The effect of mushroom components on the proliferation of HeLa cell line in vitro

  • In order to find out nutritional effects of Korean edible mushrooms on the multiplication of tissue cells, the alcohol extracts and acid bydrolysates of eleven species of mushrooms were added to Earle's ESS. A comparison of the respective multiplications of HeLa cells in this solution and in control solution of TC=199. Yielded the following result: The alcohol extracts and acid hydrolysates of a Coprinus comatur, Agaricus campestris, Agaricus bisporus, Lentinus edodes, Tricholoma matsutake, Pleurotus ostretus, Ramaria botrytis and Pholiota nameko influenced favorably the maintenance of the normal form and monolayer of HeLa cells. The growth curves of HeLa cells in the cultures containing, respectively, the alcohol extracts and acid hydrolysates of these eight mushrooms showed that five species, i.e., Coprinus comatus, Agaricus compestris, Agaricus bisporus, Lentinus edodes and Tricholoma matsutake effected an excellent multiplication and that the other three species were less effective than those five species. As to the effects on the cell multiplication, no marked difference was observed between the alcohol extracts and the acid hydrolysates of the mushrooms tested.

Effects of Paeonia Radix Rubra aqua-acupuncture on Adjuvant Arthritis in rats (적작약약침(赤芍藥藥鍼)이 Rat의 Adjuvant 관절염(關節炎)에 미치는 영향(影響))

  • To investigate effects of Paeonia Radix Rubra aqua-acupuncture on Adjuvant Athritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum were measured and histological test of the muscular tissue were done in the arthritis part. 1. After arthritis of Sprague dawley(SD) rats was induced by injecting Freund's complete adjuvant for 2 weeks, normal saline was injected for the Control group and Paeonia Radix Rubra acua-acupuncture was injected for the Exp. group during 30 days. Selected point was $D{\acute{u}}bf(ST_{35})$ in both groups. And then the inframmator volume of plantar were checked. The volume of the plantar was $0.84{\pm}0.14mm$ in Control group and $0.57{\pm}0.09mm$ in Exp. group, the swelling of plantar was restricted significanily in Exp, group(P<0.05). 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in normal group and $37.47{\pm}5.46(10^3/ml)$ in Control group. It was $25.21{\pm}3.45(10^3/ml)$ in Exp. group. This fact showed that Exp group. with Paeonia Radix Rubra acua-acupuncture was more effective than Control group in the treatment of arthritis(P<0.05). 3. The content of total protein in the blood serum were $6.14{\pm}0.43g/d{\ell}$ in normal group, $7.95{\pm}0.94g/d{\ell}$ in Control group, and $6.82{\pm}1.23g/d{\ell}$ in Exp. group. There was no significance in total protein between Exp. group and Control group from the statistical analysis. 4. The contents of albumin in the blood serum were $2.94{\pm}0.13g/d{\ell}$ in normal group, $2.01{\pm}0.48g/d{\ell}$ in Control group and $2.75{\pm}0.14g/d{\ell}$ in Exp, group. This fact showed that Exp. group had significant increase in the serum albumin from the statistical analysis, in comparison with Control group(P<0.05). 5. The contents of globulin in the blood serum were $3.19{\pm}0.48g/d{\ell}$ in normal group, $4.70{\pm}1.26g/d{\ell}$ in Control group and $3.58{\pm}0.73g/d{\ell}$ in Exp. group. There was no significance in the serum globulin between Exp. group and Control group from the statistical analysis. 6. In histological examinadon, because inflammatory reaction was active, remarkably irregular tissue and large amount of inflammatory cells were found in Control. group. But Exp. group showed that tissues were in some degree recovered, the number of inflamed cells was decreased and inflammation was restrained. From these results, it is shown Paeonia Radix Rubra aqua-acupuncture restrained inflammatory reaction and muscular tissue necrosis in SD rat's plantar was restrained by Freund's complete adjuvant.

