Comparative evaluation of NovaMin desensitizer and Gluma desensitizer on dentinal tubule occlusion: a scanning electron microscopic study

  • Purpose: In this study, the effect of calcium sodium phosphosilicate (NovaMin) desensitizing agent, which is a powder-based system, and hydroxyethyl methacrylate and glutaraldehyde (Gluma desensitizer), which is liquid-based system, on dentinal tubule occlusion was analyzed by scanning electron microscope. The effects of the above two along with one control group were compared to determine the more effective method of sealing the dentinal tubules after initial application. Methods: Twenty specimens were allocated to each of 3 groups: Control, Gluma desensitizer, and NovaMin. Two additional samples were also prepared and treated with Gluma and NovaMin; these samples were longitudinally fractured. The specimens were prepared from extracted sound human premolars and were stored in 10% formalin at room temperature. The teeth were cleaned of gross debris and then sectioned to provide one to two dentin specimens. The dentin specimens were etched with 6% citric acid for 2 minutes and rinsed in distilled water. Control discs were dried, and the test discs were treated with the desensitizing agents as per the manufacturer's instructions. The discs as well as longitudinal sections were later analyzed under the scanning electron microscope. The proportions of completely occluded, partially occluded, and open tubules within each group were calculated. The ratios of completely and partially occluded tubules to the total tubules for all the groups was determined, and the data was statistically analyzed using nonparametric tests and statistical significance was calculated. Results: NovaMin showed more completely occluded tubules ($0.545{\pm}0.051$) while Gluma desensitizer showed more partially occluded tubules ($0.532{\pm}0.075$). The differences among all the groups were statistically significant ($P{\leq}0.05$). Conclusion: Both materials were effective in occluding dentinal tubules but NovaMin appeared more promising in occluding tubules completely after initial application.

High-Performance Liquid Chromatographic Analysis of Chrysin Derivatives on A $Nova-Pak^{\circledR}C_{18}$ Column

  • A high-performance liquid chromatographic method has been developed for the separation and quantification of chrysin and synthetic chrysin derivatives (12 chrysin alkyl and 7 chrysin acyl derivatives). The chromatography was performed using a $Nova-Pak^{\circledR}C_{18}$ column. A RP-HPLC was performed by using a binary mixture (MeOH-10 mM H$_3$PO$_4$) as a mobile phase, and the column temperature was maintained at room temperature. A flow rate was 1.0 ml/min, and the effluent was monitored at a wavelenth of 280 nm. The retention times for chrysin acyl and alkyl derivatives were within 10 minutes and 20 minutes, respectively. The absolute recovery of samples were all over 96%. The detection limits were 0.1~18 ng at S/N = 3 ratio.

Cold Tolerance of Native and Introduced Evergreen Rhododendron Species According to Morphological and Physiological Changes (국내 자생종 및 도입종 만병초의 내한성과 관련된 형태 및 생리적변화)

  • Cold tolerance of the native Rhododendron species which are on the verge of extinction in Korean nature were compared with the introduced species and its mechanism were studied physiologically with the investigation of the leaf angle, leaf curling, and photosynthetic activity. The degree of cold tolerance measured with the leaf burning after winter season was higher in the native species, Rhododendron brachycarpum and Rhododendron brachycarpum var. roseum than all the introduced species. 'Nova Zembla', an introduced species, showed high sensitivity to the low temperature. Changes in leaf angle by the low temperature were bigger in 2 native species and 'Parker's Pink' than the other introduced species and small comparatively in 'Nova Zembla' and 'Cunningham's White' cultivar. Leaf curling also occurred strongly in 2 native species by the low temperature. While, it was comparatively little and mild in the other introduced species. Therefore these results suggested that the leaf movement such as leaf angle change and curling adapted to the low temperature is positively related to the cold tolerance of 2 native species. By the way, such relationship is not explainable in the cold-sensitive 'Parker's Pink' cultivar showing comparatively stronger leaf movement. Photosynthetic activity measured before the winter season was high in the cold-tolerant R. brachycarpum and its recovery after winter season was faster in the 2 native species and the introduced 'Cynosure' cultivar than the other introduced species. They were the lowest in the most cold-sensitive 'Nova Zembla'. This phenomena occurred similarly even in the stomatal conductivity, suggesting that the movement of water from the roots to the leaves is better and then the leaf burning after winter season become small in the cold-tolerant species. The recovery of photosynthetic activity and stomatal conductivity was comparatively slower in the cold-sensitive 'Parker's Pink'. From the above results, leaf behavior adapted to the low temperature during the winter season and water movement to the leaves are related collectively to the cold tolerance represented as the leaf burning in the Rhododendron species is suggested.

