicinal purpose might imply a considerable hazard in Korea. In this study, using the established method for the detoxification from the stem bark of Rhus verniciflua, an strong antioxidant substance was isolated and characterized DPPH (diphenypricryl hydrazyl) assay measures hydrogen atom-donating activity and hence provides a measure of free radical scavenging antioxidant activity. DPPH, a purple-colored stable free radical, is reduced to yellow-colored diphenylpicryl hydrazine by antioxidants to deducing agents. Antioxidative effects of the water extract from RV were measured by DPPH assay. Twenty microliters of the extract was added to 1ml of 100mM DPPH solution in ethanol The mixture was shaken and left to stand for 10min at room temperature. The crude water extracts was purified by using HPLC method with a DEAE (anionic type), CN, ODS column. The purified compound remained stable at pH 3.0-6,0, but unstable above pH 6.5. It was stable heat at 10$0^{\circ}C$ for 4 hours, but still had about 80% of residual activity after treatment at 10$0^{\circ}C$ for 5 hours. The elemental composition of the HR-EI mass spectrum at m/z 170.02 was estimated the empirical formula as $C_{7}$ $H_{6}$ $O_{5}$. $C_{10}$ $H_4$ $O_2$N$_1$, $C_{5}$ $H_4$ $O_4$N$_3$, $C_{8}$$H_2O$$_1$N$_4$. In antimicrobial test, no inhibition was observed against Gram-positive and negative bacteria. This compound was stronger than that of commercial antioxidant by DPPH test, such as BHT, BHC at the same concentration (20$\mu$g/ml).ml).

Genome Analysis and Optimization of Caproic Acid Production of Clostridium butyricum GD1-1 Isolated from the Pit Mud of Nongxiangxing Baijiu

  • Caproic acid is a precursor substance for the synthesis of ethyl caproate, the main flavor substance of nongxiangxing baijiu liquor. In this study, Clostridium butyricum GD1-1, a strain with high caproic acid concentration (3.86 g/l), was isolated from the storage pit mud of nongxiangxing baijiu for sequencing and analysis. The strain's genome was 3,840,048 bp in length with 4,050 open reading frames. In addition, virulence factor annotation analysis showed C. butyricum GD1-1 to be safe at the genetic level. However, the annotation results using the Kyoto Encyclopedia of Genes and Genomes Automatic Annotation Server predicted a deficiency in the strain's synthesis of alanine, methionine, and biotin. These results were confirmed by essential nutrient factor validation experiments. Furthermore, the optimized medium conditions for caproic acid concentration by strain GD1-1 were (g/l): glucose 30, NaCl 5, yeast extract 10, peptone 10, beef paste 10, sodium acetate 11, L-cysteine 0.6, biotin 0.004, starch 2, and 2.0% ethanol. The optimized fermentation conditions for caproic acid production by C. butyricum GD1-1 on a single-factor basis were: 5% inoculum volume, 35℃, pH 7, and 90% loading volume. Under optimal conditions, the caproic acid concentration of strain GD1-1 reached 5.42 g/l, which was 1.40 times higher than the initial concentration. C. butyricum GD1-1 could be further used in caproic acid production, NXXB pit mud strengthening and maintenance, and artificial pit mud preparation.

  • Subject : Treatment of acupuncture & moxibustion in Huangdineijing Objectives : Discover the principles and substances of acupuncture & moxibustion treatment of Huangdineijing Methods : The author reviewed the Huangdineijing: Suwen, Lingshu to gain a realistic sense of the facts and the medical book's contents that deal with the practices of acupuncture & moxibustion treatment for clinical praxis. Conclusions : 1. The acupuncture & moxibustion of Huangdineijing can be defined as follows. A treatment for correcting of meridian's drift by proper selection of surgical site, using suitable implementation and appropriate manipulation skill from a correct judgement of meridian's flowing through sensing the pulse for diagnosis when the meridian's flowing which come and go constantly driven by the tidal order of defensive Qi tactually appeared hard or soft caused by diseases, thus bring out the soft & gradual 50 laps circulation of nutrient Qi moderately driven by defensive Qi. 2. Today's acupuncture & moxibustion treatments greatly differ from those of Huangdineijing in that we do not know the substance of that treatment which is correcting of meridian's drift and do not obligatorily carry out feeling of the pulse for diagnosis which is a measure & judgement of meridian's flowing against a preliminary treatment in parallel with treatment so reduce use and power of that treatment. 3. I could form a list of general principle from the substances of acupuncture & moxibustion of Huangdineijing like as the flowing or fate of meridian Qi(tidally going in order of defensive Qi and nutrient Qi), deficiency or excess of Meridian flowing, comparing diagnosis before and after treatment, bring out the soft & gradual Qi. That was imposing "the concept of time" on recognition and treatment to human body.
