Single Crossover-Mediated Markerless Genome Engineering in Clostridium acetobutylicum

  • A novel genome-engineering tool in Clostridium acetobutylicum was developed based on single-crossover homologous recombination. A small-sized non-replicable plasmid, pHKO1, was designed for efficient integration into the C. acetobutylicum genome. The integrated pHKO1 plasmid backbone, which included an antibiotic resistance gene, can be excised in vivo by Flp recombinase, leaving a single flippase recognition target sequence in the middle of the targeted gene. Since the pSHL-FLP plasmid, the carrier of the Flp recombinase gene, employed the segregationally unstable pAMβ1 replicon, the plasmid was rapidly cured from the mutant C. acetobutylicum. Consequently, our method makes it easier to engineer C. acetobutylicum.

Real-time Measurement of Precision Displacement using Fiber Optic EFPI Sensor (광섬유 EFPI 센서를 이용한 실시간 고정밀 변위 측정)

  • Precision displacement of less than a few nm resolution was measured in real-time using fiber optic EFPI sensor. The novel method for real-time processing of analyzing EFPI output signal was developed and verified. Linearity in the mean values of interferometric light intensity among adjacent fringes was shown, and the sinusoidal approximation algorithm that estimates past and coming fringe values was verified through the linearity. Real-time signal processing program was developed, and the intensity signal of the EFPI sensor was transformed to the phase shift with this program. The resolution below 0.4 ~ 10 nm in the displacement range of $0 ~ 300\mu\textrm{m}$ was obtained by reducing the photodetector noise using low-pass filter and signal averaging. The nano-translation stage with a Piezo-electric actuator and the EFPI sensor system was designed and tested. This stage successfully reached to the desired destination in $15\mu\textrm{m}$ range within 1 nm accuracy.

Nonlinear large deflection buckling analysis of compression rod with different moduli

  • Many novel materials exhibit a property of different elastic moduli in tension and compression. One such material is graphene, a wonder material, which has the highest strength yet measured. Investigations on buckling problems for structures with different moduli are scarce. To address this new problem, firstly, the nondimensional expression of the relation between offset of neutral axis and deflection curve is derived based on the phased integration method, and then using the energy method, load-deflection relation of the rod is determined; Secondly, based on the improved constitutive model for different moduli, large deformation finite element formulations are developed and combined with the arc-length method, finite element iterative program for rods with different moduli is established to obtain buckling critical loads; Thirdly, material mechanical properties tests of graphite, which is the raw material of graphene, are performed to measure the tensile and compressive elastic moduli, moreover, buckling tests are also conducted to investigate the buckling behavior of this kind of graphite rod. By comparing the calculation results of the energy method and finite element method with those of laboratory tests, the analytical model and finite element numerical model are demonstrated to be accurate and reliable. The results show that it may lead to unsafe results if the classic theory was still adopted to determine the buckling loads of those rods composed of a material having different moduli. The proposed models could provide a novel approach for further investigation of non-linear mechanical behavior for other structures with different moduli.

  • In this paper, we newly propose a novel control strategy of a three joints-robot finger for the purpose of artificial hands. The robot finger is specifically modeled by using a 3D CAD program (CATIA), considering human fingers, and then the proposed control method is verified through the dynamic simulation tool (Simulink and Recurdyn R2). Each slider is individually controlled to be located at the optimal positions where the maximal joint torque can be generated. To prove the effectiveness of the proposed control method, we devise two cases for the reference position of sliders. By comparing the control performance of two cases, the validity of the proposed control method will be verified.

  • In this study, a novel method for sweeping automatic linearization of wavelength swept laser is proposed. Through the test performed on the implemented laser, the linear sweeping is held up well with a 97% decrease in nonlinearity, and 60 nm sweeping range, 1 kHz sweeping frequency, and 8.8 mW average optical power were obtained. The proposed method uses fiber Bragg grating array, optical-electronic conversion circuit, FPGA embedded module, and a LabVIEW program to generate new compensated wave patterns which were applied to the fiber Fabry-Perot tunable filter. Linear sweeping can reduce the cumbersome and time-consuming recalibration process required for nonlinear sweeping. Additionally, the proposed method provides more accurate measurement results for the structure safety monitoring system.

  • Sometimes, we need or try to figure out somebody's thoughts from his or her behaviors such as eye movement, facial expression, gestures, and motions. In safety-critical and complex systems such as nuclear power plants, the inference of operators' thoughts (understanding or diagnosis of a current situation) might provide a lot of opportunities for useful applications, such as development of an improved operator training program, a new type of operator support system, and human performance measures for human factor validation. In this experimental study, a novel method for inference of an operator's thoughts from his or her eye movement data is proposed and evaluated with a nuclear power plant simulator. In the experiments, about 80% of operators' thoughts can be inferred correctly using the proposed method.

A Novel Method of Improving Cache Hit-rate in Hadoop MapReduce using SSD Cache

  • The MapReduce Program of Hadoop Distributed File System operates on any unspecified nodes due to distributed-parallel process and block replicate for data stability. Since it is difficult to guarantee the cache locality when a Solid State Drive is used as a cache in hadoop, cache hit-rate is decreased. In this paper, we suggest a method to improve cache hit rate by pre-loading the input data of the MapReduce onto the SSD cache. To perform this method, we estimated the blocks that are used on each node by using capacity scheduler and block metadata. Eventually we could increase the performance of SSD cache by loading the blocks onto SSD cache before the Map Task run.

  • To enhance catalyst activity of the palladium (Pd) towards oxygen reduction reaction (ORR), iridium (Ir) and yttrium (Y) were alloyed by polyol method. Due to the low reduction potential of Y, it is hard to reduce Y ion completely by polyol method. In XPS spectra, the binding energy of the Pd is shifted to a lower value, which indicates the d-electron of Pd is filled by the electron from the Y. And other phases of Y are observed by the XPS. Among the catalysts, the $Pd_4IrY_{0.1}/C$ showed the best activity towards ORR, which indicates the metallic Y is effective for improving the catalytic activity. Thus, for further enhancing ORR activity, the novel method for complete reduction of Y is needed.

KARE Genomewide Association Study of Blood Pressure Using Imputed SNPs

  • The imputation of untyped SNPs enables researchers to validate association findings across SNP arrays and also enables them to test a large number of SNPs to reveal the fine structure of the association peak, facilitating interpretation of the results and the location of causal polymorphisms. In this study, we applied the imputation method to a genomewide association study and recapitulated the previously associated gene loci of blood pressure traits in Korean cohorts. A total of 1,827,004 SNPs were imputed by the IMPUTE program, and we conducted a genomewide association study for systolic and diastolic blood pressure. While no SNPs passed the Bonferroni correction p-value (p=$2.74{\times}10^{-8}$ for 1,827,004 SNPs), 12 novel loci for systolic blood pressure and 16 novel loci for diastolic blood pressure were detected by imputed SNPs, with $10^{-5}$ < p-value < $10^{-4}$. Moreover, 7 regions (ATP2B1, 10p15.1, ARHGEF12, ALX4, LIPC, 7q31.1, and TCF7L2) out of 14 genetic loci that were previously reported revealed that the imputed SNPs had lower p-values than those of genotyped SNPs. Moreover, a nonsynonymous SNP in the CSMD1 gene, one of the 14 genes, was found to be associated with systolic blood pressure (p<0.05). These results suggest that the imputation method can facilitate the discovery of novel SNPs as well as enhance the fine structure of the association peak in the loci.
