REGULATION OF RAT ADRENAL MEDULLARY PHENYLETHANOL AMINE N-METHYLTRANSFERASE

  • Neural regulation of phenylethanolamine N-meth-yltransferase (PNMT) was studied with reserpine as a neuronal agent in rat adrenal medulla. The enzyme activity assay and northern blot analysis were performed to determine whether the induction of PNMT activity after reserpine treatment was associated with elevation of mRNA coding for PNMT. The i.p. administration of reserpine (2.5 mg/kg) on alternate days fot 4 injections to rats brought about 30% increase of adrenal medullary PNMT activity and approximately 60% stimulation of the PNMT mRNA level in rat adrenal gland. A dose of 10 mg/kg of reserpine was chosen to perform optimum induction of PNMT activity in the rat adrenal gland based on the results of dose response curve of reserpine. Time course reserpine (10 mg/kg) effects on the rat adrenal medullary PNMT were as follows: 1. Peripheral PNMT activity reached maximum level after 7 days of drug treatment on alternate days. 2. Trans-synaptic stimulation by reserpine increased pretranslational activity of rat adrenal PNMT, but not translational activity. 3. Immunotitration of PNMT molecule after reserpine treatment indicated that reserpine produced an enzyme with greater antibody affinity than endogenous molecule in the rat adrenal gland.

Variations of Gonadotropin Subunits mRNA Levels at Different Stages of Ovarian Development in Masu Salmon, Oncorhynchus masou

  • The variations of gene expression and pituitary contents of GTH subunits during the ovarian development of masu salmon, Oncorhynchus masou, were investigated. The pituitary GTHs contents was measured by radioimmunoassays (RIAs) using purified GTH subunits and their antibodies. Pituitary contents of GTH $I\beta$ gradually increased from April through August, and reached the maximum in October. On the other hand, pituitary contents of GTH $II\beta$ remained low until August, but they rapidly increased in October. Total RNAs were prepared from pooled pituitaries and the GTH subunits mRNA in pituitaries was quantified by Northern blot hybridization using masu salmon cDNA probes for each GTH subunit. GTH $I\beta$ mRNA level increased with the progression of ovarian maturity. However, GTH $II\beta$ mRNA was detected only at a more advanced stage, and were extremly high at ovulation. A high levels for GTH a mRNA was detected only at ovulation stage. The synchronous increase in pituitary contents and mRNA levels suggested that ovarian maturity in masu salmon was regulated by both GTH I and GTH II.

Phylogeography of the Lessonia variegata species complex (Phaeophyceae, Laminariales) in New Zealand

  • A goal of phylogeography is to relate phylogenetic patterns to potential historic and contemporary geographic isolating events. Genetic breaks found in phylogeographic studies may denote boundaries between more generally applicable biogeographic regions. We investigated the distribution of Lessonia variegata, an important habitat forming alga, found on most rocky shores around New Zealand’s main islands, plus related species from surrounding waters. L. variegata has been shown to consist of four distinct cryptic species. Our aim was to compare the distribution of L. variegata with proposed bioregions; and to develop phylogeographic hypotheses to explain its present day distribution. Both a mitochondrial (atp8-sp) and plastid (RuBisCo spacer) marker, with different mutation rates, were used to gain information of the phylogenetic history of Lessonia. The data revealed high phylogeographic structuring and reciprocal endemism for all L. variegata cryptic species. One species (L. variegata / N) is confined to the northern part of the North Island of New Zealand; L. variegata / W is found at the southeast of the North Island and the northern South Island; L. variegata / K is endemic to the northeast South Island; and L. variegata / S is restricted to the southern part of the South Island. No overlapping areas of L. variegata species distribution were found. The data showed that genetic breaks in Lessonia do mostly correlate to bioregions, and highlight the importance of Cape Campbell at the northeast of the South Island and East Cape in the North Island, well known phylogeographic breaks, as a barrier between adjacent species.

