Conformational Change of Human Annexin I by the Binding of $Ca^{2+}$, ATP and cAMP

  • Human annexin I is a member of annexin family of calcium dependent phospholipid binding proteins, which have been implicated in various physiological roles including phospholipase A2(PLA2) inhibition, membrane fusion and calcium channel activity. In this work, the structure of N-terminally truncated human annexin I ({{{{ DELTA }}-annexin I) and its interactions with Ca2+, ATP and cAMP were studied at atomic level by using nuclear magnetic resonance (NMR) spectroscopy. The effect of Ca2+ binding on the structure of {{{{ DELTA }}-annexin I was investigated. The addition of Ca2+ to {{{{ DELTA }}-annexin I caused some changes in 13C NMR spectra. Carbonyl carbon resonances of some histidines were significantly broadened by Ca2+ binding. However, in the case of methionine, phenylalanine, and tyrosin, small changes could be observed. We found that ATP and cAMP bind {{{{ DELTA }}-annexin I, and the binding ratio of ATP to {{{{ DELTA }}-annexin I is 1. These results are well consistent with the report that cAMP and ATP interact with annexin I, and affect the calcium channels formed by annexin I. Because {{{{ DELTA }}-annexin I is a large protein with 35 kDa molecular weight, site-specific (carbonyl-13C) labeling technique was used to study the interaction sites of {{{{ DELTA }}-annexin I with Ca2+. NMR study was focused on the carbonyl carbon resonances of tyrosine, phenylalanine, methionine and histidine residues of {{{{ DELTA }}-annexin I because the number of these amino acids is small in the amino acid sequence of {{{{ DELTA }}-annexin I.

Clonal plant as experimental organisms - DNA mutation rate evaluation in the radiation contaminated area of Fukushima Daiichi NPP accident

  • The Fukushima Daiichi Nuclear Power Plant accident in March 2011 caused severe radioactive contamination in the surrounding environment. Since the accident, much attention has been paid to the biological and genetic consequences of organism inhabiting the contaminated area. The effect of radiation exposure on genetic mutation rates is little known, especially for low doses and in situ conditions. Evaluating DNA mutation by low levels of radiation dose is difficult due to the rare mutation event and lack of sequence information before the accident. In this study, correlations with air dose levels and somatic DNA mutation rates were evaluated using Next Generation Sequencer for the clonal plant, Phyllostachys edulis. This bamboo is known to spread an identical clone throughout Japan, and it has the advantage that we can compare genetic mutation rate among identical clone growing different air dose levels. We collected 94 samples of P. edulis from 14 sites with air dose rates from $0.04{\sim}7.80{\mu}Gy/h$. Their clonal identity was confirmed by analysis using 24 microsatellite markers, and then, sequences among samples were compared by MIG sequence. The sequence data were obtained from 2,718 loci. About ~200,000 bp sequence (80 bp X 2,718 loci) were obtained for each sample, and this corresponds to about 0.01% of the genome sequence of P. edulis. In these sequences, 442 loci showed polymorphism patterns including recent origin mutation, old mutation, and sequence errors. The number of mutations per sample ranged from 0 to 13, and did not correlate with air dose levels. This result indicated that DNA mutations have not accumulated in P. edulis living in the air doses levels less than $10{\mu}Gy/h$. Our study also suggests that mutation rates can be assessed by selecting an appropriate experimental approach and analyzing with next generation sequencer.

협력대응기반 전역네트워크 보안정보공유 시스템 (A Security Monitoring System for Security Information Sharing and Cooperative Countermeasure)

  • 최근 사이버공간의 위협이 사이버전쟁이라고 할 정도로 사실상의 위협으로 현실화되고 있다. 특히, APT 같은 특정기업이나 조직을 노린 표적공격이 치밀하게 계획된 국가 간 사이버전 공격기법의 성향을 띠고 전 세계의 사회기반시설을 통제하려는 위협으로 발전하고 있어 현재 각 ISP 및 기관에서 제공하는 통합보안관리 시스템과 같은 기존 보안장비 및 솔루션으로는 대응의 한계를 가지고 있다. 본 논문에서는 사이버테러의 체계적인 대응을 위하여 국가차원의 전역네트워크 위협 상황 대응을 위한 협력대응기반 보안정보공유 시스템을 설계 및 구현하고 이를 실 ISP 운영센터에 적용한 결과에 대하여 기술한다. 개별 ISP의 이종보안 장비에서 제공되는 다양한 보안 이벤트 통합 인터페이스의 정의 및 수집, 보안정보 전달 프로토콜 정의, 전역 네트워크 차원의 연관성분석과 시각화 인터페이스를 통한 대응 기술 등을 제공하도록 구현하였으며, 실 ISP 운영환경에 적용하여 연동시험을 수행함으로써 운영사이트 적용가능성 또한 확인하였다.