Effects of Achyranthis Radix Aqua-acupuncture on Adjuvant Athritis in Rats (우슬약침이 Rat의 Adjuvant 관절염에 미치는 영향)

  • Objectives : To investigate effects of Achyranthis Radix aqua-acupuncture on adjuvant arthritis in rats. Methods : After eliciting arthritis in Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix aqua-acupuncture was injected for the Exp. II group during 30days. The selected point was Dokbi (ST35) in both groups. Then the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum, and histological test of the muscular tissue were measured in the arthritis stricken areas. Results : I) The volume of the paws were checked. The volume of the paws was $0.84{\pm}0.l4mm$ in the Exp. I group and $0.47{\pm}0.14mm$ in the Exp. IIgroup. The swelling of the paws was restricted significantly in the Exp. II group(P<0.05). 2) The number of WBC was $10.34{\pm}0.14(l03/ml)$ in the normal group and $37.47{\pm}5.46(103/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(103/ml)$ in the Exp. II group. This fact showed that treatment with Achyranthis Radix aqua-acupuncture adminstered to the Exp. II group was more effective than the Exp. I group in the treatment of arthritis(P<0.05). 3) The content of the total protein in the blood serum was $6.14{\pm}43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4) The content of albumin in the blood serum was $2.94{\pm}0.l3g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group, which revealed a significant increase in the serum albumin from the statistical analysis of the Exp. II group compared with the Exp. I group. 5) The content of the globulin in the blood serum was $3.19{\pm}$0.48g/dl in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6) In histological findings, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed a small amount of inflammatory cells, the refrained inflammatory state and even recovering state. Conclusions : These results show that Achyranthis Radix aqua-acupuncture reduces the inflammatory reaction and muscular tissue necrosis in SD rat's paws induced by Freund's complete adjuvant.

CD30-Mediated Regulation of Cell Adhesion Molecule Expression on Murine T Cells

  • Background: CD30 is a member of TNF receptor family and expressed on lymphocytes and other hematopoietic cells following activation as well as Hodgkin and Reed-Sternberg cells in Hodgkin's lymphoma. In this study, CD30-mediated regulation of cell adhesion molecule expression on normal activated mouse T cells was investigated. Methods: Mouse T cells were activated with anti-CD3 antibody for induction of CD30, which was cross-linked by immobilized anti-CD30 antibody. Results: High level of CD30 expression on T cells was observed on day 5, but only little on day 3 even under culture condition resulting in an identical T cell proliferation, indicating that CD30 expression requires a prolonged stimulation up to 5 days. Cross-linking of CD30 alone altered neither proliferation nor apoptosis of normal activated T cells. Instead, CD30 appeared to promote cell adherence to culture substrate, and considerably upregulated ICAM-1 and, to a lesser extent, ICAM-2 expression on activated T cells, whereas CD2 and CD18 (LFA-1) expression was not affected. None of cytokines known as main regulators of ICAM-1 expression on tissue cells (IL 4, $IFN{\gamma}$ and $IFN{\alpha}$) enhanced ICAM-1 expression in the absence of CD30 signals. On the other hand, addition of $NF-{\kappa}B$ inhibitor, PDTC (0.1 mM) completely abrogated the CD30-mediated upregulation of ICAM-1 expression, but not CD2 and ICAM-2 expression. Conclusion: This results support that CD30 upregulates ICAM-1 expression of T cell and such regulation is not mediated by higher cytokine production but $NF-{\kappa}B$ activation. Therefore, CD30 may play important roles in T-T or T-B cell interaction through regulation of ICAM-1, and -2 expression.