Healthcare Systems and COVID-19 Mortality in Selected OECD Countries: A Panel Quantile Regression Analysis

  • Objectives: The pandemic caused by coronavirus disease 2019 (COVID-19) has exerted an unprecedented impact on the health of populations worldwide. However, the adverse health consequences of the pandemic in terms of infection and mortality rates have varied across countries. In this study, we investigate whether COVID-19 mortality rates across a group of developed nations are associated with characteristics of their healthcare systems, beyond the differential policy responses in those countries. Methods: To achieve the study objective, we distinguished healthcare systems based on the extent of healthcare decommodification. Using available daily data from 2020, 2021, and 2022, we applied quantile regression with non-additive fixed effects to estimate mortality rates across quantiles. Our analysis began prior to vaccine development (in 2020) and continued after the vaccines were introduced (throughout 2021 and part of 2022). Results: The findings indicate that higher testing rates, coupled with more stringent containment and public health measures, had a significant negative impact on the death rate in both pre-vaccination and post-vaccination models. The data from the post-vaccination model demonstrate that higher vaccination rates were associated with significant decreases in fatalities. Additionally, our research indicates that countries with healthcare systems characterized by high and medium levels of decommodification experienced lower mortality rates than those with healthcare systems involving low decommodification. Conclusions: The results of this study indicate that stronger public health infrastructure and more inclusive social protections have mitigated the severity of the pandemic's adverse health impacts, more so than emergency containment measures and social restrictions.

Two New Species of Hydropsychidae (Insecta: Trichoptera) from the Philippines

  • Two new Hydropsychidae (Insecta: Trichoptera) species from the Philippines are described. Hydropsyche philippinensis, n. sp., differs from other congeners in the structure of the phallic apparatus. Diplectrona lagunensis, n. sp., can be distinguished from other congeners by its distinct white band pattern in the forewings and by the structure of the phallus. Both new species were collected from a high mountain stream in the northern part of the Philippines. Paratype materials were barcoded to make the sequences readily available in the GenBank.

Effect of Hemorrhage on mRNA Expressions of Renin, Angiotensinogen and $AT_1$ Receptors in Rat Central and Peripheral Tissues

  • In an attempt to investigate whether hemorrhage affects the gene expression of the renin-angioteusin system (RAS) components in the brain and peripheral angiotensin-generating tissues, changes in mRNA levels of the RAS components in response to hemorrhage were measured in conscious unrestrained rats. Wistar rats were bled at a rate of 3 ml/kg/min for 5 min, and then decapitated 7 h after hemorrhage. Levels of mRNA for renin, angiotensinogen and angiotensin $II-AT_1$ receptor subtypes ($AT_{1A}$ and $AT_{1B}$) were determined with the methods of northern blot and reverse transcriptase-polymerase chain reaction (RT-PCR). Hemorrhage produced a profound hypotension with tachycardia, but blood pressure and heart rate recovered close to the basal level at 7 h. Plasma and renal renin levels were significantly increased at 7 h. Hemorrhage induced rapid upregulation of gene expression of both $AT_{1A}$ and $AT_{1B}$ receptor subtypes in the brainstem and hypothalamus, downregulation of them in the adrenal gland and liver. However, renin mRNA level increased in the brainstem, decreased in the liver, but was not changed in the hypothalamus, kidney and adrenals after hemorrhage. Angiotensinogen mRNA level was not significantly changed in any of the tissue except a slight increase in the liver. The kidney and liver did not show any significant change in gene expression of the RAS components. These results suggest that gene expression of the RAS in central and peripheral tissues are, at least in part, under independent control and the local RAS in each organ plays specific physiologic role.