  • 원자력시설의 비상사태 시 주변주민을 보호하기 위한 비상대응행위의 효과를 평가할 필요가 있다. 비상대응행위는 지역특성과 개인 행동특성에 따라 매우 다양하게 나타날 수 있으므로, 비상대응행위를 나타내는 변수들은 특정 값을 갖기보다는 일정구간에 분포되는 값을 갖는다. 대부분의 기존 비상대응모델에서는 계산을 단순화시키기 위하여 가정을 통해 특정 값을 사용한다. 단순화 과정중에 필연적으로 정보의 손실이 발생되어 결과적으로 비상대응 모델은 큰 불확실성을 포함하게 된다. 피지이론은 변수의 불확실성을 계산에 포함시켜 엄밀한 계산을 통해 정보손실을 최소화시키면서 계산결과를 얻어낼 수 있는 수학적인 도구를 제공해 준다. 본 연구에서는 퍼지집합, 퍼지추론, 퍼지관계 등의 이론을 응용하여 원자력시설의 비상사태 시 비상대응효과를 평가할 수 있는 방법을 개발하였다. 개발된 모델의 장점은 언어변수를 이용하여 지역특성을 표현하고 전문가의 의견을 반영하여 비상대응효과를 평가하므로, 단순화 가정중에 유발되는 정보의 손실을 줄일 수 있는데 있다. 비상대응 모델내의 불확실한 변수에 대한 퍼지이론의 응용성을 개선하기 위해서는 전문가의 의견을 반영하여 변수들에 대한 적합한 멤버쉽 함수와 퍼지조건문을 확립할 필요가 있다.

  • 본 연구는 내진설계시 많은 불확실성을 내포하는 지반의 거동특성을 규명하고, 적용대상지반의 확충 및 경제성 제고를 위하여 토사지반의 동적지반특성 평가 및 지반-구조물 거동 특성을 고찰하였다. 예제해석은 토사지반에 원전 containment 구조물이 설치된 경우를 가상하여 지진하중에 대한 지반-구조물 시스템의 거동을 반무한체해석과 유한요소해석으로 분석하였다. 이는 토사지반에 원전이 건설될 경우에 고려해야 할 안정성 및 경제성 분석의 일환으로 수행되었으며, 토사지반의 큰 비선형거동을 정확하게 해석에 반영하기 위한 해석 software와 지반입력 data의 합리적인 평가방안 등을 예제해석을 통하여 분석하였다. 예제해석결과를 종합해 볼 때 토사지반의 동적거동의 정확한 분석을 위하여 비선형 유한요소해석은 Seed & Idriss 모델이, 선형 유한요소해석은 지진하중에 대한 1차원 지반거동시 변형율에서의 동적지반특성을 이용한 방법이, 반무한체해석은 정적하중시 변형율에서의 동적지반특성을 이용한 방법이 가장 합리적으로 동적지반특성을 평가하는 것으로 추천할 수 있다.

Mutation analyses in Korean patients with MELAS (mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes)

  • The mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes (MELAS) is inherited maternally, in which the MTTL1*MELAS 3243 mutation has been most commonly found as a heteroplasmy of A to G point mutation in the $tRNA^{Leu(UUR)}$ gene. The MTTL1*MELAS 3271 mutation is known to be the second common mutation, though clinical features of both mutations are not remarkably different. Recently, a variety of minor mutations have been reported in patients with MELAS. In this study, major efforts have been made to investigate the allele frequency of major three mutations including MTTL1*MELAS 3243, 3252, 3271 in 10 Korean families with MELAS probands. The PCR and subsequent direct sequencing of the PCR product in the regions spanning these three mutation sites were employed to identify the mutation in each proband. All family members have been screened for the presence of these three mutations by PCR-RFLP assay using Apa I, Acc I and Bfr I restriction enzymes. The MTTL1*MELAS 3243 mutation was most commonly found (7 out of 10 families tested) followed by the MTTL1*MELAS 3271 which was identified in 1 out of 10 families. In the remaining 2 families none of three mutations were found, indicating the presence of either nuclear mutation or yet unidentified mitochondrial DNA mutation in these families.

Cimicifuga heracleifolia Extract Induces iNOS Expression via a Nuclear Factor-${\kappa}B$-dependent Pathway in Mouse Peritoneal Macrophages

  • Cimicifuga heracleifolia extract (CHE) was investigated for its effects on the release of nitric oxide (NO) and at the level of inducible nitric oxide synthase (iNOS) gene expression in mouse macrophages. We found that C. heracleifolia elicited a dose-dependent increase in NO production and the level of iNOS mRNA. Since, iNOS transcription has been shown to be under the control of the transcription factor $NF-{\kappa}B$, the effects of CHE on $NF-{\kappa}B$ activation were examined. Transient expression assays with $NF-{\kappa}B$ binding sites linked to the luciferase gene revealed that the increased level of iNOS mRNA, induced by CHE, was mediated by the $NF-{\kappa}B$ transcription factor complex. By using DNA fragments containing the $NF-{\kappa}B$ binding sequence, CHE was shown to activate the protein/DNA binding of $NF-{\kappa}B$ to its cognate site, as measured by electrophoretic mobility shift assay. These results demonstrate that C. heracleifolia stimulates NO production and is able to up-regulate iNOS expression through $NF-{\kappa}B$ transactivation.

