Studies on the Differentiation of Skeletal Muscle Cells in uitro : The Phosphorylation and Down Regulation of Protein Kinase C in Myoblasts of Chick Embryos (근세포 분화에 관한 연구 계배의 Myoblasts에 있어서 Protein Kinase C (PKC)의 인 산화작용과 Down Regulation)

  • In the short-term treahent of 12-0-tetradecanoylphorbol-13-acetate (TPA) or platelet-derived growth factor (PDGF), the'Wh and PDGF induced the Protein Kinase C (PKC) activation and migration from the cytoplasm to the peripheral nulcear membrane. And the activated PKC which was directly or indirectly stimulated by TPA or PDGF Phosphorylated many kinds of PKC's targeting proteins and induces various biological responses. Especially, the cytoplasmic PKC was phosphorylated within 1 hr and 10 min by TPA-and PDGF-treahent respectivelv. In the long-term treatment of TPA or PDGF, both of them induced the down-regulation and translocation of PKC in the mvoblasts. The down-regulation of PKC isozyrnes, the pattern of PKC I and ll was similar to the PKC 111 isozpnes in the cytoplasm. But in the nucleolus, the TPA did not induce and down-regulation or the inhibition of the immunoreactivity of PKC III antibody. This investigation indicates that each isozvmes of PKC mal be performed the different effects to the down-regulation of the cytoplasm or nucleolus. And douvn-regulated myoblasts contained low immunoreactivity of PKC antibodies.

Meiosis and Postmeiotic Mitosis in Boletus rubinellus (Boletus rubinellus에서 감수분열 및 감수분열후 유사분열)

  • Meiosis and postmeiotic mitosis in Boletus rubinellus were examined ultrastructurally. Meriosis occurred at the apex of the basidium. A sausage-shaped spindle pole body(SPB) was observed along with the presence of synaptonemal complexes during pachytene and a diglobular SPB was present on late pachytene or diplotene nuclei. During metaphase I, the monoglobular SPB at the spindle pole was surrounded bya membrane and the nuclear enveloope was discontinuous. At anaphase I, the chromosomes became better defined and formed a central spindle. The nucleolus was extruded from the nucleus. During anaphase I, the SPB was excluded from the chromosomal region by a membrane and both poles were fully separated to opposite sides of the basidial wall. In meiosis II, the two nuclei divided synchronously and the spindles were parallel. The spindles were smaller than in meiosis I, while the SPB was approximately the same size as that of the similar stage in meiosis I. During anaphasetelophase II, the SPB was surrounded by a cap of endoplasmic reticulum (ER) that delimited it from the spindle. The postmeiotic interphase nuclei migrated to the mid-region of the basidium before migration to the spores. The SPB at this stage was diglobular. A postmeiotic mitosis occurred within the basidiospore, and the plane of the spindle was obique to the long axis of the spore. The spindle and SPB were smaller than at meiosis I and there were fewer nonchromosomal microtubules. At anaphase, the nucleolus was present inside the nuclear envelope but lateral to the spindle.

  • The ribosomal RNA gene (rDNA) cluster was located in the nucleolus organizer and was genetically determined as one locus. We speculated by using sequence differences in the functional rDNA unit that the segregation time between Chinese and Japanese types of B. mandarina is about three million years ago. The differences of the amount of inserted non-LTR retrotransposons, R1Bm and R2Bm, in rDHA cluster were used for the identification of B.mori strains. (omitted)

Ultrastructural Studies of the Nucleus of Cancer Cells

  • To investigate ultrastructural characteristics of cancer cell nucleus, 29 cases of malignant tumors of 9 different types were examined. The common findings were marked irregularity of nuclear membrane with pronounced infoldings, clumping of heterochromatin along inner nuclear membrane, enlargement and disturbance of configuration and composition of nucleolus, and frequent observations of nuclear bodies and nuclear inclusions. These findings confirm what have been observed by light microscopy, and are also the signes that can be observed in hyperactive cells. Thus, ultrastructural characteristics of cancerous nucleus are the great variability of nuclear size, shape and composition, but none of them appear to be specific.

Differential Subcellular Localization of Ribosomal Protein L7 Paralogs in Saccharomyces cerevisiae

  • In Saccharomyces cerevisiae, ribosomal protein L7, one of the ~46 ribosomal proteins of the 60S subunit, is encoded by paralogous RPL7A and RPL7B genes. The amino acid sequence identity between RPl7a and RPl7b is 97 percent; they differ by only 5 amino acid residues. Interestingly, despite the high sequence homology, Rpl7b is detected in both the cytoplasm and the nucleolus, whereas Rpl7a is detected exclusively in the cytoplasm. A site-directed mutagenesis experiment revealed that the change in the amino acid sequence of Rpl7b does not influence its subcellular localization. In addition, introns of RPL7A and RPL7B did not affect the subcellular localization of Rpl7a and Rpl7b. Remarkably, Rpl7b was detected exclusively in the cytoplasm in rpl7a knockout mutant, and overexpression of Rpl7a resulted in its accumulation in the nucleolus, indicating that the subcellular localization of Rpl7a and Rpl7b is influenced by the intracellular level of Rpl7a. Rpl7b showed a wide range of localization patterns, from exclusively cytoplasmic to exclusively nucleolar, in knockout mutants for some rRNA-processing factors, nuclear pore proteins, and large ribosomal subunit assembly factors. Rpl7a, however, was detected exclusively in the cytoplasm in these mutants. Taken together, these results suggest that although Rpl7a and Rpl7b are paralogous and functionally replaceable with each other, their precise physiological roles may not be identical.