AN EXPERIMENTAL STUDY CN THE INFLUENCE OF FCRMCCRESOL TO THE HEALING PROCESS OF AMPUTATED PULP (Formocresol이 손상치수조직(損傷齒髓組織)의 치유(治癒)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究))

  • After a vital pulpotomy in dogs' teeth, the responses of the remaining pulp tissue under calcium hydroxide and formocresol were studied histologically. The class I and V cavities were prepared on the teeth and the pulp was amputated. Calcium hydroxide and formocresol were placed over the amputated tissue and the cavities were sealed with zine oxide eugenol cement and zinc phosphate cement. Animals. were sacrifice after 1, 2, and 3 weeks following the operation. The teeth were decalcfied, sectioned and stained with hematoxylin and eosin. Microscopic examination reveals as follows; 1. Healing of the pulp at the amputation site did not occur in the pulps treated with formocresol. 2. At one week, a thin layer of darker staining tissues just below the necrotic zone was presented in the pulps treated with formocresol. In this stage the tissues beneath the darker staining layer were normal. 3. At two weeks, the cells of the palest staining layer were showed indistinct nucleus which suggested the karyolysis and the karyorrhexis in the pulps treated with formocoresol. As reached to the middle third of the pulp, the odontoblasts were scarcely evident or missed in this stage. 4. At three weeks, the necrotic zone was reached to the middle third of the pulp canal. The cells beneath the zone showed massive infiltration of inflammatory cells in the pulps treated with formocresol. 5. Dentin bridge in the control group was deposited below the necrotic zone from the two. weeks later. 6. Normal tissues were observed ill the apical third of all. the dental pulps in all case of calcium hydroxide and formocresol.

KLK6 Promotes Growth, Migration, and Invasion of Gastric Cancer Cells

  • Purpose: Kallikrein (KLK) proteases are hormone-like signaling molecules with critical functions in different cancers. This study investigated the expression of KLK6 in gastric cancer and its potential role in the growth, migration, and invasion of gastric cancer cells. Materials and Methods: In this study, we compared protein levels of KLK6, vascular endothelial growth factor (VEGF), and matrix metallopeptidase (MMP) 9 in normal gastric epithelial and gastric cancer cell lines by western blot. Fluorescence-activated cell sorting was employed to sort 2 clones of SGC-7901 cells with distinct KLK6 expression, namely, KLK6-high ($KLK6^{high}$) and KLK6-low ($KLK6^{low}$), which were then expanded. Lastly, immunohistochemical analysis was performed to investigate KLK6 expression in gastric cancer patients. Results: The expression levels of KLK6, VEGF, and MMP 9, were significantly higher in the gastric cancer cell lines SGC-7901, BGC-823, MKN-28, and MGC-803 than in the normal gastric epithelial cell line GES-1. Compared to $KLK6^{low}$ cells, $KLK6^{high}$ cells showed enhanced viability, colony-forming ability, migration, and invasion potential in vitro. Importantly, immunohistochemical analysis of a human gastric cancer tissue cohort revealed that the staining for KLK6, VEGF, and MMP9 was markedly stronger in the cancerous tissues than in the adjacent normal tissues. KLK6 expression also correlated with that of VEGF and MMP9 expression, as well as several key clinicopathological parameters. Conclusions: Together, these results suggest an important role for KLK6 in human gastric cancer progression.

HISTOLOGICAL CHANGES OF MOUSE SPLEEN AND LYMPH NODE BY CYCLOPHOSPHAMIDE (Cyclophosphamide에 의(依)한 mouse의 비장(脾臟)과 임파절(淋巴節)의 조직학적(組織學的) 변화(變化))

  • Adult mice were injected with a single sublethal dose of cyclophosphamide. Effects of the drug on the body weight, spleen weight, and morphology of the peripheral lymphoid system have been analysed. The body weights of the mice given cyclophosphamide(300mg/kg body weight) decreased slightly and returned to normal quickly. Spleen weights, however, changed greatly by keeping the process of decrease, recovery, overshoot, and gradual return to normal only by 20 days. Histologic examinations of spleen and popliteal lymph node showed that follicles disappeared 1 to 2 days before periarteriolar lymphatic sheath or paracortex. At the peak of splenomegaly, the architectures of spleen and lymph node were replaced with the interstitial tissue composed of dense and uniform layer of lymphoid cells. With the return of spleen weight to normal range, the architecturles returned to normal. Our results clearly indicated that cyclophosphamide affected not only B cells but also T cells. These results seemed to suggest that augmentation of delayed-type hypersensitivity by cyclophosphamide may be due to the eliminateion of the suppressor T cells.