  • This study explores the Subseasonal-to-Seasonal (S2S) prediction skills of the Northern Hemisphere mid-latitude geopotential height in the Global Seasonal forecasting model version 5 (GloSea5) hindcast experiment. The prediction skills are quantitatively verified for the period of 1991~2010 by computing the Anomaly Correlation Coefficient (ACC) and Mean Square Skill Score (MSSS). GloSea5 model shows a higher prediction skill in winter than in summer at most levels regardless of verification methods. Quantitatively, the prediction limit diagnosed with ACC skill of 500 hPa geopotential height, averaged over $30^{\circ}N{\sim}90^{\circ}N$, is 11.0 days in winter, but only 9.1 days in summer. These prediction limits are primarily set by the planetary-scale eddy phase errors. The stratospheric prediction skills are typically higher than the tropospheric skills except in the summer upper-stratosphere where prediction skills are substantially lower than upper-troposphere. The lack of the summer upper-stratospheric prediction skill is caused by zonal mean error, perhaps strongly related to model mean bias in the stratosphere.

Perspectives of Korean Modernity from the 18th Century to the Present: Intellectual Struggles for Koreanity in the Age of Globalization

  • For the effective study of Korean modernization from the 18th century to the present, three areas have been investigated in my paper: the age of dawn in recognizing the necessity of modernism, the era of experimentation from recognition to practice, and the development of modernism in literature: from the 1930s to the present. Through whole process of discussing those matters, Koreanity- identifying itself to be Korean - has been emphasized. While the so-called traditional values confronted with the whole turmoil of socio-political demolitions in the name of modernization, westernization, and culturalization, Korean intellectuals tried to emphasize how important it was to keep Korean identities, namely the Koreanity. Such examples can be seen in the activities of Northern School and Moderate School. Though Koreans had to have a short hair cut in contradiction with their traditional morality to be modernized/westernized/cultivated, it was a turning point for them to take a step toward the international world. During the period of Korean modernization through the impact of Western world, Korean language-hangul- has been cultivated to the highest level in comparison with two foreign languages: Japanese and English. Those Korean linguists who were familiar with these two languages made Korean grammar systematic and they understood the importance of preserving Koreanity in the course of pursuing modem western society. In this sense, Korean modernism is related to the cultural glocalism(globalism+ localism), not to the cultural globalism. Through the help of socio-political modernization, Korean literature in modernism has been full bloomed in the early years of 1930s. One of the leading poets was Sang Lee whose poetic heritage is inherited by those groups of 1950s and I 960s. Among many others, Chunsu Kim and Sunghun Lee were the main figures in realizing the fact the poetry is written in Korean which they considered the body, the soul, and the mother land.

Susceptibility Loci Associations with Prostate Cancer Risk in Northern Chinese Men

  • Background: KLK3 gene products, like human prostate-specific antigen (PSA), are important biomarkers in the clinical diagnosis of prostate cancer (PCa). G protein-coupled receptor RFX6, C2orf43 and FOXP4 signaling plays important roles in the development of PCa. However, associations of these genes with PCa in northern Chinese men remain to be detailed. This study aimed to investigate their impact on occurrence and level of malignancy. Methods: All subjects were from Beijing and Tianjin, including 266 cases with prostate cancer and 288 normal individuals as controls. We evaluated associations between clinical covariates (age at diagnosis, prostate specific antigen, Gleason score, tumor stage and aggressive) and 6 candidate PCa risk loci, genotyped by PCR- high resolution melting curve and sequencing methods. Results: Case-control analysis of allelic frequency of PCa associated with PCa showed that one of the 6 candidate risk loci, rs339331 in the RFX6 gene, was associated with reduced risk of prostate cancer (odds ratio (OR) = 0.73, 95% confidence interval (CI) =0.57-0.94, P = 0.013) in northern Chinese men. In addition, subjects with CX (CC+TC) genotypes had a decreased risk for prostrate cancer compared to those carrying the TT homozygote (OR =0.64, 95% CI = 0.45- 0.90, P = 0.008). The TT genotype of 13q22 (rs9600079, T) was associated with tumor stage (P=0.044, OR=2.34, 95% CI=0.94-5.87). Other SNPs were not significantly associated with clinical covariates in prostate cancer (P > 0.05). Conclusions. rs339331 in the RFX6 gene may be associated with prostate cancer as a susceptibility locus in northern Chinese men.