Biology of Three Species of the Genus Tipula (Diptera: Tipulidae) in Korea (한국산 Tipula속(파리목 : 각다귀과) 3종의 생태)

  • This study was conducted to investigate the biology of three species, Tipula latemarginata Alexander, T. nova Walker and T. aino Alexander, of the Genus Tipula in Korea. Field surveys for the biology of the species were carried out at the three sites of Neaseong Stream area in Bongwha County from January 2001 to December 2005. Also many individuals of the species were reared in laboratory to investigate the biology. The larval habitats of the species were streams, rivers, paddy fields, damp earth or leaky revetments. The Tipula larvae were herbivorous. Newly-hatched first instar larvae fed on soft algae, and the larvae gradually fed on leaves, stems and roots of a variety of plants or hard leaf litters, as they grew. During the molting of the larvae their body kept moving from left to right and up and down. Molting usually took not more than 2 hours, and about four hours after molting, the larvae started to eat heavily. The Tipula larvae in aquatic places moved to drier land for pupation and went through short prepupal stage lasting 1-2 days for pupation at fourth instar larval stage. When emerging, the Tipula pupae placed their head and thorax on the earth, but the other parts in the earth. Emergence from the pupal case required about 20 min. to one hour. Mating of Tipula adults took place within 5-7min. after emergence and the duration of mating was about 40 min. The female adults of the species laid eggs by walking with 3 pairs of legs over the damp earth or algal beds. Their body was positioned vertically on the ground with their wings spread $120^{\circ}$ and legs landed on the surface. The oviposition usually took place from one day to 4 days after emergence and the number of eggs carried by female adults were an average of 501-760 per individual. Tipula adults didn't normally feed, and drank water only occasionally. For a few days after emergence, the adults reared in the laboratory rarely drank water. As they neared to death, however, they frequently drank water. The longevity of adults reared in the laboratory with only water during the summer was ca. 4-9 days and males usually survived a little longer than females. The longevity of T. nova was increased 3 times or more as much by feeding them 3% sugar water. Male adults of T. latemarginata outnumbered female adults by 2.6 : 1 in the fields.

  • A high-performance liquid chromatographic method was developed to determine the quantities of curcumin and glycyrrhizin in two different oriental herbal preparations of Kamijadowhan (KMD. NKMD). Two compounds were separated in less than 10 min with a Nova-Pak $C_{18}$ column ($3.9{\times}150$ mm ($5{\mu}$ particle size) by linear gradient elution using 0.03% (v/v) phosphoric acid-acetonitrile (60:40. v/v% at 0 min; 40:60 v/v% at 6 mini as the mobile phase at a flow-rate of 0.8 ml $min^{-1}$. A Numerical Simulation Study of Strong Wind Events at Jangbogo Station, Antarctica (남극 장보고기지 주변 강풍사례 모의 연구)

  • Jangbogo station is located in Terra Nova Bay over the East Antarctica, which is often affected by individual storms moving along nearby storm tracks and a katabatic flow from the continental interior towards the coast. A numerical simulation for two strong wind events of maximum instantaneous wind speed ($41.17m\;s^{-1}$) and daily mean wind speed ($23.92m\;s^{-1}$) at Jangbogo station are conducted using the polar-optimized version of Weather Research and Forecasting model (Polar WRF). Verifying model results from 3 km grid resolution simulation against AWS observation at Jangbogo station, the case of maximum instantaneous wind speed is relatively simulated well with high skill in wind with a bias of $-3.3m\;s^{-1}$ and standard deviation of $5.4m\;s^{-1}$. The case of maximum daily mean wind speed showed comparatively lower accuracy for the simulation of wind speed with a bias of -7.0 m/s and standard deviation of $8.6m\;s^{-1}$. From the analysis, it is revealed that the each case has different origins for strong wind. The highest maximum instantaneous wind case is caused by the approach of the strong synoptic low pressure system moving toward Terra Nova Bay from North and the other daily wind maximum speed case is mainly caused by the katabatic flow from the interiors of Terra Nova Bay towards the coast. Our evaluation suggests that the Polar WRF can be used as a useful dynamic downscaling tool for the simulation and investigation of high wind events at Jangbogo station. However, additional efforts in utilizing the high resolution terrain is required to reduce the simulation error of high wind mainly caused by katabatic flow, which is received a lot of influence of the surrounding terrain.