ADENOID CYSTIC CARCINOMA OF THE MINOR SALIVARY GLANDS (소타액선에 발생된 선양 낭포암)

  • Adenoid cystic carcinoma is a malignant salivary gland tumor with typical histologic patterns. The majority of these tumors occurs in the minor salivary glands. especially mucosa of the hard palate. The authors experienced the patients, who complained the tumor-like soft tissue masses on the palatal and mouth floor area. After careful analysis of clinical, radiological and histopathological findings, we diagnosed them as adenoid cystic carcinomas in the minor salivary glands, and obtained results were as follows : 1. Main clinical symptoms were a slow growing soft tissue mass with normal intact mucosa on the palatal area, and soft tissue mass with mild pain on the mouth floor area. 2. In the radiographic exarnminations, soft tissue masses were observed with invasion to adjacent structures, and moderate defined, heterogeneous soft tissue mass with enhanced margin, respectively. 3. In the histopathologic exarnminations, dark-stained, small uniform basaloid cells in the hyaline or fibrous stroma were observed as solid and cribriform patterns, respectively.

Serotonin and Energy Metabolism (세로토닌과 에너지 대사)

  • Serotonin, a biogenic amine widely found in many organisms, functions as both a neurotransmitter and hormone. Although serotonin is involved in various physiological processes, this study aimed to review its role in energy metabolism. Given that serotonin cannot cross the blood-brain barrier and is synthesized by two different isoforms of tryptophan hydroxylase in the central nervous system (CNS) and peripheral tissues, it is reasonable to assume that serotonin in the CNS and peripheral tissues functions independently. Recent studies have demonstrated how serotonin influences energy metabolism in metabolic target organs such as the intestines, liver, pancreas, and adipose tissue. In summary, serotonin in the CNS induces satiety and appetite suppression, stimulates thermogenesis, and reduces body weight. Conversely, serotonin in the periphery increases intestinal motility, stimulates gluconeogenesis in the liver, suppresses glucose uptake by hepatocytes, promotes fat uptake by liver cells, stimulates insulin secretion while suppressing glucagon secretion in the pancreatic islets, promotes lipogenesis in white adipose tissue, inhibits lipolysis and browning of white adipose tissue, and suppresses thermogenesis in brown adipose tissue, thereby storing energy and increasing body weight. However, considering that most experimental results were obtained using mice and conducted under specific nutritional conditions, such as high-fat diets, whether serotonin acts in the same way in humans, whether it will act similarly in individuals with normal versus obese weights, and whether its effects vary depending on the type of food consumed, remain unknown.

Anti-Oxidative Effect of Seungyangikki-tang Decoction in Spleen, Pancreas and Stomach Cells of SD Rats (승양익기탕(升陽益氣湯) 전탕액이 노화쥐의 비장, 췌장, 위장 세포의 항산화능에 미치는 영향)

  • 1. Objectives: The purpose of this study is to investigate the anti-aging and anti-oxidative effects of Seungyangikki-tang decoction(SY) in spleen, pancreas and stomach cells of Sprague-Dawley(SD) rats. 2. Methods: This experiment was used by the tissue of spleen, pancreas and stomach cells of 6, 52 and 68 weeks old SD rats. We devided each group by three. One group as normal group was non-treated cells, another group as control group was saline-treated cells, and the other group as experimental group was SY treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of kidney, bladder and spleen cells. 3. Results and Conclusions: SOD activity was significantly increased in spleen cell of 6, 68w-SY group, pancreas cell of 52,68w-SY group and in stomach cell of 52w-SY group compared with those of the control groups. GSH concentration was significantly increased in spleen cell of 6,68w-SY group and in pancreas cell of 6w-SY group compared with those of the control groups. MDA concentration was significantly decreased in spleen cell of 68w-SY group and in stomach cell of 68w-SY group compared with those of the control groups. NO concentration was significantly decreased in spleen cell of 68w-SY group, stomach cell of 68w-SY group compared with those of the control groups.