  • The observation using an electron microscope shows that the maturation of the oocyte of African giant snail, Achatina fulica, proceeds over three stages. The oocyte of stage 1 is a small elliptic cell $(220\times400{\mu}m)$ whose light nucleoplasm contains two nucleoli. In its cytoplasm, a number of mitochondria, rough endoplasmic reticula, and ribosomes are found, while yolk granules are not. The nucleus of the oocyte of stage 2 is relatively large in comparison with the volume of cytoplasm, and contains one nucleolus. In the nuclear envelope comprising inner and outer double membrane, there are found a lot of nuclear pores for materials to pass through. A number of mitochondria, Golgi complex and lipid yolk granules appears in the cytoplasm, and proteinous yolk granules begin to form and mature in the vacuoles of various sizes ($0.8\sim3.0{\mu}m$ in diameter). The oocyte of stage 3 has an enlarged nucleolus. Material transportation through nuclear pore is not found any longer. The cytoplasm in this stage is filled with proteinous and lipid yolk granules. The microvilli are developed around the egg plasma membrane.

Papillomatosis on the skin of the wild marbled sole Pleuronectes yokohamae (자연산 문치가자미, Pleuronectes yokohamae 피부의 육아종)

  • Among wild marbled sole, Pleuronectes yokohamae caught in west coastal area, Chungnam, tumor-bearing fish were found. Parasites and pathogenic bacteria were not detected and CPE was not observed. Using the light microscope, the epidermal layer of wild P. yokohamae was significantly thickened compared to the normal skin. The lesion was formed papillary folds. Hypertropical epithelial cells revealed karyolysis, marked nucleolus and cloud swelled cytoplasm. In the epidermal layer of the lesion, X-cell that is characterized by oval and small pale nucleus and prominent nucleolus was observed. Dermal layer had newly formed vessels. The size of mucous cell in the papilloma lesion was significantly increased compared to the normal. In this study, no pathogens were found, so future works for finding cause of the papilloma in the P. yokohamae are needed.

Changes of Morphology and Morphometric Characteristics of the Oocyte during Oogenesis of the Abalone Haliotis discus hannai (북방전복 Haliotis discus hannai 난자형성과정 동안 난모세포의 형태 및 계측형질 변화)

  • The developmental stage of germ cells during oogenesis can be categorized into six stages with histological features: (1) oogonium, (2) previtellogenic oocyte, (3) initial vitellogenic oocyte, (4) early active vitellogenic oocyte, (5) late active vitellogenic oocyte and (6) ripe oocyte. The size of oocyte, nucleus and nucleolus illustrated the increase tendency but size ratio of nucleolus to nucleus was decreased during oogenesis. During oogenesis the stainability in the cytoplasm of oocyte changes from basophilic to eosinophilic in H-E stain. And egg stalk and outer jelly membrane was developed in the oocyte. These histological changes are seemed to be yolk accumulation in the oocyte and preparation process for spawning.

Prognostic Significance of Two Dimensional AgNOR Evaluation in Local Advanced Rectal Cancer Treated with Chemoradiotherapy

  • The prognostic significance of AgNOR proteins in stage II-III rectal cancers treated with chemoradiotherapy was evaluated. Silver staining was applied to the $3{\mu}m$ sections of parafin blocked tissues from 30 rectal cancer patients who received 5-FU based chemoradiotherapy from May 2003 to June 2006. The microscopic displays of the cells were transferred into the computer via a video camera. AgNOR area (nucleolus organizer region area) and nucleus area values were determined as a nucleolus organizer regions area/total nucleus area (NORa/TNa). The mean NORa/TNa value was found to be $9.02{\pm}3.68$. The overall survival and disease free survival in the high NORa/TNa (>9.02) patients were 52.2 months and 39.4 months respectively, as compared to 100.7 months and 98.4 months in the low NORa/TNa (<9.02) cases. (p<0.001 and p<0.001 respectively). In addition, the prognosis in the high NORa/TNa patients was worse than low NORa/TNa patients (p<0.05). In terms of overall survival and disease-free survival, a statistically significant negative correlation was found with the value of NORa/TNa in the correlations tests. Cox regression analyses demostrated that overall survival and disease-free survival were associated with lymph node status (negative or positive) and the NORa/TNa value. We suggest that two-dimensional AgNOR evaluation may be a safe and usable parameter for prognosis and an indicator of cell proliferation instead of AgNOR dots.

Chromosome Analysis by GTG, High-Resolution, and NOR-banding Techniques in the Dog (Cams familaris) (GTG, High-Resolution, Nor-banding에 의한 개의 염색체 분석)

  • None of the numerous published canine idiograms and karyotypes has yet been generally accepted as a standard one because the dog has 76 acrocentric autosomes of similar size and shape. To establish canine banded karyotype from the 22nd chromosome to the 37th chromosome, we analyzed canine chromosomes by GTG, high resolution, and NOR-banding techniques. The GTG and high resolution banding patterns of canine chromosomes corresponded to other reports described previously except for a few chromosomes. While other researchers observed 12 bands, we observed 7 bands in the banding patterns of chromosome 24, 34 and 37. On the other hand, the banding patterns by NOR-banding technique showed that three pairs of autosomes have nucleolus organizer regions at the terminal ends of their long arm, and the Y chromosome has it in its short arm terminal. However, the X chromosome has no nucleolus organizer like other mammals.