Detection and Characterization of PCR-SSCP Markers of the Bovine Lactoferrin Gene for Clinical Mastitis

  • A total of 80 cows, including 40 top mastitis resistant and 40 top mastitis susceptible animals as Group I and Group II, were selected from a population of 520 cows based on clinical mastitis occurrence. PCR-SSCP analysis on four fragments within the 5'region and two fragments of Exons 4,15 of bovine lactoferrin (bLF) revealed that four fragments-P1,P4,E4,E15-had polymorphisms which totally included six base mutations, and only two of them had significant differences in allele frequencies between resistant and susceptible groups, P1 (53.7% vs. 70.0%, p<0.05) and P4 (55.0% vs. 68.8%, p<0.05). Further study on these two promising markers combined with the milk performance traits of cows demonstrated that their selection would result in higher fat percentage (p<0.05), lower Somatic Cell Score (SCS) (p<0.05) and Clinical Mastitis Residuals (CMR) (p<0.01) indicating higher mastitis resistance and lower milk yield (p<0.05). The putative transcription factor binding sites in the 5'region were also studied by using MatInspector 7.2.2 software, and two signal pathways regulating the expression of bLF including the NF-${\kappa}B$ pathway and nuclear hormone receptor pathway were predicted.

흉수의 혈관육종의 세포학적 소견 - 1예 보고 - (Cytologic Features of an Angiosarcoma in Pleural Fluid - A Case Report -)

  • Angiosarcomas are rare tumors, seen most commonly in the skin and soft tissue of head and neck legion. But it has been described in numerous body sites including thyroid, breast, liver, spleen, bone, etc.. Their biological behaviors depend on the microscopic grade, site of origin, and multifocality. We report the unique cytological features of an angiosarcoma in pleural fluid. A 61-year-old woman presented with a 6 month history of dyspnea on exertion and chest pain. Chest computerized tomography(CT) revealed multiple subpleural small nodules in the right lung and widespread all space consolidation and pleural effusion in the left lung. CT of liver revealed multiple small low attenuated lesion. The smears obtained from pleural fluid showed hypocellularity with a hemorrhagic background. The tumor cells were highly pleomorphic oval or spindle in shape and presented singly, in loose groups, in knitted syncytial aggregates, and in acinar pattern. Their nuclei had vesicular chromatin with delineated, thick nuclear membranes and occasionally a large eosinophilic, prominent nucleolus. The cytoplasm was plump, thin or protected in spindly fashion. Almost ail tumor cells showed variable sized intracytoplasmic vacuoles and their nuclei were sometimes crescentic by a huge vacuole. Occasional binucleated tumor cells and mitotic figures were present. Cellular debris and streaky materials were identified. Needle biopsy specimen from the pleura revealed anastomosing slit-like spaces lined by pleomorphic tumor cells. The tumor cells showed a strong reactivity for CD31 and vimentin and focal weak reactivity for factor VIII-related antigen.

온배수 확산 평가를 위한 열적외선 위성관측 비교 (The Comparison of Thermal Infrared Satellite Observation for Plume Assessment of Thermal Discharge)

  • 해수 표층 수온은 원자력발전소의 온배수 영향을 조사하기 위해서 위성원격탐사에 의해 관측되는 가장 중요한 정보들 중 하나이다. 하지만 Landsat 7 위성과 Landsat 8 위성의 열적외선 센서로부터 추출한 표층수온과 실측치를 비교한 연구는 부족하다. Landsat 8 위성은 표층수온을 추출하기 위해 열적외선 센서에 두 개의 분리된 밴드를 가지고 있지만, Landsat 7은 한 개의 밴드를 사용하고 있다. 그럼에도 불구하고 본 연구에서는 Landsat 7 ETM+센서가 Landsat 8 TIRS 보다 표층수온의 보정에 유용하다는 것을 제시하였다. 본 연구에서는 Landsat 114-36 지역의 15개 위성자료를 가지고 ENVI와 IDL을 이용한 표층수온 알고리즘을 처리하였다. 국립해양조사원으로부터 수집한 표층수온 실측자료와 위성에서 추출한 표층수온을 비교하였고, 위성관측 시계열 자료와 측정지점의 실측자료를 통해 정확도를 비교하였다.