  • There are several factors causing re-work in CMP process such as improper polish time calculation by operator. removal rate decline of the polisher, unstable in-suit pad conditioning, slurry supply module problem and wafer carrier rotation inconsistancy. And conclusively those fundimental reason for the re-work rate increasement is mainly from the cycle time delay between wafer polish and post measurement. Therefore, Wafer thickness measurement in wet condition could be able to remove those improper process conditions which may happen during the process in comparison with the conventional dried wafer measurement system and it can be able to reduce the CMP process cycle time. CMP scrap reduction by overpolish, re-work rate reduction, thickness control efficiency also can be easily achieved. CMP Equipment manufacturer also trying to develop integrated system which has multi-head & platen, cleaner, pre & post thickness measure and even control the polish time from the calculated removal rate of each polishing head by software. CMP re-work problem such as over & under polish by target thickness may result in the cycle time delay. By reducing those inefficient factors during the process and establish of the automatic process control, CLC system need to be adopted to maximize the process performance. Wafer to Wafer Polish Time Feed Back Control by measuring the wafer right after the polish shorten the polish time calculation for the next wafer and it lead to the perfact Post CMP target thickness control capability. By Monitoring all of the processed the wafer, CMP process will also be stabilize itself.

Reverse-Phase High Performance Liquid Chromatographic Determination of Amino Acids after Precolumn Derivatization with 1-Dimethyl-aminonaphthalene-5-sulfonyl chloride (Dansyl 유도체화와 역상 고성능액체크로마토그래피에 의한 아미노산의 정량)

  • This experiment was carried out to determine the separation condition of 1-dimethylaminonaphthalene-5-sulfony(Dansyl) derivatives of amino acids by reverse-phase high performance liquid chromatography with Nova-Pak C18 column. Determined solvent system was solvent A(200mA phosphate buffer pH 6.8 15%, acetonitrile 11%, water 74%) and solvent B(acetonitrile 65%, methanol 28%, water 7%). Linear gradient of solvent B was applied from 12% to 80% for 50min. Complete separation of 20 amino acids including asparagine and glutamine which constitute protein was achieved within 50min. As the detection limit was the range of picomole, the resolution power was excellent. Reproducibility of the retention time was less than mean $\pm$0.05min. According to the above optimum chromatographic conditions, the amino acid composition of some food and human blood was examined. The most affluent amino acid was alanine in human blood, aspartic acid and glutamic acid in soy sauce, alanine and threonine in soy milk and proline in milk and yoghurt.

Determination of Ceftiofur in Bovine Serum by liquid Chromatography-Electrospray Mass Spectrometry (액체크로마토그래프/질량분석기를 이용한 소의 혈청 중 Ceftiofur의 분석)

  • This study was aimed to develop a more rapid, simple and sensitive method to determine ceftiofur in bovine serum using LC/MS with electrospray interface. Separation was achieved on the Nova-Pak $C_{18}$ reverse phase column. The mobile phase consisted of 0.1% acetic add in water (A) and acetonitrile (B) and gradiently flowed at the rate of 0.4 mL/min. As a result of analysis of blank muscle samples, matrix interference was not shown. Limit of detection and limit of quantitaion was 5 ng/g and 20 ng/g, respectively. The values of precision and recovery satisfied the guideline of NVRQS. The precision and recovery developed in this method are suitable and sensitive to determine the concentration of ceftiofur in the bovine serum. These results could be applied for the confirmation and quantification in the biofluid.

  • Using a reversed-phase high performance liquid chromatography, the separation of 20(S)-, 20(R)-prosapogenin stereo-isomers of ginsenoside-$Rg_2$ and of ginsenoside-$Rg_3$ in ginseng saponins has been carried out with binary solvent system. The optimum conditions for the isomer separation are as following: Nova-$Pak^{(R)}C_{18}$ (Waters, $3.9{\times}150mm$) column, $CH_3CN/CH_3CN$ (100:8, v/v) binary solvent system and the flow rate was 1.7 mL/min. The stereoisomers were separated with change of the mixture ratio of the solvent system, the solvent elution by gradient program, and then detected at 203 nm of UV detector. The simultaneous separation of mixture that were the $Rg_2$, $Rg_3$ isomers was easily performed in nonpolar solvent for $Rg_2$, polar solvent for $Rg_3$ at the same optimum conditions.
